[Objectives] To study the effect of total flavonoids extracted from Polygonum perfoliatum L.（TFP） on immune-mediated liver injury induced by bacillus Calmette-Guerin plus lipopolysaccharide（BCG+LPS） in mice, and to explore its action mechanism. [Methods] 60 Kunming mice were divided into normal group, model group, control group（bifendate） and TFP low, medium and high dose groups according to random number table method, with 10 mice in each group. On the first day of modeling, mice were injected with 0.2 mL of BCG solution（12.5 mg/mL） through the tail vein, and on the eleventh day, 0.2 mL of LPS（37.5 μg/mL） were injected into the tail vein to prepare a mouse model of immune-mediated liver injury; from the first day of modeling, the normal group and the model group were administered intragastrically with the corresponding volume of distilled water, and the bifendate group and the TFP high, medium, and low dose groups were administered intragastrically with the corresponding doses once a day for 11 d. After the last time administration, fasting but giving water for 16 h, took blood from eyes, then collected the liver tissue. The levels of alanine transaminase（ALT） and aspartate transaminase（AST） in serum were detected by biochemical method; transforming growth factor-β1（TGF-β1）, intercellular adhesion molecule-1（ICAM-1）, interleukin-6（IL-6） and interleukin-1β（IL-1β） expression levels in liver tissue were detected by enzyme-linked immunosorbent assay（ELISA）; phosphorylated protein tyrosine kinase JAK-2（p-JAK2）, phosphorylated signal transducer and activator of transcription 3（p-STAT3） protein expression levels were detected by Western Blot method; the degree of liver tissue lesions was detected by HE staining. [Results] Compared with the model group, the levels of ALT and AST in the serum of mice in each dose group of TFP（high dose 600 mg/kg, medium dose 400 mg/kg, and low dose 200 mg/kg） were reduced, and the activities of T-SOD and GSH-Px were increased; the content or expression of β1, ICAM-1, IL-6, IL-1β decreased, and the expression of p-JAK2 and p-STAT3 protein decreased; pathological sections showed that the degree of inflammatory necrosis and the degree of lesions in the liver tissues of each dose group of TFP were reduced by varying degrees. [Conclusions] TFP has a protective effect on BCG+LPS-induced immune-mediated liver injury in mice. The mechanism may be related to regulating the phosphorylation level of JAK2 and inhibiting the inflammatory reaction, thereby regulating the TGF-β1/STAT3 signaling pathway and improving the immune-mediated liver injury. Objectives] To observe the effects of polysaccharides from Dicliptera chinensis（L.） Nees. on the expression of TLR/NF-κB pathway related proteins in HepG2 cells induced by oleic acid, and to explore the possible mechanism of polysaccharides from D. chinensis（L.） Nees. in the treatment of non-alcoholic fatty liver disease（NAFLD）. [Methods] HepG2 cells were induced with oleic acid to establish a non-alcoholic fatty liver cell model. After intervention with 0.25 and 0.5 mg/mL of D. chinensis（L.） Nees. polysaccharides, the ALT and AST activity and TG and TC contents were detected with kits, and the changes in the expression of CDK5, TLR4, p-NF-κB and NF-κB were analyzed using Western-blotting. [Results] In the HepG2 cells induced with oleic acid, the ALT and AST activity increased significantly, the TG and TC contents increased significantly, and the expression levels of CDK5, TLR4 and p-NF-κB proteins up-regulated significantly. In the HepG2 cells intervened with D. chinensis（L.） Nees. polysaccharides, the activity of ALT and AST, the contents of TG and TC, and the expression levels of CDK5, TLR4 and p-NF-κB proteins all reduced significantly. Objectives] To explore the protective effects and mechanism of Yinzhihuang combined with metformin on nonalcoholic fatty liver diseases（NAFLD） based on PPAR-α signaling pathway. [Methods] 54 male SD rats were randomly divided into 2 groups, namely normal group（12 rats） and high-fat feed group（42 rats）. After 8 weeks, 2 rats were randomly selected from each group. After pathological examination confirmed the success of the NAFLD model, the high-fat feed group was divided into model group, Yinzhihuang（9 000 mg/kg）, metformin group（200 mg/kg）, Yinzhihuang combined with metformin group（4 500 mg/kg+100 mg/kg）, 10 rats in each group. Except the normal group and the model group, the other groups were given treatment drugs at the same time, one time a day, on the fifth weekend of the experiment, at the last time of administration, fasting but giving water for 16 h, then collected blood and liver tissue. Biochemical method was used to detect AST and ALT in serum and SOD, MDA, GSH-px biochemical indicators in liver tissue; enzyme-linked immunosorbent assay（ELISA） was used to detect the levels of TNF-α, IL-6 and TGF-β in liver tissue; Western blot was used to detect the expression of peroxisome proliferators-activated receptor-α（PPAR-α） protein in rat liver tissue. [Results] Compared with the model group, the serum levels of ALT, AST and MDA in liver tissue of model group rats were significantly increased; the levels of SOD and GSH-px in liver tissue were significantly increased; TNF-α and IL-6 and TGF-β contents in liver tissue were significantly increased. [Conclusions] Both Yinzhihuang and metformin have a therapeutic effect on NAFLD rats, and the effect of combined application of the two is significantly better than the intervention effect of Yinzhihuang or metformin alone, the mechanism is possibly associated with the regulation of PPAR-α signaling pathway. [Objectives] This study aimed to explore the protective effect of Oxalis coriniculata L. on rats with acute liver injury induced by carbon tetrachloride (CC14) and related mechanism by regulating oxidative stress and the TLR-2 TLR-2/NF-kB signaling pathway. [Methods] A total of 48 female rats were randomly and evenly divided into normal group, model group, silymarin group (0.12 g/kg), and high (16 g/kg), middle (8 g/kg) and low-dose (4 g/kg) O. coriniculata L. groups. The rats in the groups were intragastrically administered with 5 mL/kg of corresponding drugs (equal-volume distilled water for normal group and control group), respectively. The administration was conducted twice a day, for 10 consecutive days. After 2 h of the last administration, the rats in all the groups except the normal group were in- traperitoneally injected with 12% carbon tetrachloride (CC14) olive oil solution (5 ml/kg), respectively to establish liver injury' rat models. After 16 h, the eyeball blood of the rats was collected, and their liver tissues were collected for preparation of HE sections. The biochemical indicators detected included aspartate aminotransferase (AST), alanine aminotransferase (ALT), total superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) content in the serum. The contents of tumor necrosis factor-a (TNF-a), interleukin-1 ß (IL-lß) and interleukin-6 (IL-6) in the serum were detected by ELISA. The expression of Toll-like receptor-2 (TLR-2) and nuclear factor-kB (NF-kB) in liver tissue was detected using Western blotting. The pathological changes of liver were observed under light microscope. [ Results] Compared with the normal group, the ALT, AST activity and MDA, IL-1 ß, IL-6, TNF-a levels in rat serum significantly increased (P <0.01), the GSH-Px, T-SOD activity in rat serum significantly decreased (P <0.01), and the expression of TLR-2 and NF-kB in liver tissue was up-regulated (P<0.01) in the model group. Compared with the model group, the ALT, AST activity and MDA, IL-1 ß, IL-6 and TNF-a levels in rat serum reduced (P<0. 05, PcO.Ol), the GSH-Px and T-SOD activity in rat serum increased (P<0.05, P <0.01), and the expression of TLR-2 and NF-kB in liver tissue was down-regulated (P<0.05, P <0.01) in the O. coriniculata L. administration groups. Pathological sections show that O. coriniculata L. had an improving effect on rats with acute liver injury' induced by CC14. [ Conclusions] O. coriniculata L. has a good protective effect on rats with acute liver injury induced by CC14. [Objectives] To study the protective effects and mechanism of POLYGONI PERFOLIATI HERBA on rats with nonalcoholic steatohepatitis (NASH). [Methods] Sixty rats were randomly divided into normal group, model group, metformin group (0.5 mg/kg), and high dose, medium dose, and low dose groups of total flavonoids of POLYGONI PERFOLIATI HERBA (600, 300, 150 mg kg). The standard feed was given to the normal group, and the model group and the total flavonoids groups were fed with high-fat diet for 13 weeks to establish the NASH rat model. At the 8th week, the metformin group and the POLYGONI PERFOLIATI HERBA total flavonoids groups were given with the corresponding drug treatment for 6 weeks, blood was taken from the eyeball to collect liver tissue. Biochemical method was used to determine ALT, AST, TC, TG, HDL-c, LDL-c, SOD, MDA, GSH-Px activity or content in serum and HOMA-IR, and ELISA was used to measure the TNF-oi, IL-1 $ and IL -6 content in liver tiisue; Western blotting was used to detect the expression levels of AMPK, p-AMPK and ACC in liver tiisue. [Results] Total flavonoids of POLYGONI PERFOLIATI HERBA could significantly decrease the activity or content of ALT, AST and MDA in serum of NASH rat (P < 0.05, P < 0.01), and enhance the activity of SOD and GSH-Px (P < 0.05, P < 0.01), reduce serumTC, TG, LDL-c levels and insulin resistance index, increase HDL-c levels (P < 0.05, P < 0.01); down-regulate liver IL-1β, IL-6, TNF-α, ACC levels and up-regulate p-AMPK expression (P < 0.05, P < 0.01). [Conclusions] The total flavonoids of POLYGONI PERFOLIATI HERBA have a good protective effect on NASH rats, and its mechanism may be related to the functons of regulating the lipid metabolism, aHeviating insulin resistance, inhibiting oxidative stress, inhibiting inflammatory reaction and regulating AMPK and ACC protein expression. [Objectives] To study the protective effects of self-made Compound Taoren Danshen Decoction (CTDD) on acute liver injury induced by D-galactosamine in rats, and to explore its mechanism. [Methods] An acute liver injury model was established by intraperitoneal injection of 500 mg/kg of D-galactosamine. The ALT, AST, MDA, SOD and GSH-Px in serum, as well as serumTNF-oo, IL-1$ and IL-6 levels were measured, and liver tissue lesions were observed under microscope. [Results] CTDD can significantly reduce ALT, AST and MDA levels, increase SOD, GSH-Px activity, and significantly reduce serum TNF-oo, IL-1$ and IL-6 levels, and improve liver tiisue lesions. [Conclusions] CTDD has a protective effect on D-galactosamine-induced acute liver injury in rats, and its mechanism may be related to inhibition of oxidative strees and inflammatory reaction. [Objectives] By comparing D. chinensis polysaccharide granule with D. chinensis polysaccharide of direct use equivalent, it is determined if the dosage form could affect hepatoprotective effects ad mechanisms of D. chinensis polysaccharide. [Methods] 50 SD rats are randomly divided into 5 groups: blank group, model group, biphenyl dimethylester positive group (50 mg/(kg µ d)), D. chinensis polysaccha ride group (200 mg (kg µ d)) ad D. chinensis polysaccharide granule group (700 mg (kg µ d), which is equivalent to 200 mg (kg µ d) of polysaccharide). Except blank group and model group, corresponding dosage of drug is irrigated to the stomach in other groups. After two weeks, except blank group, D-galactosamine (400 mg/kg)is injected into abdominal cavity of each group to establish liver injury model of rat. After anaesthesia, blood is taken from abdominal aorta, and liver is collected and fixed by 4% paraformaldehyde. The levels of glutamic-pyruvic transaminase (ALT), AST, superoxide dismutase (SOD), gutathione peroxidase (GSH-PX), malonaldehyde (MDA), tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β and interleukin-6(IL-6)in serum are detected, and microscopic observation of liver tissue pathological section is conducted. [Results] Compared with model group, the levels of ALT, AST, MDA, TNF-α, IL-β and IL-6 in rat's serum from D. chinensis polysaccharide group ad D. chinensis polysaccharide granule group significanty decline, while SOD activity significantly rises. Pathological changes of liver tissue are improved significantly, and their curative effects are equivalent. [Conclusions] D. chinensis polysaccharide granule has signifcant protection role on acute liver injury of rat caused by D-galactosamine; its utilization convenience is showed while not declining its hepatoprotective effect. [Objective] To select optimal formation process for Dicliptera chinensis polysaccharide ganules. [Methods] Forming rate, flow, and solubility of granules were used as indicators. Single factor experiment and orthogonal experiment were carried out to optimize types and volume of auxiliary materials (filling agent and binding agent) used in granule prescription. Quality of formed granules was inspected. [Results] The optimal formation process was 1:2. 5:1% for Dicliptera chinensis polysaccharide: mannitol: 3% HPMC aqueous solution. The grain size, flow, and solubility of prepared granules met requirements of Chinese Pharmacopoeia. [Objective] To study protective effect and acting mechanism of total flavonoids of POLYGONI PERFOLIATI HERBA on acute alcohol-induced liver injury in mice. [Methods] 60 mice were randomly divided into normal group, model group, Bifendate positive group (150 mg/kg), and high (600 mg/kg), medium (400 mg/kg) and low (200 mg/kg) dosage groups of PPTF, with 10 mice for each group. Except the normal group, fill the stomach with 12 mL/kg 56% liquor one time every morning for the rest groups and fill the corresponding dosage liquor in the afternoon, and fill the same volume of distilled water for normal group and model group. Continuously implement these operations for 14 days. After the end of the last time stomach filling, take fasting for 24 hours, anaesthetize them and take blood, and collect livers; measure the activity or content of alanine transaminase (ALT), aspartate transaminase (AST), malondialdehyde (MDA), and superoxide dismutase (SOD) in serum by biochemical method; measure the content of interleukin-4 (IL-4), interleukin-6 (IL-6), tumor necrosis factor -α (TNF-α) in liver tissue by enzyme-linked immunosorbent assay (ELISA) method; HE dyeing, and observe pathological changes of liver tissue by a microscope. [ Results] Compared with the model group, all dosage groups of total flavonoids of POLYGONI PERFOLIATI HERBA can effectively reduce activity and content of ALT, AST and MDA in serum of mice, can improve SOD activity, and can reduce IL-6 and TNF-α in liver tissue, and increase IL4 content. [Conclusions] Total flavonoids of POLYGONI PERFOLIATI HERBA have protective effect on acute alcohol-induced liver injury in mice and the mechanism is possibly related to anti-lipid peroxidation and inhibition of inflammation reaction in liver. [Objectives] Taking mice with acute liver injury induced by CCl4 a the model, the eefect of arabinose - mannose (w/w - 1: 1) on mice with acute liver injury induced by CCl4 was studied. [Methods] 60 experimental mice were selected and then randomly divided into norma control, model group and positive group (bifendate 120 mg/kg), high-dose arabinose - mannose group (800 mg/kg), middle-dose arabinose-t-mannose group (400 mg/kg) and low-dose arabinose - mannose group (200 mg/kg), each group had 10 mice, which were fed adaptively for 1 week. Except norma control group and model group, each treatment group was given medicine by gavage once a day and lasted for 7 days according to the dosage of 20 ml/kg. After the last drug, except normal control group, the mice of other groups were injected 10 ml/kg 0. 12% CCl4 peanut oil through enterocoelia, thereby establishing acute liver injury model. The mice were fated but not water for 24 h, after that, blood was sampled from mice eyes, then dissected rapidly. The activities of ALT and AST in the serum were determined, the expression levels of TNF-α, IL-1β and IL-6 from the hepatic tissues were detected by enzyme-linked immunosorbent assay(ELISA); then after HE dye, the changes of liver histopathology were observed. [ Results] Compared with CCl4 model, the activities of ALT and AST from the serum of mice from high-dose and middle dose groups decreased significantly(P < 0. 01); the contents of TNF-α, IL-1 β and IL-6 from the hepatic tissues of mice decreased significantly (P <0.01); the pathological section showed that the liver injury of mice from the combined drug groups showed alleviating trend to varying degrees, in which the liver injury of mice from the high-dose group was the best. [Objectives] To study the therapeutic effect and mechanism of total flavonoids extracted from Polygonum knot-weed L. on mice with liver injury induced by antituberculosis drugs. [Methods] 60 mice were randomly divided into six groups, namely normal control group, model group, glucurolactone group(200 mg/kg), high-dose (600 mg/kg), medium-dose (300 mg/kg), and low-dose (150 mg/kg) groups of total favonoids extracted from P. knotweed. Except for the mice in the normal control group, the mice in the other groups were given intragastric administration of isoniazid and rifampicin (100 mg/kg) to induce liver injury model, and were simultaneously treated with corresponding agents once a day for 30 d. On the 30th day, the blood and liver tissue were sampled, and ALT, AST, MDA, SOD and GSH-PX content were detected by biochemical methods; pathologic! changes of liver tissue were observed ater HE staining. [Results] Compared with the model group, ALT, AST and MDA content in the serum of the mice with liver injury increased, whiie SOD and GSH-PX content decreased in high-dose (600 mg/kg) and medium-dose (300 mg/kg) groups of total favonoids extracted from P. kjnotweed (P <0. 05 or P <0. 01, and the pathological chages of liver tissue of the mice were alleviated at the same time. However, the low-dose (150 mg/kg) group of total favonoids extracted from P. kjnotweed had no significat effects on liver injury caused by antituberculosis drug. [Conclusions] Total favonoids extracted from P. kjnotweed has an obvious therapeutic effect on liver injury induced by antituberculosis drugs, and the mechanism may be associated with its anti-oxidative stress and inhibition of lipid peroxidation. [Objective] Based on oxidative stress and inflammatory mechanism, the protective effects of Qiweijingganling (QWJGL) on rats liver injury induced by alcohol were researched, which contained seven Traditional Chinese Medicines. [Methods] A total of 72 rats were randomly divided into six groups according to the body weight, which were normal group, model group, bifendate group (50 mg/kg) and QWJGL dosage groups (400, 200, 100 mg/kg). Administration was carried out once each day. On the 30 day, rats were given 53% white wine (0.1 kg/d) by gavage, except the normal group. The alcoholic liver damage model was established. Effects on alanine aminotransferase (ALT), glutamic oxalacetic transaminase (AST), malondialdehyde (MDA), glutathione peroxidase (PSH-PX) and superoxide dismutase (SOD) activity were detected by colorimetry. Liver tissue tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) contents were detected by ELISA method. [Results] Compared with model group, QWJGL significantly reduced the ALT, AST and MDA actives in rats serum, enhanced the PSH-PX and SOD activities, and lowered the TNF-α and IL-6 contents in liver tissues (P <0. 05). [Conclusions] QWJGL reduced the rats liver injury induced by alcohol, the mechanism of which might be related to the reduction of oxidative stress and anti-inflammatory action. [Objective] To observe the clinical effects of Zhuang Ethnomedicine "antilipemic and liver protecting decoction" on 78 cases of nonalcoholic fatty liver diseases, and to provide new methods for the clinical treatment of nonalcoholic fatty liver diseases. [Methods] The 78 patients were randomly divided into two groups of treatment group ( antilipemic and liver protecting decoction) and control group ( fenofibrate tablets), with 39 cases in each group. After treatment for 12 weeks, changes of hepatic function, blood lipid and B-ultrasonography were observed. [Results] Alamine aminotransferase (ALT), aspartate transaminase (AST), glutamyltransferase (γ-GT), serum total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and triglyceride (TG) were all decreased in two groups after treatment. However, treatment group had significant statistical significance with control group (P <0. 05) . Results of B-ultrasonography detection after treatment showed that the fatty liver degrees improved in two groups; and treatment group was superior to control group (P < 0. 05). [Conclusions] Antilipemic and liver protecting decoction showed obvious curative effects on nonalcoholic fatty liver diseases, which should be applied and extended in clinical practice. [Objective] To research ihe effects of medicated serum of HEKBA POLYGONI PERFOLIATI on HBsAg and HBeAg secretion in 2215 cells. [Methods] Destruction rate of medicated serum of HEKBA POLYGONI PERFOLIATI to 2215 cells and HL-7702 were detected by MIT method; HBsAg and HBeAg secretion in cell supernatant was detected by ELISA method when four different concentrations of medicated serums were applied in 2215 cells for 72 h. [Results] 20% and 40% medicated serums showed inhibitory effects on HBsAg and HBeAg secretion, showing certain dose-effect relationship. At the same time, medicated serum of HERBA POLYGONI PERFOLIATI had less significant inhibitory effects on HL-7702 than that on 2215 cells. [Obbective] To reseach on the protective effects of total flavonoids in Polygonum perfoliatum L. on the acute liver injury of mice induced by CCl4 and its action mechanism. [Methods] Flavonoids were extracted from P. perfoliatum by chemical method. Acute liver injury model of mice was established by intraperitoneal injection of CCl4 peanut oil. The biochemical indexes of ALT, AST, TNF-α, IL-β and IL-6 in mice serum were detected. Histopathological examinaton of liver tissue was carried out. [Results] Flavonoids in P. perfoliatum restricted the ALT, AST, TNF-α, IL-β ad IL-6 activities in mice serum with acute liver injury (P < 0.01 , P < 0.05). Results of histopathological examination also showed that flavonoids significantly improved the liver injury of mice. [Objective] To study the microstructure characteristics of Peristrophe roxburghiana (Schult.) Bremek., so that reference can be provided for its identification, development and utilization and the establishment of its quality standards. [Methods] Microscopic identification method was used in the identification research of Peristrophe roxburghiana. [Results] The root cortex of Peristrophe roxburghiana contained a large secretory cavity; parenchymal cells of its stem cortex contained sclereids; nonglandular hair, glandular scales and cystoliths could be seen in its leaves and the leaves' powder; both upper and lower epidermis cells mianly looked irregular or were in polygon-like shapes. [Conclusions] Features above are significant in microscopic identification, and thus can be evidence for the identification of Peristrophe roxburghiana. [ABSTRACT FROM AUTHOR]

[Objective] To study the eiïect of alcohol extraction of Polygonum Perfoliatum on the expression of HBsAg and HBeAg in HepG2.2.15 cells. [Methods] The half toxic concentration (TC50) and the maximum non-toxic concentration (TC0) of PolygonumPerfoliatum extracts to HepG2. 2.15 ceHs cm be detected via methyl thiazolyl tetrazolium (MMT) method. With enzyme-linked immunosorbent assay (ELISA) method, the secretion level of HBsAg and HBeAg in ceH supernates could be measured after a 72-hour action of drugs with four different concentrations on the HepG2. 2.15 ceHs. [Results] Liquids with concentration of 0.4, 0.2 and 0.1 mg/mL have an inhibitory effect on the secretion of HBsAg and HBeAg after being cultured for 72 h in the ceH strain of HepG2.2.15 and there is andose-response relationship between the concentration and the effect. Therapeutic indexes of the liquids were all greater than 1 but lesser than 2, which showed that alcohol extract of Polygonum Perfoliatum was an effective therapeutic drug of low toxicity. [Objective] To optimize the purification technology of the total flavonoids from Polygonum knotweed L. by macroporous resins. [Methods] With rutin as the reference substance, the content of total flavonouls in P. knotweed was detected by aluminium nitrate coloration method. Taking static adsorption capacity and stalic adsorption rate as the investigation indices, the adsorption and desorption effects of four macroporous resins on the total flavonoids of P. knotweed were compared. The optimal macroporous resin was selected; and the adsorption and desorption conditions were researched. [Results] HPD-100 macroporous resin was most suitable for the purification of total flavonoids from P. knolweed. The purification technology was as follows: 2.5 g/L sample concentration, 2 BV/h sample flow rate, 2.5 BV sample volume, 70% ethanol as the eluant, 2 BV/h elution flow rate, and 5 BV eluant dosage. Under this technology, the concentration of total flavonoids in the purified products was 55.4%; and the extraction rate of total flavonoids reached 83.2% (n =3). [Conclusions] HFD-100 macroporous resin showed good purification effects on the total flavonoids of P. knotweed. The optimized technology was reasonable, feasible and stable. [ABSTRACT FROM AUTHOR]

[Objective] To reveal the anti-hepatic: fibrosis action of Perfoliate Knotweed Herb against N-Nitrosodimethylamine-indticed rat model and its mechanism. Method Eighty SD rats were randomly divided into 6 groups; normal group, model group. Perfoliate Knotweed Herb medication groups(high. medium and low doses) and colchicine group. Except normal group. rats in all other groups were medicated by introperitoneal injection with 0. 5% DMN solution (1.6 ml/kg) for continuous 3 d within a week. First week used 2/3 dose and then full dose was adopted, colchicine medication group adopted 0. 1 mg/kg colchicine; high, medium and low doses of Perfoliate Knotweed Herb medication groups applied 7.5. 5. 0 and 2.5 g/kg. Medication was performed once a day and continued for 4 weeks. At the end of 4 week. blixid collected from eyeball execution was used for detection of serum hyaluronic acid (HA). laminin(LN). procollagen III (PCIII). collagen IV (IV-C) by using ELISA. HF formation was observed by hematoxylin-eosin stain (HE stain) ; immunohistoChemical assay was employed to detect the transformation growth factorβ, (TGF-/β1). Results In comparison with model group, serum HA, LN, PCIII and IV-C expression levels in high and medium dose Perfoliate Knotweed Herb medication groups were significantly lowered (P<0.05 or P<0.01) ; immunohistochemical assay indicated that TGF-β1, level in high and medium dose groups were remarkably lowered (P <0. 05 or P < 0. 01). while that in low dose group did not (P>0.05). [Conclusion] Perfoliate Knotweed Herb is endowed with good efficacy on the treatment of N-Nitrosodimethyl-amine-induced hepatic fibrosis in rats, with the mechanism of down-regulating the expression of TGF-β1). thus inhibition the initiation of hepatic fibrosis.
