The fiber-type composition of postnatal chicken leg muscle spindles with from one to four intrafusal fibers was examined in sections incubated with monoclonal antibodies against fast and slow myosin heavy chains. In monofibral spindles the lone intrafusal fiber was almost always fast. In duofibral spindles usually one slow and one fast fiber were present. Trifibral spindles most often displayed two fast and one slow fiber, whereas quadrofibral receptors characteristically contained two slow and two fast fibers. Earlier results showed that the primary intrafusal myotube in nascent spindles has almost always a fast myosin heavy chain profile and that the proportion of slow myotubes and fibers increases as intrafusal fiber bundles grow in size. Data from postnatal chicken leg muscles collected here suggest that up to the first four fibers this proportional increase can be largely accounted for if consecutive intrafusal fibers arise in a fast-slow-fast-slow sequence. The late recognition during myogenesis of primary intrafusal myotubes and their fast myosin heavy chain profiles warrant exploring if nascent chicken muscles spindles are first seeded by fast fetal myoblasts.

The morphology and distribution of muscle spindles of jaw and tongue muscles in the mallard were examined in serial transverse sections of single muscles and in horizontal sections of a whole head. Our observations on spindle morphology are in agreement with previous descriptions of spindles in birds. Some spindles differ in their innervation and the pattern of intrafusal muscle fibers. The spindles of individual adductor and pterygoid muscles are distributed unevenly. Some adductor muscles lack spindles, whereas those of other muscles are confined to limited areas. Jaw opening muscles and extrinsic tongue muscles lack spindles. The stretch of extrafusal muscle fibers could be estimated from the difference in sarcomere length for birds with the beak open and closed. Not all muscle fiber groups are stretched evenly over the whole range of jaw opening. Only those fiber groups that are continuously stretched during jaw opening contain spindles.

The concept of parallel muscle combinations, in which spindle density is significantly higher in small muscles compared to their larger counterparts in large-small muscle combinations acting across a joint, is supported by the results of this study regardless of the joint. Analysis of the canine data as well as previously published guinea pig forelimb and human pelvic limb data revealed no significant difference in spindle density between antigravity and non-antigravity muscles. Furthermore, a gradual increase in spindle density from proximal to distal on the limb was not found, although spindle density was significantly higher in the intrinsic manus or pes muscles compared to more proximal limb muscles in all three species. The significant differences in spindle densities in parallel muscle combinations and in manus/pes versus proximal muscles are discussed relative to their possible role in the control of locomotion.

Muscle spindles were studied histochemically in serial transverse sections of 42 cat tenuissimus muscle specimens. Staining for myofibrillar adenosine triphosphatase was employed to identify nuclear bag 1, nuclear bag 2, and nuclear chain intrafusal muscle fibers. The nuclear chain fibers were further subdivided into three categories according to their polar length and the intensity of their staining for nicotinamide adenine dinucleotide tetrazolium reductase. A total of 430 spindle poles were surveyed. The mean spindle content of bag 1, bag 2, and chain fibers was established. The mean polar length of intrafusal fibers as well as that of the intracapsular and extracapsular spindle regions was determined. A cholinesterase (ChE) staining technique was used to demonstrate the termination sites of motor axons along intrafusal fibers. Two types of circumscribed ChE deposits. The "rim" and the "plate," occurred on the fibers. The nuclear chain fibers usually carried both the ChE rims and plates, while most nuclear fibers displayed only the plates. The ChE plates were assessed in term of their appearance, staining intensity, length, and location along the fibers. The mean number of ChE plates found along the fibers was established for each of the various intrafusal fiber types. These histochemical observations are discussed with regard to the current concepts of cat spindle morphology and motor innervation. The results suggest a degree of predictability in the spindle fiber content and in the distribution of motor nerve terminals along intrafusal muscle fibers, at least in the tenuissimus muscle.

Guinea pig soleus, medial gastrocnemius and vastus lateralis muscles were compared for spindle density and distribution, number of intrafusal fibers per spindle and histochemical appearance of the axial bundle. A total of 326 spindles was used in the comparisons. Spindle density was over four times greater in the soleus than in either the medial gastrocnemius or vastus lateralis. In the soleus the spindles were distributed at random, but in the other two muscles no spindles were found in those fascicles in which fast-twitch glycolytic extrafusal fibers predominated. The average number of intrafusal fibers per spindle varied by less than 5% between the three kinds of muscles. About 80% of all spindles located had four intrafusal fibers, two of the nuclear bag type and two of the nuclear chain type. The histochemical appearance of the axial bundle was the same in each kind of muscle. Based on intensities of the myofibrillar adenosine triphosphatase reaction product at polar regions nuclear bag fibers were separable into two histochemical groups; nuclear chain fibers were of only one histochemical type.

The blood supply of muscle spindles was studied in serial cross sections in macaque, cat, rabbit, guinea pig, mouse and pigeon muscles which had been incubated in a medium containing 3,3' diaminobenzidine. Lumina of blood vessels were recognized by the reaction product that was localized within erythrocytes. The outer capsule was well vascularized, but few or no capillaries were seen in the periaxial space. The inner spindle capsule, which closely invests the axial bundle, was rarely contacted by periaxial capillaries at the equator and juxtequator. Capillaries occurred more frequently adjacent to intrafusal fibers at the polar region and beyond the end of the outer capsule. Shorter diffusion distances and, usually, higher capillary densities were found at the polar region than at the spindle midsection. This suggests that transcapillary exchange at the polar segment is nearer to conditions prevalent in extrafusal muscle than elsewhere in the spindle, provided the inner and outer capsules are not less permeable at the poles than at the midsection. Differences in blood supply among mammalian species appear to be related to receptor size.

A small short muscle frequently acts across a joint in parallel with a vastly larger and longer muscle; therefore it should play a minimal role in the mechanical control of that joint. This study provides evidence suggesting that the small member of such a "parallel muscle combination" (PMC) may serve an important sensory feedback role. The spindle densities of large and small members of PMCs in man and the dog were determined and compared. Epaxial PMCs controlling canine intervertebral joints were dissected and tissue samples were embedded in paraffin, sectioned transversely to the muscles' long axis and, stained with hematoxylin-periodic acid-Schiff (PAS). Representative tissue sections were projected on to stereological grids and the percentage volume of spindles was determined. Data existing in the literature were used to ascertain spindle densities in human PMCs controlling joints in the cervico-occipital region and the extremities. The spindle density for each muscle in a group of PMCs controlling a particular motion was listed, and the mean spindle densities were determined for both the large and the small members of the group. Student's unpaired t test was used to determine the significance of the differences between mean spindle densities. Linear regression was calculated and the data were plotted graphically. In all PMCs examined, the spindle density of the small muscles was significantly higher than that of their large counterparts. It is therefore proposed that the small muscles of PMCs may function as "kinesiological monitors" generating important proprioceptive feedback to the central nervous system.
