Magnetoencephalography (MEG) is a noninvasive imaging technique used in neuroscience and clinical research. The source estimation of MEG involves solving a highly underdetermined inverse problem, which requires additional constraints to restrict the solution space. Traditional methods tend to obscure the extent of the sources. However, an accurate estimation of the source extent is important for studying brain activity or preoperatively estimating pathogenic regions. To improve the estimation accuracy of the extended source extent, the spatial constraint of sources is employed in the Bayesian framework. For example, the source is decomposed into a linear combination of validated spatial basis functions, which is proved to improve the source imaging accuracy. In this work, we further construct the spatial properties of the source using the diagonal covariance bases (DCB), which we summarize as the source imaging method SI-DCB. In this approach, specifically, the covariance matrix of the spatial coefficients is modeled as a weighted combination of diagonal covariance basis functions. The convex analysis is used to estimate noise and model parameters under the Bayesian framework. Extensive numerical simulations showed that SI-DCB outperformed five benchmark methods in accurately estimating the location and extent of patch sources. The effectiveness of SI-DCB was verified through somatosensory stimulation experiments performed on a 31-channel OPM-MEG system. The SI-DCB correctly identified the source area where each brain response occurred. The superior performance of SI-DCB suggests that it can provide a template approach for improving the accuracy of source extent estimations under a sparse Bayesian framework.

Previous studies of vicarious touch suggest that we automatically simulate observed touch experiences in our own body representation including primary and secondary somatosensory cortex (SCx). However, whether these early sensory areas are activated in a reflexive manner and the extent with which such SCx activations represent touch qualities, like texture, remains unclear. We measured event-related potentials (ERPs) of SCx's hierarchical processing stages, which map onto successive somatosensory ERP components, to investigate the timing of vicarious touch effects. In the first experiment, participants (n = 43) merely observed touch or no-touch to a hand; in the second, participants saw different touch textures (soft foam and hard rubber) either touching a hand (other-directed) or they were instructed that the touch was self-directed and to feel the touch. Each touch sequence was followed by a go/no-go task. We probed SCx activity and isolated SCx vicarious touch activations from visual carry over effects. We found that vicarious touch conditions (touch versus no-touch and soft versus hard) did not modulate early sensory ERP components (i.e. P50, N80); but we found effects on behavioural responses to the subsequent go/no-go stimulus consistent with post-perceptual effects. When comparing other- with self-directed touch conditions, we found that early and mid-latency components (i.e. P50, N80, P100, N140) were modulated consistent with early SCx activations. Importantly, these early sensory activations were not modulated by touch texture. Therefore, SCx is purposely recruited when participants are instructed to attend to touch; but such activation only situates, rather than fully simulates, the seen tactile experience in SCx.

Spaceflight has numerous untoward effects on human physiology. Various countermeasures are under investigation including artificial gravity (AG). Here, we investigated whether AG alters resting-state brain functional connectivity changes during head-down tilt bed rest (HDBR), a spaceflight analog. Participants underwent 60 days of HDBR. Two groups received daily AG administered either continuously (cAG) or intermittently (iAG). A control group received no AG. We assessed resting-state functional connectivity before, during, and after HDBR. We also measured balance and mobility changes from pre- to post-HDBR. We examined how functional connectivity changes throughout HDBR and whether AG is associated with differential effects. We found differential connectivity changes by group between posterior parietal cortex and multiple somatosensory regions. The control group exhibited increased functional connectivity between these regions throughout HDBR whereas the cAG group showed decreased functional connectivity. This finding suggests that AG alters somatosensory reweighting during HDBR. We also observed brain-behavioral correlations that differed significantly by group. Control group participants who showed increased connectivity between the putamen and somatosensory cortex exhibited greater mobility declines post-HDBR. For the cAG group, increased connectivity between these regions was associated with little to no mobility declines post-HDBR. This suggests that when somatosensory stimulation is provided via AG, functional connectivity increases between the putamen and somatosensory cortex are compensatory in nature, resulting in reduced mobility declines. Given these findings, AG may be an effective countermeasure for the reduced somatosensory stimulation that occurs in both microgravity and HDBR.

Proprioception is the sense of body position and movement that relies on afference from the proprioceptors in muscles and joints. Proprioceptive responses in the primary sensorimotor (SM1) cortex can be elicited by stimulating the proprioceptors using evoked (passive) limb movements. In magnetoencephalography (MEG), proprioceptive processing can be quantified by recording the movement evoked fields (MEFs) and movement-induced beta power modulations or by computing corticokinematic coherence (CKC) between the limb kinematics and cortical activity. We examined whether cortical proprioceptive processing quantified with MEF peak strength, relative beta suppression and rebound power and CKC strength is affected by the movement range of the finger.MEG activity was measured from 16 right-handed healthy volunteers while movements were applied to their right-index finger metacarpophalangeal joint with an actuator. Movements were either intermittent, every 3000 ± 250 ms, to estimate MEF or continuous, at 3 Hz, to estimate CKC. In both cases, 4 different ranges of motion of the stimuli were investigated: 15, 18, 22 and 26 mm for MEF and 6, 7, 9 and 13 mm for CKC. MEF amplitude, relative beta suppression and rebound as well as peak CKC strength at the movement frequency were compared between the movement ranges in the source space. Inter-individual variation was also compared between the MEF and CKC strengths.As expected, MEF and CKC responses peaked at the contralateral SM1 cortex. MEF peak, beta suppression and rebound and CKC strengths were similar across all movement ranges. Furthermore, CKC strength showed a lower degree of inter-individual variation compared with MEF strength.Our result of absent modulation by movement range in cortical responses to passive movements of the finger indicates that variability in movement range should not hinder comparability between different studies or participants. Furthermore, our data indicates that CKC is less prone to inter-individual variability than MEFs, and thus more advantageous in what pertains to statistical power.

The exact somatotopy of the human facial representation in the primary somatosensory cortex (S1) remains debated. One reason that progress has been hampered is due to the methodological challenge of how to apply automated vibrotactile stimuli to face areas in a manner that is: (1) reliable despite differences in the curvatures of face locations; and (2) MR-compatible and free of MR-interference artefacts when applied in the MR head-coil. Here we overcome this challenge by using soft pneumatic actuator (SPA) technology. SPAs are made of a soft silicon material and can be in- or deflated by means of airflow, have a small diameter, and are flexible in structure, enabling good skin contact even on curved body surfaces (as on the face). To validate our approach, we first mapped the well-characterised S1 finger layout using this novel device and confirmed that tactile stimulation of the fingers elicited characteristic somatotopic finger activations in S1. We then used the device to automatically and systematically deliver somatosensory stimulation to different face locations. We found that the forehead representation was least distant from the representation of the hand. Within the face representation, we found that the lip representation is most distant from the forehead representation, with the chin represented in between. Together, our results demonstrate that this novel MR compatible device produces robust and clear somatotopic representational patterns using vibrotactile stimulation through SPA-technology.

Identical sensory stimuli can lead to different neural responses depending on the instantaneous brain state. Specifically, neural excitability in sensory areas may shape the brain´s response already from earliest cortical processing onwards. However, whether these dynamics affect a given sensory domain as a whole or occur on a spatially local level is largely unknown. We studied this in the somatosensory domain of 38 human participants with EEG, presenting stimuli to the median and tibial nerves alternatingly, and testing the co-variation of initial cortical responses in hand and foot areas, as well as their relation to pre-stimulus oscillatory states. We found that amplitude fluctuations of initial cortical responses to hand and foot stimulation – the N20 and P40 components of the somatosensory evoked potential (SEP), respectively – were not related, indicating local excitability changes in primary sensory regions. In addition, effects of pre-stimulus alpha (8-13 Hz) and beta (18-23 Hz) band amplitude on hand-related responses showed a robust somatotopic organization, thus further strengthening the notion of local excitability fluctuations. However, for foot-related responses, the spatial specificity of pre-stimulus effects was less consistent across frequency bands, with beta appearing to be more foot-specific than alpha. Connectivity analyses in source space suggested this to be due to a somatosensory alpha rhythm that is primarily driven by activity in hand regions while beta frequencies may operate in a more hand-region-independent manner. Altogether, our findings suggest spatially distinct excitability dynamics within the primary somatosensory cortex, yet with the caveat that frequency-specific processes in one sub-region may not readily generalize to other sub-regions.

Intracortical mapping in monkeys revealed a full body map in all four cytoarchitectonic subdivisions of the contralateral primary somatosensory cortex (S1), as well as positive associations between spatio-tactile acuity performance of the fingers and their representation field size especially within cytoarchitectonic Area 3b and Area 1. Previous non-invasive investigations on these associations in humans assumed a monotonous decrease of representation field size from index finger to little finger although the field sizes are known to change in response to training or in disease. Recent developments improved noninvasive functional mapping of S1 by a) adding a cognitive task during repetitive stimulation to decrease habituation to the stimuli, b) smaller voxel size of fMRI-sequences, c) surface-based analysis accounting for cortical curvature, and d) increase of spatial specificity for fMRI data analysis by avoidance of smoothing, partial volume effects, and pial vein signals.We here applied repetitive pneumatic stimulation of digit 1 (D1; thumb) and digit 5 (D5; little finger) on both hands to investigate finger/hand representation maps in the complete S1, but also in cytoarchitectonic Areas 1, 2, 3a, and 3b separately, in 21 healthy volunteers using 3T fMRI. The distances between activation maxima of D1 and D5 were evaluated by two independent raters, blinded for performance parameters. The fingertip representations showed a somatotopy and were localized in the transition region between the crown and the anterior wall of the post central gyrus agreeing with Area 1 and 3b. Participants were comprehensively tested for tactile performance using von Freyhair filaments to determine cutaneous sensory thresholds (CST) as well as grating orientation thresholds (GOT) and two-point resolution (TPD) for spatio-tactile acuity testing. Motor performance was evaluated with pinch grip performance (Roeder test). We found bilateral associations of D1-D5 distance for GOT thresholds and partially also for TPD in Area 3b and in Area 1, but not if using the complete S1 mask. In conclusion, we here demonstrate that 3T fMRI is capable to map associations between spatio-tactile acuity and the fingertip representation in Area 3b and Area 1 in healthy participants.

Working memory (WM) representations are generally known to be influenced by task demands, but it is not clear whether this extends to the somatosensory domain. One way to investigate the influence of task demands is with categorization paradigms, wherein either a single stimulus or an associated category is maintained in WM. In the somatosensory modality, category representations have been identified in the premotor cortex (PMC) and the intraparietal sulcus (IPS). In this study we used multivariate-pattern-analysis with human fMRI data to investigate whether the WM representations in the PMC, IPS or other regions are influenced by changing task demands. We ensured the task-dependent, categorical WM information was decorrelated from stimulus features by (1) teaching participants arbitrary, non-rule based stimulus groupings and (2) contrasting identical pairs of stimuli across experimental conditions, where either a single stimulus or the associated group was maintained in WM. Importantly, we also decoupled the decision and motor output from the WM representations. With these experimental manipulations, we were able to pinpoint stimulus-specific WM information to the left frontal and parietal cortices and context-dependent, group-specific WM information to the left IPS. By showing that grouped stimuli are represented more similarly in the Group condition than in the Stimulus condition, free from stimulus and motor output confounds, we provide novel evidence for the adaptive nature of somatosensory WM representations in the IPS with changing task-demands.

Sensory gating (SG) is a neurophysiological phenomenon whereby the response to the second stimulus in a repetitive pair is attenuated. This filtering of irrelevant or redundant information is thought to preserve neural resources for more behaviorally-relevant stimuli and thereby reflect the functional inhibition of sensory input. Developing a SG paradigm in which optimal suppression of sensory input is achieved requires investigators to consider numerous parameters such as stimulus intensity, time between stimulus pairs, and the inter-stimulus interval (ISI) within each pair. While these factors have been well defined for the interrogation of auditory gating, the precise parameters for eliciting optimal gating in the somatosensory domain are far less understood. To address this, we investigated the impact of varying the ISI within each identical pair of stimuli on gating using magnetoencephalography (MEG). Specifically, 25 healthy young adults underwent paired-pulse electrical stimulation of the median nerve with increasing ISIs between 100 and 1000 ​ms (in 100 ​ms increments). Importantly, for correspondence with previous studies of somatosensory gating, both time-domain and oscillatory neural responses to somatosensory stimulation were evaluated. Our results indicated that gating of somatosensory input was optimal (i.e., best suppression) for trials with an ISI of 200–220 ​ms, as evidenced by the smallest gating ratios and through statistical modeling estimations of optimal suppression. Importantly, this was true irrespective of whether oscillatory or evoked neural activity was used to calculate SG. Interestingly, oscillatory metrics of gating calculated using peak gamma (30-75 ​Hz) power and frequency revealed more robust gating (i.e., smaller ratios) than those calculated using time-domain neural responses, suggesting that high frequency oscillations may provide a more sensitive measure of SG. These findings have important implications for the development of optimal protocols and analysis pipelines to interrogate SG and inhibitory processing with a higher degree of sensitivity and accuracy.

The cerebellum is known to contain a double somatotopic body representation. While the anterior lobe body map has shown a robust somatotopic organization in previous fMRI studies, the representations in the posterior lobe have been more difficult to observe and are less precisely characterized. In this study, participants went through a simple motor task asking them to move either the eyes (left-right guided saccades), tongue (left-right movement), thumbs, little fingers or toes (flexion). Using high spatial resolution fMRI data acquired at ultra-high field (7T), with special care taken to obtain sufficient B1 over the entire cerebellum and a cerebellar surface reconstruction facilitating visual inspection of the results, we were able to precisely map the somatotopic representations of these five distal body parts on both subject- and group-specific cerebellar surfaces. The anterior lobe (including lobule VI) showed a consistent and robust somatotopic gradient. Although less robust, the presence of such a gradient in the posterior lobe, from Crus II to lobule VIIIb, was also observed. Additionally, the eyes were also strongly represented in Crus I and the oculomotor vermis. Overall, crosstalk between the different body part representations was negligible. Taken together, these results show that multiple representations of distal body parts are present in the cerebellum, across many lobules, and they are organized in an orderly manner.

The "Dual-Core Beamformer" (DCBF) is a new lead-field based MEG inverse-modeling technique designed for localizing highly correlated networks from noisy MEG data. Conventional beamformer techniques are successful in localizing neuronal sources that are uncorrelated under poor signal-to-noise ratio (SNR) conditions. However, they fail to reconstruct multiple highly correlated sources. Though previously published dual-beamformer techniques can successfully localize multiple correlated sources, they are computationally expensive and impractical, requiring a priori information. The DCBF is able to automatically calculate optimal amplitude-weighting and dipole orientation for reconstruction, greatly reducing the computational cost of the dual-beamformer technique. Paired with a modified Powell algorithm, the DCBF can quickly identify multiple sets of correlated sources contributing to the MEG signal. Through computer simulations, we show that the DCBF quickly and accurately reconstructs source locations and their time-courses under widely varying SNR, degrees of correlation, and source strengths. Simulations also show that the DCBF identifies multiple simultaneously active correlated networks. Additionally, DCBF performance was tested using MEG data in humans. In an auditory task, the DCBF localized and reconstructed highly correlated left and right auditory responses. In a median-nerve stimulation task, the DCBF identified multiple meaningful networks of activation without any a priori information. Altogether, our results indicate that the DCBF is an effective and valuable tool for reconstructing correlated networks of neural activity from MEG recordings.

Previous studies using functional magnetic resonance imaging (fMRI) and event-related potentials (ERPs) of the brain have found that a distributed parietal-frontal neuronal network is activated in normals during both auditory and visual oddball tasks. The common cortical regions in this network are inferior parietal lobule (IPL)/supramarginal gyrus (SMG), anterior cingulate cortex (ACC), and dorsolateral prefrontal cortex (DLPFC). It is not clear whether the same network is activated by oddball tasks during somatosensory stimulation. The present study addressed this question by testing healthy adults as they performed a novel median-nerve oddball paradigm while undergoing magnetoencephalography (MEG). An automated multiple dipole analysis technique, the Multi-Start Spatio-Temporal (MSST) algorithm, localized multiple neuronal generators, and identified their time-courses. IPL/SMG, ACC, and DLPFC were reliably localized in the MEG median-nerve oddball responses, with IPL/SMG activation significantly preceding ACC and DLPFC activation. Thus, the same parietal-frontal neuronal network that shows activation during auditory and visual oddball tests is activated in a median-nerve oddball paradigm. Regions uniquely related to somatosensory oddball responses (e.g., primary and secondary somatosensory, dorsal premotor, primary motor, and supplementary motor areas) were also localized. Since the parietal-frontal network supports attentional allocation during performance of the task, this study may provide a novel method, as well as normative baseline data, for examining attention-related deficits in the somatosensory system of patients with neurological or psychiatric disorders.

Background: The present study aims at evaluating and comparing electrical and magnetic distributed source imaging methods applied to high-density Electroencephalography (hdEEG) and Magnetoencephalography (MEG) data. We used resolution matrices to characterize spatial resolution properties of Minimum Norm Estimate (MNE), dynamic Statistical Parametric Mapping (dSPM), standardized Low-Resolution Electromagnetic Tomography (sLORETA) and coherent Maximum Entropy on the Mean (cMEM, an entropy-based technique). The resolution matrix provides information of the Point Spread Functions (PSF) and of the Crosstalk functions (CT), this latter being also called source leakage, as it reflects the influence of a source on its neighbors. Methods: The spatial resolution of the inverse operators was first evaluated theoretically and then with real data acquired using electrical median nerve stimulation on five healthy participants. We evaluated the Dipole Localization Error (DLE) and the Spatial Dispersion (SD) of each PSF and CT map. Results: cMEM showed the smallest spatial spread (SD) for both PSF and CT maps, whereas localization errors (DLE) were similar for all methods. Whereas cMEM SD values were lower in MEG compared to hdEEG, the other methods slightly favored hdEEG over MEG. In real data, cMEM provided similar localization error and significantly less spatial spread than other methods for both MEG and hdEEG. Whereas both MEG and hdEEG provided very accurate localizations, all the source imaging methods actually performed better in MEG compared to hdEEG according to all evaluation metrics, probably due to the higher signal-to-noise ratio of the data in MEG. Conclusion: Our overall results show that all investigated methods provide similar localization errors, suggesting very accurate localization for both MEG and hdEEG when similar number of sensors are considered for both modalities. Intrinsic properties of source imaging methods as well as their behavior for well-controlled tasks, suggest an overall better performance of cMEM in regards to spatial resolution and spatial leakage for both hdEEG and MEG. This indicates that cMEM would be a good candidate for studying source localization of focal and extended generators as well as functional connectivity studies.

Survival in a suddenly-changing environment requires animals not only to detect salient stimuli, but also to promptly respond to them by initiating or revising ongoing motor processes. We recently discovered that the large vertex brain potentials elicited by sudden supramodal stimuli are strongly coupled with a multiphasic modulation of isometric force, a phenomenon that we named cortico-muscular resonance (CMR). Here, we extend our investigation of the CMR to the time-frequency domain. We show that (i) both somatosensory and auditory stimuli evoke a number of phase-locked and non-phase-locked modulations of EEG spectral power. Remarkably, (ii) some of these phase-locked and non-phase-locked modulations are also present in the Force spectral power. Finally, (iii) EEG and Force time-frequency responses are correlated in two distinct regions of the power spectrum. An early, low-frequency region (∼4 Hz) reflects the previously-described coupling between the phase-locked EEG vertex potential and force modulations. A late, higher-frequency region (beta-band, ∼20 Hz) reflects a second coupling between the non-phase-locked increase of power observed in both EEG and Force. In both time-frequency regions, coupling was maximal over the sensorimotor cortex contralateral to the hand exerting the force, suggesting an effect of the stimuli on the tonic corticospinal drive. Thus, stimulus-induced CMR occurs across at least two different types of cortical activities, whose functional significance in relation to the motor system should be investigated further. We propose that these different types of corticomuscular coupling are important to alter motor behaviour in response to salient environmental events. • We delivered salient stimuli to humans exerting a constant isometric force. • The stimuli evoked several modulations of both EEG and Force spectral power. • Some of these modulations were coupled across EEG and Force. • The coupled modulations were both phase-locked (∼4 Hz) and non-phase-locked (∼20 Hz). • Stimulus-induced corticomuscular coupling involves two types of cortical activity. Abstract Magnetoencephalography (MEG) is a non-invasive neuroimaging technique that provides whole-head measures of neural activity with millisecond temporal resolution. Over the last three decades, MEG has been used for assessing brain activity, most commonly in adults. MEG has been used less often to examine neural function during early development, in large part due to the fact that infant whole-head MEG systems have only recently been developed. In this review, an overview of infant MEG studies is provided, focusing on the period from birth to three years. The advantages of MEG for measuring neural activity in infants are highlighted (See Box 1), including the ability to assess activity in brain (source) space rather than sensor space, thus allowing direct assessment of neural generator activity. Recent advances in MEG hardware and source analysis are also discussed. As the review indicates, efforts in this area demonstrate that MEG is a promising technology for studying the infant brain. As a noninvasive technology, with emerging hardware providing the necessary sensitivity, an expected deliverable is the capability for longitudinal infant MEG studies evaluating the developmental trajectory (maturation) of neural activity. It is expected that departures from neuro-typical trajectories will offer early detection and prognosis insights in infants and toddlers at-risk for neurodevelopmental disorders, thus paving the way for early targeted interventions. • Artificial gravity alters somatosensory functional connectivity during a spaceflight analog. • Increased connectivity between the putamen and somatosensory cortex was associated with preserved mobility. • Somatosensory stimulation during artificial gravity resulted in compensatory somatosensory connectivity changes. Spaceflight has numerous untoward effects on human physiology. Various countermeasures are under investigation including artificial gravity (AG). Here, we investigated whether AG alters resting-state brain functional connectivity changes during head-down tilt bed rest (HDBR), a spaceflight analog. Participants underwent 60 days of HDBR. Two groups received daily AG administered either continuously (cAG) or intermittently (iAG). A control group received no AG. We assessed resting-state functional connectivity before, during, and after HDBR. We also measured balance and mobility changes from pre- to post-HDBR. We examined how functional connectivity changes throughout HDBR and whether AG is associated with differential effects. We found differential connectivity changes by group between posterior parietal cortex and multiple somatosensory regions. The control group exhibited increased functional connectivity between these regions throughout HDBR whereas the cAG group showed decreased functional connectivity. This finding suggests that AG alters somatosensory reweighting during HDBR. We also observed brain-behavioral correlations that differed significantly by group. Control group participants who showed increased connectivity between the putamen and somatosensory cortex exhibited greater mobility declines post-HDBR. For the cAG group, increased connectivity between these regions was associated with little to no mobility declines post-HDBR. This suggests that when somatosensory stimulation is provided via AG, functional connectivity increases between the putamen and somatosensory cortex are compensatory in nature, resulting in reduced mobility declines. Corticokinematic coherence (CKC) between limb kinematics and magnetoencephalographic (MEG) signals reflects cortical processing of proprioceptive afference. However, it is unclear whether strength of CKC is reproducible across measurement sessions. We thus examined reproducibility of CKC in a follow-up study. Thirteen healthy right-handed volunteers (7 females, 21.7 ± 4.3 yrs) were measured using MEG in two separate sessions 12.6 ± 1.3 months apart. The participant was seated and relaxed while his/her dominant or non-dominant index finger was continuously moved at 3 Hz (4 min for each hand) using a pneumatic movement actuator. Finger kinematics were recorded with a 3-axis accelerometer. Coherence was computed between finger acceleration and MEG signals. CKC strength was defined as the peak coherence value at 3 Hz form a single sensor among 40 pre-selected Rolandic gradiometers contralateral to the movement. Pneumatic movement actuator provided stable proprioceptive stimuli and significant CKC responses peaking at the contralateral Rolandic sensors. In the group level, CKC strength did not differ between the sessions in dominant (Day-1 0.40 ± 0.19 vs. Day-2 0.41 ± 0.17) or non-dominant (0.35 ± 0.16 vs. 0.36 ± 0.17) hand, nor between the hands. Intraclass-correlation coefficient (ICC) values indicated excellent inter-session reproducibility for CKC strength for both dominant (0.86) and non-dominant (0.97) hand. However, some participants showed pronounced inter-session variability in CKC strength, but only for the dominant hand. CKC is a promising tool to study proprioception in long-term longitudinal studies in the group level to follow, e.g. , integrity of cortical proprioceptive processing with motor functions after stroke. Working memory (WM) studies have been essential for ascertaining how the brain flexibly handles mentally represented information in the absence of sensory stimulation. Most studies on the memory of sensory stimulus features have focused, however, on the visual domain. Here, we report a human WM study in the tactile modality where participants had to memorize the spatial layout of patterned Braille-like stimuli presented to the index finger. We used a whole-brain searchlight approach in combination with multi-voxel pattern analysis (MVPA) to investigate tactile WM representations without a priori assumptions about which brain regions code tactospatial information. Our analysis revealed that posterior and parietal cortices, as well as premotor regions, retained information across the twelve-second delay phase. Interestingly, parts of this brain network were previously shown to also contain information of visuospatial WM. Also, by specifically testing somatosensory regions for WM representations, we observed content-specific activation patterns in primary somatosensory cortex (SI). Background The present study aims at evaluating and comparing electrical and magnetic distributed source imaging methods applied to high-density Electroencephalography (hdEEG) and Magnetoencephalography (MEG) data. We used resolution matrices to characterize spatial resolution properties of Minimum Norm Estimate (MNE), dynamic Statistical Parametric Mapping (dSPM), standardized Low-Resolution Electromagnetic Tomography (sLORETA) and coherent Maximum Entropy on the Mean (cMEM, an entropy-based technique). The resolution matrix provides information of the Point Spread Functions (PSF) and of the Crosstalk functions (CT), this latter being also called source leakage, as it reflects the influence of a source on its neighbors. Methods The spatial resolution of the inverse operators was first evaluated theoretically and then with real data acquired using electrical median nerve stimulation on five healthy participants. We evaluated the Dipole Localization Error (DLE) and the Spatial Dispersion (SD) of each PSF and CT map. Results cMEM showed the smallest spatial spread (SD) for both PSF and CT maps, whereas localization errors (DLE) were similar for all methods. Whereas cMEM SD values were lower in MEG compared to hdEEG, the other methods slightly favored hdEEG over MEG. In real data, cMEM provided similar localization error and significantly less spatial spread than other methods for both MEG and hdEEG. Whereas both MEG and hdEEG provided very accurate localizations, all the source imaging methods actually performed better in MEG compared to hdEEG according to all evaluation metrics, probably due to the higher signal-to-noise ratio of the data in MEG. Conclusion Our overall results show that all investigated methods provide similar localization errors, suggesting very accurate localization for both MEG and hdEEG when similar number of sensors are considered for both modalities. Intrinsic properties of source imaging methods as well as their behavior for well-controlled tasks, suggest an overall better performance of cMEM in regards to spatial resolution and spatial leakage for both hdEEG and MEG. • Movement range did not affect MEF strength or beta modulation in SM1 cortex. • Movement range did not affect the CKC strength in SM1 cortex. • Studies or participants with moderate variation in movement range are comparable. Proprioception is the sense of body position and movement that relies on afference from the proprioceptors in muscles and joints. Proprioceptive responses in the primary sensorimotor (SM1) cortex can be elicited by stimulating the proprioceptors using evoked (passive) limb movements. In magnetoencephalography (MEG), proprioceptive processing can be quantified by recording the movement evoked fields (MEFs) and movement-induced beta power modulations or by computing corticokinematic coherence (CKC) between the limb kinematics and cortical activity. We examined whether cortical proprioceptive processing quantified with MEF peak strength, relative beta suppression and rebound power and CKC strength is affected by the movement range of the finger. MEG activity was measured from 16 right-handed healthy volunteers while movements were applied to their right-index finger metacarpophalangeal joint with an actuator. Movements were either intermittent, every 3000 ± 250 ms, to estimate MEF or continuous, at 3 Hz, to estimate CKC. In both cases, 4 different ranges of motion of the stimuli were investigated: 15, 18, 22 and 26 mm for MEF and 6, 7, 9 and 13 mm for CKC. MEF amplitude, relative beta suppression and rebound as well as peak CKC strength at the movement frequency were compared between the movement ranges in the source space. Inter-individual variation was also compared between the MEF and CKC strengths. As expected, MEF and CKC responses peaked at the contralateral SM1 cortex. MEF peak, beta suppression and rebound and CKC strengths were similar across all movement ranges. Furthermore, CKC strength showed a lower degree of inter-individual variation compared with MEF strength. Our result of absent modulation by movement range in cortical responses to passive movements of the finger indicates that variability in movement range should not hinder comparability between different studies or participants. Identical sensory stimuli can lead to different neural responses depending on the instantaneous brain state. Specifically, neural excitability in sensory areas may shape the brain´s response already from earliest cortical processing onwards. However, whether these dynamics affect a given sensory domain as a whole or occur on a spatially local level is largely unknown. We studied this in the somatosensory domain of 38 human participants with EEG, presenting stimuli to the median and tibial nerves alternatingly, and testing the co-variation of initial cortical responses in hand and foot areas, as well as their relation to pre-stimulus oscillatory states. We found that amplitude fluctuations of initial cortical responses to hand and foot stimulation – the N20 and P40 components of the somatosensory evoked potential (SEP), respectively – were not related, indicating local excitability changes in primary sensory regions. In addition, effects of pre-stimulus alpha (8-13 Hz) and beta (18-23 Hz) band amplitude on hand-related responses showed a robust somatotopic organization, thus further strengthening the notion of local excitability fluctuations. However, for foot-related responses, the spatial specificity of pre-stimulus effects was less consistent across frequency bands, with beta appearing to be more foot-specific than alpha. Connectivity analyses in source space suggested this to be due to a somatosensory alpha rhythm that is primarily driven by activity in hand regions while beta frequencies may operate in a more hand-region-independent manner. Altogether, our findings suggest spatially distinct excitability dynamics within the primary somatosensory cortex, yet with the caveat that frequency-specific processes in one sub-region may not readily generalize to other sub-regions. In non-human primates, opioid-receptor blockade increases social grooming, and the endogenous opioid system has therefore been hypothesized to support maintenance of long-term relationships in humans as well. Here we tested whether social touch modulates opioidergic activation in humans using in vivo positron emission tomography (PET). Eighteen male participants underwent two PET scans with [11C]carfentanil, a ligand specific to μ-opioid receptors (MOR). During the social touch scan, the participants lay in the scanner while their partners caressed their bodies in a non-sexual fashion. In the baseline scan, participants lay alone in the scanner. Social touch triggered pleasurable sensations and increased MOR availability in the thalamus, striatum, and frontal, cingulate, and insular cortices. Modulation of activity of the opioid system by social touching might provide a neurochemical mechanism reinforcing social bonds between humans. [ABSTRACT FROM AUTHOR]

Touch can be highly emotional, and depending on the environment, it can be perceived as pleasant and comforting or disgusting and dangerous. Here, we studied the impact of context on the processing of tactile stimuli using a functional magnetic resonance imaging (fMRI) paradigm. This was achieved by embedding tactile stimulation in a variable olfactory environment. Twenty people were scanned with BOLD fMRI while receiving the following stimulus blocks: Slow stroking Touch, Civette odor (feces like), Rose odor, Touch + Civette, and Touch + Rose. Ratings of pleasantness and intensity of tactile stimuli and ratings of disgust and intensity of olfactory stimuli were collected. The impact of the olfactory context on the processing of touch was studied using covariance analyses. Coupling between olfactory processing and somatosensory processing areas was assessed with psychophysiological interaction analysis (PPI). A subjectively disgusting olfactory environment significantly reduced the perceived pleasantness of touch. The touch fMRI activation in the secondary somatosensory cortex, operculum 1 (OP1), was positively correlated with the disgust towards the odors. Decreased pleasantness of touch was related to decreased posterior insula activity. PPI analysis revealed a significant interaction between the OP1, posterior insula, and regions processing the disgust of odors (orbitofrontal cortex and amygdala). We conclude that the disgust evaluation of the olfactory environment moderates neural reactivity in somatosensory regions by upregulation of the OP1 and downregulation of the posterior insula. This adaptive regulation of affective touch processing may facilitate adaptive reaction to a potentially harmful stimulus. [ABSTRACT FROM AUTHOR]

Recent evidence from blind participants suggests that visual areas are task-oriented and sensory modality input independent rather than sensory-specific to vision. Specifically, visual areas are thought to retain their functional selectivity when using non-visual inputs (touch or sound) even without having any visual experience. However, this theory is still controversial since it is not clear whether this also characterizes the sighted brain, and whether the reported results in the sighted reflect basic fundamental a-modal processes or are an epiphenomenon to a large extent. In the current study, we addressed these questions using a series of fMRI experiments aimed to explore visual cortex responses to passive touch on various body parts and the coupling between the parietal and visual cortices as manifested by functional connectivity. We show that passive touch robustly activated the object selective parts of the lateral–occipital (LO) cortex while deactivating almost all other occipital–retinotopic-areas. Furthermore, passive touch responses in the visual cortex were specific to hand and upper trunk stimulations. Psychophysiological interaction (PPI) analysis suggests that LO is functionally connected to the hand area in the primary somatosensory homunculus (S1), during hand and shoulder stimulations but not to any of the other body parts. We suggest that LO is a fundamental hub that serves as a node between visual-object selective areas and S1 hand representation, probably due to the critical evolutionary role of touch in object recognition and manipulation. These results might also point to a more general principle suggesting that recruitment or deactivation of the visual cortex by other sensory input depends on the ecological relevance of the information conveyed by this input to the task/computations carried out by each area or network. Activity-induced manganese-dependent MRI (AIM-MRI) is a powerful tool to track system-wide neural activity using high resolution, quantitative T 1 -weighted MRI in animal models and has significant advantages for investigating neural activity over other modalities including BOLD fMRI. With AIM-MRI, Mn 2+ ions enter neurons via voltage-gated calcium channels preferentially active during the time of experimental exposure. A broad range of AIM-MRI studies using different species studying different phenomena have been performed, but few of these studies provide a systematic evaluation of the factors influencing the detection of Mn 2+ such as dosage and the temporal characteristics of Mn 2 + uptake. We identified an optimal dose of Mn 2+ (25 mg/kg, s.c.) in order to characterize the time-course of Mn 2+ accumulation in active neural regions in the rabbit. T 1 -weighted MRI and functional MRI were collected 0–3, 6–9, and 24–27 h post-Mn 2+ injection while the vibrissae on the right side were vibrated. Significant BOLD activation in the left somatosensory (SS) cortex and left ventral posteromedial (VPM) thalamic nucleus was detected during whisker vibration. T 1 -weighted signal intensities were extracted from these regions, their corresponding contralateral regions and the visual cortex (to serve as controls). A significant elevation in T 1 -weighted signal intensity in the left SS cortex (relative to right) was evident 6–9 and 24–27 h post-Mn 2+ injection while the left VPM thalamus showed a significant enhancement (relative to the right) only during the 24–27 h session. Visual cortex showed no hemispheric difference at any timepoint. Our results suggest that studies employing AIM-MRI would benefit by conducting experimental manipulations 6–24 h after subcutaneous MnCl 2 injections to optimize the concentration of contrast agent in the regions active during the exposure. One of the most striking demonstrations of plasticity in the adult human brain follows peripheral injury, such as amputation. In the primary sensorimotor cortex, arm amputation results in massive local remapping of the missing hands' cortical territory. However, little is known about the consequences of sensorimotor deprivation on global brain organisation. Here, we used resting-state fMRI to identify large-scale reorganisation beyond the primary sensorimotor cortex in arm amputees, compared with two-handed controls. Specifically, we characterised changes in functional connectivity between the cortical territory of the missing hand in the primary sensorimotor cortex (‘missing hand cortex’) and two networks of interest: the sensorimotor network, which is typically strongly associated with the hand cortex, and the default mode network (DMN), which is normally dissociated from it. Functional connectivity values between the missing hand cortex and the sensorimotor network were reduced in amputees, and connectivity was weaker in individuals amputated for longer periods. Lower levels of functional coupling between the missing hand cortex and the sensorimotor network were also associated with emerged coupling of this cortex with the DMN. Our results demonstrate that plasticity following arm amputation is not restricted to local remapping occurring within the sensorimotor homunculus of the missing hand but rather produces a cascade of cortical reorganisation at a network-level scale. These findings may provide a new framework for understanding how local deprivation following amputation could elicit complex perceptual experiences of phantom sensations, such as phantom pain. Abstract: What are the spectral signatures of somatosensory attention? Here we show that the answer to this question depends critically on the sensory context in which attention is deployed. We recorded magnetoencephalography (MEG) in humans and investigated tactile spatial attention in two different sensory contexts: in anticipation and during the processing of sustained tactile stimuli. We observe a double dissociation between these contexts and two key electrophysiological correlates of attention: in anticipation we primarily observe an attentional suppression of contralateral alpha and beta oscillations (8–12 and 15–30Hz, respectively), whereas during stimulus processing we primarily observe an attentional amplification of contralateral gamma oscillations (55–75Hz). This dissociation is well explained by the different neural states that occur prior and during the stimulus, and on which attention can exert its influence. In line with analogous observations in the visual modality, this suggests that the neural implementation of attention must be understood in relation to context and existing brain states. Consequently, different signatures of attention may contribute to perception in different contexts and, as our data reveals for the attentional modulation of alpha oscillations, these are not always required for attention to improve perception. At the same time, these data demonstrate that the attentional modulations of alpha and gamma oscillations (during, respectively, attentional orienting and attentional selection), are generalizable phenomena across the different sensory modalities. Abstract: Oscillatory interactions within functionally specialized but distributed brain regions are believed to be central to perceptual and cognitive functions. Here, using human scalp electroencephalography (EEG) recordings combined with source reconstruction techniques, we study how oscillatory activity functionally organizes different neocortical regions during a tactile discrimination task near the limit of spatial acuity. While undergoing EEG recordings, blindfolded participants felt a linear three-dot array presented electromechanically, under computer control, and reported whether the central dot was offset to the left or right. The average brain response differed significantly for trials with correct and incorrect perceptual responses in the timeframe approximately between 130 and 175ms. During trials with correct responses, source-level peak activity appeared in the left primary somatosensory cortex (SI) at around 45ms, in the right lateral occipital complex (LOC) at 130ms, in the right posterior intraparietal sulcus (pIPS) at 160ms, and finally in the left dorsolateral prefrontal cortex (dlPFC) at 175ms. Spectral interdependency analysis of activity in these nodes showed two distinct distributed networks, a dominantly feedforward network in the beta band (12–30Hz) that included all four nodes and a recurrent network in the gamma band (30–100Hz) that linked SI, pIPS and dlPFC. Measures of network activity in both bands were correlated with the accuracy of task performance. These findings suggest that beta and gamma band oscillatory networks coordinate activity between neocortical regions mediating sensory and cognitive processing to arrive at tactile perceptual decisions. Abstract: The operculo-insular cortex has been termed the ‘homeostatic control center’ or ‘general magnitude estimator’ of the human mind. In this study, somatosensory, nociceptive and caloric vestibular stimuli were applied to reveal, whether there are mainly common, or possibly specific regions activated by one modality alone and whether lateralization effects, time pattern differences or influences of the aversive nature of the stimuli could be observed. Activation of the dorsal posterior insula was caused by all stimuli alike thus terming this area multimodal. Early phases of the noxious heat and caloric vestibular stimulation led to responses in the anterior insula. Using conjunction analyses we found that left- and right-sided tactile stimulation, but not nociceptive stimulation, caused a joint activation of the cytoarchitectonic area OP1 and nociceptive but not tactile stimulation of the anterior insula bilaterally. Tactile activation in the parietal operculum (SII, OP1) was distinct from nociceptive activation (OP3 and frontal operculum). The joint activation by all three stimuli located in the dorsal posterior insula argues for the presence of multisensory structures. The distinct activation of the anterior insula by aversive stimuli and the posterior insula by multisensory signals supports the concept of a partitioned insular cortex recently introduced based on connectivity studies and meta-analyses. Abstract: Paired-pulse stimulation has been used previously to evaluate cortical excitability and sensory gating. To help elucidate the neural network involved in paired-pulse suppression of somatosensory cortical processing, magnetoencephalographic (MEG) responses to paired-pulse electrical stimulation of the left median nerve of the wrists of 13 healthy males were recorded using an intra-pair interstimulus interval (ISI) of 500ms and an inter-pair ISI of 8s. Minimum norm estimates showed the presence of cortical activation in the bilateral primary somatosensory cortex, the post-central sulcus and the supplementary motor areas. Compared with the responses to the first stimulation, the responses to the second stimulation were attenuated in these areas with gating ratios (the amplitude ratios of the second response to the first response) of 0.54–0.69. By spectral power dynamic analysis, beta frequency oscillations were found to be associated with an early-latency (30–36ms) gating process in the contralateral primary somatosensory cortex and post-central sulcus, whereas theta and alpha oscillations were correlated with paired-pulse suppression of activations at 98–136ms in the ipsilateral primary somatosensory cortex, the bilateral post-central sulcus and the supplementary motor areas. In summary, it can be concluded that differential oscillatory activities are involved in the pair-pulse suppression in various somatosensory regions in response to repetitive external stimulations. Abstract: The representation of the human body in the human cerebellum is still relatively unknown, compared to the well-studied homunculus in the primary somatosensory cortex. The investigation of the body representation in the cerebellum and its somatotopic organisation is complicated because of the relatively small dimensions of the cerebellum, compared to the cerebrum. Somatotopically organised whole-body homunculi have previously been reported in both humans and rats. However, whether individual digits are represented in the cerebellum in a somatotopically organised way is much less clear. In this study, the high spatial resolution and high sensitivity to the blood oxygenation level dependent (BOLD) signal of 7T fMRI were employed to study the BOLD responses in the human cerebellum to the stroking of the skin of individual digits, the hand and forearm. For the first time, a coarse somatotopic organisation of the digits, ordered from D1–D5, could be visualised in individual human subjects in both the anterior (lobule V) and the posterior (lobule VIII) lobes of the cerebellum using a somatosensory stimulus. The somatotopic gradient in lobule V was found consistently in the posterior to anterior direction, with the thumb most posterior, while the direction of the somatotopic gradient in lobule VIII differed between subjects. No somatotopic organisation was found in Crus I. A comparison of the digit patches with the hand patch revealed that the digit regions are completely covered by the hand region in both the anterior and posterior lobes of the cerebellum, in a non-somatotopic manner. These results demonstrate the promise of ultra-high field, high-resolution fMRI for studies of the cerebellum. [Copyright &y& Elsevier]

Abstract: Studies have shown that the periodic repetition of a stimulus induces, at certain stimulation frequencies, a sustained electro-cortical response of corresponding frequency, referred to as steady-state evoked potential (SSEP). Using infrared laser stimulation, we recently showed that SSEPs can be used to explore nociceptive cortical processing. Here, we implemented a novel approach to elicit such responses, using a periodic intra-epidermal electrical stimulation of cutaneous Aδ-nociceptors (Aδ-SSEPs). Using a wide range of frequencies (3–43Hz), we compared the scalp topographies and temporal dynamics of these Aδ-SSEPs to the Aβ-SSEPs elicited by non-nociceptive transcutaneous electrical stimulation, as well as to the transient ERPs elicited by the onsets of the 10-s stimulation trains, applied to the left and right hand. At 3Hz, we found that the topographies of Aβ- and Aδ-SSEPs were both maximal at the scalp vertex, and resembled closely that of the late P2 wave of transient ERPs, suggesting activity originating from the same neuronal populations. The responses also showed marked habituation, suggesting that they were mainly related to unspecific, attention-related processes. In contrast, at frequencies >3Hz, the topographies of Aβ- and Aδ-SSEPs were markedly different. Aβ-SSEPs were maximal over the contralateral parietal region, whereas Aδ-SSEPs were maximal over midline frontal regions, thus indicating an entrainment of distinct neuronal populations. Furthermore, the responses showed no habituation, suggesting more obligatory and specific stages of sensory processing. Taken together, our results indicate that Aβ- and Aδ-SSEPs offer a unique opportunity to study the cortical representation of nociception and touch. [Copyright &y& Elsevier]

Abstract: Connectivity-based segmentation has been used to identify functional gray matter subregions that are not discernable on conventional magnetic resonance imaging. However, the accuracy and reliability of this technique has only been validated using indirect means. In order to provide direct electrophysiologic validation of connectivity-based thalamic segmentations within human subjects, we assess the correlation of atlas-based thalamic anatomy, connectivity-based thalamic maps, and somatosensory evoked thalamic potentials in two adults with medication-refractory epilepsy who were undergoing intracranial EEG monitoring with intrathalamic depth and subdural cortical strip electrodes. MRI with atlas-derived localization was used to delineate the anatomic boundaries of the ventral posterolateral (VPL) nucleus of the thalamus. Somatosensory evoked potentials with intrathalamic electrodes physiologically identified a discrete region of phase reversal in the ventrolateral thalamus. Finally, DTI was obtained so that probabilistic tractography and connectivity-based segmentation could be performed to correlate the region of thalamus linked to sensory areas of the cortex, namely the postcentral gyrus. We independently utilized these three different methods in a blinded fashion to localize the “sensory” thalamus, demonstrating a high-degree of reproducible correlation between electrophysiologic and connectivity-based maps of the thalamus. This study provides direct electrophysiologic validation of probabilistic tractography-based thalamic segmentation. Importantly, this study provides an electrophysiological basis for using connectivity-based segmentation to further study subcortical anatomy and physiology while also providing the clinical basis for targeting deep brain nuclei with therapeutic stimulation. Finally, these direct recordings from human thalamus confirm early inferences of a sensory thalamic component of the N18 waveform in somatosensory evoked potentials. [Copyright &y& Elsevier]

Abstract: Egocentric tactile perception is crucial for skilled hand motor control. In order to better understand the brain functional underpinnings related to this basic sensorial perception, we performed a tactile perception functional magnetic resonance imaging (fMRI) experiment with two aims. The first aim consisted of characterizing the neural substrate of two types of egocentric tactile discrimination: the spatial localization (SLD) and simultaneity succession discrimination (SSD) in both hands to define hemispheric dominance for these tasks. The second goal consisted of characterizing the brain activation related to the spatial attentional load, the functional changes and their connectivity patterns induced by the psychometric performance (PP) during SLD. We used fMRI in 25 right-handed volunteers, applying pairs of sinusoidal vibratory stimuli on eight different positions in the palmar surface of both hands. Subjects were required either to identify the stimulus location with respect to an imaginary midline (SLD), to discriminate the simultaneity or succession of a stimuli pair (SSD) or to simply respond to stimulus detection. We found a fronto-parietal network for SLD and frontal network for SSD. During SLD we identified right hemispheric dominance with increased BOLD activation and functional interaction of the right supramarginal gyrus with contralateral intra-parietal sulcus for right and left hand independently. Brain activity correlated to spatial attentional load was found in bilateral structures of intra-parietal sulcus, precuneus extended to superior parietal lobule, pre-supplementary motor area, frontal eye fields and anterior insulae for both hands. We suggest that the right supramarginal gyrus and its interaction with intra-parietal lobule may play a pivotal role in the phenomenon of tactile neglect in right fronto-parietal lesions. Abstract: A single brief visual stimulus accompanied by two brief tactile stimuli is frequently perceived incorrectly as two flashes, a phenomenon called double-flash illusion (DFI). We investigated whether the DFI is accompanied by changes in rhythmic neuronal activity, using magnetoencephalography in human subjects. Twenty-two subjects received visuo-tactile stimulation and reported the number of perceived visual stimuli. We sorted trials with identical physical stimulation according to the reported subjective percept and assessed differences in spectral power in somatosensory and occipital sensors. In DFI trials, occipital sensors displayed a contralateral enhancement of gamma-band (80–140Hz) activity in response to stimulation. In somatosensory sensors, the DFI was associated with an increase of spectral power for low frequencies (5–17.5Hz) around stimulation and a decrease of spectral power in the 22.5–30Hz range between 450 and 750ms post-stimulation. In summary, several components of rhythmic activity predicted variable subjective experience for constant physical stimulation. Notably, the enhanced occipital gamma-band activity during DFI was similar in time and frequency extent to the somatosensory gamma-band response to tactile stimulation. We speculate that the DFI might therefore occur when the somatosensory gamma-response is transmitted to visual cortex. Abstract: Advances in non-invasive neuroimaging technology now provide a means of directly observing learning within the brain. Classical conditioning serves as an ideal starting point for examining the dynamic expression of learning within the human brain, since this paradigm is well characterized using multiple levels of analysis in a broad range of species. We used MEG to expand the characterization of conditioned responses (CR) recorded from the human brain with a simultaneous examination of their spatial, temporal and spectral properties. We paired an auditory conditioned stimulus (CS+) with a somatosensory unconditioned stimulus (US). We found that when the US was randomly omitted, presentations of CS+ alone, elicited greater desynchronization of beta-band activity in contralateral somatosensory cortex compared to presentations of an auditory stimulus that was never paired with the US (CS−), and compared the CS+ following a non-reinforced extinction session. This differentiation was largest between 150 and 350ms following US omission. We show that cross-modal CRs in the primary sensorimotor system are predominantly characterized by modulation of ongoing cortical oscillations. [ABSTRACT FROM AUTHOR]

Abstract: It is well known that primary and non-primary areas of human somatosensory cortex are involved in the processing of adequate deviant/rare stimuli and omission of frequent stimuli. However, the relative weight and interaction of these variables is poorly known. This functional magnetic resonance imaging (fMRI) study tested the hypothesis that somatosensory stimulus processing and attention especially interact in non-primary somatosensory areas including secondary somatosensory cortex (SII) and insula. To test this hypothesis, responses of somatosensory cortex were mapped during four conditions of an oddball paradigm: DELIVERED COUNT and IGNORE (count or ignore deviant/rare electrical stimuli, respectively); OMITTED COUNT and IGNORE (count or ignore the rare omission of frequent electrical stimuli, respectively). The deviant/rare and frequent electrical stimuli were delivered to median and ulnar nerve, respectively. It was observed that contralateral (left) primary somatosensory responses were not markedly modulated by the mentioned deviant/rare events. Furthermore, contralateral SII and insula responded to all but not OMITTED IGNORE (purely attentive) condition, whereas ipsilateral (right) SII responded to all conditions. Finally, ipsilateral insula responded to the COUNT (attentive) conditions, regardless of the physical presence of the deviant/rare stimuli. The results suggest that in somatosensory modality, bilateral SII and left (contralateral) insula reflect complex integrative processes of stimulus elaboration and attention, whereas right (ipsilateral) insula mainly sub-serves active attention to deviance within a sequence of somatosensory stimuli. Abstract: We describe a neuroimaging protocol that utilizes an anatomical atlas of the human head to guide diffuse optical tomography of human brain activation. The protocol is demonstrated by imaging the hemodynamic response to median-nerve stimulation in three healthy subjects, and comparing the images obtained using a head atlas with the images obtained using the subject-specific head anatomy. The results indicate that using the head atlas anatomy it is possible to reconstruct the location of the brain activation to the expected gyrus of the brain, in agreement with the results obtained with the subject-specific head anatomy. The benefits of this novel method derive from eliminating the need for subject-specific head anatomy and thus obviating the need for a subject-specific MRI to improve the anatomical interpretation of diffuse optical tomography images of brain activation. Abstract: The use of minimally and non-invasive neuroimaging methods in animal models has sharply increased over the past decade. Such studies have enhanced understanding of the neural basis of the physical signals quantified by these tools, and have addressed an assortment of fundamental and otherwise intractable questions in neurobiology. To date, these studies have almost exclusively utilized positron-emission tomography or variants of magnetic resonance based imaging. These methods provide largely indirect measures of brain activity and are strongly reliant on intact vasculature and normal blood-flow, which is known to be compromised in many clinical conditions. The current study provides the first demonstration of whole-head magnetoencephalography (MEG), a non-invasive and direct measure of neuronal activity, in a rhesus monkey, and in the process supplies the initial data on systems-level dynamics in somatosensory cortices. An adult rhesus monkey underwent three separate studies of tactile stimulation on the pad of the right second or fifth digit as whole-head MEG data were acquired. The neural generators of the primary neuromagnetic components were localized using an equivalent-current-dipole model. Second digit stimulation produced an initial cortical response peaking ∼16 ms after stimulus onset in the contralateral somatosensory cortices, with a later response at ∼96 ms in an overlapping or nearby neural area with a roughly orthogonal orientation. Stimulation of the fifth digit produced similar results, the main exception being a substantially weaker later response. We believe the 16 ms response is likely the monkey homologue of the human M50 response, as both are the earliest cortical response and localize to the contralateral primary somatosensory area. Thus, these data suggest that mechanoreception in nonhuman primates operates substantially faster than that in adult humans. More broadly, these results demonstrate that it is feasible to use current human whole-head MEG instrumentation to record neuromagnetic responses in adult rhesus monkeys. Nonhuman primate models of human disease provide the closest phylogenetic link to humans. The present, non-invasive imaging study could promote exciting translational integration of invasive animal studies and non-invasive human studies, allowing experimentally induced deficits and pharmacological treatments to be interpreted in light of resulting brain network interactions. Abstract: Human superior temporal sulcus (STS) is thought to be a key brain area for multisensory integration. Many neuroimaging studies have reported integration of auditory and visual information in STS but less is known about the role of STS in integrating other sensory modalities. In macaque STS, the superior temporal polysensory area (STP) responds to somatosensory, auditory and visual stimulation. To determine if human STS contains a similar area, we measured brain responses to somatosensory, auditory and visual stimuli using blood-oxygen level dependent functional magnetic resonance imaging (BOLD fMRI). An area in human posterior STS, STSms (multisensory), responded to stimulation in all three modalities. STSms responded during both active and passive presentation of unisensory somatosensory stimuli and showed larger responses for more intense vs. less intense tactile stimuli, hand vs. foot, and contralateral vs. ipsilateral tactile stimulation. STSms showed responses of similar magnitude for unisensory tactile and auditory stimulation, with an enhanced response to simultaneous auditory–tactile stimulation. We conclude that STSms is important for integrating information from the somatosensory as well as the auditory and visual modalities, and could be the human homolog of macaque STP. Abstract: In the human somatosensory system, the contralateral primary somatosensory cortex (SI) is presumed to process and encode type and intensity of the sensory inputs, whereas the bilateral secondary somatosensory cortex (SII) is believed to perform higher order functions including sensorimotor integration, integration of information from the two body halves, attention, learning and memory. In this fMRI study we investigated the effect of attention on the activation of SI and SII, as induced by nonpainful and painful rare deviant electric stimuli during somatosensory oddball tasks. The working hypothesis is of stronger effects of attention on SII with respect to SI. Four runs were acquired according to an oddball scheme. Frequent nonpainful electrical stimuli were delivered to the ulnar nerve at motor threshold, whereas rare/deviant stimuli were delivered to median nerve in four conditions (one condition per run): nonpainful, painful, counting nonpainful, and counting painful. Results showed a statistically significant fMRI activation in bilateral SII but not in contralateral SI when the rare/deviant median nerve stimuli were delivered at nonpainful and painful levels as well as at the two levels of attention considered (i.e., associated with counting and non-counting tasks). Furthermore, fMRI activation in SII did not differ across the different levels of stimulus intensity (nonpainful, painful) and attention (non-counting, counting). These results corroborate the notion that SII is the target of independent pathways for the processing and integration of nonpainful and painful somatosensory stimuli salient for further high-order elaborations. Abstract: In serial sensory processing, information flows from the thalamus via primary sensory cortices to higher-order association areas. However, association cortices also receive, albeit weak, direct thalamocortical sensory inputs of unknown function. For example, while information proceeds from primary (SI) to secondary (SII) somatosensory cortex in a serial fashion, both areas are known to receive direct thalamocortical sensory input. The present study examines the potential roles of such parallel input arrangements. The subjects were presented with median nerve somatosensory stimuli with the instruction to respond with the contralateral hand. The locations and time courses of the activated brain areas were first identified with magnetoencephalography (MEG). In a subsequent session, these brain areas were modulated with single-pulse transcranial magnetic stimulation (TMS) at 15–210 ms after the somatosensory stimulus while electroencephalography (EEG) was recorded. TMS pulses at 15–40 ms post-stimulus significantly speeded up reaction times and somatosensory-evoked responses, with largest facilitatory effects when the TMS pulse was given to contralateral SII at about 20 ms. To explain the results, we propose that the early somatosensory-evoked physiological SII activation exerts an SII→SI influence that facilitates the reciprocal SI→SII pathway – with TMS to SII we apparently amplified this mechanism. The results suggest that the human brain may utilize parallel inputs to facilitate long-distance cortico-cortical connections, resulting in accelerated processing and speeded reaction times. Abstract: Inhibiting inappropriate behavior and thoughts is an essential ability for humans, but the regions responsible for inhibitory processing are a matter of continuous debate. This is the first study of somatosensory go/nogo tasks using event-related functional magnetic resonance imaging (fMRI). Fifteen subjects preformed two different types of go/nogo task, i.e. (1) Movement and (2) Count, to compare with previous studies using visual go/nogo tasks, and confirm whether the inhibitory processing is dependent on sensory modalities. Go and nogo stimuli were presented with an even probability. Our data indicated that the response inhibition network involved the dorsolateral (DLPFC) and ventrolateral (VLPFC) prefrontal cortices, pre-supplementary motor area (pre-SMA), anterior cingulate cortex (ACC), inferior parietal lobule (IPL), insula, and temporoparietal junction (TPJ), which were consistent with previous results obtained using visual go/nogo tasks. These activities existed in both Movement and Count Nogo trials. Therefore, our results suggest that the network for inhibitory processing is not dependent on sensory modalities but reflects common neural activities. In addition, there were differences of activation intensity between Movement and Count Nogo trials in the prefrontal cortex, temporal lobe, and ACC. Abstract: Somatotopic mapping of human body surface using fMRI is challenging. First, it is difficult to deliver tactile stimuli in the scanner. Second, multiple stimulators are often required to cover enough area of the complex-shaped body surface, such as the face. In this study, a computer-controlled pneumatic system was constructed to automatically deliver air puffs to 12 locations on the body surface through an MR-compatible manifold (Dodecapus) mounted on a head coil inside the scanner bore. The timing of each air-puff channel is completely programmable and this allows systematic and precise stimulation on multiple locations on the body surface during functional scans. Three two-condition block-design “Localizer” paradigms were employed to localize the cortical representations of the face, lips, and fingers, respectively. Three “Phase-encoded” paradigms were employed to map the detailed somatotopic organizations of the face, lips, and fingers following each “Localizer” paradigm. Multiple somatotopic representations of the face, lips, and fingers were localized and mapped in primary motor cortex (MI), ventral premotor cortex (PMv), polysensory zone (PZ), primary (SI) and secondary (SII) somatosensory cortex, parietal ventral area (PV) and 7b, as well as anterior and ventral intraparietal areas (AIP and VIP). The Dodecapus system is portable, easy to setup, generates no radio frequency interference, and can also be used for EEG and MEG experiments.
