Abstract

Measurements of the atmospheric carbon (C) and oxygen (O) relative to hydrogen (H) in hot Jupiters (relative to their host stars) provide insight into their formation location and subsequent orbital migration1,2. Hot Jupiters that form beyond the major volatile (H2O/CO/CO2) ice lines and subsequently migrate post disk-dissipation are predicted have atmospheric carbon-to-oxygen ratios (C/O) near 1 and subsolar metallicities2, whereas planets that migrate through the disk before dissipation are predicted to be heavily polluted by infalling O-rich icy planetesimals, resulting in C/O

Abstract

The clinical development of an inhibitor of cellular proteasome function suggests that compounds targeting other components of the ubiquitin-proteasome system might prove useful for the treatment of human malignancies. NEDD8-activating enzyme (NAE) is an essential component of the NEDD8 conjugation pathway that controls the activity of the cullin-RING subtype of ubiquitin ligases, thereby regulating the turnover of a subset of proteins upstream of the proteasome. Substrates of cullin-RING ligases have important roles in cellular processes associated with cancer cell growth and survival pathways. Here we describe MLN4924, a potent and selective inhibitor of NAE. MLN4924 disrupts cullin-RING ligase-mediated protein turnover leading to apoptotic death in human tumour cells by a new mechanism of action, the deregulation of S-phase DNA synthesis. MLN4924 suppressed the growth of human tumour xenografts in mice at compound exposures that were well tolerated. Abstract

The role of genomic imprinting in the development of embryonal tumors was investigated. Imprinted genes are implicated in the development in mice of Wilms' tumor and Beckman-Wiedemann syndrome, but the exact underlying mechanism is not well understood. The study protocol involved the study of the expression of insulin-like growth factor II (IGF2) gene which is highly expressed in Wilms' tumors. The results showed that IGF2 is expressed in fetal tissues from the paternal allelle, but is expressed biallelically in Wilms' tumor. This indicates that relaxation of imprinting could be one mechanism in carcinogenesis.

Abstract

The National Vibrating Probe Facility of the National Institutes of Health developed the non-selective vibrating voltage and ion-selective probe to provide physiologists a tool that can directly observe the flux of a single ion species across the plasma membrane of a cell. These probes use ionophores, which are liphophilic complexing agents that selectively transports ions across a membrane, to eliminate difficulties caused by drifts and differential signals. They are now also used to measure flux of other ions such as hydrogen, potassium, calcium and chloride.
