Microplastics (MPs) have become a ubiquitous emerging pollutant in the global marine environment. The potential toxic effects of MPs and interactions of MP pollution with other stressors such as food limitation on marine organisms’ health are not yet well understood. This study investigated the effects of three-week exposure to different MPs and food shortage on the physical defense mechanisms (byssus production and properties) of Mytilus coruscus. Starvation significantly reduced the number of byssus threads, and combined exposure to MPs and food shortage suppressed the adhesion ability and condition index of mussels. The length of the byssus threads was not affected by all experimental exposures. Transcript levels of genes encoding key proteins involved in byssus formation (the mussel foot proteins mfp-1, -2, -3, -4, -5 and -6, and prepolymerized collagen proteins preCOL-D, -P and -NG) were altered by interactions between the MPs and food shortage. These findings show that insufficient food supply can exacerbate the adverse effects of MPs on mussel defense which might have implications for survival and fitness of mussels under food limited conditions (e.g. in winter) in polluted coastal habitats.

Understanding the variability in organism's immunological response is crucial for predicting changes at population or community level. The present study investigated the immunoecology of a commercially valuable clam Paphia malabarica in a tropical monsoon-influenced estuary. Clams were collected monthly during a year cycle, which coincided with pre-monsoon (February-May), monsoon (June-September) and post-monsoon seasons (October-January). For assessment of immune functioning, selected hemocyte parameters (total hemocyte concentration, hemocyte mortality, lysosomal content, esterase activity, reactive oxygen species production, and phagocytic activity) were analyzed using flow cytometry. Simultaneously, clam's condition index, nutrients, chlorophyll a, dissolved oxygen, pH, temperature and bacterial density were also measured at the sampling site. Our results exhibited seasonal patterns in hemocyte functioning with the highest activity during the pre-monsoon season (suggestive of a suitable harvesting period) and lowest during monsoon (suggestive of a critical biological period). The critical biological period for P. malabarica was marked with compromised immune parameters inflicted by low salinity, food availability, and possibly high bacterial abundance. Also, the involvement of reproductive stress altering the hematological functioning in P. malabarica cannot be ruled out. Nutrients, dissolved oxygen, pH and temperature could not explain much of the hemocyte variability. The present study has further validated the usefulness of hemocyte as a suitable marker for understanding immunoecology of P. malabarica which is of prime importance, especially in a monsoon-influenced tropical estuarine environment. The findings of our research will be constructive in monitoring natural as well as cultivated bivalve populations of economic and ecological relevance.

The combined effect of ocean acidification and warming on the common cockle Cerastoderma edule was investigated in a fully crossed laboratory experiment. Survival of the examined adult organisms remained high and was not affected by elevated temperature (+3 °C) or lowered pH (−0.3 units). However, the morphometric condition index of the cockles incubated under high pCO2 conditions (i.e. combined warming and acidification) was significantly reduced after six weeks of incubation. Respiration rates increased significantly under low pH, with highest rates measured under combined warm and low pH conditions. Calcification decreased significantly under low pH while clearance rates increased significantly under warm conditions and were generally lower in low pH treatments. The observed physiological responses suggest that the reduced food intake under hypercapnia is insufficient to support the higher energy requirements to compensate for the higher costs for basal maintenance and growth in future high pCO2 waters.

International audience; Coastal waters corresponding to macrotidal systems are among the most variable marine biotopes. Sessile animals as bivalve mollusks may however be found forming intertidal beds at high densities, as allowed by full adaptation to local conditions. A better knowledge of adaptive responses to environmental factors is required to foresee possible adverse effects of global change. At the sub-cellular level, transcriptional responses are among the earliest signals of environmental disturbances and they can reveal subtle and meaningful changes in organism exposed to stress. Three blue mussel (Mytilus spp.) populations inhabiting the Bay of Brest (France) in sites exposed to different levels of chronic pollution, from low to moderate, were surveyed upon a seasonal schedule, with special attention to the reproductive cycle. Major seawater parameters were monitored over a full-year in the framework of the S! RANO project, based on an automatic high frequency acquisition system installed aboard a ship of opportunity. The health status of mussels has been assessed by measuring a condition index and gametogenesis has been followed by histology. Selected biological responses to environmental stress were detected using a multimarker approach including expression of genes involved in chemical stress response and energetic metabolism, and cellular immune parameters. Environmental parameters showed deep seasonal variations which differed among sites. Most biological responses followed a seasonal pattern. Late winter and spring corresponded to an active reproduction period in the Bay of Brest. Earlier spawning was observed in harbor areas compared to the oceanic site and an altered physiological state was assumed in commercial harbor mussels during the reproductive period, suggesting that their health is compromised at this time of year. However, no signs of severe chemical stress were detected in both harbor mussel populations, which could reflect adaptive responses to adverse environmental conditions.

Understanding strategies for energy allocation to maintenance, reproduction and growth is essential to evaluate the state of the fish stocks. In this study, we address the energetics dynamics of the annual cycle of anchovies (Engraulis encrasicolus) and sardines (Sardina pilchardus) in the north-western Mediterranean Sea using indirect and direct condition indices. We assessed and validated the use of morphometric, biochemical and energetic indices for both species. Annual patterns of the relative condition index (Kn), gonadosomatic index (GSI), lipid content (% lipids) and energy density (ED) were linked to the energy allocation strategy. Our results highlight that anchovy mainly rely on income energy to reproduce, while sardine accumulate the energy during the resting period to be used in the reproduction period. Consequently, variability in the lipid content and ED between seasons was lower in anchovy than in sardine. In both species, we observed an early decline in energy reserves in late summer-early fall, which may be related to unfavourable environmental conditions during spring and summer. Regarding the use of different condition indices, both direct indices, lipid content and ED, were highly correlated with Kn for sardine. ED was better correlated with Kn than lipid content for anchovy. For the first time, a relationship between ED of gonads and GSI for sardine and anchovy was provided, highlighting the importance of the energy invested in reproduction. This work provides new insights into the energy dynamics of sardine and anchovy. Bay scallops (Argopecten irradians) are an economically valuable species whose populations have declined in recent decades due in part to harmful algal and cyanobacterial blooms. Nantucket, Massachusetts hosts one of the last remaining bay scallop fisheries in the U.S., but recently documented the occurrence of a non-native cyanobacterium (Hydrocoleum sp.). Hydrocoleum can form dense mats in seagrass beds, the primary habitat of scallops, but is also diazotrophic, potentially augmenting bioavailable nitrogen to primary producers and fueling secondary production. We conducted surveys to explore the relationships between Hydrocoleum and scallop condition, reproductive potential, and density in eelgrass beds in Nantucket Harbor as well as effects of other habitat characteristics (e.g., eelgrass cover) on these same scallop traits. We found low Hydrocoleum cover during our sampling, but found fewer large scallops in plots with Hydrocoleum, suggesting that this size class may be especially vulnerable to negative effects of Hydrocoleum. Contrary to expectation, we found a positive correlation between Hydrocoleum cover and scallop condition. These patterns suggest that Hydrocoleum may enhance scallop condition, but also affect habitat use, highlighting the need for manipulative experiments to clarify mechanisms driving these relationships. Understanding how non-native species such as Hydrocoleum impact fishery species will help advance conservation and resource management efforts.

Globally, non-native species (NNS) have been introduced and now often entirely replace native species in captive aquaculture; in part, a result of a perceived greater resilience of NSS to climate change and disease. Here, the effects of ocean acidification and warming on metabolic rate, feeding rate, and somatic growth was assessed using two co-occurring species of oysters – the introduced Pacific oyster Magallana gigas (formerly Crassostrea gigas), and native flat oyster Ostrea edulis. Biological responses to increased temperature and pCO2 combinations were tested, the effects differing between species. Metabolic rates and energetic demands of both species were increased by warming but not by elevated pCO2. While acidification and warming did not affect the clearance rate of O. edulis, M. gigas displayed a 40% decrease at 750 ppm pCO2. Similarly, the condition index of O. edulis was unaffected, but that of M. gigas was negatively impacted by warming, likely due to increased energetic demands that were not compensated for by increased feeding. These findings suggest differing stress from anthropogenic CO2 emissions between species and contrary to expectations, this was higher in introduced M. gigas than in the native O. edulis. If these laboratory findings hold true for populations in the wild, then continued CO2 emissions can be expected to adversely affect the functioning and structure of M. gigas populations with significant ecological and economic repercussions, especially for aquaculture. Our findings strengthen arguments in favour of investment in O. edulis restoration in UK waters.

The use of imidacloprid (IMI) and its formulated products in agriculture is a risk to aquatic organisms due to deposition into waterways from runoff and aerial spraying. However, there is limited information on the potential effects of this pesticide on commercially important shellfish, such as oysters. We investigated the impacts of IMI and Spectrum 200SC (IMI formulation) on the activity of the enzymes Glutathione-S-transferase (GST), Catalase (CAT) and Acetylcholinesterase (AChE), in different oyster tissues including the gill, adductor muscle and digestive gland. We also investigated the condition index and fatty acid composition of the flesh of oysters after 2 weeks exposure. The concentrations of IMI in the different tissues was assessed using Liquid Chromatography-Mass Spectrometry (LC-MS) after QuEChERS extraction. Higher concentrations of IMI residues were detected in the adductor muscle of the oysters, followed by the gills and with the lowest amounts recovered from the digestive gland across all the concentrations tested. IMI and Spectrum 200SC significantly affected the gill AChE activity at 2 mg/L, but digestive gland CAT, and gill and digestive gland GST were impacted at environmentally relevant concentrations (0.01 and 0.05 mg/L). In the whole oyster, 2 weeks exposure to IMI (≥0.01 mg/L) resulted in a proportional increase in saturated fatty acids (SFA), altered the polyunsaturated fatty acid (PUFA) to SFA ratio and altered the omega 3 fatty acids (n-3) to omega 6 fatty acids (n-6) ratio, but there were no effects on the condition index of the oyster. Although the oysters responded differently to the formulated product, there was no consistent difference in the sublethal effects of analytical IMI and Spectrum 200SC. This study showed that exposure to IMI and Spectrum 200SC can significantly affect the biochemical processes and metabolites in oysters, with implications for food quality and safety.

A short fasting-refeeding experience was applied to specimens of red porgy, Pagrus pagrus (Teleostei, Sparidae) to assess its effects on some physiological parameters. Haematological (haematocrit), biochemical (serum cortisol and glucose) and immunological (lysozyme, haemolytic and haemagglutinating activities) parameters were measured. For this study, two fish groups were considered: one was fasted for 14 days and then refed to satiation during further 7 and 15 days (indicated as fasted/refed group), the other was fed throughout the study and was taken as a control group. Significantly lower values were recorded for the condition index, the hepato-somatic index and viscero-somatic index in the fasted/refed group compared to the fed one. Fasting did not affect significantly the examined parameters, except for cortisol; refeeding for 7 days induced a significant increase in the haemoagglutinating titre and the spontaneous haemolytic activity, but when refeeding was extended to 14 days haemagglutinating and haemolytic values remained lower than those measured in fed fish.

In a pilot field study the proteome response of Mytilus sp. was analyzed in relation to the concentration of different trace metal contaminants. Over a period of eight month test organisms have been exposed at a near-shore station in the anthropogenic impacted estuary of the river Elbe and at an off-shore station in the vicinity of the Island of Helgoland in the German Bight (North Sea). The stations differ in their hydrological as well as chemical characteristics. The physiological biomarkers, such as condition index which have been continuously monitored during the experiment clearly indicate the effects of the different environmental conditions. Multiple protein abundance changes were detected utilizing the techniques of two dimensional gel electrophoresis (2dGE) and consequently proteins arising as potential candidates for ecotoxicological monitoring have been identified by MALDI-ToF and ToF/ToF mass spectrometry. Different cytoskeletal proteins, enzymes of energy metabolism, stress proteins and one protein relevant for metal detoxification have been pointed out.

To assess the biological effects of the Prestige oil spill (November 2002) on coastal ecosystems, mussels (Mytilus galloprovincialis) were sampled in 22 locations along the coast of Galicia and the Bay of Biscay in April, July and September 2003 and in February, April, July and October 2004. Several cell and tissue-level biomarkers were measured in digestive gland. Flesh condition index and gamete development stages were assessed as supporting parameters. AOX activity, a marker of exposure to peroxisome proliferating compounds including PAHs, was particularly low in Galicia in 2003 and further was markedly increased in several localities in 2004. Values of the labilization period (LP) of the lysosomal membrane were low in all the studied localities, especially in Galicia in 2003. In 2004, LP values raised, evidencing a certain recovery in mussel's health. In agreement, the volume density of basophilic cells was markedly high in 2003 and showed a decreasing trend throughout 2004. Parameters defining the structure of digestive alveoli showed few variations between 2003 and 2004. Significant correlations between several biomarkers and total polycyclic aromatic hydrocarbons (PAHs) were found. In conclusion, employed biomarkers detected highest degree of disturbance in areas most impacted by the oil spill (Galicia) and were able to evidence a recovery trend during 2004, related to a decrease in total PAH concentrations in mussels.

A multiple biomarker approach was adopted in a seasonal study carried out in the Lagoon of Venice, with the double aim of evaluating the natural and anthropic stresses influencing the biological responses of mussel ( Mytilus galloprovincialis ) and of assessing the effects due to spatial rather than temporal variations. Biochemical (aldehyde dehydrogenase and catalase activities), cellular (neutral red retention time) and physiological (survival in air and condition index) biomarkers were determined in mussels collected in four differently impacted lagoon areas. Multivariate analysis showed that samples were distributed mainly according to temperature and four seasonal groups were identified. The combination of spatial and temporal information enabled us to distinguish physiological variations due to natural causes from those due to anthropic stress, and to identify the sampling period when several biomarkers are less influenced by both natural and endogenous factors.

Variation in glycogen concentration, condition index (CI) and filtration activity were measured in the bivalve Macoma balthica buried in sediment and experimentally exposed to cadmium (Cd). The stress due to elevated but sub-lethal concentrations (300 ppb Cd) affected the overall fitness of the organism as all parameters monitored responded significantly. Lower concentrations tested (10, 30 and 100 ppb) only induced a significant decrease in filtration activity, which may play a protective role, enabling the organism to slow down its metabolic activity and preserving the integrity of its reserves (reflected by stable CI and glycogen levels). Hence, the various endpoints selected show different thresholds. Our results also demonstrate that under high exposure, small individuals loose proportionally more glycogen per unit of weight than larger ones, thus confirming the higher sensitivity of small individuals to metal contamination. Furthermore, exposure to intermediate concentration (30 ppb) seems to be beneficial to the small individuals as indicated by their high CI values compared to the control. These results showed thus that non-sigmoidal concentration-response relationship and sizes of individuals should be considered in monitoring programmes and risk assessment.

Using a comprehensive approach, intertidal, near- and offshore sites in the German Bight were analysed for their environmental quality by assessing the health of blue mussels (Mytilus edulis). During a ten month sampling period mussels were studied with a set of biomarkers comprising lysosomal membrane stability and accumulation of lipofuscin, supplemented by biomarkers indicating nutritional status such as neutral lipids and glycogen in the cells of the digestive gland. Data were analysed in relation to sex, gonadal status, condition index and for the presence of parasites, to determine the overall health status of mussels at the respective sites. Mussels from all sites showed clear signs of stress, indicating an inferior environmental quality throughout the southern German Bight. Further, habitat characteristics such as inundation time and growing on- or off-bottom, as well as seasonal factors, can clearly influence the response of biomarkers in mussels exposed to similar levels of chemical environmental stress.

With the construction of wind farms all across the North Sea, numerous artificial reefs are created. These windmill artificial reefs (WARs) harbour high abundances of fish species which can be attracted from elsewhere or can be the result of extra production induced by these wind farms. To resolve the attractioneproduction debate in suddenly altered ecosystems (cf. wind farms), the possible consequences of attraction should be assessed; thereby bearing in mind that ecological traps may arise. In this paper we investigated whether the wind farms in the Belgian part of the North Sea act as ecological traps for pouting and Atlantic cod. Length-at-age, condition and diet composition of fish present at the windmill artificial reefs was compared to local and regional sandy areas. Fish data from the period 2009e2012 were evaluated. Mainly I- and II-group Atlantic cod were present around the WARs; while the 0- and I-group dominated for pouting. For Atlantic cod, no differences in length were observed between sites, indicating that fitness was comparable at the WARs and in sandy areas. No significant differences in condition index were observed for pouting. At the WARs, they were slightly larger and stomach fullness was enhanced compared to the surrounding sandy areas. Also diet differed considerably among the sites. The outcome of the proxies indicate that fitness of pouting was slightly enhanced compared to the surrounding sandy areas. No evidence was obtained supporting the hypothesis that the WARs act as an ecological trap for Atlantic cod and pouting.

The common guillemots, Uria aalge, found stranded at the Belgian coast, display high levels of Cu in both liver and kidneys. The condition index of the animals, defined as the ratio of liver to kidneys mass (Wenzel & Adelung, 1996, The suitability of oiled Guillemots (Uria aalge) as monitoring organisms for geographical comparisons of trace element contaminants. Archives of Environmental Contamination and Toxicology, 31, 368-377), influences both the metal concentration and its binding to metallothioneins (MT): the lower the condition index, the more emaciated the animals, and the higher the total Cu concentration and the concentration of Cu bound to MT. In less robust individuals, our results suggest that Cu could displace Zn from MT, rendering the Zn ions available to induce a new MT synthesis. Sex-related effects also emerged as significantly higher hepatic MT as well as Cu- and Zn-MT concentrations were found in emaciated male guillemots compared to females. In both organs, Cd concentrations remained low and typically demonstrated an age-dependent renal accumulation, with no noticeable effect of the condition index. As a whole, these results suggest that, for guillemots found stranded at the Belgian coast, Cu binding to hepatic and renal MT could function as a protective mechanism, rendering the metal ions unavailable to exert any cytotoxic activity.

Dogwhelks Nucella lapillus feed mainly on mussels and barnacles, and may experience periods of starvation. We report effects of nutritional state and prey type on the survival, growth, cadmium (Cd) accumulation, metallothionein (MT) induction and glycogen stores in N. lapillus exposed to Cd in water. Adult dogwhelks, with similar shell length (30.0+/-1.5 mm), were either starved or fed to satiation with barnacles Semibalanus balanoides, mussels Mytilus edulis or Cd-dosed M. edulis, and kept in filtered natural seawater (0.01 microg Cd 1(-1)) or Cd-contaminated (400 microg Cd 1(-1)) seawater for 80 days. Mortality and individual growth rate were determined. Cd, MT and glycogen were measured in different tissues. Prolonged starvation and exposure to Cd significantly reduced the survivorship of N. lapillus, but feeding could help dogwhelks to combat Cd toxicity and minimise mortality. Extended starvation also caused tissue wastage, leading to higher concentrations of Cd and MT in tissues, whereas fed animals increased in weight and had lower Cd and MT concentrations because of the tissue dilution effect. Prey type significantly affected growth rate of dogwhelks and indirectly influenced Cd accumulation, MT induction and glycogen stores. Eating mussels promoted better growth and higher glycogen reserves than eating barnacles. Individual growth rate decreased with increasing Cd accumulation. Cd-exposed survivors grew faster and consumed more than control animals, implying that these survivors may have better fitness and greater tolerance to Cd toxicity. The use of growth, condition index, MT and glycogen as biomarkers of environmental pollution are discussed. These results indicate a need to incorporate biological data including growth (or at least condition index) and prey type into biomonitoring programmes to allow sound interpretation.

Juvenile mussels (Mytilus edulis) were transplanted to Howe Sound, British Columbia, Canada, along an apparent pollution gradient of acid mine drainage (AMD) from an abandoned copper (Cu) mine. Cages containing 75 mussels each were placed at a total of 15 stations and were exposed to concentrations of dissolved Cu in surface waters ranging from 5 to 1009 micrograms/l for a period of 41 days. Mussels located at stations closer to the source of AMD at the mouth of Britannia Creek bioaccumulated higher concentrations of Cu and zinc (Zn) in their tissues. Mussel growth was adversely affected by Cu tissue concentrations above 20 micrograms/g dry wt., while declines in survival and condition index occurred in mussels that bioaccumulated greater than 40 micrograms/g dry wt. Cu. Tissue Zn concentrations (117-192 micrograms/g dry wt.) were likely not high enough to have a direct impact on mussel health. Reduced survival of transplanted mussels was supported by an absence of natural mussels in contaminated areas. Phytoplankton was also severely reduced in areas contaminated by mine waters. Based on the weight of evidence, AMD from the Britannia mine had a deleterious impact on mussel survival in a zone extending at least 2.1 km to the north and 1.7 km to the south of Britannia Creek on the east shore of Howe Sound.

Protothaca staminea and Macoma inquinata were exposed to sediment contaminated with 1237 ppm Prudhoe Bay crude oil in the field. Eighty-five per cent of the Protothaca and 17% of the Macoma survived 54 days' exposure. Body burdens of saturated and di- and tri-aromatic hydrocarbons were less than 2 ppm and quite variable. The condition index of Protothaca was reduced by 6% by exposure to oil. The level of free glycine in the mantle, gills and adductor muscle did not change significantly, but the taurine level fell, leading to a decrease in the taurine:glycine ratio. It was concluded that Protothaca , a filter feeder, is affected less severely by oil pollution than Macoma , a detritivore, perhaps because the feeding activity of filter feeders is less inhibited.

Whilst foraging in the water column for zooplankton, sand lance, Ammodytes hexapterus , are under heavy predation from marine birds and fish. To avoid predation, sand lance bury in the sand when not foraging and during overwintering. We did two experiments to determine whether oil contamination of the sand would reduce the amount of time that sand lance spent buried. In the first experiment (June, 1980) sand lance significantly decreased by 20% the time spent buried in oiled sand (306 ppm). In the second experiment (June, 1981) sand lance did not decrease time spent buried in oiled sand at 28 and 256 ppm but did at 3384 ppm. The higher condition index of the 1981 fish suggested that nutritional state may have influenced how sand lance used the sediment as a refuge and how they responded to contamination of that refuge.

This paper discusses the following topics related to polynuclear aromatic hydrocarbon pollution in estuaries: (1) the use of oysters (Crassostrea virginica), hard clams (Mercenaria mercenaria) and brackish water clams (Rangia cuneata) in residue monitoring; (2) the effects of elevated PAH residues on oyster condition; (3) three years of field studies relating PAH sediment contamination to abnormalities in fishes; and (4) laboratory bioassays for effects and bioconcentration modeling. Oysters, hard clams and Rangia have been shown, from 3 years of field studies, to be good monitors of pollution inputs as one proceeds along salinity gradients from 25‰ to 0·5‰. Effects of increased body burdens of PAHs are shown by a lowering of the oysters' condition index, as measured by lipid levels. Fishes inhabiting the Elizabeth River, VA, which is highly contaminated with PAHs, have abnormalities (cataracts, skin lesions, abnormal fins, etc.). These abnormalities have higher incidence in regions of the river where the sediments are more heavily contaminated. Laboratory studies utilizing contaminated sediments have reproduced some of the abnormalities observed in the field. Bioconcentration of PAHs from sediments has been studied with oysters and hard clams. Oysters generally accumulate three times the body burdens of clams exposed to the same suspensions. A faster depuration rate (k2) for clams appears to be responsible for the higher equilibrium body burden of oysters.

The bivalves Protothaca staminea and Mytilus edulis were exposed to five depths of decomposing bark from an active log transfer facility for 13–96 days in a field experiment at Auke Bay, Alaska. Survival and condition of both species was inversely related to depth and duration of bark coverage. As little as 6 cm of bark reduced survival; survival declined most rapidly between 10 and 15 cm of bark. The depths of bark under which half the animals died after 96 days of exposure were 12·8 ± 1·6 cm for P. staminea and 10·9 ± 1·5 cm for M. edulis . Interstitial water in the bark deposits had low concentrations of dissolved O 2 (2·5–5·3 mg/liter) and elevated concentrations of hydrogen sulfide (1·0–8·1 mg/liter total sulfide) and ammonia (46–131 μg-at N/liter) compared to the surface water from the control treatment. Survival of P. staminea and M. edulis was inversely related to concentrations of hydrogen sulfide and ammonia and directly related to dissolved O 2 concentrations.
