Ectomycorrhizal fungi are known to synthesize antifungal compounds both in vitro and in symbiosis with the host-plants. Culture filtrates of the ectomycorrhizal fungus Suillus bovinus (at pHs of 2.5-6) showed antifungal activity towards saprotrophs Trichoderma harzianum, and Trichoderma virens and the pathogen Heterobasidion annosum, by significantly suppressing their growth relative to sterile liquid medium at the same pHs. In the presence of the culture filtrates, hyphae of the saprotrophs and the pathogen were characterized by distensions, irregular and frequent branching, tip damage and cytoplasm coagulation. Since hyphal abnormalities may be evoked by disruptions in the cytoskeleton and mitochondria, their structural changes were also examined. Depolymerization of microtubules was confirmed for all of the fungi. Serious damage to mitochondria morphology may cause significant functional impairment. Growth of mycelia was inhibited in the lower pH S. bovinus culture filtrate, and the mitochondrial morphology was altered. This suggests that the activity of antifungal compounds synthesized by ectomycorrhizal fungus is significantly affected by pH.

Harpellales (Legeriomycetaceae, Zygomycota) or 'trichomycetes' are fungi that inhabit the digestive tracts of arthropods such as insects, millipedes, and crustaceans. In the current study we examined changes in 5 morphological characters of Smittium culisetae (Harpellales: Legeriomycetaceae) between the two dipteran (mosquito, black fly) hosts reared under 3 different temperatures (17, 22, 30 degrees C). Both host and temperature had a pervasive effect on the linear dimension of trichospores, their generative cells and hyphae width. At 30 degrees C the mean size of all 5 morphological characters were consistently larger in fungus taken from the mosquito host than from the black fly host. At 17 degrees C and 22 degrees C, however, there were no consistent patterns. The effect of host was so pronounced that it could be accurately determined which host S. culisetae colonised based on differences in linear morphology. Such changes in fungal morphology between hosts have important ramifications for the morphologically based taxonomy of this group.

Root endophytic fungi are seen as promising alternatives to replace chemical fertilizers and pesticides in sustainable and organic agriculture systems. Fungal endophytes structure formations play key roles in symbiotic intracellular association with plant-roots. To compare the morphologies of Ascomycete endophytic fungi in wheat, we analyzed growth morphologies during endophytic development of hyphae within the cortex of living vs. dead root cells. Confocal laser scanning microscopy (CLSM) was used to characterize fungal cell morphology within lactofuchsin-stained roots. Cell form regularity Ireg and cell growth direction Idir, indexes were used to quantify changes in fungal morphology. Endophyte fungi in living roots had a variable Ireg and Idir values, low colonization abundance and patchy colonization patterns, whereas the same endophyte species in dead (gamma-irradiated) roots had consistent form of cells and mostly grew parallel to the root axis. Knot, coil and vesicle structures dominated in living roots, as putative symbiotic functional organs. Finally, an increased hypha septation in living roots might indicate local specialization within endophytic Ascomycota. Our results suggested that the applied method could be expanded to other septate fungal symbionts (e.g. Basidiomycota). The latter is discussed in light of our results and other recent discoveries.

Growth and organelle morphology in the wood rotting basidiomycete fungus Phanerochaete velutina were examined in Petri dishes, on agar-coated slides, and in submerged cultures, using DIC, fluorescence and four-dimensional (4-D; x,y,z,t) confocal microscopy, with several fluorescent probes. Phanerochaete is ideal for this work because of its fast growth, robustness, and use in a wide range of other studies. The probe carboxy-DFFDA, widely used for labelling vacuoles, has no effect either on hyphal tip extension or colony growth at the concentrations usually applied in labelling experiments. Carboxy-DFFDA labels the vacuoles and these form a tubular reticulum in hyphal tip cells. The probe also labels extremely small vesicles (punctate fluorescence) in the apex of tip cells, the Spitzenkörper, and short tubules that undergo sequences of characteristic movements and transformations to produce various morphologies, including ring-like structures. Their location and behaviour suggest that they are a distinct group of structures, possibly a subset of vacuoles, but as yet to be fully identified. Regular incursions of tubules extending from these structures and from the vacuolar reticulum into the apical dome indicate the potential for delivery of material to the apex via tubules as well as vesicles. Such structures are potential candidates for delivering chitin synthases to the apex. Spitzenkörper behaviour has been followed as hyphal tips with linear growth encounter obstacle hyphae and, as the hydrolysis product of carboxy-DFFDA only accumulates in membrane-enclosed compartments, it can be inferred that the labelled structures represent the Spitzenkörper vesicle cloud. Mitochondria also form a reticular continuum of branched tubules in growing hyphal tips, and dual localisation with DiOC6(3) and CMAC allows this to be distinguished from the vacuolar reticulum. Like vacuolar tubules, mitochondrial tubules also span the septa, indicating that they may also be a conduit for intercellular transport.

Approximately 50 single-basidiospore isolates (SBIs) obtained from each of 16 field isolates of Thanatephorus cucumeris AG-1 IC were examined for heterokaryon formation. All SBIs obtained from each field isolate were divided into two mating groups (SBIs-M1 and SBIs-M2), and tufts of mycelia were formed in the contact zone between colonies of paired SBIs-M1 and -M2 based on 0.5 % charcoal agar medium. Tufts were produced from all possible pairing between SBIs from non-parental field isolates. Hyphal anastomosis reactions indicated no cell death and random cell death at the contact cell, and was not related to tuft formation. AFLP phenotypes of SBIs from each field isolate were not identical to each other and were different from their parental field isolate. AFLP phenotypes of the tuft isolates formed from SBIs-M1 and SBIs-M2 from each field isolate were heterokaryotic. Moreover, several SBIs also formed tufts with their parental and non-parental field isolates. AFLP phenotypes of these tuft isolates suggested that they were all heterokaryotic. Results of these experiments suggest that T. cucumeris AG-1 IC is heterothallic and bipolar, and that genetic exchange can occur between homokaryotic and heterokaryotic isolates (Buller phenomenon).

Coniolariella hispanica, isolated as an endophyte from leaves of Eryngium campestre on the Iberian Peninsula, is established as a new cleistocarpous species. The perithecial species Rosellinia limoniispora is transferred to Coniolariella on morphological and molecular evidence. R. australis is transferred to Coniolariella as a variety of C. limoniispora. The type species of Coniolariella, C. gamsii, is considered to be a variety of C. limoniispora. The taxa discussed here have soft stromata, deliquescent asci that lack an iodine-positive apical ring, and more or less symmetrical ascospores. They lack subicula in natural conditions. C. hispanica and C. limoniispora var. limoniispora have Sporothrix- or Geniculosporium-like anamorphs associated with stromata at different stages of development, but anamorphs are not usually evident in cultures. Coniolariella limoniispora var. gamsii produces the anamorph abundantly in culture. It is not known whether or not C. limoniispora var. australis possesses an anamorph. An emendation of the description of Coniolariella is necessitated by these changes.

Fomitiporia punicata sp. nov., collected from Shaanxi Province, northern China, and so far exclusively on Punica granatum (Punicaceae), is described and illustrated, and its preliminary phylogenetic relationships are inferred based on sequence data from the ribosomal ITS-5.8S regions. Fomitiporia punicata belongs to the F. robusta-F. punctata complex, but is characterized by an effused-reflexed to pileate basidioma with triquetrous to ungulate pilei, pale yellowish brown to cinnamon brown pore surface, moderately thick-walled skeletal hyphae in the context, which are partly collapsed on drying, lack of setae, presence of fusoid to subulate cystidioles, and growth on Punica. It also forms a distinct clade within Fomitiporia, and is related to some species from the temperate or Mediterranean zone of the northern hemisphere. An identification key to the species of Fomitiporia recorded in China is provided. Fomitiporia tibetica is transferred to Pseudoinonotus.

The closely related genera Carestiella, Schizoxylon and Stictis (Stictidaceae, Ostropales, Ascomycota) are revised in northern Scandinavia, and a revised phylogenetic hypothesis, including all seven species of this group in the area, is presented. A key to the species of Stictis s. lat. (including Carestiella, Schizoxylon and Stictis) in Northern Scandinavia is presented. S. albescens and S. confusum are described as new to science, and the new name S. brunnescens is introduced for S. mollis ssp. populorum, when treated as a distinct species. C. socia and S. albescens are the only representatives of their genera in northern Scandinavia, but a total of five species of Stictis are present in the area (S. brunnescens, S. confusum, S. mollis, S. populorum and S. radiata). Substantial within-species variation in ascoma morphology and wall pigmentation is revealed and discussed, and found to correlate with fungal lifestyles; optional lichenization apparently affects several details in fungal anatomy and morphology.

Cord-forming basidiomycetes are important decomposers of dead wood in forest ecosystems but the impact of mycophagous soil invertebrates on their mycelia are little known. Here we investigate the effects of different grazing intensities of Collembola (Folsomia candida) on mycelial foraging patterns of the saprotrophic cord-forming basidiomycetes Hypholoma fasciculare, Phanerochaete velutina and Resinicium bicolor growing from beech (Fagus sylvatica) wood block inocula in dishes of non-sterile soil. Mycelial extension rate and hyphal coverage decreased with increased grazing intensity. R. bicolor was most affected, high grazing density resulting in only a few major cords remaining. Grazing of H. fasciculare often resulted in points of more rapid outgrowth as cords with a fanned margin. In grazed mycelia of P. velutina the main cords had fanned tips and lateral cords became branched. These results suggest that mycophagy by Collembola may hinder the growth of cord-forming fungi in woodlands, which might impact on the ability of these fungi to forage for and decompose dead organic material.

The Spitzenkörper is a dynamic structure present at the tips of hyphal cells with a single highly polarized growth site. It is closely connected with cell morphogenesis and polar growth, and is only present at actively growing sites. Morphogenesis of such highly polarized cells is complex, and requires the coordinated action of multiple protein complexes. We discuss the relevance of these complexes for the structure and function of the Spitzenkörper.

Although Plasmopara viticola causes grape downy mildew in most, if not all, wine producing countries, many basic biological or chemical aspects are still unknown and thus the histopathological changes during development of this oomycete pathogen were studied. The fluorochromes Aniline blue and Uvitex 2B were successfully used in whole leaf stainings to visualise intercellular hyphae and to investigate septal-development in sporangiophores. The occurrence and transfer of cytoplasm in sporangiophores was studied with the aid of Chlorazole Black E and Phloxin-B. Application of Chlorazole Black E was the most reliable method to differentiate between cytoplasm and septa in sporangiophores due to a high contrast between the dark cytoplasm and the pale septa. In P. viticola, septa were found in the stem and branches of the sporangiophores, but not in the intercellular hyphae, which was in contrast to other oomycetes, such as P. tabacina, Pseudoperonospora cubensis and P. humuli, where septa were frequently found in the mycelium. In order to verify chemical composition of septa, sporangiophores were digested using chitinase and beta-1,3-glucanase. After staining with Aniline blue and Uvitex 2B, the intense fluorescence of septa was conserved after the application of chitinase but not after beta-1,3-glucanase, indicating that septa are mainly composed of beta-1,3-glucans. Pretreatments of infected vine leaves with 2-deoxy-D-glucose (2-DOG) at low concentrations (1-5 mM) led to a disorganisation of the sporangiophore structure of P. viticola, whereas the production of septa was unaffected. Application of 2-DOG at higher concentrations resulted in a reduced length of intercellular hyphae (10 mM) or total inhibition (50 mM) of the intercellular mycelium. Thus, 2 DOG or its analogues interferes with morphogenesis in Plasmopara viticola and may have a deleterious effect on the spread of this downy mildew between plants.

Dikaryotic and haploid mycelia of more than 150 gilled species of euagarics were studied morphologically and by molecular phylogenetic methods. The morphological investigations revealed anamorphs in more than 90 species that were often specific at the genus or family level. Thallic conidiogenesis dominated and varied from fragmentation of normally branched hyphae to the formation of differentiated sympodially branched conidiophores. Secession modes, coiling of the conidiogenous hyphae or the swelling of the conidia were additional distinguishing features. Phylogenetic analysis of the D1-D3 domains of the nuclear gene for the ribosomal large subunit using a Bayesian Markov chain Monte Carlo approach resulted in several well-supported groups that are consistent with anamorph morphology. These results indicate that the anamorphs provide valuable characters for a natural classification of the Agaricales.

Colletotrichum lindemuthianum is a pathogen of the common bean plant (Phaseolus vulgaris) causing anthracnose. Large numbers of isolates can rapidly arise with different genetic and chromosomal compositions but their origin is unknown since sexual fruit bodies have only been found in the laboratory. We have recently described the occurrence of special kinds of hyphae that create anastomoses directly between conidia. In this work we show that conidial anastomoses can occur between two different Colletotrichum species. The implications of this observation on the generation of genetic diversity in these species are discussed.
