Monocystis perplexa n. sp., a parasite of an important invasive Japanese earthworm in North America, Amynthas agrestis, is described from a site in Vermont. An improved standard for Monocystis species descriptions is proposed including a standard nomenclature to reduce synonymies, a standard set of biometrics and shape descriptions for living cells, and a DNA genomic sequence for the 18S rRNA (∼1,700 base pairs). Comparing morphologies of Monocystis parasites in sympatric earthworm species indicates that M. perplexa is specific to A. agrestis in the study region. Also, polymerase chain reaction primers specific to M. perplexa amplified samples of A. agrestis earthworms taken from several sites in Japan. This suggests the parasite entered North America from Japan, the origin of the invasive Amynthas earthworm, and thus M. perplexa would be the first Monocystis described from the diverse Japanese Amynthas earthworms and the first from East Asia. Monocystis perplexa was found in every population of A. agrestis surveyed in Vermont, always reaching 100% prevalence by late summer (the host has an annual life cycle in Vermont). The 18S gene sequence differed from that of Monocystis agilis from the sympatric earthworm Lumbricus terrestris (the only other sequence available for Monocystis), and a genetic similarity tree places them closest among other gregarines. Many of the 95 described species of Monocystis are very similar in morphology (based on species descriptions), so the 18S gene can act as a barcode for Monocystis species and thus will help to eliminate both synonymies and reveal cryptic species., (© American Society of Parasitologists 2020.)

Within Oligochaeta, Chaetogaster limnaei is unusual in exhibiting a parasitic relationship with freshwater pulmonate snails. Taxonomic confusion has been caused by differences in what have been considered 2 subspecies of this worm: Chaetogaster limnaei limnaei is an ectosymbiont and is present inside the mantle cavity of the snail, whereas Chaetogaster limnaei vaghini is parasitic and lives in the kidney of the snail. This study explored the distribution of these annelids in central New York and used mitochondrial DNA sequence data from the COI locus to examine the relationship, evolution, and species status of the ectosymbiotic and parasitic forms of C. limnaei. Snails ( Physa gyrina ) were collected from 6 streams and lakes in central New York, with additional specimens collected from a lake in Massachusetts for comparison. One hundred and forty snails were examined, and at least 1 form of Chaetogaster was present in 88 specimens, a prevalence of 62.9%. COI sequence data from New York and Massachusetts did not reveal separate ectosymbiotic and parasitic lineages. Instead, all parasitic forms were part of a mixed clade that included both ectosymbiotic and parasitic forms. This mixed clade was nested within clades of ectosymbiotic forms only, suggesting that a plastic lineage of C. limnaei, able to be both ectosymbionts and parasites, evolved from ectoparasitic ancestors.

Although widespread throughout the continental United States, Myxobolus cerebralis, the myxozoan parasite that causes whirling disease in salmonids, has not been reported from the state of Alaska. As part of a risk assessment for the introduction and establishment of M. cerebralis into Alaska, the distribution of the invertebrate host Tubifex tubifex was surveyed, and its genetic composition and susceptibility to the parasite were determined. Many oligochaetes, but no T. tubifex, were collected from southeastern Alaska; however, 4 lineages of T. tubifex (I, III, IV, and VI) were identified from south-central Alaska. Lineage IV had not been previously described in North America, and its susceptibility to M. cerebralis was unknown. When lineage IV T. tubifex and 3 mixed lineage (I, III, IV, and VI) groups were exposed to M. cerebralis, only lineage III became infected under our experimental conditions. Infection occurred in this lineage even when it made up just 3% of the population. Implications of the presence of nonsusceptible lineages of T. tubifex on Alaskan salmonids would be significant in areas where these lineages dominate T. tubifex populations.

We analyzed the geographic distribution of Tubifex tubifex from various river drainages in central Colorado by genetic screening with specific mitochondrial 16S ribosomal DNA (mt 16S rDNA) markers. Four distinct mt 16S rDNA lineages are evident. The sites varied with respect to land- and water-use practices. All sites represented habitats presumed capable of supporting oligochaetes. At the locations where whirling disease has had the greatest impact on resident rainbow trout, T. tubifex, representing lineages I and III (genotypes known to be susceptible to Mxyobolus cerebralis), were most commonly found. In contrast, at sites less affected by whirling disease, T. tubifex of lineages V and VI that are more resistant to M. cerebralis infections were more abundant. The predominance of resistant T. tubifex worms (lineages V and VI) at low-impact sites supports the conclusion that when these genotypes are in greater abundance, the potential for more severe effects of whirling disease on wild rainbow trout populations may be diminished.

Whirling disease, caused by the parasite Myxobolus cerebralis, has infected rainbow trout (Oncorhynchus mykiss) and other salmonid fish in the western United States, often with devastating results to native populations but without a discernible spatial pattern. The parasite develops in a complex 2-host system in which the aquatic oligochaete Tubifex tubifex is an obligate host. Because substantial differences in whirling disease severity in different areas of North America did not seem explainable by environmental factors or features of the parasite or its fish host, we sought to determine whether ecological or genetic variation within oligochaete host populations may be responsible. We found large differences in compatibility between the parasite and various laboratory strains of T. tubifex that were established from geographic regions with different whirling disease histories. Moreover, 2 closely related species of tubificids, Limnodrilus hoffmeisteri and Ilyodrilus templetoni, which occur naturally in mixed species assemblages with T. tubifex, were incompatible with M. cerebralis. Virulence of the parasite was directly correlated with the numbers of triactinomyxon spores that developed within each strain of T. tubifex. Thus, the level of virulence was directly related to the compatibility between the host strain and the parasite. Genetic analyses revealed relationships that were in agreement with the level of parasite production. Differences in compatibilities between oligochaetes and M. cerebralis may contribute to the spatial variance in the severity of the disease among salmonid populations.

There is little ecological information on actinosporean parasites and their oligochaete hosts. Between 1995 and 1997, about 14,100 oligochaetes belonging to 19 species were collected from Lake Sasajewun. The oligochaete fauna consisted of 5 tubificid, 10 naidid, 3 enchytraeid, and 1 lumbriculid species. The most widely distributed species in the lake were Limnodrilus hoffmeisteri, Dero nivea, and Stylaria lacustris, with L. hoffmeisteri being the most prevalent. The diversity and abundance of worms decreased with increased water depth. Four species, L. hoffmeisteri, Tubifex tubifex, Rhyacodrilus coccineus, and Lumbriculus variegatus, harbored actinosporean parasites and were distributed along shallow areas of the lake shore where the sediment was comprised mainly of clay-mud, with the shoregrass, Littorella americana. The actinosporean parasites of the oligochaetes belong to 25 forms of 8 collective groups. Triactinomyxon 'F' was the most prevalent form, whereas triactinomyxon, raabeia, and echinactinomyxon were the most speciose groups. The overall prevalence of actinosporeans was about 1%. Waterborne spores of different actinosporean forms showed distinct temporal patterns. Spores of triactinomyxon 'F' occurred throughout the sampling season, whereas spores of other forms occurred only at certain times. The presence of waterborne spores peaked from late June to August when water temperatures ranged from 18 to 24 C and coincided with the feeding and growing season of larval fish.
