Water availability plays a critical role in shaping terrestrial ecosystems, particularly in low- and mid-latitude regions. The sensitivity of vegetation growth to precipitation strongly regulates global vegetation dynamics and their responses to drought, yet sensitivity changes in response to climate change remain poorly understood. Here we use long-term satellite observations combined with a dynamic statistical learning approach to examine changes in the sensitivity of vegetation greenness to precipitation over the past four decades. We observe a robust increase in precipitation sensitivity (0.624% yr-1) for drylands, and a decrease (-0.618% yr-1) for wet regions. Using model simulations, we show that the contrasting trends between dry and wet regions are caused by elevated atmospheric CO2 (eCO2). eCO2 universally decreases the precipitation sensitivity by reducing leaf-level transpiration, particularly in wet regions. However, in drylands, this leaf-level transpiration reduction is overridden at the canopy scale by a large proportional increase in leaf area. The increased sensitivity for global drylands implies a potential decrease in ecosystem stability and greater impacts of droughts in these vulnerable ecosystems under continued global change.

Abstract Interferon-gamma (IFN-γ) signaling is necessary for the proinflammatory activation of macrophages but IFN-γ-independent pathways, for which the initiating stimuli and downstream mechanisms are lesser known, also contribute. Here we identify, by high-content screening, SEPTIN2 (SEPT2) as a negative regulation of IFN-γ-independent macrophage autoactivation. Mechanistically, endoplasmic reticulum (ER) stress induces the expression of SEPT2, which balances the competition between acetylation and ubiquitination of heat shock protein 5 at position Lysine 327, thereby alleviating ER stress and constraining M1-like polarization and proinflammatory cytokine release. Disruption of this negative feedback regulation leads to the accumulation of unfolded proteins, resulting in accelerated M1-like polarization, excessive inflammation and tissue damage. Our study thus uncovers an IFN-γ-independent macrophage proinflammatory autoactivation pathway and suggests that SEPT2 may play a role in the prevention or resolution of inflammation during infection.

Global warming is expected to cause wet seasons to get wetter and dry seasons to get drier, which would have broad social and ecological implications. However, the extent to which this seasonal paradigm holds over land remains unclear. Here we examine seasonal changes in surface water availability (precipitation minus evaporation, P-E) from CMIP5 and CMIP6 projections. While the P-E seasonal cycle does broadly intensify over much of the land surface, ~20% of land area experiences a diminished seasonal cycle, mostly over subtropical regions and the Amazon. Using land-atmosphere coupling experiments, we demonstrate that 63% of the seasonality reduction is driven by seasonally varying soil moisture (SM) feedbacks on P-E. Declining SM reduces evapotranspiration and modulates circulation to enhance moisture convergence and increase P-E in the dry season but not in the wet season. Our results underscore the importance of SM-atmosphere feedbacks for seasonal water availability changes in a warmer climate.

Weather extremes have widespread harmful impacts on ecosystems and human communities with more deaths and economic losses from flash floods than any other severe weather-related hazards. Flash floods attributed to storm runoff extremes are projected to become more frequent and damaging globally due to a warming climate and anthropogenic changes, but previous studies have not examined the response of these storm runoff extremes to naturally and anthropogenically driven changes in surface temperature and atmospheric moisture content. Here we show that storm runoff extremes increase in most regions at rates higher than suggested by Clausius-Clapeyron scaling, which are systematically close to or exceed those of precipitation extremes over most regions of the globe, accompanied by large spatial and decadal variability. These results suggest that current projected response of storm runoff extremes to climate and anthropogenic changes may be underestimated, posing large threats for ecosystem and community resilience under future warming conditions.
