Abstract

The first single-molecule (SM) fluorescence studies of cytochromes P450 (CYPs) are used for measuring residence times of the fluorescent dye Nile Red (NR) bound to CYP isoform 3A4 (CYP3A4) incorporated in surface-immobilized lipid Nanodiscs, with and without the effector [alpha]-naphthoflavone (ANF). The SM methods have provided superior sensitivity and complementary detail to ensemble methods, thus providing novel insights into CYP3A4 allosteric mechanisms.
