The plasmon-driven chemistry of ferri-/ferrocyanide ions inside surface-enhanced Raman spectroscopy (SERS) active hot spots associated with gold nanoparticle oligomers is studied with continuous wave (CW) pump-probe SERS. By comparing with solution-phase normal Raman spectra, the characteristic spectral variations observed upon 532 nm optical pumping can be attributed to an oxidation process that occurs on the surface species followed by desorption of the oxidized surface species from the gold nanoparticles. Interrogating the plasmon-driven processes over a wide range of temperatures reveals that neither process is purely driven by the thermal effects associated with the optical pumping, and the apparent activation energies of both steps are estimated based on semiquantitative SERS analysis. Our observation identifies a more detailed reaction pathway for this classic model system under considerably simplified reaction conditions, adding to the current mechanistic background for future plasmon-driven chemistry studies and applications.

Electrochemical tip-enhanced Raman spectroscopy (EC-TERS) has been implemented to investigate the structure and activity of iron(II) phthalocyanine (FePc)-a model catalyst for the oxygen reduction reaction (ORR). Using EC-TERS, both reversible change and irreversible degradation to FePc have been observed during ORR. The reversible change in the Raman spectrum of FePc can be related to the FePc molecules that adapt a nonplanar geometry during catalysis. In contrast, the irreversible degradation of FePc is a consequence of FePc demetalation, leading to the subsequent formation of free base phthalocyanine. This observation affirms that FePc demetalation during ORR proceeds via a direct loss of Fe 2+ and that carbon corrosion is not the operative mechanism. Importantly, the FePc demetalation process can be correlated with a loss of ORR activity suggesting that Fe-containing sites are essential for FePc to achieve high catalytic activity. This study establishes EC-TERS as a promising technique for the operando characterization of electrocatalytic reactions at the molecular scale.

Nanoparticle encapsulation inside zirconium-based metal-organic frameworks (NP@MOF) is hard to control, and the resulting materials often have nonuniform morphologies with NPs on the external surface of MOFs and NP aggregates inside the MOFs. In this work, we report the controlled encapsulation of gold nanorods (AuNRs) by a scu-topology Zr-MOF, via a room-temperature MOF assembly. This is achieved by functionalizing the AuNRs with poly(ethylene glycol) surface ligands, allowing them to retain colloidal stability in the precursor solution and to seed the MOF growth. Using this approach, we achieve core-shell yields exceeding 99%, tuning the MOF particle size via the solution concentration of AuNRs. The functionality of AuNR@MOFs is demonstrated by using the AuNRs as embedded probes for selective surface-enhanced Raman spectroscopy (SERS). The AuNR@MOFs are able to both take-up or block molecules from the pores, thereby facilitating highly selective sensing at the AuNR ends. This proof-of-principle study serves to present both the outstanding level of control in the synthesis and the high potential for AuNR@Zr-MOF composites for SERS.

Atomic layer deposition (ALD) of TiO 2 was performed in tandem with in situ surface-enhanced Raman spectroscopy (SERS) to monitor changes in the transient surface species across multiple ALD cycles. A self-assembled monolayer of 3-mercaptopropionic acid was used as a capture agent to ensure that nucleation of the titanium precursor (titanium tetraisopropoxide [TTIP]) occurs. Comparisons between the Raman spectra of the neat precursor and the SER spectra of the first ALD cycle of TiO 2 reveal typical ligand exchange chemistry taking place, with self-limiting behavior and intact isopropoxide ligands. However, subsequent cycles show drastically different chemistry, with no isopropoxide ligands remaining at any point during the second and third cycles. Continuous exposure of either TTIP or isopropyl alcohol after the first cycle shows unlimited chemical vapor deposition (CVD)-type growth. Comparisons with alternative precursors (aluminum isopropoxide, titanium tert-butoxide, and titanium propoxide) and DFT calculations reveal that, for the TTIP precursor, isolated TiO 2 sites play a role in the dehydration of off-gassing isopropyl alcohol. The resulting propene then undergoes oligomerization into six-carbon olefins before polymerizing into indistinguishable carbon products that accumulate on the surface. The emergence of the dehydration chemistry is expected to be exclusively the result of these isolated TiO 2 sites and, as such, is expected to occur on other surfaces where TiO 2 ALD is feasible. This work showcases how seemingly innocuous ALD can evolve into a CVD process when the products can participate in various side reactions with newly made surface sites.

Singlet fission (SF) in two or more electronically coupled organic chromophores converts a high-energy singlet exciton into two low-energy triplet excitons, which can be used to increase solar cell efficiency. Many known SF chromophores are unsuitable for device applications due to chemical instability and low triplet state energies. The results described here show that efficient SF occurs in polycrystalline thin films of 9,10-bis(phenylethynyl)anthracene (BPEA), a commercial dye that has singlet and triplet energies of 2.40 and 1.11 eV, respectively, in the solid state. BPEA crystallizes into two polymorphs with space groups C2/ c and Pbcn, which undergo SF with k SFA = (109 ± 4 ps) -1 and k SFB = (490 ± 10 ps) -1 , respectively. The high triplet energy and efficient SF evidenced from the 180 ± 20% triplet yield make BPEA a promising candidate for enhancing solar cell performance.

Jarosite, a mineral with a kagomé lattice, displays magnetic frustration yet orders magnetically below 65 K. As magnetic frustration can engender exotic physical properties, understanding the complex magnetism of jarosite comprises a multidecade interdisciplinary challenge. Unraveling the nature of the disparate magnetic coupling interactions that lead to magnetic order in jarosite remains an open question. Specifically, there is no observed trend in the interlayer spacing with magnetic order. Similarly, the relationship between metal-ligand bond distance and magnetic order remains uninvestigated. Here, we use applied pressure to smoothly vary jarosite's structure without manipulating the chemical composition, enabling a chemically invariant structure-function study. Using single-crystal and powder X-ray diffraction, we show that high applied pressures alter both the interlayer spacing and the metal-ligand bond distances. By harnessing a suite of magnetic techniques under pressure, including SQUID-based magnetometry, time-resolved synchrotron Mössbauer spectroscopy, and X-ray magnetic circular dichroism, we construct the magnetic phase diagram for jarosite up to 40 GPa. Notably, we demonstrate that the magnetic ordering temperature increases dramatically to 240 K at the highest pressures. Additionally, we conduct X-ray emission spectroscopy, Mössbauer spectroscopy, and UV-visible absorption spectroscopy experiments to comprehensively map the magnetic and electronic structures of jarosite at high pressure. We use these maps to construct chemically pure magnetostructural correlations which fully explain the nature and role of the disparate magnetic coupling interactions in jarosite.

Continuous wave (CW) pump-probe surface-enhanced Raman spectroscopy (SERS) is used to examine a range of plasmon-driven chemical behavior in the molecular SERS signal of trans-1,2-bis(4-pyridyl)ethylene (BPE) adsorbed on individual Au nanosphere oligomers (viz., dimers, trimers, tetramers, etc.). Well-defined new transient modes are caused by high fluence CW pumping at 532 nm and are monitored on the seconds time scale using a low intensity CW probe field at 785 nm. Comparison of time-dependent density functional theory (TD-DFT) calculations with the experimental data leads to the conclusion that three independent chemical processes are operative: (1) plasmon-driven electron transfer to form the BPE anion radical; (2) BPE hopping between two adsorption sites; and (3) trans-to- cis-BPE isomerization. Resonance Raman and electron paramagnetic resonance (EPR) spectroscopy measurements provide further substantiation for the observation of an anion radical species formed via a plasmon-driven electron transfer reaction. Applications of these findings will greatly impact the design of novel plasmonic devices with the future ability to harness new and efficient energetic pathways for both chemical transformation and photocatalysis at the nanoscale level.

Ultrahigh vacuum tip-enhanced Raman spectroscopy (UHV-TERS) is used to investigate adsorption of molecular oxygen (O 2 ) on cobalt(II) phthalocyanine (CoPc) supported on Ag(111) single crystal surfaces, which is the initial step for the oxygen reduction reaction (ORR) using metal Pc catalysts. Two adsorption configurations are primarily observed, assigned as O 2 /CoPc/Ag(111) and O/CoPc/Ag(111) based on scanning tunneling microscopy (STM) imaging, TERS, isotopologue substitution, and density functional theory (DFT) calculations. Distinct vibrational features are observed for different adsorption configurations such as the 18 O- 18 O stretching frequency at 1151 cm -1 for O 2 /CoPc/Ag(111), and Co- 16 O and Co- 18 O vibrational frequencies at 661 and 623 cm -1 , respectively, for O/CoPc/Ag(111). DFT calculations show vibrational mode coupling of O-O and Co-O vibrations to the Pc ring, resulting in different symmetries of oxygen-related normal modes. This study establishes UHV-TERS as a chemically sensitive tool for probing catalytic systems at the molecular scale.

Ultrahigh vacuum tip-enhanced Raman spectroscopy (UHV-TERS) combines the atomic-scale imaging capability of scanning probe microscopy with the single-molecule chemical sensitivity and structural specificity of surface-enhanced Raman spectroscopy. Here, we use these techniques in combination with theory to reveal insights into the influence of intermolecular interactions on the vibrational spectra of a N-N'-bis(2,6-diisopropylphenyl)-perylene-3,4:9,10-bis(dicarboximide) (PDI) self-assembled monolayer adsorbed on single-crystal Ag substrates at room temperature. In particular, we have revealed the lifting of a vibrational degeneracy of a mode of PDI on Ag(111) and Ag(100) surfaces, with the most strongly perturbed mode being that associated with the largest vibrational amplitude on the periphery of the molecule. This work demonstrates that UHV-TERS enables direct measurement of molecule-molecule interaction at nanoscale. We anticipate that this information will advance the fundamental understanding of the most important effect of intermolecular interactions on the vibrational modes of surface-bound molecules.

We clarify mechanistic questions regarding plasmon-driven chemistry and nanoscale photocatalysis within optically confined near-field plasmonic systems. Using surface-enhanced Raman scattering (SERS), we directly monitor the photoinduced reaction dynamics of 4,4'-bipyridine molecules, localized in plasmonic hot spots within individual gold nanosphere oligomers. Our experiment generates surface electrons from the gold nanoparticle using an intense off-molecular resonance continuous wave pump field, and detects radical anion products via SERS. This is done by adopting a dual-wavelength spectroscopic approach. Empirical evidence of plasmon-driven electron transfer is provided for the first time by direct detection of the 4,4'-bipyridine radical anion species localized in the plasmonic hot spots of individual gold nanosphere oligomers, corroborated by open-shell density functional theory calculations. An isotopologue approach using both protonated and deuterated 4,4'-bipyridine molecules demonstrates the single molecule response of plasmon-driven electron transfer occurring in single nanosphere oligomer systems with a 3% yield, a phenomenon unobserved in ensemble measurements under analogous experimental conditions. This mechanism has broad applicability to using nanoscale chemical reactors for surface redox reactions on the subnanometer scale.

Operando surface-enhanced Raman spectroscopy (SERS) was used to successfully identify hitherto unknown dimeric methylalumina surface species during atomic layer deposition (ALD) on a silver surface. Vibrational modes associated with the bridging moieties of both trimethylaluminum (TMA) and dimethylaluminum chloride (DMACl) surface species were found during ALD. The appropriate monomer vibrational modes were found to be absent as a result of the selective nature of SERS. Density functional theory (DFT) calculations were also performed to locate and identify the expected vibrational modes. An operando localized surface plasmon resonance (LSPR) spectrometer was utilized to account for changes in SER signal as a function of the number of ALD cycles. DMACl surface species were unable to be measured after multiple ALD cycles as a result of a loss in SERS enhancement and shift in LSPR. This work highlights how operando optical spectroscopy by SERS and LSPR scattering are useful for probing the identity and structure of the surface species involved in ALD and, ultimately, catalytic reactions on these support materials.

This paper demonstrates the direct sensing of glucose at physiologically relevant concentrations with surface-enhanced Raman spectroscopy (SERS) on gold film-over-nanosphere (AuFON) substrates functionalized with bisboronic acid receptors. The combination of selectivity in the bisboronic acid receptor and spectral resolution in the SERS data allow the sensors to resolve glucose in high backgrounds of fructose and, in combination with multivariate statistical analysis, detect glucose accurately in the 1-10 mM range. Computational modeling supports assignments of the normal modes and vibrational frequencies for the monoboronic acid base of our bisboronic acids, glucose and fructose. These results are promising for the use of bisboronic acids as receptors in SERS-based in vivo glucose monitoring sensors.

We demonstrate that incorporation of a redox-active benzoquinoid ligand into a one-dimensional chain compound can give rise to a material that exhibits simultaneous solid-state redox switching of optical, magnetic, and electronic properties. Metalation of the ligand 4,5-bis(pyridine-2-carboxamido)-1,2-catechol ((N,O)LH4) with Mn(III) affords the chain compound Mn((N,O)L)(DMSO). Structural and spectroscopic analysis of this compound show the presence of Mn(II) centers bridged by (N,O)L(2-) ligands, resulting partially from a spontaneous ligand-to-metal electron transfer. Upon soaking in a solution of the reductant Cp2Co, Mn((N,O)L)(DMSO) undergoes a ligand-centered solid-state reduction to [Mn((N,O)L)](-), as revealed by a suite of techniques, including Raman and X-ray absorption spectroscopy. The ligand-based reduction engenders a dramatic modulation of the physical properties of the chain compound. An electrochromic response, evidenced by a color change from dark green to dark purple is accompanied by a nearly 40-fold increase in magnetic coupling strength, from J = -0.38(1) to -15.6(2) cm(-1), and a 10,000-fold increase in electronic conductivity, from σ = 2.33(1) × 10(-12) S/cm (Ea = 0.64(1) eV) to 8.61(1) × 10(-8) S/cm (Ea = 0.39(1) eV). Importantly, the chemical reduction is reversible: treatment of the reduced compound with [Cp2Fe](+) regenerates the oxidized chain. Taken together, these results highlight the ability of benzoquinoid ligands to facilitate solid-state ligand-based redox reactions in nonporous coordination solids, giving rise to reversible switching of optical properties, magnetic exchange interactions, and electronic conductivity.

The incorporation of tetraoxolene radical bridging ligands into a microporous magnetic solid is demonstrated. Metalation of the redox-active bridging ligand 2,5-dichloro-3,6-dihydroxy-1,4-benzoquinone (LH2) with Fe(II) affords the solid (Me2NH2)2[Fe2L3]·2H2O·6DMF. Analysis of X-ray diffraction, Raman spectra, and Mössbauer spectra confirm the presence of Fe(III) centers with mixed-valence ligands of the form (L3)(8-) that result from a spontaneous electron transfer from Fe(II) to L(2-). Upon removal of DMF and H2O solvent molecules, the compound undergoes a slight structural distortion to give the desolvated phase (Me2NH2)2[Fe2L3], and a fit to N2 adsorption data of this activated compound gives a BET surface area of 885(105) m(2)/g. Dc magnetic susceptibility measurements reveal a spontaneous magnetization below 80 and 26 K for the solvated and the activated solids, respectively, with magnetic hysteresis up to 60 and 20 K. These results highlight the ability of redox-active tetraoxolene ligands to support the formation of a microporous magnet and provide the first example of a structurally characterized extended solid that contains tetraoxolene radical ligands.

Confirmatory, nondestructive, and noninvasive identification of colorants in situ is of critical importance for the understanding of historical context and for the long-term preservation of cultural heritage objects. Although there are several established techniques for analyzing cultural heritage materials, there are very few analytical methods that can be used for molecular characterization when very little sample is available, and a minimally invasive approach is required. Tip-enhanced Raman spectroscopy (TERS) is a powerful analytical technique whose key features include high mass sensitivity, high spatial resolution, and precise positioning of the tip. In the current proof-of-concept study we utilized TERS to identify indigo dye and iron gall ink in situ on Kinwashi paper. In addition, TERS was used to identify iron gall ink on a historical document with handwritten text dated to the 19th century. We demonstrate that TERS can identify both of these colorants directly on paper. Moreover, vibrational modes from individual components of a complex chemical mixture, iron gall ink, can be identified. To the best of our knowledge, this is the first demonstration of in situ TERS for colorants of artistic relevance directly on historical materials. Overall, this work demonstrates the great potential of TERS as an additional spectroscopic tool for minimally invasive compositional characterization of artworks in situ and opens exciting new possibilities for cultural heritage research.

Tip-enhanced Raman spectroscopy (TERS) provides chemical information for adsorbates with nanoscale spatial resolution, single-molecule sensitivity, and, when combined with scanning tunneling microscopy (STM), Ångstrom-scale topographic resolution. Performing TERS under ultrahigh-vacuum conditions allows pristine and atomically smooth surfaces to be maintained, while liquid He cooling minimizes surface diffusion of adsorbates across the solid surface, allowing direct STM imaging. Low-temperature TER (LT-TER) spectra differ from room-temperature TER (RT-TER), RT surface-enhanced Raman (SER), and LT-SER spectra because the vibrational lines are narrowed and shifted, revealing additional chemical information about adsorbate-substrate interactions. As an example, we present LT-TER spectra for the rhodamine 6G (R6G)/Ag(111) system that exhibit such unique spectral shifts. The high spectral resolution of LT-TERS provides intramolecular insight in that the shifted modes are associated with the ethylamine moiety of R6G. LT-TERS is a promising approach for unraveling the intricacies of adsorbate-substrate interactions that are inaccessible by other means.

Surface-enhanced spatially offset Raman spectroscopy (SESORS) is a label-free vibrational spectroscopy that has the potential for in vivo imaging. Previous SESORS experiments have been limited to acquiring spectra using SERS substrates implanted under the skin or from nanoparticles embedded in tissue. Here we present SESORS measurements of SERS active nanoparticles coated with a Raman reporter molecule (nanotags) acquired, for the first time, through bone. We demonstrate the ability of SESORS to measure spectra through various thicknesses (3-8 mm) of bone. We also show that diluted nanotag samples (~2 × 10(12) particles) can be detected through the bone. We apply a least-squares support vector machine analysis to demonstrate quantitative detection. It is anticipated that these through-bone SESORS measurements will enable real-time, non-invasive spectroscopic measurement of neurochemicals through the skull, as well as other biomedical applications.

An explanation of the relative intensity fluctuations observed in single-molecule Raman experiments is described utilizing both single-molecule tip-enhanced Raman spectroscopy and time-dependent density functional theory calculations. No correlation is observed in mode to mode intensity fluctuations indicating that the changes in mode intensities are completely independent. Theoretical calculations provide convincing evidence that the fluctuations are not the result of diffusion, orientation, or local electromagnetic field gradients but rather are the result of subtle variations of the excited-state lifetime, energy, and geometry of the molecule. These variations in the excited-state properties will provide information on adsorbate-adsorbate and adsorbate-substrate interactions and may allow for inversion of experimental results to obtain these excited-state properties.

Determining the existence of any direct spectral relationship between the far-field scattering properties and the near-field Raman-enhancing properties of surface-enhanced Raman spectroscopy (SERS) substrates has been a challenging task with only a few significant results to date. Here, we prove that hot spot dominated systems show little dependence on the far-field scattering properties because of differences between near- and far-field localized surface plasmon resonance (LSPR) effects as well as excitation of new plasmon modes via a localized emitter. We directly probe the relationship between the near- and far-field light interactions using a correlated LSPR-transmission electron microscopy (TEM) surface-enhanced Raman excitation spectroscopy (SERES) technique. Fourteen individual SERS nanoantennas, Au nanoparticle aggregates ranging from dimers to undecamers, coated in a reporter molecule and encased in a protective silica shell, were excited using eight laser wavelengths. We observed no correlation between the spectral position of the LSPR maxima and the maximum enhancement factor (EF). The single nanoantenna data reveal EFs ranging from (2.5 ± 0.6) × 10(4) to (4.5 ± 0.6) × 10(8) with maximum enhancement for excitation wavelengths of 785 nm and lower energy. The magnitude of maximum EF was not correlated to the number of cores in the nanoantenna or the spectral position of the LSPR, suggesting a separation between near-field SERS enhancement and far-field Rayleigh scattering. Computational electrodynamics confirms the decoupling of maximum SERS enhancement from the peak of the scattering spectrum. It also points to the importance of a localized emitter for radiating Raman photons to the far-field which, in nonsymmetric systems, allows for the excitation of radiative plasmon modes that are difficult to excite with plane waves. Once these effects are considered, we are able to fully explain the hot spot dominated SERS response of the nanoantennas.

Single-particle tracking experiments were carried out with gold nanoparticle-labeled solid supported lipid bilayers (SLBs) containing increasing concentrations of ganglioside (GM(1)). The negatively charged nanoparticles electrostatically associate with a small percentage of positively charged lipids (ethyl phosphatidylcholine) in the bilayers. The samples containing no GM(1) show random diffusion in 92% of the particles examined with a diffusion constant of 4.3(±4.5) × 10(-9) cm(2)/s. In contrast, samples containing 14% GM(1) showed a mixture of particles displaying both random and confined diffusion, with the majority of particles, 62%, showing confined diffusion. Control experiments support the notion that the nanoparticles are not associating with the GM(1) moieties but instead most likely confined to regions in between the GM(1) clusters. Analysis of the root-mean-squared displacement plots for all of the data reveals decreasing trends in the confined diffusion constant and diameter of the confining region versus increasing GM(1) concentration. In addition, a linearly decreasing trend is observed for the percentage of randomly diffusing particles versus GM(1) concentration, which offers a simple, direct way to measure the percolation threshold for this system, which has not previously been measured. The percolation threshold is found to be 22% GM(1) and the confining diameter at the percolation threshold only ∼50 nm.

This work characterizes the adsorption, structure, and binding mechanism of oxygenated organic species from cyclohexane solution at the liquid/solid interface of optically flat alumina-supported palladium nanoparticle surfaces prepared by atomic layer deposition (ALD). The surface-specific nonlinear optical vibrational spectroscopy, sum-frequency generation (SFG), was used as a probe for adsorption and interfacial molecular structure. 1-Hexanoic acid is an overoxidation product and possible catalyst poison for the aerobic heterogeneous oxidation of 1-hexanol at the liquid/solid interface of Pd/Al(2)O(3) catalysts. Single component and competitive adsorption experiments show that 1-hexanoic acid adsorbs to both ALD-prepared alumina surfaces and alumina surfaces with palladium nanoparticles, that were also prepared by ALD, more strongly than does 1-hexanol. Furthermore, 1-hexanoic acid adsorbs with conformational order on ALD-prepared alumina surfaces, but on surfaces with palladium particles the adsorbates exhibit relative disorder at low surface coverage and become more ordered, on average, at higher surface coverage. Although significant differences in binding constant were not observed between surfaces with and without palladium nanoparticles, the palladium particles play an apparent role in controlling adsorbate structures. The disordered adsorption of 1-hexanoic acid most likely occurs on the alumina support, and probably results from modification of binding sites on the alumina, adjacent to the particles. In addition to providing insight on the possibility of catalyst poisoning by the overoxidation product and characterizing changes in its structure that result in only small adsorption energy changes, this work represents a step toward using surface science techniques that bridge the complexity gap between fundamental studies and realistic catalyst models.

Single-molecule surface-enhanced Raman spectroscopy (SMSERS) of crystal violet (CV) has been reported since 1997, yet others have offered alternative explanations that do not necessarily imply SMSERS. Recently, the isotopologue approach, a statistically significant method to establish SMSERS, has been implemented for members of the rhodamine dye family. We provide the first demonstration of SMSERS of a triphenylmethane dye using the isotopologue approach. Two isotopologues of CV are employed to create chemically identical yet vibrationally distinct probe molecules. Experimental spectra were compared extensively with computational simulations to assign changes in mode frequencies upon deuteration. More than 90 silver nanoparticle clusters dosed with a 50:50 mixture of CV isotopologues were spectroscopically characterized, and the vibrational signature of only deuterated or undeuterated CV was observed 79 times, demonstrating that the isotopologue approach for proving SMSERS is applicable to both the CV and the rhodamine systems. The use of CV, a minimally fluorescent dye, allowed direct evaluation of enhancement factors (EF), which are reported herein. Through experiment and theory, we show that molecular electronic resonance Raman (RR) and surface-enhanced Raman effects combine synergistically in SMSERS. Excluding RR effects, the EF(SERS) is ∼10(9). Variations and relationships between substrate morphology and optical properties are further characterized by correlated SMSERS-localized surface plasmon resonance (LSPR)-high-resolution transmission electron microscopy (HRTEM) studies. We did not observe SMSERS from individual nanoparticles; further, SMSERS-supporting dimers are heterodimers of two disparately sized particles, with no subnanometer gaps. We present the largest collection to date of HRTEM images of SMSERS-supporting nanoparticle assemblies.

We report the first inert gas sensing and characterization studies based on high-resolution localized surface plasmon resonance (HR-LSPR) spectroscopy. HR-LSPR was used to detect the extremely small changes (<3 × 10(-4)) in bulk refractive index when the gas was switched between He(g) and Ar(g) or He(g) and N2(g). We also demonstrate submonolayer sensitivity to adsorbed water from exposure of the sensor to air (40% humidity) versus dry N2(g). These measurements significantly expand the applications space and characterization tools for plasmonic nanosensors.

Understanding the detailed relationship between nanoparticle structure and activity remains a significant challenge for the field of surface-enhanced Raman spectroscopy. To this end, the structural and optical properties of individual plasmonic nanoantennas comprised of Au nanoparticle assemblies that are coated with organic reporter molecules and encapsulated by a SiO(2) shell have been determined using correlated transmission electron microscopy (TEM), dark-field Rayleigh scattering microscopy, surface-enhanced Raman scattering (SERS) microscopy, and finite element method (FEM) calculations. The distribution of SERS enhancement factors (EFs) for a structurally and optically diverse set of nanoantennas is remarkably narrow. For a collection of 30 individual nanoantennas ranging from dimers to heptamers, the EFs vary by less than 2 orders of magnitude. Furthermore, the EFs for the hot-spot-containing nanoparticles are uncorrelated to aggregation state and localized surface plasmon resonance (LSPR) wavelength but are crucially dependent on the size of the interparticle gap. This study demonstrates that the creation of hot spots, where two particles are in subnanometer proximity or have coalesced to form crevices, is paramount to achieving maximum SERS enhancements.

The surface-enhanced Raman excitation profiles (REPs) of rhodamine 6G (R6G) on Ag surfaces are studied using a tunable optical parametric oscillator excitation source and versatile detection scheme. These experiments afford the ability to finely tune the excitation wavelength near the molecular resonance of R6G (i.e., approximately 500-575 nm) and perform wavelength-scanned surface-enhanced Raman excitation measurements of a single molecule. The ensemble-averaged surface-enhanced REPs are measured for collections of molecules on Ag island films. The relative contributions of the 0-0 and 0-1 vibronic transitions to the surface-enhanced REPs vary with vibrational frequency. These results highlight the role of excitation energy in determining the resonance Raman intensities for R6G on surface-enhancing nanostructures. Single-molecule measurements are obtained from individual molecules of R6G on Ag colloidal aggregates, where single-molecule junctions are located using the isotope-edited approach. Overall, single-molecule surface-enhanced REPs are narrow in comparison to the ensemble-averaged excitation profiles due to a reduction in inhomogeneous broadening. This work describes the first Raman excitation spectroscopy studies of a single molecule, revealing new information previously obscured by the ensemble.

Molecular electronics is partly driven by the goal of producing active electronic elements that rival the performance of their solid-state counterparts, but on a much smaller size scale. We investigate what constitutes an ideal switch or molecular device, and how it can be designed, by analyzing transmission plots. The interference features in cross-conjugated molecules provide a large dynamic range in electron transmission probability, opening a new area for addressing electronic functionality in molecules. This large dynamic range is accessible through changes in electron density alone, enabling fast and stable switching. Using cross-conjugated molecules, we show how the width, depth, and energetic location of the interference features can be controlled. In an example of a single molecule transistor, we calculate a change in conductance of 8 orders of magnitude with an applied gate voltage. Using multiple interference features, we propose and calculate the current/voltage behavior of a molecular rectifier with a rectification ratio of >150,000. We calculate a purely electronic negative differential resistance behavior, suggesting that the large dynamic range in electron transmission probability caused by quantum interference could be exploited in future electronic devices.

We calculate that significant quantum interference effects can be observed in elastic electron transport through acyclic molecules. Interference features are evident in the transmission characteristics calculated for cross-conjugated molecules; significantly, these effects dominate the experimentally observable conduction range. The unusual transport characteristics of these molecules are highlighted through comparison with linearly conjugated and nonconjugated systems. The cross-conjugated molecules presented here show a large dynamic range in conductance. These findings represent a new motif for electron transfer through molecules that exhibit both very high and very low tunneling conductance states accessible at low bias without nuclear motion. In designing single molecule electronic components, a large dynamic range allows a high on/off ratio, a parameter of fundamental importance for switches, transistors, and sensors.

We present here a detailed study of the specific nanoparticle structures that give rise to single-molecule surface-enhanced Raman scattering (SMSERS). A variety of structures are observed, but the simplest are dimers of Ag nanocrystals. We chose one of these structures for detailed study using electrodynamics calculations and found that the electromagnetic SERS enhancement factors of 10(9) are easily obtained and are consistent with single-molecule SERS activity.

We present an interesting consequence of the differences between cross-conjugated and linearly conjugated molecules: the breakdown of conventional understanding of trends in molecular electron transfer. Interference effects are dominant in cross-conjugated molecules with unusual results: long molecules may have faster rates of electron transfer than short molecules, saturated molecules may have faster rates of electron transfer than conjugated molecules of the same length, and the rate of electron transfer cannot be correlated with energy gaps between the donor and acceptor states and the energy levels of the bridging molecule.

A plasmonic switch based on the calcium-induced conformational changes of calmodulin is shown to exhibit reversible wavelength modulations in response to changing calcium concentration. The extinction maximum (lambdamax) of a localized surface plasmon resonance (LSPR) sensor functionalized with a novel calmodulin construct, cutinase-calmodulin-cutinase (CutCaMCut), reversibly shifts by 2-3 nm. A high-resolution (HR) LSPR spectrometer with a wavelength resolution (3sigma) of 1.5 x 10-2 nm was developed to detect these wavelength modulations in real-time, providing information about the dynamics and structure of the protein. The rate of conversion from open (Ca2+-bound) to closed (Ca2+-free) calmodulin is shown to be 4-fold faster than the reverse process, with a closing rate of 0.127 s-1 and opening rate of 0.034 s-1. As far as we are aware, this plasmonic switch marks the first use of LSPR spectroscopy to detect reversible conformational changes in an unlabeled protein.

The existence of single-molecule surface-enhanced Raman spectroscopy (SMSERS) is proven by employing a frequency-domain approach. This is demonstrated using two isotopologues of Rhodamine 6G that offer unique vibrational signatures. When an average of one molecule was adsorbed per silver nanoparticle, only one isotopologue was typically observed under dry N2 environment. Additionally, the distribution of vibrational frequencies hidden under the ensemble average is revealed by examining the single-molecule spectra. Correlation with transmission electron microscopy reveals that SMSERS active aggregates are composed of multiple randomly sized and shaped nanoparticles. At higher coverage and in a humid environment, adsorbate interchange was detected. Using 2D cross correlation, vibrational modes from different isotopologues were anti-correlated, indicating that the dynamic behavior was from multiple molecules competing for a single hot spot. This allows hot-spot diffusion to be directly observed without analyzing the peak intensity fluctuations.

A new detection mechanism has been developed for low molecular weight substrate binding to heme proteins based on resonance localized surface plasmon spectroscopy. Cytochrome P450 has strong electronic transitions in the visible wavelength region. Upon binding of a substrate molecule (e.g., camphor), the absorption band of cytochrome P450 shifts to shorter wavelength. The event of camphor binding to a nanoparticle surface modified with cytochrome P450 protein receptors is monitored using UV-vis spectroscopy. It is observed for the first time that the binding of the substrate molecules to the protein receptor induces a blue-shift in the localized surface plasmon resonance (LSPR) of the nanosensors. The coupling between the molecular resonance of the substrate-free and substrate-bound cytochrome P450 proteins and the nanoparticles' LSPR leads to a highly wavelength-dependent LSPR response. When the LSPR of the nanoparticles is located at a wavelength distant from the cytochrome P450 resonance, an average of approximately 19 nm red-shift is observed upon cytochrome P450 binding to the nanoparticles and a approximately 6 nm blue-shift is observed upon camphor binding However, this response is significantly amplified approximately 3 to 5 times when the LSPR of the nanoparticles is located at a slightly longer wavelength than the cytochrome P450 resonance, that is, a 66.2 nm red-shift upon cytochrome P450 binding and a 34.7 nm blue-shift upon camphor binding. This is the first example of the detection of small molecules binding to a protein modified nanoparticle surface on the basis of LSPR.

The peak location of the localized surface plasmon resonance (LSPR) of noble metal nanoparticles is highly dependent upon the refractive index of the nanoparticles' surrounding environment. In this study, new phenomena are revealed by exploring the influence of interacting molecular resonances and nanoparticle resonances. The LSPR peak shift and line shape induced by a resonant molecule vary with wavelength. In most instances, the oscillatory dependence of the peak shift on wavelength tracks with the wavelength dependence of the real part of the refractive index, as determined by a Kramers-Kronig transformation of the molecular resonance absorption spectrum. A quantitative assessment of this shift based on discrete dipole approximation calculations shows that the Kramers-Kronig index must be scaled in order to match experiment.

A new method to stabilize and functionalize surfaces for surface-enhanced Raman spectroscopy (SERS) is demonstrated. Atomic layer deposition (ALD) is used to deposit a sub-1-nm alumina layer on silver film-over-nanosphere (AgFON) substrates. The resulting overlayer maintains and stabilizes the SERS activity of the underlying silver while presenting the surface chemistry of the alumina overlayer, a commonly used polar adsorbent in chromatographic separations. The relative affinity of analytes for alumina-modified AgFON substrates can be determined by their polarity. On the basis of SERS measurements, dipicolinic acid displays the strongest binding to the ALD alumina-modified AgFON among a set of pyridine derivatives with varying polarity. This strong affinity for carboxylate groups makes the SERS substrate an ideal candidate for bacillus spores detection using the dipicolinate biomarker. The SERS signal from extracted dipicolinate was measured over the spore concentration range 10(-14)-10(-12) M to determine the saturation binding capacity of the alumina-modified AgFON surface. The adsorption constant was determined to be Kspore = 9.0 x 10(13) M(-1). A 10-s data collection time is capable of achieving a limit of detection of approximately 1.4 x 10(3) spores. The shelf life of prefabricated substrates is at least 9 months prior to use. In comparison to the bare AgFON substrates, the ALD-modified AgFON substrates demonstrate twice the sensitivity with 6 times shorter data acquisition time and 7 times longer temporal stability. ALD expands the palette of available chemical methods to functionalize SERS substrates, which will enable improved and diverse chemical control over the nature of analyte-surface binding for biomedical, homeland security, and environmental applications.

A rapid detection protocol suitable for use by first-responders to detect anthrax spores using a low-cost, battery-powered, portable Raman spectrometer has been developed. Bacillus subtilis spores, harmless simulants for Bacillus anthracis, were studied using surface-enhanced Raman spectroscopy (SERS) on silver film over nanosphere (AgFON) substrates. Calcium dipicolinate (CaDPA), a biomarker for bacillus spores, was efficiently extracted by sonication in nitric acid and rapidly detected by SERS. AgFON surfaces optimized for 750 nm laser excitation have been fabricated and characterized by UV-vis diffuse reflectance spectroscopy and SERS. The SERS signal from extracted CaDPA was measured over the spore concentration range of 10(-14)-10(-12) M to determine the saturation binding capacity of the AgFON surface and to calculate the adsorption constant (Kspore=1.7 x 10(13) M(-1)). At present, an 11 min procedure is capable of achieving a limit of detection (LOD) of approximately 2.6 x 10(3) spores, below the anthrax infectious dose of 10(4) spores. The data presented herein also demonstrate that the shelf life of prefabricated AgFON substrates can be as long as 40 days prior to use. Finally, these sensing capabilities have been successfully transitioned from a laboratory spectrometer to a field-portable instrument. Using this technology, 10(4) bacillus spores were detected with a 5 s data acquisition period on a 1 month old AgFON substrate. The speed and sensitivity of this SERS sensor indicate that this technology can be used as a viable option for the field analysis of potentially harmful environmental samples.

A nanoscale optical biosensor based on localized surface plasmon resonance (LSPR) spectroscopy has been developed to monitor the interaction between the antigen, amyloid-beta derived diffusible ligands (ADDLs), and specific anti-ADDL antibodies. Using the sandwich assay format, this nanosensor provides quantitative binding information for both antigen and second antibody detection that permits the determination of ADDL concentration and offers the unique analysis of the aggregation mechanisms of this putative Alzheimer's disease pathogen at physiologically relevant monomer concentrations. Monitoring the LSPR-induced shifts from both ADDLs and a second polyclonal anti-ADDL antibody as a function of ADDL concentration reveals two ADDL epitopes that have binding constants to the specific anti-ADDL antibodies of 7.3 x 10(12) M(-1) and 9.5 x 10(8) M(-1). The analysis of human brain extract and cerebrospinal fluid samples from control and Alzheimer's disease patients reveals that the LSPR nanosensor provides new information relevant to the understanding and possible diagnosis of Alzheimer's disease.

A comparative analysis of the properties of two optical biosensor platforms: (1) the propagating surface plasmon resonance (SPR) sensor based on a planar, thin film gold surface and (2) the localized surface plasmon resonance (LSPR) sensor based on surface confined Ag nanoparticles fabricated by nanosphere lithography (NSL) are presented. The binding of Concanavalin A (ConA) to mannose-functionalized self-assembled monolayers (SAMs) was chosen to highlight the similarities and differences between the responses of the real-time angle shift SPR and wavelength shift LSPR biosensors. During the association phase in the real-time binding studies, both SPR and LSPR sensors exhibited qualitatively similar signal vs time curves. However, in the dissociation phase, the SPR sensor showed an approximately 5 times greater loss of signal than the LSPR sensor. A comprehensive set of nonspecific binding studies demonstrated that this signal difference was not the consequence of greater nonspecific binding to the LSPR sensor but rather a systematic function of the Ag nanoparticle's nanoscale structure. Ag nanoparticles with larger aspect ratios showed larger dissociation phase responses than those with smaller aspect ratios. A theoretical analysis based on finite element electrodynamics demonstrates that this results from the characteristic decay length of the electromagnetic fields surrounding Ag nanoparticles being of comparable dimensions to the ConA molecules. Finally, an elementary (2 x 1) multiplexed version of an LSPR carbohydrate sensing chip to probe the simultaneous binding of ConA to mannose and galactose-functionalized SAMs has been demonstrated.

This work presents the first step toward a glucose biosensor using surface-enhanced Raman spectroscopy (SERS). Historically, glucose has been extremely difficult to detect by SERS because it has a small normal Raman cross section and adsorbs weakly or not at all to bare silver surfaces. In this paper, we report the first systematic study of the direct detection of glucose using SERS. Glucose is partitioned into an alkanethiol monolayer adsorbed on a silver film over nanosphere (AgFON) surface and thereby, it is preconcentrated within the 0-4 nm thick zone of electromagnetic field enhancement. The experiments presented herein utilize leave-one-out partial least-squares (LOO-PLS) analysis to demonstrate quantitative glucose detection both over a large (0-250 mM) and clinically relevant (0-25 mM) concentration range. The root-mean-squared error of prediction (RMSEP) of 1.8 mM (33.1 mg/dL) in the clinical study is near that desired for medical applications (1 mM, 18 mg/dL). Future studies will advance toward true in vivo, real time, minimally invasive sensing.

Triangular silver nanoparticles ( approximately 100 nm wide and 50 nm high) have remarkable optical properties. In particular, the peak extinction wavelength, lambda(max) of their localized surface plasmon resonance (LSPR) spectrum is unexpectedly sensitive to nanoparticle size, shape, and local ( approximately 10-30 nm) external dielectric environment. This sensitivity of the LSPR lambda(max) to the nanoenvironment has allowed us to develop a new class of nanoscale affinity biosensors. The essential characteristics and operational principles of these LSPR nanobiosensors will be illustrated using the well-studied biotin-streptavidin system. Exposure of biotin-functionalized Ag nanotriangles to 100 nM streptavidin (SA) caused a 27.0 nm red-shift in the LSPR lambda(max). The LSPR lambda(max) shift, DeltaR/DeltaR(max), versus [SA] response curve was measured over the concentration range 10(-)(15) M < [SA] < 10(-)(6) M. Comparison of the data with the theoretical normalized response expected for 1:1 binding of a ligand to a multivalent receptor with different sites but invariant affinities yielded approximate values for the saturation response, DeltaR(max) = 26.5 nm, and the surface-confined thermodynamic binding constant K(a,surf) = 10(11) M(-)(1). At present, the limit of detection (LOD) for the LSPR nanobiosensor is found to be in the low-picomolar to high-femtomolar region. A strategy to amplify the response of the LSPR nanobiosensor using biotinylated Au colloids and thereby further improve the LOD is demonstrated. Several control experiments were performed to define the LSPR nanobiosensor's response to nonspecific binding as well as to demonstrate its response to the specific binding of another protein. These include the following: (1) electrostatic binding of SA to a nonbiotinylated surface, (2) nonspecific interactions of prebiotinylated SA to a biotinylated surface, (3) nonspecific interactions of bovine serum albumin to a biotinylated surface, and (4) specific binding of anti-biotin to a biotinylated surface. The LSPR nanobiosensor provides a pathway to ultrasensitive biodetection experiments with extremely simple, small, light, robust, low-cost instrumentation that will greatly facilitate field-portable environmental or point-of-service medical diagnostic applications.
