Piezocatalytic materials have attracted widespread attention in the fields of clean energy and water treatment because of their ability to convert mechanical energy directly into chemical energy. In this study, γ-AlON particles synthesised using carbothermal reduction and nitridation (CRN) were used for the first time as a novel piezocatalytic material to degrade dye solutions under ultrasonic vibration. The γ-AlON particles exhibited good performance as a piezocatalytic material for the degradation of organic pollutants. After 120 min under ultrasonic vibration, 40 mg portions of γ-AlON particles in 50 mL dye solutions (10 mg/L) achieved 78.06%, 67.74%, 74.29% and 64.62% decomposition rates for rhodamine B (RhB), methyl orange (MO), methylene blue (MB) and crystal violet (CV) solutions, respectively; the fitted k values were 13.35 × 10−3, 10.79 × 10−3, 12.09 × 10−3 and 8.00 × 10−3 min−1, respectively. The piezocatalytic mechanism of γ-AlON particles in the selective degradation of MO was further analysed in free-radical scavenging activity experiments. Hydroxyl radicals (•OH), superoxide radicals (•O2−), holes (h+) and electrons (e−) were found to be the main active substances in the degradation process. The use of antimicrobial peptides (AMPs) covalently grafted on surfaces has been recognized in recent years as a promising strategy to fight against biofilm formation. However, after grafting, the understanding of AMP–bacteria interactions is still debated in the literature. In this study, Nisin, a cyclic AMP, was grafted onto gold surfaces via an indirect grafting on acidic thiol self-assembled monolayers using succinimide linkers. The physical and chemical properties of these SAMs were then finely characterized by XPS and FT-IR to confirm the covalent grafting of Nisin. The antiadhesion and bactericidal effects were then studied for Escherichia coli ATCC25922, Staphylococcus aureus ATCC 25923, and Listeria ivanovii Li4(pVS2) by a posteriori analysis of the culture supernatants (i.e., indirect technique) and ex situ by optical microscopy following crystal violet staining (i.e., direct technique). Statistical analysis reveals that the Nisin coating has bactericidal and antiadhesive properties towards Gram-positive bacteria, while no significant results were obtained for Gram-negative bacteria. Lithium, a natural element, has been employed as a mental stabilizer in psychiatric treatments; however, some reports indicate it has an anticancer effect, prompting the consideration of repurposing lithium for cancer treatment. The potential anticancer use of lithium may depend on its form (salt type) and the type of cancer cells targeted. Little is known about the effects of Li2CO3 or LiCl on cancer cells, so we focused on exploring their effects on proliferation, apoptosis, migration, and cell cycle as part of the hallmarks of cancer. Firstly, we established the IC50 values on HeLa, SiHa, and HaCaT cells with LiCl and Li2CO3 and determined by crystal violet that cell proliferation was time-dependent in the three cell lines (IC50 values for LiCl were 23.43 mM for SiHa, 23.14 mM for HeLa, and 15.10 mM for HaCaT cells, while the IC50 values for Li2CO3 were 20.57 mM for SiHa, 11.52 mM for HeLa, and 10.52 mM for HaCaT cells.) Our findings indicate that Li2CO3 and LiCl induce DNA fragmentation and caspase-independent apoptosis, as shown by TUNEL, Western Blot, and Annexin V/IP assay by flow cytometry. Also, cell cycle analysis showed that LiCl and Li2CO3 arrested the cervical cancer cells at the G1 phase. Moreover, lithium salts displayed an anti-migratory effect on the three cell lines observed by the wound-healing assay. A nanostructured material, ordered mesoporous carbon (OMC), was synthesised in metal- and halide-free form and its use for the sequestration of crystal violet, a hazardous triphenylmethane dye, is reported for the first time. The OMC material is characterised using scanning transmission electron microscopy with energy-dispersive spectroscopy for chemical analysis, by Fourier-transform infrared spectroscopy, and by nitrogen gas physisorption. The ideal conditions for the uptake of crystal violet dye were determined in batch experiments covering the standard parameters: pH, concentration, contact time, and adsorbent dosage. Experimental data are validated by applying Langmuir, Freundlich, Dubinin–Radushkevich, and Temkin isotherms. The thermodynamic parameters, ΔH°, ΔG°, and ΔS°, are calculated and it has been found that the adsorption process is spontaneous and endothermic with increasing disorder. An in-depth analysis of the kinetics of the adsorption process, order of the reaction and corresponding values of the rate constants was performed. The adsorption of crystal violet over OMC has been found to follow pseudo-second-order kinetics through a film diffusion process at all temperatures studied. Continuous flow column operations were performed using fixed bed adsorption. Parameters including percentage saturation of the OMC bed are evaluated. Zinc cobalt oxide-zinc hydroxide (ZnCo2O4-Zn(OH)2) microspheres were successfully fabricated on carbon cloth via a sample hydrothermal method. The surface morphology of these microspheres and their efficacy in degrading methyl violet were further modulated by varying the thermal annealing temperatures. Adjusting the thermal annealing temperatures was crucial for controlling the porosity of the ZnCo₂O₄-Zn(OH)₂ microspheres, enhancing their photocatalytic performance. Various analytical techniques were utilized to evaluate the physical and chemical properties of the ZnCo2O4-Zn(OH)2 microspheres, including field-emission scanning electron microscopy, energy-dispersive spectroscopy, X-ray diffraction, field-emission transmission electron microscopy, X-ray photoelectron spectroscopy, and UV-vis spectroscopy. Compared to untreated ZnCo2O4-Zn(OH)2 microspheres, those subjected to thermal annealing exhibited increased specific surface area and light absorption capacity, rendering them highly effective photocatalysts under UVC light exposure. Subsequent studies have confirmed the superior performance of ZnCo2O4-Zn(OH)2 microspheres as a reusable photocatalyst for degrading methyl violet and tetracycline. A scalable and cost-effective solution for removing pollutants from water is to use biodegradable and eco-friendly sorbents that are readily available such as starch. The current research explored the removal of crystal violet (CV) dye from water using chemically modified potato starch. The adsorbent was prepared by cross-linking potato starch with sodium trimetaphosphate (STMP). The impact of various operating factors including pH, temperature, contact time, initial CV concentration, and adsorbent dosage on the removal of CV were investigated using batch experiments. The adsorption data were analyzed using a fuzzy regression approach, which provided a range-based representation of the model's output. The cross-linked starch adsorbent was mesoporous, with a mean pore diameter of 9.8 nm and a specific surface area of 2.7 m2/g. The adsorption of CV by the STMP cross-linked potato starch was primarily influenced by the adsorbent dosage, followed by the solution pH, temperature, initial CV concentration, and contact time. The fuzzy regression model accurately predicted the independent experimental data of CV removal with an R2 of 0.985, demonstrating its value as a tool for the continuous monitoring of CV removal as well as optimizing water treatment conditions. Acid mine drainage (AMD) is one of the main environmental problems associated with mining activity, whether the mine is operational or abandoned. In this work, several precipitates from this mine drainage generated by the oxidation of sulfide minerals, when exposed to weathering, were used as adsorbents. Such AMD precipitates from abandoned Portuguese mines (AGO, AGO-1, CF, and V9) were compared with two raw materials from Morocco (ClayMA and pyrophyllite) in terms of their efficiency in wastewater treatment. Different analytical techniques, such as XRD diffraction (XRD), Fourier Transform Infrared spectroscopy (FTIR), N2 adsorption isotherms, and Scanning Electron Microscopy (SEM) with Energy Dispersive X-ray (EDX) were used to characterize these natural materials. The adsorption properties were studied by optimizing different experimental factors, such as type of adsorbent, adsorbent mass, and dye concentration by the Box–Behnken Design model, using methylene blue (MB) and crystal violet (CV) compounds as organic pollutants. The obtained kinetic data were examined using the pseudo-first and pseudo-second order equations, and the equilibrium adsorption data were studied using the Freundlich and Langmuir models. The adsorption behavior of the different adsorbents was perfectly fitted by the pseudo-second order kinetic model and the Langmuir isotherm. The most efficient adsorbent for both dyes was AGO-1 due to the presence of the cellulose molecules, with qm equal to 40.5 and 16.0 mg/g for CV and MB, respectively. This study confirms the possibility of employing AMD precipitates to adsorb organic pollutants in water, providing valuable information for developing future affordable solutions to reduce the wastes associated with mining activity. Gac fruit (Momordica cochinchinensis Spreng.) is a prominent source of carotenoids, renowned for its exceptional concentration of these compounds. This study focuses on optimizing the extraction of active components from the aril of gac fruit by evaluating the effects of extraction temperature, solid–liquid ratio, and extraction time. The primary objective is to maximize the yield of gac oil while assessing its antioxidant capacity. To analyze the kinetics of the solid–liquid extraction process, both first-order and second-order kinetic models were employed, with the second-order model providing the best fit for the experimental data. In addition, the potential of gac fruit peel as a precursor for biochar production was investigated through carbonization. The resultant biochars were evaluated for their efficacy in adsorbing crystal violet (CV) dye from aqueous solutions. The adsorption efficiency of the biochars was found to be dependent on the carbonization temperature, with the highest efficiency observed for BCMC550 (91.72%), followed by BCM450 (81.35%), BCMC350 (78.35%), and BCMC250 (54.43%). The adsorption isotherm data conformed well to the Langmuir isotherm model, indicating monolayer adsorption behavior. Moreover, the adsorption kinetics were best described by the pseudo-second-order model. These findings underscore the potential of gac fruit and its byproducts for diverse industrial and environmental applications, highlighting the dual benefits of optimizing gac oil extraction and utilizing the peel for effective dye removal. Efficient and thorough treatment of dye wastewater is essential to achieve ecological harmony. In this study, a new type of calcium-based modified coal gangue (Ca-CG) was prepared by using solid waste coal gangue as raw material and a CaCl2 modifier, which was used for the removal of malachite green, methylene blue, crystal violet, methyl violet and other dyes in water. When the dosage of Ca-CG was 1–5 g/L, the dosage of Ca-CG was the main factor affecting the dye adsorption effect. The adsorption effects of Ca-CG on four dyes were as follows: malachite green > crystal violet > methylene blue > methyl violet. Kinetics, isotherms and thermodynamic analysis showed that the adsorption of malachite green, methyl blue, crystal violet and methyl violet by Ca-CG fitted the second-order kinetic model, and adsorption with chemical reaction is the main process. The adsorption of four dyes by Ca-CG conformed to the Freundlich model, which is dominated by multi-molecular layer adsorption, and the adsorption was easy to carry out. The adsorption process of Ca-CG on the four dyes was spontaneous. The results of FTIR, XRD and SEM showed that the calcium-based materials such as lipscombite and dolomite were the key to the adsorption of malachite green by Ca-CG, and the main mechanisms for the adsorption of malachite green by Ca-CG are surface precipitation, electrostatic action, and chelation reaction. The aqueous instability of halide perovskite seriously hinders its direct application in water as a potential photocatalyst. Here, we prepared a new type of polyvinylpyrrolidone (PVP) passivated δ-CsPbI3 (δ-CsPbI3@PVP) microcrystal by a facile method. This material can be uniformly dispersed in water and stably maintain its crystal structure for a long time, breaking through the bottleneck of halide perovskite photocatalysis in water. Under visible light, δ-CsPbI3@PVP can almost completely photodegrade organic dyes (including Rhodamine B, methylene blue, and crystal violet) in only 20 min. The efficient photocatalytic activity is attributed to the enhanced visible light absorption arising from PbI2 defects in δ-CsPbI3@PVP and the intrinsic low photoluminescence quantum yield of δ-CsPbI3, which induces efficient light absorption and photocatalytic activity. We highlight δ-CsPbI3@PVP as an effective aqueous photocatalyst, and this study provides new insights into how to exploit the potential of halide perovskite in photocatalytic applications. The rapid growth of the dye and textile industry has raised significant public concerns regarding the pollution caused by dye wastewater, which poses potential risks to human health. In this study, we successfully improved the adsorption efficiency of activated carbon derived from pomegranate peel waste (PPAC) through a single-step and surface modification approach using 5-sulfonate-salicylaldehyde sodium salt. This innovative and effective sulfonation approach to produce sulfonated activated carbon (S-PPAC) proved to be highly effective in removing crystal violet dye (CV) from polluted water. The prepared PPAC and S-PPAC were characterized via FESEM, EDS, FTIR and BET surface area. Characterization studies confirmed the highly porous structure of the PPAC and its successful surface modification, with surface areas reaching 1180.63 m2/g and 740.75 m2/g for the PPAC and S-PPAC, respectively. The maximum adsorption capacity was achieved at 785.53 mg/g with the S-PPAC, an increase of 22.76% compared to the PPAC at 45 °C. The isothermic adsorption and kinetic studies demonstrated that the adsorption process aligned well with the Freundlich isotherm model and followed the Elovich kinetic model, respectively. Fungicides are widely used in agriculture for crop protection. Succinate dehydrogenase inhibitors (SDHIs) and strobilurins inhibit mitochondria electron transport chain (ETC) in fungi, by blocking complex II and complex III, respectively. Questions regarding their selectivity of action for fungi have been raised in the literature, and we previously showed that boscalid and bixafen (SDHIs) alter the mitochondrial function of human hepatocytes. Here, we analyzed the impact of the exposure of human hepatocytes to pyraclostrobin, a fungicide belonging to the class of strobilurins. Using electron paramagnetic resonance (EPR), we observed a decrease in oxygen consumption rate (OCR) and an increase in mitochondrial superoxide levels after 24 h exposure to 0.5 µM concentration. As a consequence, the content in ATP amount in the cells was reduced, the ratio reduced/oxidized glutathione was decreased, and a decrease in cell viability was observed using three different assays (PrestoBlue, crystal violet, and annexin V assays). In addition, as SDHIs and strobilurins are commonly associated in commercial preparations, we evaluated a potential "cocktail" toxic effect. We selected low concentrations of boscalid (0.5 µM) and pyraclostrobin (0.25 µM) that did not induce a mitochondrial dysfunction in liver cells when used separately. In sharp contrast, when both compounds were used in combination at the same concentration, we observed a decrease in OCR, an increase in mitochondrial superoxide production, a decrease in the ratio reduced/oxidized glutathione, and a decrease in cell viability in three different assays. Coordination polymers (CPs) are a diverse class of multi-dimensional compounds that show promise as photocatalysts for degrading dyes in polluted water. Herein, a new 1D Cd(II)-based coordination polymer with the formula [Cd(bpyp)(nba)2] (1) (bpyp = 2,5-bis(pyrid-4-yl)pyridine and Hnba = 4-nitrobenzoic acid) is synthesized and characterized. In 1, the two carboxyl groups of two different nba− ligands show μ2-η1:η1 and μ1-η1:η1 coordination modes to connect the CdII centers and sit on either side of the chain along the b direction. The produced CP 1 was utilized as the photocatalyst in the process of the photodegradation of methyl blue (MB), methyl orange (MO), rhodamine B (RhB), and methyl violet (MV) dyes when exposed to UV light. The photocatalytic degradation activities of CP 1 were analyzed, and the results suggest that it exhibits an extraordinary efficiency in the degradation of MB, MV, MO, and RhB. RhB has a 95.52% efficiency of degradation, whereas MV has a 58.92% efficiency, MO has 35.44%, and MB has 29.24%. In the present study, the ability of an adsorbent (SLM Stem) obtained from the stem of the Silybum Marianum plant to treat wastewater containing the cationic dyes basic blue 3 (BB3) and basic yellow 28 (BY28) from aqueous solutions was investigated using a batch method. Then, the SLM Stem (SLM Stem-Natural) adsorbent was carbonized at different temperatures (200–900 °C) and the removal capacity of the products obtained for both dyes was examined again. The investigation continued with the product carbonized at 800 °C (SLM Stem-800 °C), the adsorbent with the highest removal capacity. The dyestuff removal studies were continued with the SLM Stem-Natural and SLM Stem-800 °C adsorbents because they had the highest removal values. The surface properties of these two adsorbents were investigated using IR, SEM, and XRD measurements. It was determined that the SLM Stem-Natural has mainly non-porous material, and the SLM Stem-800 °C has a microporous structure. The optimal values for various parameters, including adsorbent amount, initial dye solution concentration, contact time, temperature, pH, and agitation speed, were investigated for BY28 dye and were 0.05 g, 15 mg/L, 30 min, 40 °C, pH 6 and 100 rpm when SLM Stem-Natural adsorbent was used and, 0.15 g, 30 mg/L, 30 min, 40 °C, pH 10, and 150 rpm when SLM Stem-800 °C adsorbent was used. For BB3 dye, optimal parameter values of 0.20 g, 10 mg/L, 30 min, 25 °C, pH 7, and 100 rpm were obtained when SLM Stem-Natural adsorbent was used and 0.15 g, 15 mg/L, 40 min, 40 °C, pH 10, and 100 rpm when SLM Stem-800 °C adsorbent was used. The Langmuir isotherm described the adsorption process best, with a value of r2 = 0.9987. When SLM Stem-800 °C adsorbent was used for BY28 dye at 25 °C, the highest qm value in the Langmuir isotherm was 271.73 mg/g. When the study was repeated with actual water samples under optimum conditions, the highest removal for the BY28 dye was 99.9% in tap water with the SLM Stem-800 °C adsorbent. Determining the approximate dates that written documents were drawn up based on the chemical composition of the ink is not a simple process. It is very demanding in terms of legal requirements. Various studies have succeeded in dating manuscripts by analyzing the temporal evolutions of the concentrations of dyes and solvents in documents based on the original formulations of the ink pens. These analyses were carried out simultaneously by HPLC-DAD for dyes and by GC-MS for solvents. This study aims, for the first time, to evaluate novel ink compounds and the temporal evolution of the concentrations of the degradation products of the dyes used by most suppliers and which are present in almost all types of ballpoint inks, i.e., Crystal Violet (CV). CV degrades through two parallel pathways: on the one hand, it undergoes progressive demethylation until it becomes pararosaniline, and on the other, it undergoes a breakdown of the molecule obtaining, among other by-products, the compound N,N′-Dimethyl-4-aminophenol (NNAPH), that was experimentally verified using four different inks (e.g., Inoxcrom® and Sigma® brands, in blue and black). For the NNAPH compound, we observed that four of the inks under analysis displayed the same temporary behavior despite having different initial chemical compositions. These initial results show the high potential for both CV and NNAPH, together with the rest of the pararosaniline family, as age tracers for dated/old documents. In this study, activated carbons were produced through the chemical activation of caraway seeds using three different activators: Na2CO3, K2CO3, and H3PO4. A 1:2 weight ratio of precursor to activator was maintained in every instance. Comprehensive analyses were conducted on the resultant activated carbons, including elemental analysis, textural parameters determination, Boehm titration for surface oxygen functional groups, pH assessment of aqueous extracts, and quantification of ash content. The produced materials were subjected to adsorption tests for methylene blue and methyl red sodium salt from the liquid phase and the effects of adsorbent dosage, pH of the aqueous dye solution, process temperature, and adsorbent–adsorbate contact time on sorption capacity obtained. To characterize the adsorption model of the examined pollutants, both the Langmuir and Freundlich equations were employed. In addition, the sorption capacity of the obtained carbon materials against an iodine aqueous solution was assessed. The specific surface area of the obtained adsorbents ranged from 269 to 926 m2/g. By employing potassium carbonate to chemically activate the starting substance, the resulting activated carbons show the highest level of specific surface area development and the greatest sorption capacity against the tested impurities—296 mg/g for methylene blue and 208 mg/g for methyl red sodium salt. The adsorption rate for both dyes was determined to align with a pseudo-second-order kinetic model. The experimental adsorption data for methylene blue were well-described by the Langmuir model, whereas the Freundlich model was found to be congruent with the data pertaining to methyl red sodium salt. Globularia alypum L. (GA) is a Mediterranean plant of the Globulariaceae family which is widely used in traditional Tunisian medicine. The main goal of this study was to evaluate the phytochemical composition, antioxidant, antibacterial, and antibiofilm activities, and the antiproliferative potential of different extracts of this plant. The identification and the quantification of the different constituents of extracts were determined using gas chromatography–mass spectrometry (GC-MS). The antioxidant activities were evaluated using spectrophotometric methods and chemical tests. The antiproliferative study was based on the use of colorectal cancer SW620 cells, including an antibacterial assessment with the microdilution method and analysis of the antibiofilm effects via the crystal violet assay. All extracts presented several components, mainly sesquiterpenes, hydrocarbon, and oxygenated monoterpenes. The results revealed that the maceration extract had the most important antioxidant effect (IC50 = 0.04 and 0.15 mg/mL), followed by the sonication extract (IC50 = 0.18 and 0.28 mg/mL). However, the sonication extract demonstrated significant antiproliferative (IC50 = 20 µg/mL), antibacterial (MIC = 6.25 mg/mLand MBC > 25 mg/mL), and antibiofilm (35.78% at 25 mg/mL) properties against S. aureus. Klebsiella is a common dangerous pathogen for humans and animals and is widely present in the digestive system. The genus Klebsiella is ubiquitous, as it is endemic to surface water, soil, and sewage. In this study, 70 samples were obtained from soil-dwelling invertebrates from September 2021 to March 2022 from Taif and Shafa in different altitudinal regions of Saudi Arabia. Fifteen of these samples were identified as Klebsiella spp. The Klebsiella isolates were genetically identified as Klebsiella pneumoniae using rDNA sequencing. The antimicrobial susceptibility of the Klebsiella isolates was determined. Amplification of virulence genes was performed using PCR. In this study, 16S rDNA sequencing showed a similarity from 98% to 100% with related K. pneumonia from the NCBI database, and the sequences were deposited in the NCBI GenBank under accession numbers ON077036 to ON077050. The growth inhibition properties of ethanolic and methanolic extracts of the medicinal plant Rhazya stricta's leaves against K. pneumoniae strains using the minimum inhibitory concentration (MIC) method and disc diffusion were evaluated. In addition, the biofilm inhibitory potential of these extracts was investigated using crystal violet. HPLC analysis identified 19 components divided into 6 flavonoids, 11 phenolic acids, stilbene (resveratrol), and quinol, and revealed variations in the number of components and their quantities between extracts. Both extracts demonstrated interesting antibacterial properties against K. pneumoniae isolates. The 2 extracts also showed strong biofilm inhibitory activities, with percentages of inhibition extending from 81.5% to 98.7% and from 35.1% to 85.8% for the ethanolic and methanolic extracts, respectively. Rhazya stricta leaf extract revealed powerful antibacterial and antibiofilm activities against K. pneumoniae isolates and could be a good candidate for the treatment or prevention of K. pneumonia-related infections. This study includes the characterization and exploitation of an abundant agricultural waste in Algeria, Alfa (Stipa tenacissima L.) leaf powder (ALP) as a biosorbent for the removal of hazardous triphenylmethane dyes, malachite green (basic green 4) and crystal violet (basic violet 3), from aqueous media under various operating conditions in batch mode. The effect of experimental parameters such as initial dye concentration (10–40 mg/L), contact time (0–300 min), biosorbent dose (2.5–5.5 g/L), initial pH (2–8), temperature (298–328 K), and ionic strength on dye sorption was investigated. The results of both dyes show that the increase in initial concentration, contact time, temperature, and initial pH of solution leads to an increase in biosorbed quantity, unlike the effect of ionic strength. The biosorption kinetics for triphenylmethane dyes on ALP was analyzed by pseudo-first-order, pseudo-second-order, Elovich, and intraparticle diffusion models proposed by the Weber–Morris equation. Equilibrium sorption data were analyzed by six isotherms, namely the Langmuir, Freundlich, Harkins–Jura, Flory–Huggins, Elovich, and Kiselev isotherms. Flavonoids are the most abundant class of secondary metabolites that are ubiquitously involved in plant development and resistance to biotic and abiotic stresses. Flavonoid biosynthesis involves multiple channels of orchestrated molecular regulatory factors. Methyl jasmonate (MeJA) has been demonstrated to enhance flavonoid accumulation in numerous plant species; however, the underlying molecular mechanism of MeJA-induced flavonoid biosynthesis in safflower is still not evident. In the present study, we revealed the underlying molecular basis of a putative F3′5′H gene from safflower imparting MeJA-induced flavonoid accumulation in transgenic plants. The constitutive expression of the CtF3′5′H1 gene was validated at different flowering stages, indicating their diverse transcriptional regulation through flower development in safflower. Similarly, the CtF3′5′H1-overexpressed Arabidopsis plants exhibit a higher expression level, with significantly increased anthocyanins and flavonoid content, but less proanthocyanidins than wild-type plants. In addition, transgenic plants treated with exogenous MeJA revealed the up-regulation of CtF3′5′H1 expression over different time points with significantly enhanced anthocyanin and flavonoid content as confirmed by HPLC analysis. Moreover, CtF3′5′H1- overexpressed Arabidopsis plants under methyl violet and UV-B irradiation also indicated significant increase in the expression level of CtF3′5′H1 with improved anthocyanin and flavonoid content, respectively. Noticeably, the virus-induced gene silencing (VIGS) assay of CtF3′5′H1 in safflower leaves also confirmed reduced anthocyanin accumulation. However, the CtF3′5′H1 suppression in safflower leaves under MeJA elicitation demonstrated significant increase in total flavonoid content. This study aimed to investigate the antibacterial [minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC)] and antibiofilm activity [log10 colony forming unit/mL (CFU/mL) and biofilm disruption] of copper-doped phosphate glass (CDPG) against Streptococcus oralis, Enterococcus faecalis, Lactobacillus casei, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. Methods: the antibacterial activity was determined using microbroth dilution and time-kill assay. The antibiofilm activity was investigated using crystal violet and confocal laser scanning microscopy. Bacteria growing in absence of CDPG were used as controls. Results: the MIC was ≥125 mg of CPDG/mL; the log10 CFU/mL reduction ranged from 2.66–3.14 to 6.23–9.65 after 4 and 24 h respectively. Generally, no growth was observed after 24 h of treatment with CDPG; the MBC was 250 mg/mL for L. casei and S. oralis while 500 mg/mL for the rest of the bacteria. The highest and lowest antibiofilm activity was observed against S. oralis and E. coli respectively. Three patterns of complete biofilm disruption were seen: (i) large areas with E. fecalis and S. oralis, (ii) medium-size pockets with S. aureus and P. aeruginosa, or (iii) small areas with E. coli and L. casei. Antibiotic water contamination is a growing environmental problem in the present day. As a result, water treatment is required for its reduction and elimination. Due to their important role in resolving this issue, photocatalysts have drawn a great deal of interest over the past few decades. When non-biodegradable organic matter is present in polluted water, the photo catalytic process, which is both environmentally friendly and an improved oxidation method, can be an effective means of remediation. In this regard, we report the successful synthesis of pure phased rare earth doped ZnO nanoparticles for tetracycline degradation. The prepared catalysts were systematically characterized for structural, optical, and magnetic properties. The optical band gap was tailored by rare earth doping, with redshift for Sm and Dy doped nanoparticles and blueshift for Nd doped ZnO nanoparticles. The analysis of photoluminescence spectra revealed information about the defect chemistry of all synthesised nanoparticles. Magnetic studies revealed that all synthesized diluted magnetic semiconductors exhibit room temperature ferromagnetism and can be employed for spintronic applications. Moreover, Dy doped ZnO nanoparticles were found to exhibit a maximum degradation efficiency of 74.19% for tetracycline (TCN) removal. The synthesized catalysts were also employed for the degradation of Malachite green (MG), and Crystal violet (CV) dyes. The maximum degradation efficiency achieved was 97.18% for MG and 98% for CV for Dy doped ZnO nanoparticles. The pathogenicity of foodborne Vibrio parahaemolyticus is a major concern for global public health. This study aimed to optimize the liquid–solid extraction of Wu Wei Zi extracts (WWZE) against Vibrio parahaemolyticus, identify its main components, and investigate the anti-biofilm action. The extraction conditions optimized by the single-factor test and response surface methodology were ethanol concentration of 69%, temperature at 91 °C, time of 143 min, and liquid–solid ratio of 20:1 mL/g. After high performance liquid chromatography (HPLC) analysis, it was found that the main active ingredients of WWZE were schisandrol A, schisandrol B, schisantherin A, schisanhenol, and schisandrin A–C. The minimum inhibitory concentration (MIC) of WWZE, schisantherin A, and schisandrol B measured by broth microdilution assay was 1.25, 0.625, and 1.25 mg/mL, respectively, while the MIC of the other five compounds was higher than 2.5 mg/mL, indicating that schisantherin A and schizandrol B were the main antibacterial components of WWZE. Crystal violet, Coomassie brilliant blue, Congo red plate, spectrophotometry, and Cell Counting Kit-8 (CCK-8) assays were used to evaluate the effect of WWZE on the biofilm of V. parahaemolyticus. The results showed that WWZE could exert its dose-dependent potential to effectively inhibit the formation of V. parahaemolyticus biofilm and clear mature biofilm by significantly destroying the cell membrane integrity of V. parahaemolyticus, inhibiting the synthesis of intercellular polysaccharide adhesin (PIA), extracellular DNA secretion, and reducing the metabolic activity of biofilm. Injectable platelet-rich fibrin (iPRF) is a frequently used platelet concentrate used for various medical purposes both in veterinary and human medicine due to the regenerative potential of hard and soft tissues, and also because of its antimicrobial effectiveness. This in vitro study was carried out to assess the cumulative antimicrobial and antibiofilm effect of iPRF functionalized with a multifunctional glycoprotein, human lactoferrin (Lf). Thus, the ability to potentiate cell proliferation was tested on keratinocytes and evaluated by the CCK8 test. The combinations of iPRF and Lf induced an increase in the proliferation rate after 24 h. The average cell viability of treated cultures (all nine variants) was 102.87% ± 1.00, and the growth tendency was maintained even at 48 h. The highest proliferation rate was observed in cultures treated with 7% iPRF in combination with 50 µg/mL of Lf, with an average viability of 102.40% ± 0.80. The antibacterial and antibiofilm activity of iPRF, of human lactoferrin and their combination were tested by agar-well diffusion (Kirby–Bauer assay), broth microdilution, and crystal violet assay against five reference bacterial strains. iPRF showed antimicrobial and antibiofilm potential, but with variations depending on the tested bacterial strain. The global analysis of the results indicates an increased antimicrobial potential at the highest concentration of Lf mixed with iPRF. The study findings confirmed the hypothesized enhanced bioactive properties of functionalized iPRF against both Gram-positive and Gram-negative biofilm-producing bacteria. Three new monosquaramides (3a–c) were synthesized, characterized by IR, NMR and X-ray, and evaluated for inhibitory activity against deoxyribonuclease I (DNase I) and xanthine oxidase (XO) in vitro. The target compounds inhibited DNase I with IC50 values below 100 μM, being at the same time more potent DNase I inhibitors than crystal violet, used as a positive control. 3-Ethoxy-4-((1-(pyridin-3-yl)propan-2-yl)amino)cyclobut-3-ene-1,2-dione (3c) stood out as the most potent compound, exhibiting a slightly better IC50 value (48.04 ± 7.98 μM) compared to the other two compounds. In order to analyze potential binding sites for the studied compounds with DNase I, a molecular docking study was performed. A novel malachite green molecularly imprinted membrane (MG-MIM) with specific selectivity for malachite green (MG) and leucomalachite green (LMG) was prepared using a hydrophobic glass fiber membrane as the polymer substrate, methyl violet as a template analog, 4-vinyl benzoic acid as the functional monomer, and ethyleneglycol dimethacrylate as the crosslinking agent. MG-MIM and non-imprinted membrane (NIM) were structurally characterized using scanning electron microscopy, surface area analyzer, Fourier-transform infrared spectrometer and synchronous thermal analyzer. The results showed that MG-MIM possessed a fluffier surface, porous and looser structure, and had good thermal stability. Adsorption properties of MG-MIM were investigated under optimal conditions, and adsorption equilibrium was reached in 20 min. The saturated adsorption capacities for MG and LMG were 24.25 ng·cm−2 and 13.40 ng·cm−2, and the maximum imprinting factors were 2.41 and 3.20, respectively. Issues such as "template leakage" and "embedding" were resolved. The specific recognition ability for the targets was good and the adsorption capacity was stable even after five cycles. The proposed method was successfully applied for the detection of MG and LMG in real samples, and it showed good linear correlation in the range of 0 to 10.0 μg·L−1 (R2 = 0.9991 and 0.9982), and high detection sensitivity (detection limits of MG and LMG of 0.005 μg/kg and 0.02 μg·kg−1 in shrimp, and 0.005 μg/kg and 0.02 μg/kg in fish sample). The recoveries and relative standard deviations were in the range of 76.31–93.26% and 0.73–3.72%, respectively. Magnetic metal–organic frameworks (MMOFs) are gaining increased attention as emerging adsorbents/water remediation agents. Herein, a facile development of novel MMOFs comprised of coated ferrite nanoparticles (MNPs) and UiO-66 metal–organic framework is reported. In specific, coated Co- and Zn-doped ferrite magnetic nanoparticles were synthesized as building block while the metal–organic framework was grown in the presence of MNPs via a semi-self-assembly approach. The utilization of coated MNPs facilitated the conjugation and stands as a novel strategy for fabricating MMOFs with increased stability and an explicit structure. MMOFs were isolated with 13–25 nm crystallites sizes, 244–332 m2/g specific surface area (SSA) and 22–42 emu/g saturation magnetization values. Establishing the UiO-66 framework via the reported semi-self-assembly resulted in roughly 70% reduction in both magnetic properties and SSA, compared with the initial MNPs building blocks and UiO-66 framework, respectively. Nonetheless, the remaining 30% of the magnetization and SSA was adequate for successful and sufficient adsorption of two different pesticides, 2,4-Dichlorophenoxyacetic acid (2,4-D) and 2,4,5-Trichlorophenoxyacetic acid (2,4,5-T), while the recovery with a commercial magnet and reuse were also found to be effective. An anti-biofilm that can inhibit the matrix of biofilm formation is necessary to prevent recurrent and chronic Pseudomonas aeruginosa infection. This study aimed to design compounds with a new mechanism through competitive inhibitory activity against phosphomannomutase/phosphoglucomutase (PMM/PGM), using in vitro assessment and a computational (in silico) approach. The active site of PMM/PGM was assessed through molecular redocking using L-tartaric acid as the native ligand and other small molecules, such as glucaric acid, D-sorbitol, and ascorbic acid. The docking program set the small molecules to the active site, showing a stable complex formation. Analysis of structural similarity, bioavailability, absorption, distribution, metabolism, excretion, and toxicity properties proved the potential application of ligands as an anti-biofilm. In vitro assessment with crystal violet showed that the ligands could reach up to 95.87% inhibition at different concentrations. Green propolis may represent a promising therapeutic alternative against dental anaerobic pathogens because of its antimicrobial action. This study aimed to evaluate the antimicrobial and antibiofilm actions of Brazilian green propolis aqueous extract (BGP-AqExt) against dental anaerobic bacteria. The minimum inhibitory concentration (MIC) and minimum microbicide concentration (MMC) of the extract were determined against the standard strains (ATCC) of Fusobacterium nucleatum, Parvimonas micra, Prevotella intermedia, Porphyromonas gingivalis and Porphyromonas endodontalis. BGP-AqExt was chemically characterized by high-performance liquid chromatography with diode-array detection (HPLC-DAD) analysis. Antibiofilm action was measured by MTT and crystal violet tests. The data were statistically analyzed by ANOVA and Tukey (5%) tests. The extract had antimicrobial action against all tested anaerobic bacteria, with an MIC value of 55 mg/mL for all bacteria, an MMC of 27.5 mg/mL for F. nucleatum and P. micra and 55 mg/mL for P. intermedia. Chemically, BGP-AqExt is composed of quercetin, gallic acid, caffeic and p-coumaric acid, drupani, kaempferol and Artepillin C. Significant reductions in biomass and metabolic action of biofilms were found after BGP-AqExt application. Organic dyes are widely used in the textile, biological, medical and other fields. However, a serious environmental problem has appeared because of the presence of organic dyes in industrial aqueous effluents. Thus, the efficient treatment of organic dyes in industrial wastewaters is currently in real demand. The current study investigated the oxidative degradation of the organic dye gentian violet by meso-tetra(carboxyphenyl) porphyriniron(III), [FeIII(TCPP)] as a cytochrome P450 model and iodosylbenzene (PhIO) as an oxidant at room temperature. The degradation reaction was monitored by UV–vis absorption spectroscopy via the observation of UV–vis spectral changes of the gentian violet. The results showed that the efficiency of catalyzed degradation reached more than 90% in 1 h, indicating the remarkable oxidative degradation capacity of the [FeIII(TCPP)]/PhIO system, which provided an efficient approach for the treatment of dyeing wastewater. HPLC-UV was used to compare the major constituents of two Pelargonium × hortorum cultivars and Pelargonium sidoides root extract. It revealed the presence of catechin and gallic acid in high concentrations and the absence of umckalin in P. × hortorum root extracts. The antibacterial activity of these extracts was screened against 19 Pseudomonas aeruginosa clinical isolates. P. × hortorum root extracts showed the lowest MIC values (512–1024 µg/mL). This activity was concluded to be attributable to the high concentrations of catechin and gallic acid. The anti-biofilm activity of catechin, gallic acid, and their combination was examined by a crystal violet assay. The combination reduced the percentage of strong and moderate biofilm-forming isolates from 52.63% to 5.26%. The impact on lasI and lasR genes expression using qRT-PCR and simultaneous docking against LasR protein was explored. The combination downregulated lasI and lasR gene expression in eight and six P. aeruginosa isolates, respectively, and showed the greatest docking score. Additionally, the in vivo protection capability of this combination in infected mice showed enhancement in the survival rate. Our study revealed the potential biofilm and quorum-sensing-inhibitory activity of the catechin and gallic acid combination as a novel alternative to inhibit bacterial pathogenicity. A series of copper(II) complexes of 2-imino-2H-chromen-3-yl-1,3,5-triazines 2a-h, 3-(benzoxazol-2-yl)-2H-chromen-2-imines 4a-b, and 3-(benzothiazol-2-yl)-2H-chromen-2-imines 6a-c were obtained by reacting of appropriate 2-iminocoumarin ligands L1a-h, L3a-b, and L5a-c with 3-fold molar excess of copper(II) chloride. The structure of these compounds was confirmed by IR spectroscopy, elemental analysis, and single-crystal X-ray diffraction data (2f, 2g, 2h, and 6c). All the synthesized complexes were screened for their activity against five human cancer cell lines: DAN-G, A-427, LCLC-103H, SISO, and RT-4 by using a crystal violet microtiter plate assay and relationships between structure and in vitro cytotoxic activity are discussed. The coordination of 2-iminocoumarins with copper(II) ions resulted in complexes 2a-h, 4a-b, and 6a-c with significant inhibitory properties toward tested tumor cell lines with IC50 values ranging from 0.04 μM to 15.66 μM. In comparison to the free ligands L1a-h, L3a-b, and L5a-c, the newly prepared Cu(II) complexes often displayed increased activity. In the series of copper(II) complexes of 2-imino-2H-chromen-3-yl-1,3,5-triazines 2a-h the most potent compound 2g contained a 4-phenylpiperazine moiety at position 6 of the 1,3,5-triazine ring and an electron-donating diethylamino group at position 7′ of the 2-iminocoumarin scaffold. Among the Cu(II) complexes of 3-(benzoxazol-2-yl)-2H-chromen-2-imines 4a-b and 3-(benzothiazol-2-yl)-2H-chromen-2-imines 6a-c the most active was benzoxazole-2-iminocoumarin 4b that also possessed a diethylamino group at position 7′ of the 2-iminocoumarin moiety. Photocatalysis has been recognized as a feasible method in water and wastewater treatment. Compared to other methods such as adsorption and chemical oxidation, the use of photocatalyst in the advanced oxidation processes gives benefits such as a longer lifetime of the catalyst and less consumable chemicals. Currently, explorations into low-cost, effective photocatalysts for organic contaminated water are being developed. Within this scheme, an easily separated photocatalyst with other functionality, such as high adsorption, is important. In this research, preparation of a magnetic nanocomposite photocatalyst based on agricultural waste, palm leaves biochar impregnated nickel nanoparticles (Ni/BC), was investigated. The nanocomposite was prepared by direct pyrolysis of palm leaves impregnated with nickel (II) chloride precursor. Furthermore, the physicochemical characterization of the material was performed by using an X-ray diffractometer (XRD), scanning electron microscopy-energy dispersive X-ray fluorescence (SEM-EDX), transmission electron microscopy (TEM), gas sorption analysis, X-ray photoelectron spectroscopy (XPS) and vibrating sample magnetometer (VSM). The photocatalytic activity of Ni/BC was evaluated for methyl violet (MV) photocatalytic oxidation. The results from XRD, XPS and TEM analyses identified single nickel nanoparticles dispersed on the biochar structure ranging from 30–50 nm in size. The dispersed nickel nanoparticles increased the BET specific surface area of biochar from 3.92 m2/g to 74.12 m2/g oxidation. High photocatalytic activity of the Ni/BC was exhibited by complete MV removal in 30 min for the concentration ranging from 10–80 mg/L. In addition, the Ni/BC showed stability in the pH range of 4–10 and reusability without any activity change until fifth usage. The separable photocatalyst is related to magnetism of about 13.7 emu/g. Hesperetin is the aglycone of citrus flavonoid hesperidin. Due to the limited information regarding hesperetin antimicrobial potential and emerging need for novel antimicrobials, we have studied its antimicrobial activity (microdilution assay), antibiofilm activity with different assays in two models (mono- and polymicrobial biofilm), and toxicity (MTT and brine shrimp lethality assays). Hesperetin inhibited growth of all Candida isolates (minimal inhibitory concentration, MIC, 0.165 mg/mL), while it's inhibitory potential towards Staphylococcus aureus was lower (MIC 4 mg/mL). Hesperetin (0.165 mg/mL) reduced ability of Candida to form biofilms and moderately reduced exopolysaccharide levels in biofilm matrix. Effect on the eradication of 24 h old C. albicans biofilms was promising at 1.320 mg/mL. Inhibition of staphylococcal biofilm formation required higher concentrations of hesperetin (<50% inhibition with MIC 4 mg/mL). Establishment of polymicrobial C. albicans-S. aureus biofilm was significantly inhibited with the lowest examined hesperetin concentration (1 mg/mL) in crystal violet and CFU assays. Hesperetin toxicity was examined towards MRC-5 fibroblasts (IC50 0.340 mg/mL) and in brine shrimp lethality assay (LC50 > 1 mg/mL). Hesperetin is efficient in combating growth and biofilm formation of Candida species. The discharge of large amounts of effluents contaminated with gentian violet (GV) and phenol red (PR) threatens aquatic flora and fauna as well as human health, which is why these effluents must be treated before being discarded. This study seeks the removal of dyes, using water lily (Eichhornia crassipes) as an adsorbent with different pretreatments. PR and GV were analyzed by a UV-visible spectrophotometer. Equilibrium experimental data showed that Freundlich is the best model to fit PR and SIPS for GV, showing that the adsorption process for both dyes was heterogeneous, favorable, chemical (for GV), and physical (for PR). The thermodynamic analysis for the adsorption process of both dyes depends directly on the increase in temperature and is carried out spontaneously. The Pseudo first Order (PFO) kinetic model for GV and PR is the best fit for the dyes having an adsorption capacity of 91 and 198 mg/g, respectively. The characterization of the materials demonstrated significant changes in the bands of lignin, cellulose, and hemicellulose, which indicates that the functional groups could participate in the capture of the dyes together with the electrostatic forces of the medium, from which it be concluded that the adsorption process is carried out by several mechanisms. The focus of this work was on developing a green, low-cost, and efficient biosorbent based on the biological structure and properties of MT and applying it to the remediation of cationic dyes in dye wastewater. The adsorption performance and mechanism of MT on methylene blue (MB) and crystal violet (CV) were investigated by batch adsorption experiments. The results demonstrated that the highest adsorption values of MT for MB (411 mg/g) and CV (553 mg/g) were greatly higher than the reported values of other biosorbents. In addition, the adsorption behaviors of methylene blue (MB) and crystal violet (CV) by MT were spontaneous exothermic reactions and closely followed the pseudo-second-order (PSO) kinetics and Langmuir isotherm. Further, the depleted MT was regenerated using pyrolysis mode to convert depleted MT into MT-biochar (MBC). The maximum adsorption of Cu2+ and Pb2+ by MBC was up to 320 mg/g and 840 mg/g, respectively. In this research, a new biodegradable and eco-friendly adsorbent, starch-grafted polymethyl methacrylate (St-g-PMMA) was synthesized. The St-g-PMMA was synthesized by a free radical polymerization reaction in which methyl methacrylate (MMA) was grafted onto a starch polymer chain. The reaction was performed in water in the presence of a potassium persulfate (KPS) initiator. The structure and different properties of the St-g-PMMA was explored by FT-IR, 1H NMR, TGA, SEM and XRD. After characterization, the St-g-PMMA was used for the removal of MB dye. Different adsorption parameters, such as effect of adsorbent dose, effect of pH, effect of initial concentration of dye solution, effect of contact time and comparative adsorption study were investigated. The St-g-PMMA showed a maximum removal percentage (R%) of 97% towards MB. The other parameters, such as the isothermal and kinetic models, were fitted to the experimental data. The results showed that the Langmuir adsorption and pseudo second order kinetic models were best fitted to experimental data with a regression coefficient of R2 = 0.93 and 0.99, respectively. Glioblastoma multiforme (GBM) is considered to be one of the most serious version of primary malignant tumors. Temozolomide (TMZ), an anti-cancer drug, is the most common chemotherapeutic agent used for patients suffering from GBM. However, due to its inherent instability, short biological half-life, and dose-limiting characteristics, alternatives to TMZ have been sought. In this study, the TMZ-loaded PLGA nanoparticles were prepared by employing the emulsion solvent evaporation technique. The prepared TMZ-PLGA-NPs were characterized using FT-IR, zeta potential analyses, XRD pattern, particle size estimation, TEM, and FE-SEM observations. The virotherapy, being safe, selective, and effective in combating cancer, was employed, and TMZ-PLGA-NPs and oncolytic Newcastle Disease Virus (NDV) were co-administered for the purpose. An AMHA1-attenuated strain of NDV was propagated in chicken embryos, and the virus was titrated in Vero-slammed cells to determine the infective dose. The in vitro cytotoxic effects of the TMZ, NDV, and the TMZ-PLGA-NPs against the human glioblastoma cancer cell line, AMGM5, and the normal cell line of rat embryo fibroblasts (REFs) were evaluated. The synergistic effects of the nano-formulation and viral strain combined therapy was observed on the cell lines in MTT viability assays, together with the Chou–Talalay tests. The outcomes of the in vitro investigation revealed that the drug combinations of NDV and TMZ, as well as NDV and TMZ-PLGA-NPs exerted the synergistic enhancements of the antitumor activity on the AMGM5 cell lines. The effectiveness of both the mono, and combined treatments on the capability of AMGM5 cells to form colonies were also examined with crystal violet dyeing tests. Quorum sensing (QS) plays an important role in the production of virulence factors and pathogenicity in pathogenic bacteria and is, therefore, a hopeful target to fight against bacterial infections. During our search for natural QS inhibitors, two new xanthonolignoids (1 and 2), each existing as a racemic mixture, one new simple oxygenated xanthone (7), and eight known analogs (3–6, 8–11) were isolated from Hypericum scabrum Linn. Chiral separation of 1 yielded a pair of enantiomers 1a and 1b. The structures of these compounds were elucidated by spectroscopic analysis and ECD (electrostatic circular dichroism) calculations. All isolates were evaluated for their QS inhibitory activity against Chromobacterium violaceum. Both 9 and 10 exhibited the most potent QS inhibitory effects with minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values of 31.25 and 62.5 μM, respectively. Crystal violet staining was used to further evaluate the biofilm inhibition potential of compounds 7, 9 and 10, and the formation of biofilms increased with decreasing drug concentration in a classic dose-dependent manner. In this study, a polysaccharide-based hydrogel microsphere (SFP/SA) was prepared using S. fusiforme polysaccharide (SFP) and sodium alginate (SA). Fourier transform infrared spectroscopy (FT-IR) demonstrated that SFP was effectively loaded onto the hydrogel microsphere. Texture profile analysis (TPA) and differential scanning calorimetry (DSC) showed that, with the increase of SFP concentration, the hardness of SFP/SA decreased, while the springiness and cohesiveness of SFP/SA increased, and the thermal stability of SFP/SA improved. The equilibrium adsorption capacity of SFP/SA increased from 8.20 mg/g (without SFP) to 67.95 mg/g (SFP accounted 80%) without swelling, and from 35.05 mg/g (without SFP) to 81.98 mg/g (SFP accounted 80%) after 24 h swelling. The adsorption of crystal violet (CV) dye by SFP/SA followed pseudo-first order and pseudo-second order kinetics (both with R2 > 0.99). The diffusion of intraparticle in CV dye was not the only influencing factor. Moreover, the adsorption of CV dye for SFP/SA (SFP accounted 60%) fit the Langmuir and Temkin isotherm models. SFP/SA exhibited good regenerative adsorption capacity. Its adsorption rate remained at > 97% at the 10th consecutive cycle while SFP accounted for 80%. The results showed that the addition of Sargassum fusiforme polysaccharide could increase the springiness, cohesiveness and thermal stability of the hydrogel microsphere, as well as improve the adsorption capacity of crystal violet dye. In response to the need for novel therapeutic strategies to combat the development of microbial resistance, plant essential oils may represent a promising alternative source. This study set out to characterize the chemical composition and assess the antibacterial potential of Myriactis nepalensis Less. essential oil (MNEO). Essential oil isolated from M. nepalensis by hydrodistillation was analyzed using a GC–MS technique. The antibacterial properties of MNEO alone and combined with antibiotics (chloramphenicol and streptomycin) were tested via the disc diffusion, microbroth dilution, and checkerboard methods. MNEO was represented by oxygenated sesquiterpenes (60.3%) and sesquiterpene hydrocarbons (28.6%), with caryophyllene oxide, spathulenol, humulene epoxide II, β-elemene, neointermedeol, and β-caryophyllene as the main compounds. MNEO exhibited a strong antibacterial effect against Gram-positive bacteria, with MIC and MBC values of 0.039 mg/mL and 0.039–0.156 mg/mL, respectively, and synergistic effects were observed in both combinations with chloramphenicol and streptomycin. Furthermore, the antibiofilm and cytotoxic activities of MNEO were also evaluated. The crystal violet assay was used for quantification of Staphylococcus aureus biofilm formation, and an MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay was conducted to determine cell viability. The results revealed MNEO could dose-dependently inhibit Staphylococcus aureus biofilm formation and possessed potential cytotoxic on both normal and cancer cells (IC50 values from 13.13 ± 1.90 to 35.22 ± 8.36 μg/mL). The purpose of this study was to evaluate the potential of a newly modified cyclodextrin derivative, water-soluble β-cyclodextrin–epichlorohydrin (β-CD), as an effective drug carrier to enhance the poor solubility and bioavailability of galangin (GAL), a poorly water-soluble model drug. In this regard, inclusion complexes of GAL/β-CDP were prepared. UV-VIS spectrophotometry, Fourier-transform infrared spectroscopy (FTIR), X-ray crystallography (XRD), zeta potential analysis, particle size analysis, field emission scanning electron microscopy (FESEM), and transmission electron microscopy (TEM) were applied to characterize the synthesized GAL/β-CD. Michigan Cancer Foundation-7 (MCF-7; human breast cancer cells) and rat embryo fibroblast (REF; normal cells) were employed to examine the in vitro cytotoxic effects of GAL/β-CD using various parameters. The dye-based tests of MTT and crystal violet clearly exhibited that GAL/β-CD-treated cells had a reduced proliferation rate, an influence that was not found in the normal cell line. The cells' death was found to follow apoptotic mechanisms, as revealed by the dye-based test of acridine orange/ethidium bromide (AO/EtBr), with the involvement of the mitochondria via caspase-3-mediated events, as manifested by the Rh 123 test. We also included a mouse model to examine possible in vivo toxic effects of GAL/β-CD. It appears that the inclusion complex does not have a significant influence on normal cells, as indicated by serum levels of kidney and liver enzymatic markers, as well as thymic and splenic mass indices. Listeria monocytogenes is a major foodborne pathogen that can cause listeriosis in humans and animals. Andrographolide is known as a natural antibiotic and exhibits good antibacterial activity. We aimed to investigate the effect of andrographolide on two quorum-sensing (QS) systems, LuxS/AI-2 and Agr/AIP of L. monocytogenes, as well as QS-controlled phenotypes in this study. Our results showed that neither luxS expression nor AI-2 production was affected by andrographolide. Nevertheless, andrographolide significantly reduced the expression levels of the agr genes and the activity of the agr promoter P2. Results from the crystal violet staining method, confocal laser scanning microscopy (CLSM), and field emission scanning electron microscopy (FE-SEM) demonstrated that andrographolide remarkably inhibited the biofilm-forming ability of L. monocytogenes 10403S. The preformed biofilms were eradicated when exposed to andrographolide, and reduced surviving cells were also observed in treated biofilms. L. monocytogenes treated with andrographolide exhibited decreased ability to secrete LLO and adhere to and invade Caco-2 cells. Several diseases, including atherosclerosis, are characterized by inflammation, which is initiated by leukocyte migration to the inflamed lesion. Hence, genes implicated in the early stages of inflammation are potential therapeutic targets to effectively reduce atherogenesis. Algal-derived polysaccharides are one of the most promising sources for pharmaceutical application, although their mechanism of action is still poorly understood. The present study uses a computational method to anticipate the effect of fucoidan and alginate on interactions with adhesion molecules and chemokine, followed by an assessment of the cytotoxicity of the best-predicted bioactive compound for human monocytic THP-1 macrophages by lactate dehydrogenase and crystal violet assay. Moreover, an in vitro pharmacodynamics evaluation was performed. Molecular docking results indicate that fucoidan has a greater affinity for L-and E-selectin, monocyte chemoattractant protein 1 (MCP-1), and intercellular adhesion molecule-1 (ICAM-1) as compared to alginate. Interestingly, there was no fucoidan cytotoxicity on THP-1 macrophages, even at 200 µg/mL for 24 h. The strong interaction between fucoidan and L-selectin in silico explained its ability to inhibit the THP-1 monocytes migration in vitro. MCP-1 and ICAM-1 expression levels in THP-1 macrophages treated with 50 µg/mL fucoidan for 24 h, followed by induction by IFN-γ, were shown to be significantly suppressed as eight- and four-fold changes, respectively, relative to cells treated only with IFN-γ. These results indicate that the electrostatic interaction of fucoidan improves its binding affinity to inflammatory markers in silico and reduces their expression in THP-1 cells in vitro, thus making fucoidan a good candidate to prevent inflammation. The presence of cationic dyes, even in a tiny amount, is harmful to aquatic life and pollutes the environment. Therefore, it is essential to remove these hazardous dyes to protect the life of marine creatures from these pollutants. In this research, crystal violet (CV) dye elimination was performed using a lignin copper ferrite (LCF) adsorbent. The adsorbent was synthesized and characterized using FTIR, Raman, SEM, EDX with mapping, and VSM, which proved the successful formation of magnetic LCF. Adsorption experiments were performed using different effective parameters. The highest adsorption potential (97%) was executed at mild operating conditions, with a 5 min contact time at room temperature and pH 8. The adsorption kinetic study utilized four kinetic models: first-order, second-order, intraparticle diffusion, and Elovich. The results revealed that the adsorption process complies with the pseudo-first-order with a maximum adsorption capacity of 34.129 mg/g, proving that the adsorption process mechanism is a physical adsorption process. Three isotherm models, Langmuir, Freundlich, and Temkin, were examined. The adsorption mechanism of CV onto LCF was also followed by the Langmuir and Freundlich models. The thermodynamic parameters were examined and revealed that the adsorption onto LCF was an exothermic process. It was proposed that the adsorption process is a spontaneous exothermic process. Acetylcholinesterase (AChE) is an essential enzyme that terminates cholinergic transmission by a rapid hydrolysis of the neurotransmitter acetylcholine. AChE is an important target for treatment of various cholinergic deficiencies, including Alzheimer's disease and myasthenia gravis. In a previous high throughput screening campaign, we identified the dye crystal violet (CV) as an inhibitor of AChE. Herein, we show that CV displays a significant cooperativity for binding to AChE, and the molecular basis for this observation has been investigated by X-ray crystallography. Two monomers of CV bind to residues at the entrance of the active site gorge of the enzyme. Notably, the two CV molecules have extensive intermolecular contacts with each other and with AChE. Computational analyses show that the observed CV dimer is not stable in solution, suggesting the sequential binding of two monomers. Guided by the structural analysis, we designed a set of single site substitutions, and investigated their effect on the binding of CV. Only moderate effects on the binding and the cooperativity were observed, suggesting a robustness in the interaction between CV and AChE. Taken together, we propose that the dimeric cooperative binding is due to a rare combination of chemical and structural properties of both CV and the AChE molecule itself. [ABSTRACT FROM AUTHOR]

Screening of anti-biofilm compounds from the burdock leaf based on metabolomics is reported here. The crystal violet assay indicated 34% ethanol elution fraction of burdock leaf could completely inhibit biofilm formation of Pseudomonas aeruginosa at 1 mg·mL-1. Then, the chemical composition of burdock leaf fraction was analyzed by ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) and 11 active compounds (chlorogenic acid, caffeic acid, p-coumaric acid, quercetin, ursolic acid, rutin, cynarin, luteolin, crocin, benzoic acid, and Tenacissoside I) were identified. Lastly, UPLC-MS analysis was employed to obtain the metabolic fingerprints of burdock leaf fractions before and after inhibiting the biofilm of Pseudomonas aeruginosa. The metabolic fingerprints were transformed to data, analyzed with PLS-DA (partial least squares discriminant analysis) and the peaks whose area was significantly changed were found out. Thus, 81 compounds were screened as potential anti-biofilm ingredients. Among them, rutin, ursolic acid, caffeic acid, p-coumaric acid and quercetin were identified and confirmed as the main anti-biofilm compounds in burdock leaf. The study provided basic anti-biofilm profile data for the compounds in burdock leaf, as well as provided a convenient method for fast screening of anti-biofilm compounds from natural plants.
