α-Boryl ethers, carbonates, and acetals, readily prepared from the corresponding alcohols that are accessed through ketone diboration, react rapidly with hydrogen peroxide to release alcohols, aldehydes, and ketones through the collapse of hemiacetal intermediates. Experiments with α-boryl acetals containing a latent fluorophore clearly demonstrate that cargo can be released inside cells in the presence of exogenous or endogenous hydrogen peroxide. These experiments show that this protocol can be used for drug activation in an oxidative environment without generating toxic byproducts.

Chiral, neutral H-bond donors have found widespread use as catalysts in enantioselective reactions involving ion-pair intermediates. Herein, a systematic mechanistic study of a prototypical anion-binding reaction, the thiourea-catalyzed enantioselective alkylation of α-chloroethers, is detailed. This study reveals that the catalyst resting state is an inactive dimeric aggregate that must dissociate and then reassemble to form a 2:1 catalyst-substrate complex in the rate-determining transition structure. Insight into this mode of catalyst cooperativity sheds light on the practical limitations that have plagued many of the H-bond donor-catalyzed reactions developed to date and suggests design strategies for new, highly efficient catalyst structures.

Cupriavidus metallidurans CH34 is a bacterium that is resistant to high metal concentrations in the environment. Increased copper resistance is associated with the cop cluster on the large plasmid pMOL30 that is composed of at least 21 genes. The copK gene encodes a 74 residue periplasmic protein whose expression is strongly upregulated in the presence of copper. CopK was previously shown to cooperatively bind Cu(I) and Cu(II) in distinct, specific sites. The solution structure of Cu(I)-CopK and the characterization of the Cu(I) site by X-ray absorption spectroscopy and NMR are reported here. EXAFS spectra are in agreement with a tetrathioether Cu(I) site, providing so far unique spectral information on a 4S-coordinated Cu(I) in a protein. The methionine residues forming the Cu(I) site, M28, M38, M44, and M54, are identified by NMR. We propose the chemical shift of the methionine C(epsilon) as a new and sensitive probe for the detection of Cu(I) bound to thioether groups. The solution structure of Cu(I)-CopK demonstrates that Cu(I) binding induces a complete structural modification with the disruption of the second beta-sheet and a rotation of the C-terminal part of nearly 180 degrees around a hinge formed by asparagine 57. This conformational change is directly related to the loss of the dimer interface and most probably to the formation of the Cu(II) site involving histidine 70. The solution structure of Cu(I)-CopK therefore provides the molecular basis for the understanding of the Cu(I)/Cu(II) binding cooperativity.

We have developed low-valent titanium-mediated 1,3-transpositive cross-coupling reactions of acyclic and cyclic allylic alcohols for the stereoselective introduction of ethyl, 2-silylethyl, 2-phenethyl, and alkenyl groups. Cross-coupling of an allylic alcohol with a vinylsilane or styrene derivative is particularly noteworthy, as an efficient cross-selective coupling of two alkenes has been elusive. The stereochemistry of the cross-coupling alkylation is consistent with syn addition/beta-elimination.

The reaction of thallium ethoxide with [H(OEt2)2][H2N{B(C6F5)3}2] in diethyl ether afforded [Tl(OEt2)3][H2N{B(C6F5)3}2] (2a), [Tl(OEt2)4][H2N{B(C6F5)3}2] (2b), or [Tl(OEt2)2][H2N{B(C6F5)3}2].CH2Cl2 (2c), depending on the reaction conditions. The dication in the hydrolysis product [Tl4(mu3-OH)2][H2N{B(C6F5)3}2]2.4CH2Cl2 consists of two bridging and two terminal Tl+ ions bound to triply bridging hydroxides. Heating Et2O complexes in toluene afforded [Tl(eta6-toluene)n][H2N{B(C6F5)3}2] (4, n = 2, 3), while C6Me6 addition gave the first thallium-C6Me6 adduct, [Tl(eta6-C6Me6)2][H2N{B(C6F5)3}2].1.5CH2Cl2 (5a), a bent sandwich complex with very short Tl...centroid distances. These arene complexes show no close contacts between cations and anions. Displacement of toluene ligands by ferrocene gave [Tl2(FeCp2)3][H2N{B(C6F5)3}2]2.5CH2Cl2 (6) which contains the multidecker cations [Tl(FeCp2)]+ and [Tl(FeCp2)2]+ in a 1:1 ratio. By contrast, decamethylferrocene leads to electron transfer; the isolable thallium-ferrocene complexes may therefore be viewed as precursor complexes for this redox step. With 18-crown-6 the complexes [Tl(18-crown-6)2][H2N{B(C6F5)3}2] (11a) and [Tl(18-crown-6)][H2N{B(C6F5)3}2].2CH2Cl2 (11b) were isolated. The structure of the latter shows an eight-coordinate thallium ion, where the coordination to the six oxygen donors in equatorial positions is completed by axial contacts to two F atoms of the counter anions. The bonding between thallium(I) and arenes was explored by density-functional theory (DFT) calculations. The optimized geometry of [Tl(tol)3]+ converged to a structure very similar to that obtained experimentally. Calculations on [Tl(C6Me6)2]+ (5b) to establish whether a linear or bent geometry is the most stable revealed a very flat potential-energy surface for distortions of the Ctr(3)-Tl-Ctr(4) angle. Overall, there is very little energetic preference for one particular geometry over another above about 140 degrees , in good agreement with the crystallographic geometry. The calculated Tl-arene interaction energies increase from 73.7 kJ mol-1 for toluene to 121.7 kJ mol-1 for C6Me6.

Soluble methane monooxygenase (sMMO) isolated from Methylococcus capsulatus (Bath) utilizes a carboxylate-bridged diiron center and dioxygen to catalyze the conversion of methane to methanol. Previous studies revealed that a di(mu-oxo)diiron(IV) intermediate termed Q is responsible for the catalytic activity with hydrocarbons. In addition, the peroxodiiron(III) intermediate (H(peroxo)) that precedes Q formation in the catalytic cycle has been demonstrated to react with propylene, but its reactivity has not been extensively investigated. Given the burgeoning interest in the existence of multiple oxidants in metalloenzymes, a more exhaustive study of the reactivity of H(peroxo) was undertaken. The kinetics of single turnover reactions of the two intermediates with ethyl vinyl ether and diethyl ether were monitored by single- and double-mixing stopped-flow optical spectroscopy. For both substrates, the rate constants for reaction with H(peroxo) are greater than those for Q. An analytical model for explaining the transient kinetics is described and used successfully to fit the observed data. Activation parameters were determined through temperature-dependent studies, and the kinetic isotope effects for the reactions with diethyl ether were measured. The rate constants indicate that H(peroxo) is a more electrophilic oxidant than Q. We propose that H(peroxo) reacts via two-electron transfer mechanisms, and that Q reacts by single-electron transfer steps.
