This study aimed to analyze the morphology of the tongue, which varies among fish species and has not been studied in turbot (Scophthalmus maximus), using macro-anatomical, light and scanning electron microscopy (SEM). As research materials, the tongues of eight adult turbot, which were used for consumption, were examined. The roughly triangular-shaped tongue of the turbot consisted of three parts: apex, body, and root. The thickness of the tongue continued to increase from the apex to the root. Although papillae were not observed in the macroscopic examination of the tongue, microscopic examination revealed the presence of cone-like shaped papillae on the submucosa. The tongue was composed of mucosa, submucosa, and hyaline cartilage. By SEM the presence of the taste buds (Types I and III), taste pores and papillae on the dorsal surface were demonstrated. Taste buds are the first descriptions in S. maximus. Therefore our results could add new data to the fish tongue anatomy. HIGHLIGHTS: Morphological and morphometric data of the turbot (Scophthalmus maximus) tongue were obtained in this study and compared with other species. In addition, the dorsal surface of the turbot tongue was described for the first time using SEM.

The current work is designed to give the first trial to characterize the ultrastructural lineaments of the oral cavity floor in juvenile and adult white grouper. The present work depends on 10 oral cavities floor from each age. The common features joined the oral cavities floor of the juvenile and adult fish summarized in; oral cavity floor had two teeth bands (lower incisive and lower molar), tongue with its two lateral spinated lines, lower velum, ridges, the lower lip divided into a single anterior part and two lateral part, and the absence of lower canine teeth. The oral surface of semilunar lower velum had round fungiform papillae that carried taste buds type I. The non-protrusible elongated tongue had a clear apex, body, and root with the absence of any taste buds. The dorsal lingual surface of the body had two lateral spinated lines, a single ridge and microtubercles. The smooth dorsal lingual surface of the root did not carry any ridges. The notched lower velum at the middle of the free border was observed in juveniles and adults. Meanwhile, there are some variations between juvenile and adult as; absence of lower incisive ridge only in juvenile, pointed tongue with sublingual ridge observed in juvenile while round without sublingual ridge in adult fish, moreover the presence of velvar ridges observed only in adult fish. The obtained findings provided essential data to aquaculture of this fish species in Egypt by determining the food particle types that are favorite to this fish. Recent literature has demonstrated only adult avian palate, albeit there has been only limited focus on the postnatal development of the avian oropharyngeal cavity roof. Hence, the current investigation was designed to obtain the full ultrastructure postnatal description of the oropharyngeal roof during the five developmental age-stages of Coturnix coturnix by employing assessments using gross morphometric analysis and stero and scanning electron microscopy. The elongated triangular oropharyngeal roof has a spoonful rounded beak tip. The palate region is subdivided into the rostral ridged area and the choanal area. The palate has eight longitudinal palatine ridges (seven nonpapillated and one papillated median) and four transverse papillary rows (one slightly oblique row and three transverse papillary crests). The median palatine ridge continuous caudally and is then divided into three ridges: one median and two paramedian ridges (forming the lateral boundaries of the choanal field). The choanal field had three regions (rostral, middle, and caudal). The finger-like projection papillary region has five papillae. The choanal cleft has two unequal parts (rostral and caudal). The rostral nonpapillated short choanal part is subdivided by transverse papillary row into rostral narrow straight and caudal diamond portions. The caudal wide papillated choanal part is further divided by a second transverse crest into rostral long (encircled by interdigitated papillae) and caudal short wider part (not encircled by interdigitated papillae). The infundibular cleft is not bordered by any papillae, while the pharyngeal region has numerous papillae and openings of the salivary glands. Moreover, the morphometric analysis revealed a higher value with increasing age for all dimensions. Our findings indicated a higher degree of functional adaptation between the five developmental age stages of quail. Our observations suggest that adaptations such as these may increase the efficiency of food prehension with increasing age.

The current investigation gave a full ultrastructural and histological description of the oropharyngeal cavity of the rock-pigeon Columba livia dakhlae. Our study carried on 10 heads of adult rock pigeons by gross, scanning, and light microscopic examination, in addition to the immunohistochemical analysis of the cytokeratin of the dorsal surface mucosa epithelium and the proliferating cell nuclear antigen expression (PCNA) immunoreactivity. The pointed apex of the elongated tongue covered rostrally and laterally with numerous caudally directed scales-like papillae. The dorsal surface of the apex and body was covered with numerous caudally directed scales-like filiform papillae. The U-shaped transverse papillary crest carried caudally directed pointed triangular conical papillae with two giant papillae. The elevated caudal area of the lingual body contains little numbers of the opening of the lingual gland. The laryngeal mound is divided into two symmetrical halves by rostral fissure and caudal laryngeal fissure into the right and left half. There were four palatine ridges: two lateral, middle, and median ridges. The choanal field bounded by the two lateral palatine ridges and containing the median choanae. The rostral part of the palatine cleft bounded laterally by a line of the small caudally directed conical papillae, while the caudal part was free from these papillae. There was a little number of taste buds in the palate. The high magnification of the rostral part of the beak is covered with directed rods-like projections. In conclusion, the obtained results described the adaptations of the tongue and its belonged structures with the feeding demand of the rock-pigeon.

The current study was designed to describe the prenatal developmental stages of the rabbit tongue. Thirty-five embryos of prenatal ages (E16, E19, E23, E25, E28, and Postnatal) were obtained from New Zealand white (NZW) rabbit. The primordia of the tongue were observed on the foundation as early as day 16 prenatal age (E16). The first primordia of lingual papillae were observed that including the primordia of circumvallate papillae at day 19 prenatal (E19). Thickenings of lingual epithelium developed at 19 day prenatal of embryo. At day 23-25 pc, the presence of primordia of foliate papillae was shown in addition to primordia of the fungiform and filiform papillae were found. The development of the final shape of the tongue is a longstanding process that occurs during the prenatal and postnatal life. The first primordia of gustatory papillae appear, and only later primordia of mechanical papillae are formed. In conclusion, the morphogenesis of the tongue and its lingual papillae discussed its herbivorous nature of feeding.

The morphology of different lingual papillae of the Egyptian goats was studied by gross observation, scanning electron microscopy, and light microscopy. According to function, two types of papillae were present; mechanical (filiform, conical and lenticular papillae) and gustatory papillae (fungiform and vallate papillae). Two types of filiform papillae were detected with different shape and position. Moreover, two types of conical and lenticular papillae could be recognized on the lingual torus. Abundant fungiform papillae were found on the dorsal and ventral surfaces of the tip of the tongue. The vallate papillae had also different shapes. Taste buds were demonstrated within the epithelium of the dorsal surfaces of the fungiform papillae and the lateral surfaces of the vallate papillae. In addition, a longitudinal row of large papillae was located on the lateral aspect of the tongue in relation to the filiform papillae. These papillae were suggested to be called elongated conical papillae and may be considered as ones of the lingual papillae. In conclusion, the current study provides a detailed description of the different lingual papillae of the Egyptian goat's tongue. Presence of different types and shapes of mechanical and gustatory lingual papillae in goats' tongue suggests a high degree of functional adaptation to their diet.

The current study was done to provide comprehensive information on the anatomical features of the lips and cheeks of the goat by gross examination and morphometric analysis in addition to scanning electron microscope (SEM). Samples from 12 normal healthy adult goat's heads of both sexes were collected directly after slaughtering. The lips and cheeks were dissected, and specimens were collected for both light and SEM. The lips of goat were soft and mobile. The free border of both lips was characterized rostrally by the presence of labial projections. The number, size, and arrangement of labial projections differed in the upper and lower lips. On the other hand, the buccal papillae were arranged into 6-8 longitudinal rows parallel to the cheek teeth. The length of these papillae decreased caudally while they were absent on the most caudal part of the cheek. Presence of several types and shapes of labial projections and papillae, and buccal papillae suggest a high degree of mechanical adaptation of the lips and cheeks of the goat. This study provides baseline data for clinical studies. This study is the first report to shed light on the morphology of the lips and cheeks of the goat by gross and scanning electron microscopy.

This study was conducted with the tongue samples of different life stages of hilsa, that is, adult Marine hilsa, adult Riverine hilsa, and Riverine juvenile hilsa, respectively. Three types of taste buds (Types I, II, and III based on their elevation from the epithelium at different levels) of the tongue, which may be to ensure full utilization of the gustatory ability of the fish were rocorded. Presence of specific taste buds indicate that the fish hilsa dwells in turbid waters with a possible preference toward diatom like planktonic food source. Enhanced expression of taste receptors (T1R1 and T1R3) and associated stimulatory G-proteins subunits on tongue also indicate occurrence of amino acid like substances that guided sensory cues for feeding by this fish. A firm regularity or stringency of the free surface of the epithelial cells may be attributed to compactly arranged microridges. These structures protect against physical abrasions potentially caused during food manoeuvring and swallowing. In our present observations, the surface architectures of the tongue of hilsa are discussed within the background of migratory adaptation of the species in the context of feeding and habitat preferences.

The current work gives concern to study the morphology of the Merluccius merluccius gills by using gross morphology, scanning electron microscopy (SEM), and light microscopy. The findings of the present study revealed that the gill system consisted of four pairs of gill arches which carry the gill filaments on the convex border and gill rakers on the concave border of them. SEM results revealed that the rakers and the spines distribution on the first gill arch differed from that of the other three gill arches on the lateral and medial surfaces. On the surface the gill filaments, there were longitudinal ridges that carried pores of chloride cells and mucous cells. The histological examination revealed that, the gill arch composed of hyaline cartilage that presented in the form of cups. Each cup consisted of central cartilagenous core and peripheral cartilagenous matrix. The gill filaments composed of cartilaginous bar of peripheral cartilaginous matrix and central cartilaginous core extended from the gill arches and covered by an epithelial layers with a few mucous cells permeate it, and chloride cells were straggly in the interlamellar epithelium. Each gill filament carried several leaves like secondary lamellae on both sides of it. The epithelium, which lined the secondary lamellae, composed of epithelial pavement cells, some mucous cells, and pillar cells.

While epiglottis is essentially a mammalian structure, studying its microstructure in any placental model will add an important information to the field of comparative anatomy and the related branches of biology. The aim of this study was to describe the structure of the epiglottis in dromedary camels using light and scanning electron microscopy (SEM), with reference to the possible functions. A total of 11 epiglottis cartilages from 11 larynges were used. The study revealed unusual, deeply situated glands just beneath the cartilage plate. They have unusually, wide surface-openings, while their ducts were partly located within the cartilage. This is presumed to be an adaptation to the need for rapid and efficient mucosal surface hydration in the arid conditions. The possible secretion transport mechanisms in these glands were also discussed. Furthermore, the SEM revealed for the first time, the presence of taste buds in camel epiglottis. However, in histological sections, visibility of taste buds was dependent upon the staining techniques. The taste buds were not seen with standard H& E stain, as they blended imperceptibly with the surrounding epithelium. Conversely, Mallory's trichrome showed contrasting colors, and taste buds were visible. In conclusion, camel epiglottis has an unusual structure, which may be correlated to environmental adaptation and important for the general health of upper respiratory tract in this species.

Bagrus Bayad (Forskal, 1775) is a bottom feeder fish feeds on different food items. Its oral cavity anatomy has many types of teeth adapted to the capacity and varieties of feeding. The aim of the present study was to use the gross anatomy and scanning electron microscopy to identify the anatomical characteristic, especially of the teeth and taste buds distributed on the roof and floor of the oropharyngeal cavity. The results clarified that Bagrus Bayad has two types of teeth; curved pointed and blunt teeth of different sizes. The teeth appeared on epithelial protrusions. The pharyngeal region had upper and lower pharyngeal teeth, the upper arranged into two oval prominences while the lower ones arranged into two triangular areas. The higher number of the pointed teeth is prove the carnivorous nature of the Bagrus Bayad. Three types of taste bud (I, II, & III) were found in the oropharyngeal cavity. The tongue present in the floor and free from teeth and characterized by the presence of taste bud type I and II. Many microridges of different arrangement observed on the surface epithelium of oropharyngeal cavity. The upper and lower valve characterized by the microridges, pores and taste buds. The knowledge of the nature of the oropharyngeal cavity of the Bagrus Bayad helps in identifying new and better methods for feeding in aquaculture.

The current work considers the first anatomical description of oropharyngeal cavity of Garganey, which was performed on eight heads with the help of scanning electron microscopy (SEM). The round apex of elongated tongue has anterior spatula-like named lingual nail. SEM of dorsal surface of lingual nail carry microtubercles and micropores on its rostral part, while its caudal part carry numerous microridges and micropores, while lateral apical surface only without lingual nail carrying filiform papillae, but its ventral surface carry exfoliated scales-like projections. Lateral lingual tip carries numerous laterally directed hairs-like structures. Rostral part of body carry region of small conical papillae in between them small hairs-like papillae on its lateral surface. Middle part of body carry lingual comb on its dorsal surface, while its lateral surfaces carry region of small conical papillae in between them hairs-like papillae in addition to filiform papillae, and large conical with small filiform papillae begin to appear laterally to the heads of lingual comb. Lateral surface of lingual prominence carry region of large conical and small filiform papillae on its rostral part, while its caudal part occupied by laterally situated spinated border from the root. Lingual root has two triangular smooth middle and spinated lateral and caudal. Laryngeal entrance divided into papillary and nonpapillary regions. Roof of oropharyngeal cavity divided into lamellar and papillary regions. Choanal cleft divided into rostral narrow ¼ and wide caudal ¾ parts. In conclusion, feeding process depend on the filter feeding mechanism that performed by the help of lateral situated papillae with lingual prominence.

PubMedID: 30575207 In this study, it was aimed to perform light microscopy and scanning electron microscopy (SEM) investigation of the tongue and papilla belonging to two wolves. Light microscopy and SEM images of tongues were taken. It was observed that papillae filiformes concentrated in apex of these investigated tongues. In addition to, these papillae were observed in the whole tongues. It was also determined that papillae fungiformes were distributed rarely in between papillae filiformes. Papillae foliatae were placed in the lateral side of tongues. There were two papillae vallatae in the median part of tongues. Papillae vallatae was determined in radix linguae. It was observed that papillae vallatae formed circular extensions in many different dimensions and a hole was present in a circular structure in the center. Papillae conicae were seen on dorsal surface of radix linguae. Papillae foliatae were seen four laminal structure and placed on the each lateral side of tongues. To the best of our knowledge, this is the first study that presents of light microscopy and SEM findings related with the tongues of wolves. © 2018 Wiley Periodicals, Inc.

Surface ultrastructure of the gills of the angler catfish Chaca chaca was investigated to unravel the adaptive modifications associated with the feeding ecology of the fish. The fish is often found in mud or in soft substrates where they remain buried both for protection and to feed. Gill rakers present on the gill arch in most fish species are absent in this fish. The absence of gill rakers are associated with the feeding habit of the fish and is considered to facilitate the swallowing of captured prey smoothly without any hindrance. Highly corrugated surface of the gill arch and gill filaments could be associated to retain water/mucus to prevent dessicassion of the fish. Papillae like epithelial protuberances each bearing a taste bud at its summit toward the pharyngeal side of the gill arch is associated with the sorting of the food. Large number of mucous goblet cells on the gill arch epithelium are considered to secret copious mucus to lubricate the prey for easy swallowing. In C. chaca the gill septa between gill filaments are reduced. This could enhance the flexibility and permit the free movement of the gill filaments. Extensive secondary lamellae and infrequent mucous goblet cells on secondary lamellae are associated to increase the surface area to enhance efficiency of gaseous exchange.

The aim of this study was to compare the anatomical and functional characteristics of the lingual papilla among the Camelidae. For this purpose, tongues of alpaca, guanaco, and llama were used. Numerous long and thin filiform papillae were located in the median groove and none were detected on the rest of the dorsal surface of the lingual apex in alpaca. Secondary papillae originated from the base of some filiform papillae on the ventral surface of alpaca tongue. The bases of some filiform papillae of the lateral surface of the lingual apex were inserted into conspicuous grooves in guanaco and tips of filiform papillae on the dorsal surface of the lingual body were ended by bifurcated apex. On the dorsal surface of the lingual apex of llama, there were no filiform papillae but there were numerous filiform papillae on both the lateral margins of the ventral surface of the lingual apex. Fungiform papillae were distributed randomly on dorsal lingual surface and ventral margins of the tongues of all camelid species. Lenticular papillae were located on the lingual torus and varied in size and topographical distribution for each species. Circumvallate papillae had irregular surfaces in llama and alpaca, and smooth surface in guanaco. In conclusion, llama and alpaca tongues were more similar to each other, and tongues of all camelid species displayed more similarities to those of Bactrian and dromedary camels in comparison with other herbivores and ruminants.

On the basis of our previous report that protein gene product 9.5 (PGP 9.5)-immunoreactive nerve fibers and taste cells and calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers are found in guinea pig vallate papillae [Huang and Lu (1996b) Arch. Histol. Cytol. 59:433–441]. We speculated that PGP 9.5 might be a marker for taste receptor cells and that CGRP might play an important role in taste transmission. We, therefore, performed an immunohistochemical and ultrastructural analysis of taste cells and related nerves in guinea pig vallate papillae. In the connective tissue of the vallate papilla, the ultrastructural data revealed that the PGP 9.5-immunoreactive nerve fibers were both myelinated and unmyelinated. The CGRP-immunoreactive nerve fibers were unmyelinated and surrounded by the cytoplasm of Schwann cells as were the non-immunoreactive fibers. In the vallate taste buds, only type III cells, which make synaptic contacts with intragemmal nerves, were PGP 9.5-immunoreactive, while the nerve terminals making synaptic contact with the underlying type III cells were CGRP-immunoreactive. From these observations, we conclude that: (1) PGP 9.5 might be a useful specific marker for type III cells in guinea pig vallate taste buds; and (2) CGRP-containing nerve fibers might be primarily involved in the neural transmission of taste stimuli. This study represents the first definitive anatomical description of the tongue and lingual papillae of the pampas deer and compares the different information on the morphology of the other ruminant species available in the literature. In this study, the tongues of four adult and one fetal deer were used. The tongue was elongated with an oval or rounded apex. The filiform papillae on the lingual apex were smaller than the ones on the dorsal and lateral surfaces of the lingual body. Two very thin secondary papillary projections were found to emerge from the bilateral sides of some filiform papillae. Spherical fungiform papillae were randomly distributed among filiform papillae on dorsal surface of the lingual body and ventral surface of the apex. More developed conical papillae were observed in the caudal half of the lingual torus, whereas the rostral half of the torus had smaller conical papillae. Each conical papilla included shallow longitudinal groove on its anterior surface and some conical papillae on the lingual torus had bifid sharp apices. On the caudal portion of the torus, we also observed a few fungiform papillae, which were larger than those located on the lingual body. There were no lenticular papillae on the lingual torus, and five to nine round or oval circumvallate papillae were situated on each caudolateral side of the lingual torus. Morphological features of the tongue in the pampas deer were more similar to wild ruminant species than they were to domestic species.

The lingual taste buds of mammals are complex organs containing dozens of cells of varying morphology and numerous nerve fibers that are intermingled among the cellular processes. Some of the taste bud cells form synaptic contacts with these nerve fibers. Important questions remain to be answered regarding the structure and function of the cells of various types within taste buds and the means by which responses to gustatory stimuli are transmitted to the nerve fibers that communicate with the brain. Using both conventional and high voltage electron microscopy, we have examined serially sectioned taste buds from the tongues of mice and rabbits in order to address these issues and to obtain more complete information than that available from sampling of sections. The technique of computer-assisted 3-D reconstruction was used to generate models of whole taste buds and individual cellular and neural elements within taste buds from the serial sections. Analysis of serially sectioned taste buds from mice and rabbits has revealed that in both of these species relatively few (30% or less) of the cells within the taste buds form synaptic contacts with nerve fibers. In the foliate taste buds of rabbits, all of the cells that are presynaptic to nerve fibers are of a single morphological type (type III). The cells that are presynaptic to nerve fibers within the taste buds of mice are morphologically diverse. A pattern of synaptic connectivity exists within murine taste buds such that a given nerve fiber receives synaptic input only from taste cells that are ultrastructurally similar. In the taste buds of both mice and rabbits, we have observed both divergence and convergence of synaptic input from the putative taste receptor cells onto nerve fibers, suggesting that at the level of the taste bud there is some integration of the information generated by individual receptor cells. In addition to typical chemical synapses, other cytoplasmic specializations (such as subsurface cisternae and atypical mitochondria) may be involved in interactions between taste bud cells and nerve fibers. © 1994 Wiley-Liss, Inc.

Unilateral interruption of the chorda-lingual nerve led to a loss of most epithelial axons and to the deterioration of fungiform taste buds in the anterior portion of the tongue of albino rats, mongolian gerbils, and golden hamsters. By three weeks after surgery the following percentages of fungiform taste buds had completely disappeared: 71% in gerbils, 28% in rats, and 26% in hamsters. Residual taste buds were classified into two groups: atrophic taste buds and taste bud remnants. Atrophic taste buds were smaller than normal and typically had no visible taste pore, although they retained the characteristic oval shape of a taste bud and numerous elongated cells. Taste bud remnants were non-oval fragments of taste buds with few elongated cells. Specific markers for elongated taste cells (monoclonal antibodies to keratin 19) confirmed that atrophic taste buds, as well as some taste bud remnants, had elongated taste cells. By 180 days after chorda-lingual nerve transection, 44% of rat fungiform taste buds had disappeared; morphometric analysis of the 311 residual taste buds established that 241 atrophic taste buds and 69 taste bud remnants were, respectively, 50% and 75% smaller than the average volume of 480 normal taste buds. The aggregate loss of gustatory tissue, calculated from the shrinkage of residual taste buds and the volume lost by the outright disappearance of many taste buds, was 88% for gerbils, 72% for rats, and 65% for hamsters. Evaluation in gerbils of the co-occurrence of taste buds and axons suggests residual taste buds were neurotrophically supported. We have used high voltage electron microscopy and computer-generated three-dimensional reconstructions from serial sections to elucidate the structure of taste bud cells and their associated synapses in fungiform taste buds of the mouse. Five fungiform taste buds (two of which were serially sectioned) were examined with the high-voltage electron microscope (HVEM). We identified the synaptic connections from taste cells onto sensory nerve fibers and classified the presynaptic taste cells based on previously established ultrastructural criteria. From those data we have distinguished dark, intermediate, and light cells in murine fungiform taste buds. Synapses in murine fungiform taste buds are fewer in number, but contain many more vesicles than synapses in either foliate or circumvallate taste buds. Synapses in mouse circumvallate and foliate taste buds typically contain a few to several synaptic vesicles per section, whereas fungiform synapses may have in excess of 100 vesicles per profile. The significance of these differences in the numbers of synapses and synaptic structure between fungiform and circumvallate/foliate synapses is not known. Based on the small number of synapses observed in fungiform taste buds, we speculate that fungiform taste buds have only a few cells transducing sensory stimuli at any given time. Alternatively, communication of sensory information from the taste receptor cells to the afferent nerve fibers may be mediated by some other mechanism(s) in addition to classical chemical synapses. © Wiley-Liss, Inc.

Thaumatin is a protein that tastes intensely sweet only to Old World monkeys and to higher primates, including man. Here we used pre-embedding ultrastructural methods to study the distribution of thaumatin in apical regions of Rhesus monkey foliate papillae, using thaumatin conjugated to 5 nm gold particles. With freeze-substitution we saw that gold-labeled thaumatin bound to an electron-opaque, sponge-like secretory substance inside the taste bud pores. Labeled thaumatin was found at the surface of the secretory substance even deep inside the pore, where other, unlabeled cellular structures surrounded the substance. With freeze-fracture deep-etching the secretory substance that bound the thaumatin-gold particles appeared coarsely granular. There was no labeling of any other taste bud pore structure, including microvilli and small membranelined vesicles. Pre-incubation with an excess of unlabeled thaumatin inhibited binding with goldlabeled thaumatin. The results suggest that the secretory substance had the greatest affinity of all taste pore structures to the sweet-tasting compound under our experimental conditions. Therefore, gustatory reception probably involves various taste compound binding structures, microvilli, and also secretory substances like the one described here which bound thaumatin. We speculate that the secretory substance may bind taste stimuli and serve as an intermediate between stimuli and receptors. The epithelial downgrowth arises in the presumptive foliate papilla region of the tongue approximately at prenatal day 22, and the distal portion of the primary epithelial cell cords transforms to the bifurcated serous gland and excretory duct between prenatal day 30 and postnatal day 2. The excretory ducts extend upward through the primary epithelial cell cords at rather random intervals and are open to the tongue surface between postnatal days 1 and 2. In this process, successive connections between the adjacent ascending excretory ducts occur mainly due to desquamation of the keratinized lining cells of the ducts, resulting in the formation of the foliate gutter. Taste bud primordia appear in the primary epithelial cell cords by nerve penetration through the basal lamina from prenatal day 30 and nearly fully formed taste buds facing the ascending excretory ducts have been already observed before the foliate gutter formation is completed. In the differentiation process of chemoreceptor (type III) cells from less-differentiated basal (type IV) cells, subsurface cisterns of endoplasmic reticulum are occasionally present where nerve endings make contact. Since subsurface cisterns have been considered to be involved in reciprocal or efferent synaptic transmission, it is reasonable to consider that these morphological specializations may take a role in neurotrophic signals for differentiation of the chemoreceptor cells.

The eight current species of bear (Ursidae) are widely distributed throughout Europe, Asia, and America. They are mainly encountered in the northern hemisphere, except for the spectacled bear and the sun bear, which are also found in the south of the equator. Adaptations of the masticatory apparatus (teeth, tongue, and musculature) to diet are one of the factors that imply the greatest structural changes in the cranium. This diet may be carnivorous, herbivorous, melliferous, or insectivorous, with one type of food predominating according to the time of year. The way in which food is eaten determines the morphology of the lingual surface; generally speaking, all bears put their mouth to the food, which, initially, they lick or they let the food stick to their tongue, as occurs when insects are eaten. As in all mammals, a distinction can be made between mechanical and gustatory papillae and the development and distribution of which depend on the species and their eating habits. In this study of the complete tongues of four species of adult bears, we describe the morphology of the lingual surfaces, the different types of papillae, their characteristics, and topographic distribution. It was seen that there were five main types of lingual papillae: filiform, conical, fungiform, foliate, and vallate. Morphology of the microgrooves and pores was similar to that observed in other mammals. In general, there were no great differences among the four species of bears studied, perhaps due to the similarity in the kind of food they consume in captivity.

A clear understanding of the role of different papillae in the pig may permit the development of a more palatable diet, thus, better utilizing anatomical structures and morphological characteristics to enhance health and productivity in this domestic species. The study used ten healthy market-size, mixed-breed pigs to properly describe the surface structure of the tongue papillae using standard scanning electron microscopic techniques. Two types of sharp and blunt filiform papillae were identified. Fungiform papillae, located on the lateral side of the tongue, were circular and large with surface taste pores. The majority of oval papillae on the dorsal surface of the tongue contained multidirectional surface grooves and lacked taste pores. The foliate papillae, located principally on the caudal third and lateral part of the tongue, were comprised of four to five leaves and they had additional pseudopapillae. The two vallate papillae, found in the caudal third and dorsum of the tongue, had a well-defined vallum and pseudopapillae on their surface. At the root of the tongue, two types of conical papillae with or without apex were observed. The entire area covered by conical papillae was deeply occupied by lingual tonsils. The lingual tonsils were scattered with varying sized lymph nodules in the propria mucosa of the conical papillae deep to epithelium. Functionally, the filiform and conical papillae appeared to have a mechanical role whereas fungiform, foliate, and vallate papillae apparently perform a gustatory role in the pig. The tonsillar location at the root of the tongue provides an ideal experimental model for immunohistological and immunological studies on the lymphoid tissue in man.

The barbel taste buds of catfish are widely used as a model system for investigating the structure and function of vertebrate taste buds. We have examined the ultrastructure of the taste buds of the channel catfish, Ictalurus punctatus, as part of a comparative study of the morphology of taste buds in various mammalian and non-mammalian vertebrates. Since conventional chemical fixation methods have limited usefulness for certain kinds of ultrastructural studies (i.e., localization of diffusible substances or labeling techniques requiring retention of biological activity), we have developed methods for fixing catfish barbel taste buds by high-pressure freezing followed by freeze-substitution (HPF/FS) and have compared the ultrastructure of taste buds fixed by this technique and by chemical fixation procedures. The morphological details of the cells within taste buds are significantly affected by the method of fixation employed. In general, membrane contours are smoother and intracellular organelles more regular in shape in HPF/FS samples as compared with the chemically fixed specimens. Absolute and relative electron-densities of various tissue components are also affected by the fixation method employed. Certain ultrastructural features are more clearly visualized by one or the other of the fixation procedures. Fixation of barbel taste buds by HPF/FS not only provides an alternative view of the ultrastructure of taste bud cells but also offers a method of tissue preparation that may prove to be preferable to chemical methods for ultrastructural investigations involving procedures such as immunohistochemical labeling.

HRP histochemistry and microelectrode mapping were combined to study the sizes, shapes, and orientations of neuronal cell bodies and dendrites located at sites of taste-elicited single unit activity in the nucleus of the solitary tract (NST). Cells responding to sapid stimulation of the anterior tongue were extracellularly recorded using micropipettes containing HRP. Iontophoretic injection of the marker at the recording sites resulted in small (50-200 microns diameter) opaque zones bordered by a small number (2-15) of neurons with Golgi-like filling of their cell bodies, dendrites, and to some extent, their axons. The cell bodies were near (50-250 microns) the injection sites into which they sent labelled dendrites. Two broad categories of neurons were typically filled. Elongate cells had oval- to spindle-shaped cell bodies oriented mediolaterally. Two primary dendrites extended 100-300 microns from the cell body, one medially and one laterally, and branched within a cylindrical dendritic field oriented mediolaterally. A minority of the HRP-filled elongate cells had unusually long rostrally or caudally directed dendritic branches. Stellate cells had oval, round, triangular, or polygonal cell bodies and 3-5 primary dendrites coursing 200-300 microns in all directions and branching as unoriented, spheroidal fields. A minority of stellate cells had relatively unbranched wavy dendrites, resembling tentacles, while others had unusually small cell bodies (10-15 microns diameter), small dendrites, and locally arborizing axons. Of 151 labelled cells, all but 12 were remarkably confined to the rostral NST. Nearly 90% were concentrated in the rostral central cytoarchitectonic subdivision, where stellate cells predominated, or in the rostral lateral subdivision, where elongate cells predominated. These morphological types of neurons, filled at neurophysiological recording sites, are compared with cell types identified in previous light and electron microscopic studies of the cytoarchitecture, connections, and synaptic organization of the gustatory NST.

The presence of glutamate immunoreactivity (glu-IR) in the nerve fibers of the mudpuppy taste bud was investigated by electron microscopy. Pre-embedding staining with avidin-biotin-peroxidase complex (ABC) and post-embedding staining with 5 nm colloid gold conjugates were used separately to identify immuno-stained structures. We have found the following: 1) the majority of the nerve fibers innervating the mudpuppy taste bud are unmyelinated; 2) about 85% of nerve fibers located at the base of the taste bud and about 60% of the nerve fibers located between the taste cells show glu-IR by pre-embedding staining; 3) there is a preferential staining of the glu-IR in the nerve fibers of the mudpuppy taste bud; and 4) the distribution of the colloidal gold particles in the nerve fibers is 1.5 to 2 times denser than that of the staining in the connective tissue background or cellular profiles of taste cells. From the distribution and pattern of the nerve fibers obtained in the thick and thin sections, we conclude that the mudpuppy taste bud is innervated by glutamate-containing unmyelinated nerve fibers.

The fine structure of the taste buds of circumvallate papillae of two strains of mice was studied by electron microscopy. Mice anesthetized with ketamine were perfused through the heart with a double aldehyde mixture in cacodylate buffer and the tissues embedded in Epon. Semi-serial sections were employed. The morphology and relationships of cell types are consistent with the majority of descriptions of mammalian taste buds served by the ninth cranial nerve. Cells of type II are particularly well documented, as the stages in their origin, maturation and degeneration could be followed. Significant differences, however, relate to cell type I. These cells contain large dense-cored granules, contrasted with the more irregular and somewhat larger dark granules of the type I cells in the rabbit. These granules do not produce a dense homogenous product for the pore, as seen in the rabbit. Rather the pore substance consists of small, empty vesicles in a diffuse dark matrix. These granules are only moderately larger than the dense-cored vesicles of the type III cells. All features of the type III cell were demonstrated, although no instance of a complete cell was seen in any section. No significant differences were noted between the two strains of mice. Intimate proximity of a nerve to a cell nucleolus, suggestive of a trophic pathway, is illustrated. © 1993 Wiley-Liss, Inc.
