The article presents a new approach to use the engineered DNA Holliday junctions (HJs) for reporting the protein-DNA interactions in various experiments. The method is highly efficient, as the changes in DNA due to the MerR-family regulator, PbrR691 give accurate results.

Nanovesicle trapping and single-molecule fluorescence resonance energy transfer (smFRET) measurements are used for probing the transient interactions between Hah1 and the fourth metal-binding domains (MBD4) of Wilson disease protein (WDP) in real time. The single-molecule approach has offered a means to detect the [Cu.sup.1} transfer process and identify the productive interaction complex.

Materials exhibiting aggregation-induced emission (AIE) are both highly emissive in the solid state and prompt a strongly red-shifted emission and should therefore pose as good candidates toward emerging near-infrared (NIR) applications of organic semiconductors (OSCs). Despite this, very few AIE materials have been reported with significant emissivity past 700 nm. In this work, we elucidate the potential of ortho -carborane as an AIE-active component in the design of NIR-emitting OSCs. By incorporating ortho -carborane in the backbone of a conjugated polymer, a remarkable solid-state photoluminescence quantum yield of 13.4% is achieved, with a photoluminescence maximum of 734 nm. In contrast, the corresponding para and meta isomers exhibited aggregation-caused quenching. The materials are demonstrated for electronic applications through the fabrication of nondoped polymer light-emitting diodes. Devices employing the ortho isomer achieved nearly pure NIR emission, with 86% of emission at wavelengths longer than 700 nm and an electroluminescence maximum at 761 nm, producing a significant light output of 1.37 W sr -1 m -2 .

Oligomeric species populated during α-synuclein aggregation are considered key drivers of neurodegeneration in Parkinson's disease. However, the development of oligomer-targeting therapeutics is constrained by our limited knowledge of their structure and the molecular determinants driving their conversion to fibrils. Phenol-soluble modulin α3 (PSMα3) is a nanomolar peptide binder of α-synuclein oligomers that inhibits aggregation by blocking oligomer-to-fibril conversion. Here, we investigate the binding of PSMα3 to α-synuclein oligomers to discover the mechanistic basis of this protective activity. We find that PSMα3 selectively targets an α-synuclein N-terminal motif (residues 36-61) that populates a distinct conformation in the mono- and oligomeric states. This α-synuclein region plays a pivotal role in oligomer-to-fibril conversion as its absence renders the central NAC domain insufficient to prompt this structural transition. The hereditary mutation G51D, associated with early onset Parkinson's disease, causes a conformational fluctuation in this region, leading to delayed oligomer-to-fibril conversion and an accumulation of oligomers that are resistant to remodeling by molecular chaperones. Overall, our findings unveil a new targetable region in α-synuclein oligomers, advance our comprehension of oligomer-to-amyloid fibril conversion, and reveal a new facet of α-synuclein pathogenic mutations.

Peptide-induced transmembrane pore formation is commonplace in biology. Examples of transmembrane pores include pores formed by antimicrobial peptides (AMPs) and cell-penetrating peptides (CPPs) in bacterial membranes and eukaryotic membranes, respectively. In general, however, transmembrane pore formation depends on peptide sequences, lipid compositions, and intensive thermodynamic variables and is difficult to observe directly under realistic solution conditions, with structures that are challenging to measure directly. In contrast, the structure and phase behavior of peptide-lipid systems are relatively straightforward to map out experimentally for a broad range of conditions. Cubic phases are often observed in systems involving pore-forming peptides; however, it is not clear how the structural tendency to induce negative Gaussian curvature (NGC) in such phases is quantitatively related to the geometry of biological pores. Here, we leverage the theory of anisotropic inclusions and devise a facile method to estimate transmembrane pore sizes from geometric parameters of cubic phases measured from small-angle X-ray scattering (SAXS) and show that such estimates compare well with known pore sizes. Moreover, our model suggests that although AMPs can induce stable transmembrane pores for membranes with a broad range of conditions, pores formed by CPPs are highly labile, consistent with atomistic simulations.

In their Comment (DOI: 10.1021/jacs.2c02965) on two related publications by our group ( J. Am. Chem. Soc. 2022 , 144 , 1380-1388; DOI: 10.1021/jacs.1c11754) and another ( J. Am. Chem. Soc. 2021 , 143 , 20884-20890; DOI: 10.1021/jacs.1c09455), Huang and Granick refer to the diffusion NMR measurements of molecules during a copper-catalyzed azide-alkyne cycloaddition (CuAAC) "click" reaction. Here we respond to their comments and maintain that no measurable diffusion enhancement was observed during the reaction. We expand on the physical arguments presented in our original JACS Article regarding the appropriate reference state for the diffusion coefficient and present new data showing that the use of other reference states, as suggested by Huang and Granick, will still support our conclusion that the two reactants and one product of the CuAAC reaction do not exhibit boosted mobility during the reaction.

Micrometer-sized objects are widely known to exhibit chemically driven motility in systems away from equilibrium. Experimental observation of reaction-induced motility or enhancement in diffusivity at the much shorter length scale of small molecules is, however, still a matter of debate. Here, we investigate the molecular diffusivity of reactants, catalyst, and product of a model reaction, the copper-catalyzed azide-alkyne cycloaddition click reaction, and develop new NMR diffusion approaches that allow the probing of reaction-induced diffusion enhancement in nanosized molecular systems with higher accuracy than the state of the art. Following two different approaches that enable the accounting of time-dependent concentration changes during NMR experiments, we closely monitored the diffusion coefficient of reaction components during the reaction. The reaction components showed distinct changes in the diffusivity: while the two reactants underwent a time-dependent decrease in their diffusivity, the diffusion coefficient of the product gradually increased and the catalyst showed only slight diffusion enhancement within the range expected for reaction-induced sample heating. The decrease in diffusion coefficient of the alkyne, one of the two reactants of click reaction, was not reproduced during its copper coordination when the second reactant, azide, was absent. Our results do not support the catalysis-induced diffusion enhancement of the components of the click reaction and, instead, point to the role of a relatively large intermediate species within the reaction cycle with diffusivity lower than that of both the reactants and product molecule.

The promising field of nanomedicine stimulates a continuous search for multifunctional nanotheranostic systems for imaging and drug delivery. Herein, we demonstrate that application of supramolecular chemistry's concepts in dendritic assemblies can enable the formation of advanced dendrimer-based nanotheranostic devices. A dendrimer bearing 81 triazolylferrocenyl terminal groups adopts a more compact shell-like structure in polar solvents with the ferrocenyl peripheral groups backfolding toward the hydrophobic dendrimer interior, while exposing the more polar triazole moieties as the dendritic shell. Akin to lipids, the compact dendritic structure self-assembles into uniform nanovesicles that in turn self-assemble into larger vesosomes in water. The vesosomes emit green nontraditional intrinsic fluorescence (NTIL), which is an emerging property as there are no classical fluorophores in the dendritic macromolecular structure. This work confirms the hypothesis that the NTIL emission is greatly enhanced by rigidification of the supramolecular assemblies containing heteroatomic subluminophores (HASLs) and by the presence of electron rich functional groups on the periphery of dendrimers. This work is the first one detecting NTIL in ferrocenyl-terminated dendrimers. Moreover, the vesosomes are stable in biological medium, are uptaken by cells, and show cytotoxic activity against cancer cells. Accordingly, the self-organization of these dendrimers into tertiary structures promotes the emergence of new properties enabling the same component, in this case, ferrocenyl group, to function as both antitumoral drug and fluorophore.

Defect passivation constitutes one of the most commonly used strategies to fabricate highly efficient perovskite solar cells (PSCs). However, the durability of the passivation effects under harsh operational conditions has not been extensively studied regardless of the weak and vulnerable secondary bonding between the molecular passivation agents and perovskite crystals. Here, we incorporated strategically designed passivating agents to investigate the effect of their interaction energies on the perovskite crystals and correlated these with the performance and longevity of the passivation effects. We unraveled that the passivation agents with a stronger interaction energy are advantageous not only for effective defect passivation but also to suppress defect migration. The prototypical PSCs treated with the optimal passivation agent exhibited superior performance and operational stability, retaining 81.9 and 85.3% of their initial performance under continuous illumination or nitrogen at 85 °C after 1008 h, respectively, while the reference device completely degraded during that time. This work provides important insights into designing operationally durable defect passivation agents for perovskite optoelectronic devices.

We present herein the development of a new polycationic molecular receptor, inspired by the ubiquitous cyclobis(paraquat- p-phenylene)cyclophane ("blue box"). Our analogue, the "white box", has been easily self-assembled on a preparative scale in water, using a template-assisted process by acyl hydrazone bonding of complementary bis(pyridinium)xylylene tweezers, followed by kinetic trapping of the empty receptor. The obtained macrocycle was found to display a marked pH responsiveness in water, because of an abnormal acidity of the amide protons within its structure. Consequently, and because of the concurrence of rotational isomerism under acidic conditions (fixed at higher pH values), the compound was found to display a dual behavior as a conformationally locked/flexible molecular host, being able to recognize appropriate aromatic substrates, in a lock and key or induced fit fashion, by a conjunction of π-π, C-H···π, and, crucially, the hydrophobic effect.

Piperidine scaffolds are present in a wide range of bioactive natural products and are therefore considered as highly valuable, privileged synthetic targets. In this manuscript, we describe a gold-catalyzed annulation strategy that allows a straightforward assembly of piperidines and piperidine-containing aza-bridged products from readily available alkene-tethered oxime ethers (or esters) and N-allenamides. Importantly, we demonstrate the advantages of using oxime derivatives over imines, something pertinent to the whole area of gold catalysis, and provide relevant mechanistic experiments that shed light into the factors affecting the annulation processes. Moreover, we also describe preliminary experiments demonstrating the viability of enantioselective versions of the above reactions.

Ammonia borane hydrolysis is considered as a potential means of safe and fast method of H 2 production if it is efficiently catalyzed. Here a series of nearly monodispersed alloyed bimetallic nanoparticle catalysts are introduced, optimized among transition metals, and found to be extremely efficient and highly selective with sharp positive synergy between 2/3 Ni and 1/3 Pt embedded inside a zeolitic imidazolate framework (ZIF-8) support. These catalysts are much more efficient for H 2 release than either Ni or Pt analogues alone on this support, and for instance the best catalyst Ni 2 Pt@ZiF-8 achieves a TOF of 600 mol H 2 ·mol catal -1 ·min -1 and 2222 mol H 2 ·mol Pt -1 ·min -1 under ambient conditions, which overtakes performances of previous Pt-base catalysts. The presence of NaOH boosts H 2 evolution that becomes 87 times faster than in its absence with Ni 2 Pt@ZiF-8, whereas NaOH decreases H 2 evolution on the related Pt@ZiF-8 catalyst. The ZIF-8 support appears outstanding and much more efficient than other supports including graphene oxide, active carbon and SBA-15 with these nanoparticles. Mechanistic studies especially involving kinetic isotope effects using D 2 O show that cleavage by oxidative addition of an O-H bond of water onto the catalyst surface is the rate-determining step of this reaction. The remarkable catalyst activity of Ni 2 Pt@ZiF-8 has been exploited for successful tandem catalytic hydrogenation reactions using ammonia borane as H 2 source. In conclusion the selective and remarkable synergy disclosed here together with the mechanistic results should allow significant progress in catalyst design toward convenient H 2 generation from hydrogen-rich substrates in the close future.

The first example of near-room-temperature α-arylation of benzo[ b]thiophenes is reported. The discovery rests on the observation of a switch in α-/β-regioselectivity at different loadings of Pd 2 (dba) 3 ·CHCl 3 in the coupling between benzo[ b]thiophene and 4-iodotoluene. We show that this unprecedented regioselectivity switch is driven by a Ag(I)-mediated C-H activation at the α-C-H position, which becomes the dominant mode of reactivity at low concentrations of Pd. Competition experiments, kinetic studies, KIE, and D/H scrambling experiments have been carried out supporting this mechanism.

Non-noble metal nanoparticles are notoriously difficult to prepare and stabilize with appropriate dispersion, which in turn severely limits their catalytic functions. Here, using zeolitic imidazolate framework (ZIF-8) as MOF template, catalytically remarkably efficient ligand-free first-row late transition-metal nanoparticles are prepared and compared. Upon scrutiny of the catalytic principles in the hydrolysis of ammonia-borane, the highest total turnover frequency among these first-row late transition metals is achieved for the templated Ni nanoparticles with 85.7 mol H2 mol cat -1 min -1 at room temperature, which overtakes performances of previous non-noble metal nanoparticles systems, and is even better than some noble metal nanoparticles systems. Mechanistic studies especially using kinetic isotope effects show that cleavage by oxidative addition of an O-H bond in H 2 O is the rate-determining step in this reaction. Inspired by these mechanistic studies, an attractive and effective "on-off" control of hydrogen production is further proposed.

The ever growing increase of CO2 concentration in the atmosphere is one of the main causes of global warming. Thus, CO2 activation and conversion toward valuable added compounds is a major scientific challenge. A new set of Au/δ-MoC and Cu/δ-MoC catalysts exhibits high activity, selectivity, and stability for the reduction of CO2 to CO with some subsequent selective hydrogenation toward methanol. Sophisticated experiments under controlled conditions and calculations based on density functional theory have been used to study the unique behavior of these systems. A detailed comparison of the behavior of Au/β-Mo2C and Au/δ-MoC catalysts provides evidence of the impact of the metal/carbon ratio in the carbide on the performance of the catalysts. The present results show that this ratio governs the chemical behavior of the carbide and the properties of the admetal, up to the point of being able to switch the rate and mechanism of the process for CO2 conversion. A control of the metal/carbon ratio paves the road for an efficient reutilization of this environmental harmful greenhouse gas.

The stereochemical outcome of cis-trans isomerization of optically pure [60], [70], and endohedral H2O@C60 fulleropyrrolidines reveals that the electronic nature of substituents, fullerene size, and surprisingly the incarcerated water molecule plays a crucial role in this rearrangement process. Theoretical DFT calculations are in very good agreement with the experimental findings. On the basis of the experimental results and computational calculations, a plausible reaction mechanism involving the hydrogen-bonding assistance of the inner water molecule in the carbanion stabilization of endofullerene is proposed.

Charge transport through an insulating layer was probed using ferrocenyl-terminated dendrimers and scanning electrochemical microscopy. Experiments show that the passage through the layer is considerably enhanced when the transferred charges are brought globally to the surface by the ferrocenyl dendrimer instead of a single ferrocene molecule. This result shows that charge tunneling through an insulator could be promoted by a purely molecular nano-object.

Synthesizing unprecedented diamagnetic adducts of an endohedral metallofullerene was achieved by using 1,3-dipolar cycloaddition reaction of paramagnetic La@C(s)-C82 with a simultaneous hydrogen addition. The selective formation of two main products, La@C(s)-C82HCMe2NMeCHPh (2a and 2b), was first detected by HPLC analysis and MALDI-TOF mass spectrometry. 2a and 2b-O, which was readily formed by the oxidation of 2b, were isolated by multistep HPLC separation and were fully characterized by spectroscopic methods, including 1D and 2D-NMR, UV-vis-NIR measurements and electrochemistry. The hydrogen atom was found to be connected to the fullerene cage directly in the case of 2a, and the redox behavior indicated that the C-H bond can still be readily oxidized. The reaction mechanism and the molecular structures of 2a and 2b were reasonably proposed by the interplay between experimental observations and DFT calculations. The feasible order of the reaction process would involve a 1,3-dipolar cycloaddition followed by the hydrogen addition through a radical pathway. It is concluded that the characteristic electronic properties and molecular structure of La@C(s)-C82 resulted in a site-selective reaction, which afforded a unique chemical derivative of an endohedral metallofullerene in high yields. Derivative 2a constitutes the first endohedral metallofullerene where the direct linking of a hydrogen atom has been structurally proven.

Polymers containing triazolylbiferrocene are synthesized by ROMP or radical chain reactions and react with HAuCl4 to provide class-2 mixed-valent triazolylbiferrocenium polyelectrolyte networks (observed inter alia by TEM and AFM) that encapsulate gold nanoparticles (AuNPs). With triazolylbiferrocenium in the side polymer chain, the intertwined polymer networks form nanosnakes, unlike with triazolylbiferrocenium in the main polymer chain. By contrast, simple ferrocene-containing polymers do not form such a ferricenium network upon reaction with Au(III), but only small AuNPs, showing that the triazolyl ligand, the cationic charge, and the biferrocenium structure are coresponsible for such network formations.

The electronic properties of Pt nanoparticles deposited on CeO(2)(111) and CeO(x)/TiO(2)(110) model catalysts have been examined using valence photoemission experiments and density functional theory (DFT) calculations. The valence photoemission and DFT results point to a new type of "strong metal-support interaction" that produces large electronic perturbations for small Pt particles in contact with ceria and significantly enhances the ability of the admetal to dissociate the O-H bonds in water. When going from Pt(111) to Pt(8)/CeO(2)(111), the dissociation of water becomes a very exothermic process. The ceria-supported Pt(8) appears as a fluxional system that can change geometry and charge distribution to accommodate adsorbates better. In comparison with other water-gas shift (WGS) catalysts [Cu(111), Pt(111), Cu/CeO(2)(111), and Au/CeO(2)(111)], the Pt/CeO(2)(111) surface has the unique property that the admetal is able to dissociate water in an efficient way. Furthermore, for the codeposition of Pt and CeO(x) nanoparticles on TiO(2)(110), we have found a transfer of O from the ceria to Pt that opens new paths for the WGS process and makes the mixed-metal oxide an extremely active catalyst for the production of hydrogen.

The promising properties of anatase TiO(2) nanocrystals exposing specific surfaces have been investigated in depth both theoretically and experimentally. However, a clear assessment of the role of the crystal faces in photocatalytic processes is still under debate. In order to clarify this issue, we have comprehensively explored the properties of the photogenerated defects and in particular their dependence on the exposed crystal faces in shape-controlled anatase. Nanocrystals were synthesized by solvothermal reaction of titanium butoxide in the presence of oleic acid and oleylamine as morphology-directing agents, and their photocatalytic performances were evaluated in the phenol mineralization in aqueous media, using O(2) as the oxidizing agent. The charge-trapping centers, Ti(3+), O(-), and O(2)(-), formed by UV irradiation of the catalyst were detected by electron spin resonance, and their abundance and reactivity were related to the exposed crystal faces and to the photoefficiency of the nanocrystals. In vacuum conditions, the concentration of trapped holes (O(-) centers) increases with increasing {001} surface area and photoactivity, while the amount of Ti(3+) centers increases with the specific surface area of {101} facets, and the highest value occurs for the sample with the worst photooxidative efficacy. These results suggest that {001} surfaces can be considered essentially as oxidation sites with a key role in the photoxidation, while {101} surfaces provide reductive sites which do not directly assist the oxidative processes. Photoexcitation experiments in O(2) atmosphere led to the formation of Ti(4+)-O(2)(-) oxidant species mainly located on {101} faces, confirming the indirect contribution of these surfaces to the photooxidative processes. Although this work focuses on the properties of TiO(2), we expect that the presented quantitative investigation may provide a new methodological tool for a more effective evaluation of the role of metal oxide crystal faces in photocatalytic processes.

A Cu(111) surface displays a low activity for the oxidation of carbon monoxide (2CO + O(2) → 2CO(2)). Depending on the temperature, background pressure of O(2), and the exposure time, one can get chemisorbed O on Cu(111) or a layer of Cu(2)O that may be deficient in oxygen. The addition of ceria nanoparticles (NPs) to Cu(111) substantially enhances interactions with the O(2) molecule and facilitates the oxidation of the copper substrate. In images of scanning tunneling microscopy, ceria NPs exhibit two overlapping honeycomb-type moiré structures, with the larger ones (H(1)) having a periodicity of 4.2 nm and the smaller ones (H(2)) having a periodicity of 1.20 nm. After annealing CeO(2)/Cu(111) in O(2) at elevated temperatures (600-700 K), a new phase of a Cu(2)O(1+x) surface oxide appears and propagates from the ceria NPs. The ceria is not only active for O(2) dissociation, but provides a much faster channel for oxidation than the step edges of Cu(111). Exposure to CO at 550-750 K led to a partial reduction of the ceria NPs and the removal of the copper oxide layer. The CeO(x)/Cu(111) systems have activities for the 2CO + O(2) → 2CO(2) reaction that are comparable or larger than those reported for surfaces of expensive noble metals such as Rh(111), Pd(110), and Pt(100). Density-functional calculations show that the supported ceria NPs are able to catalyze the oxidation of CO due to their special electronic and chemical properties. The configuration of the inverse oxide/metal catalyst opens new interesting routes for applications in catalysis.

A family of rigid ferrocenyl-terminated redox stars has been synthesized--by Negishi coupling, including hexa(ferrocenethynyl)benzene complexes, a dodecaferrocenyl star, and stars with extended rigid tethers--and fully characterized. Cyclic voltammetry (CV) studies of the parent complex hexa(ferrocenylethynyl)benzene, 1, show a single wave for the six-electron oxidation of 1 using Nn-Bu(4)PF(6) as the supporting electrolyte on a Pt anode in CH(2)Cl(2), whereas three distinct two-electron reversible CV waves are observed using Nn-Bu(4)BAr(F)(4) (Ar(F) = 3,5-C(6)H(3)-(CF(3))(2)). The CV of 1,3,5-tris(ferrocenylethynyl)benzene, 11, also shows only one wave for the three-electron transfer with Nn-Bu(4)PF(6) and three one-electron waves using Nn-Bu(4)BAr(F)(4). This confirms the lack of electronic communication between the ferrocenyl groups and a significant electrostatic effect among the oxidized ferrocenyl groups. This effect is not significant between para-ferrocenyl groups in 1,4-bis(ferrocenylethynyl)benzene for which only a single wave is observed even with Nn-Bu(4)BAr(F)(4) as the supporting electrolyte. The para-ferrocenyl substituents are quite independent, which explains that two para-ferrocenyl groups are oxidized at about the same potential in a single CV wave of 1. With the additional steric bulk introduced with a methyl substituent on the ferrocenyl group, however, even the para-methylferrocenyl groups are submitted to a small electrostatic effect splitting the six-electron transfer into six single-electron waves, probably because of the overall steroelectronic constraints. Contrary to 11, 1,3-bis(ferrocenylethynyl)benzene and related complexes with a third, different substituent in the remaining meta position different from a ferrocenylethynyl only show a single two-electron wave using Nn-Bu(4)BAr(F)(4), which is attributed to the transoïd conformation of the ferricinium groups minimizing the electrostatic effect. This shows that, in 11, it is the steric frustration that is responsible for the electrostatic effect, and the same occurs in 1. In several cases, ΔE(p) is much larger than the expected 60 mV value, characterizing a quasi-reversible (i.e., relatively slow) redox process. It is suggested that this slower electron transfer be attributed to conformational rearrangement of the ferrocenyl groups toward the transoïd position in the course of electron transfer. Thus both the thermodynamic and kinetic aspects of the electrostatic factor (isolated from the electronic factor), including the frustration effect, are characterized. The distinction between the electronic communication and through-space electrostatic effect was made possible in all of these complexes in which the absence of wave splitting using a strongly ion-pairing electrolyte shows the absence of significant electronic communication, and was confirmed by the new frustration phenomenon.

Water-soluble arene-cored "clicked" and non-"clicked" dendrimers terminated by 27, 81, and 243 triethyleneglycol (TEG) tethers (respectively generations G0, G1, and G2) have been synthesized and shown to form dendrimer-encapsulated gold nanoparticles (DEAuNPs) and dendrimer-stabilized gold nanoparticles (DSAuNPs). The dendrimers have been characterized by IR, (1)H NMR, (13)C NMR, size-exclusion chromatography, elemental analysis, MALDI-TOF mass spectroscopy, DOSY NMR, and dynamic light scattering. The AuNPs have been generated and stabilized by these PEGylated dendrimers using a variety of reduction modes, including NaBH(4) in methanol, various single-electron metallocene-type reductants, and even in the absence of additional reductants. The active role of the "clicked" triazole rings, dendrimer generation, stoichiometry of Au precursor, and nature of the reductant and of the solvent are delineated, leading to DSAuNPs with the G0 dendrimer and smaller DEAuNPs with the G1 and G2 dendrimers. Altogether, AuNPs in the size range from 1.8 to 42 nm were formed and characterized by transmission electron microscopy (TEM), high resolution TEM (HRTEM) and UV-vis spectroscopy. Both 1,2,3-triazole and PEGylated Percec-type dendrons are required in the dendrimer structure for the stabilization of AuNPs upon NaBH(4) reduction of HAuCl(4) in methanol. On the other hand, in the absence of other reductant in water, only PEGylated Percec-type dendrons in dendrimers were found to be indispensable, because of their semicavitand shape, for the spontaneous reduction of HAuCl(4) and stabilization of DSAuNPs.

At small coverages of ceria on TiO(2)(110), the CeO(x) nanoparticles have an unusual coordination mode. Scanning tunneling microscopy and density-functional calculations point to the presence of Ce(2)O(3) dimers, which form diagonal arrays that have specific orientations of 0, 24, and 42 degrees with respect to the [1 -1 0] direction of the titania substrate. At high coverages of ceria on TiO(2)(110), the surface exhibits two types of terraces. In one type, the morphology is not very different from that observed at low ceria coverage. However, in the second type of terrace, there is a compact array of ceria particles with structures that do not match the structures of CeO(2)(111) or CeO(2)(110). The titania substrate imposes on the ceria nanoparticles nontypical coordination modes, enhancing their chemical reactivity. This phenomenon leads to a larger dispersion of supported metal nanoparticles (M = Au, Cu, Pt) and makes possible the direct participation of the oxide in catalytic reactions. The M/CeO(x)/TiO(2)(110) surfaces display an extremely high catalytic activity for the water-gas shift reaction that follows the sequence Au/CeO(x)/TiO(2)(110) < Cu/CeO(x)/TiO(2)(110) < Pt/CeO(x)/TiO(2)(110). For low coverages of Cu and CeO(x), Cu/CeO(x)/TiO(2)(110) is 8-12 times more active than Cu(111) or Cu/ZnO industrial catalysts. In the M/CeO(x)/TiO(2)(110) systems, there is a strong coupling of the chemical properties of the admetal and the mixed-metal oxide: The adsorption and dissociation of water probably take place on the oxide, CO adsorbs on the admetal nanoparticles, and all subsequent reaction steps occur at the oxide-admetal interface. The high catalytic activity of the M/CeO(x)/TiO(2)(110) surfaces reflects the unique properties of the mixed-metal oxide at the nanometer level.

Giant redox dendrimers were synthesized with ferrocenyl and pentamethylferrocenyl termini up to a theoretical number of 3(9) tethers (seventh generation). Lengthening of the tethers proved to be a reliable strategy to overcome the bulk constraint at the dendrimers periphery. These redox metallodendrimers were characterized by (1)H, (13)C, and (29)Si NMR; MALDI-TOF mass spectrometry (for the low generations); elemental analysis; UV-vis spectroscopy; dynamic light scattering (DLS); atomic force microscopy (AFM); electron-force microscopy (EFM) for half- or fully oxidized dendrimers; cyclic voltammetry; and coulometry. UV-vis spectroscopy, coulometry, and analytical data are consistent with an increasing amount of defects as the generation number increases, with this amount remaining relatively weak up to G(5). AFM shows that the dendrimers form aggregates of discrete size on the mica surface, recalling the agglomeration of metal atoms in monodisperse nanoparticles. Cyclic voltammetry reveals full chemical and electrochemical reversibility up to G(7), showing that electron transfer is fast among the flexible peripheral redox sites. Indeed, the redox stability of these new electrochromic dendrimers, i.e., a battery behavior, was established by complete chemical oxido-reduction cycles, and the blue 17-electron ferrocenium and deep-green mixed-valence Fe(III)/Fe(II) dendritic complexes were isolated and characterized. AFM studies also show the reversible dendrimer size changes from upon redox switching between Fe(II) and Fe(III), suggesting a breathing mechanism controlled by the redox potential. Considerable adsorption of high-generation dendrimers on Pt electrodes such as G(7)-Fc allows the easy formation of modified electrodes that sense the ATP anion only involving the electrostatic factor even in the absence of any other type of interaction with the redox tethers.

Inclusion bodies are insoluble aggregates that are formed by bacteria to store excess recombinant protein produced during expression. The structure of the protein in inclusion bodies is poorly understood but it has been hypothesized that the protein may form misfolded beta sheet aggregates. This paper presents an isotopic labeling and solid-state nuclear magnetic resonance approach to determine the secondary structure of individual residues within a recombinant influenza virus "FHA2" protein in inclusion bodies. The inclusion bodies were studied either in the context of the unlysed hydrated E. coli cells or in the hydrated pellet formed from centrifugation of the material insoluble in the cell lysate. The native structure of FHA2 is predominantly helical and native helical structure was also observed for several specific residues in the inclusion body FHA2. This approach will be applicable to structural analysis of many inclusion body proteins and should provide useful information for optimizing solubilization and purification protocols of these proteins.

N-doping of titania makes photocatalytic activity possible for the splitting of water, and other reactions, under visible light. Here, we show from both theory and experiment that Au preadsorption on TiO2 surfaces significantly increases the reachable amount of N implanted in the oxide. The stabilization of the embedded N is due to an electron transfer from the Au 6s levels toward the N 2p levels, which also increases the Au-surface adhesion energy. Theoretical calculations predict that Au can also stabilize embedded N in other metal oxides with photocatalytic activity, such as SrTiO3 and ZnO, producing new states above the valence band or below the conduction band of the oxide. In experiments, the Au/TiN(x)O(2-y) system was found to be more active for the dissociation of water than TiO2, Au/TiO2, or TiO(2-y). Furthermore, the Au/TiN(x)O(2-y) surfaces were able to catalyze the production of hydrogen through the water-gas shift reaction (WGS) at elevated temperatures (575-625 K), displaying a catalytic activity superior to that of pure copper (the most active metal catalysts for the WGS) or Cu nanoparticles supported on ZnO.

Olefin cross metathesis was used to efficiently functionalize polyolefin dendrimers, polymers, and gold nanoparticles using the second-generation Grubbs catalyst. In these structures, the tethers were lengthened to prevent the facile cross metathesis that otherwise predominates in polyolefin dendrimers having short tethers. This synthetic strategy allows the one-step access to polyacid, polyester, and polyferrocenyl structures from polyolefins. Cross metathesis is also used to efficiently achieve an iterative divergent dendritic construction. All the cross metathesis reactions were monitored by 1H NMR showing the chemo-, regio-, and stereoselectivity. MALDI-TOF mass spectrometry was a very useful technique to confirm the efficiency of this synthetic strategy.

Structure-based drug discovery requires the iterative determination of protein-ligand costructures in order to improve the binding affinity and selectivity of potential drug candidates. In general, X-ray and NMR structure determination methods are time consuming and are typically the limiting factor in the drug discovery process. The application of molecular docking simulations to filter and evaluate drug candidates has become a common method to improve the throughput and efficiency of structure-based drug design. Unfortunately, molecular docking methods suffer from common problems that include ambiguous ligand conformers or failure to predict the correct docked structure. A rapid approach to determine accurate protein-ligand costructures is described based on NMR chemical shift perturbation (CSP) data routinely obtained using 2D 1H-15N HSQC spectra in high-throughput ligand affinity screens. The CSP data is used to both guide and filter AutoDock calculations using our AutoDockFilter program. This method is demonstrated for 19 distinct protein-ligand complexes where the docked conformers exhibited an average rmsd of 1.17 +/- 0.74 A relative to the original X-ray structures for the protein-ligand complexes.

A key factor in the potential clinical utility of membrane-active antibiotics is their cell selectivity (i.e., prokaryote over eukaryote). Cationic steroid antibiotics were developed to mimic the lipid A binding character of polymyxin B and are shown to bind lipid A derivatives with affinity greater than that of polymyxin B. The outer membranes of Gram-negative bacteria are comprised primarily of lipid A, and a fluorophore-appended cationic steroid antibiotic displays very high selectivity for Gram-negative bacterial membranes over Gram-positive bacteria and eukaryotic cell membranes. Giant pentane-soluble organo-silicon dendrimers have been synthesized using a triallylphenol brick according to a new divergent construction that uses a hydrosilylation-nucleophilic substitution sequence up to the ninth generation (G(9)). All the reactions were monitored by (1)H, (13)C, and (29)Si NMR until G(9), indicating that they were clean at the NMR accuracy until this last generation. MALDI TOF mass spectra were recorded for G(1) to G(4) and show the nature and amounts of defects that are intrinsic to the divergent construction. This technique and SEC (recorded up to G(5)) confirm the monodispersity (1.00 to 1.02) from G(1) to G(5). HRTEM and AFM images recorded for the high generations disclose the expected smooth dendrimer size progression and the globular shape. At G(9), the theoretical number of termini (TNT) is 177 407 branches (abbreviation: G(9)-177 047). It is estimated that more than 10(5) terminal branches are actually present in the G(9) dendrimer, far beyond the De Gennes "dense-packing" limit (6000 branches), and it is believed that the branch termini turn inside the dendrimer toward the core. This is corroborated by lower reaction rates and yields for the highest generation numbers presumably due to intradendritic reactions. It is probable that the dendritic construction is limited by the density of branches inside the dendrimer, i.e., far beyond the dense-packing limit.

Tri- and nonaferrocenyl thiol dendrons have been synthesized and used to assemble dendronized gold nanoparticles either by the ligand-substitution method from dodecanethiolate-gold nanoparticles (AB(3) units) or Brust-type direct synthesis from a 1:1 mixture of dodecanethiol and dendronized thiol (AB(9) units). The dendronized colloids are a new type of dendrimers with a gold colloidal core. Two colloids containing a nonasilylferrocenyl dendron have been made; they bear respectively 180 and 360 ferrocenyl units at the periphery. These colloids selectively recognize the anions H(2)PO(4)(-) and adenosine-5'-triphosphate (ATP(2)(-)) with a positive dendritic effect and can be used to titrate these anions because of the shift of the CV wave even in the presence of other anions such as Cl(-) and HSO(4)(-). Recognition is monitored by the appearance of a new wave at a less positive potential in cyclic voltammetry (CV). The anion HSO(4)(-) is also recognized and titrated by the dendronized colloid containing the tris-amidoferrocenyl units, because of the progressive shift of the CV wave until the equivalence point. These dendronized colloids can form robust modified electrodes by dipping the naked Pt electrode into a CH(2)Cl(2) solution containing the colloids. The robustness is all the better as the dendron is larger. These modified electrodes can recognize H(2)PO(4)(-), ATP(2)(-) and HSO(4)(-), be washed with minimal loss of adsorbed colloid, and be reused.

The DSM polyamine dendrimers dend-DAB-(NH2)x of generations 1 (x = 4) to 4 (x = 32) form H-bonded dendritic assemblies with the phenol AB3 units p-HOC6H4C(CH2CHCH2)3 and p-HOC6H4C{(CH2)3SiCH2NHCOFc}3 (Fc = ferrocenyl), as shown by the shifts of the NH2 and OH signals giving a concentration-dependent common signal between 2.4 and 4.1 ppm in CDCl3. The supramolecular dendrimers efficiently recognize H2PO4- anions with positive and unusual dendritic effects upon electrochemical titration involving half-stoichiometry for G1, a sudden cyclovoltammetry wave change at the equivalent point, and a dramatic intensity decrease of the new wave.

Stable, CH(2)Cl(2)-soluble mixed dodecanethiol/(amidoferrocenyl)alkanethiol (AFAT) gold colloids were synthesized by ligand substitution reactions from Brust's dodecanethiol gold colloids and the AFAT ligands to study the recognition and titration of oxoanions. Gold colloids were obtained with various chain lengths (C(11) and C(6) chains) of the AFAT ligand and different proportions of AFAT ligands in the colloids. Modification of the amidoferrocenyl structure [replacement of the free C(5)H(5) ferrocene ring by C(5)Me(5) (Cp) or C(5)H(4)COCH(3)] has been achieved to investigate the stereoelectronic effects on the recognition. The cyclic voltammetry of these colloids in CH(2)Cl(2) on Pt electrode shows a reversible Fe(II/III) wave with some adsorption. With AFAT ligands, a new, less electrochemically reversible wave (with some adsorption) at a potential 220 +/- 20 mV less positive than that of the initial wave appears upon titration of [n-Bu(4)N][H(2)PO(4)], and the initial wave completely disappears after addition of 1 equiv of anion, which allows its titration. The potential shift does not depend on the AFAT proportion nor on its chain length but is reduced with Cp and enhanced with C(5)H(4)COCH(3), showing the key role of the hydrogen bonding between the -NH-amido group and a terminal oxygen atom of the oxoanions. According to the Echegoyen-Kaifer model, the potential shift leads to the ratio K((+))/K((0)) of apparent association constants. In the presence of both [n-Bu(4)N][HSO(4)] and [n-Bu(4)N]Cl, a shift of the initial wave (rather than its replacement) allows an easy titration, ideally with 20-Fc. Upon addition of [n-Bu(4)N][HSO(4)] alone, a weak wave shift (30 mV) of the colloids is also observed, allowing the titration of the HSO(4)(-) anion by the colloids containing a low percentage of AFAT ligand. The Echegoyen-Kaifer model provides access to the apparent association constant K((+)) in this case for which the interaction between the anion and the neutral form of the host is not significant. With the C(5)H(4)COCH(3) modification of the amidoferrocenyl branch, a new wave appears at a potential 70 mV more positive than the initial wave, signifying a stronger interaction with this modified ligand than with the parent AFAT ligand. These colloids favorably compare with ferrocenyl dendrimers in terms of rapid synthesis and selectivity of H(2)PO(4-) over HSO(4-) and with gold surfaces for the recognition of HSO(4)(-).
