Physiological studies indicate that the output neurons in the multisensory (i.e. intermediate and deep) laminae of the cat superior colliculus receive converging information from widespread regions of the neuraxis, integrate this information, and then relay the product to regions of the brainstem involved in the control of head and eye movements. Yet, an understanding of the neuroanatomy of these converging afferents has been hampered because many terminals contact distal dendrites that are difficult to label with the neurochemical markers generally used to visualize superior colliculus output neurons. Here we show that the SMI-32 antibody, directed at the non-phosphorylated epitopes of high molecular weight neurofilament proteins, is an effective marker for these superior colliculus output neurons. It is also one that can label their distal dendrites. Superior colliculus sections processed for SMI-32 revealed numerous labeled neurons with varying morphologies within the deep laminae. In contrast, few labeled neurons were observed in the superficial laminae. Neurons with large somata in the lateral aspects of the deep superior colliculus were particularly well labeled, and many of their secondary and tertiary dendrites were clearly visible. Injections of the fluorescent biotinylated dextran amine into the pontine reticular formation revealed that approximately 80% of the SMI-32 immunostained neurons also contained retrogradely transported biotinylated dextran amine, indicating that SMI-32 is a common cytoskeletal component expressed in descending output neurons. Superior colliculus output neurons also are known to express the calcium-binding protein parvalbumin, and many SMI-32 immunostained neurons also proved to be parvalbumin immunostained. These studies suggest that SMI-32 can serve as a useful immunohistochemical marker for detailing the somatic and dendritic morphology of superior colliculus output neurons and for facilitating evaluations of their input/output relationships.

The main purpose of this study was to investigate the role of N-methyl-D-aspartate receptors in host-graft synaptic transmission in the neocortex. The effects of low extracellular magnesium, the glutamate agonist N-methyl-D-aspartate and N-methyl-D-aspartate antagonists on the synaptic activation of connections between embryonic neocortical graft tissue and the surrounding host tissue were studied in 17 perfused slices of rat neocortex. In standard artificial cerebrospinal fluid, stimulation of the host white matter evoked field potentials in four of 17 grafts. However, in Mg(2+)-free medium, the same stimulation evoked field potentials in an additional six grafts, with significant increases in the mean duration of the evoked responses in the 10 responsive grafts. In five of these slices stimulation of the graft also evoked field potentials in the host tissue, suggesting reciprocal interaction between graft and host. Simultaneous extracellular recordings from graft and host tissues in Mg(2+)-free medium showed that spontaneous epileptiform discharges developed in the graft and host tissue synchronously. In Mg(2+)-free medium, application of N-methyl-D-aspartate induced a shift of the baseline with superimposed epileptiform discharges in both graft and host. Application of the non-competitive N-methyl-D-aspartate antagonist ketamine and the competitive antagonist D,L-2-amino-5-phosphonovaleric acid attenuated or reversibly blocked both the spontaneous epileptiform discharges and the evoked field potentials. Our data provides evidence that N-methyl-D-aspartate receptors are present at synapses created between fetal graft and host neocortex, and that the N-methyl-D-aspartate-activated receptor-channel complex plays an active role in mediating excitatory synaptic transmission in host-graft circuitry.

Sera of Alzheimer's disease and Down's syndrome patients contain antibodies which bind specifically to the high molecular weight neurofilament protein of Torpedo cholinergic neurons. We have recently shown that prolonged immunization of rats with this antigen results in the accumulation of IgG in neurons in the septum and hippocampus of the immunized rats and in cognitive impairments. This animal model is termed experimental autoimmune dementia. In the present study we examined whether the anti-cholinergic high molecular weight neurofilament subunit immune response of the experimental autoimmune dementia rats affects forebrain cholinergic neurons. This was performed immunohistochemically utilizing a monoclonal antibody to nerve growth factor receptor, a specific marker of cholinergic neurons in the forebrain. The results obtained revealed significant decreases in the density of cholinergic neurons in the medial septal nucleus and diagonal band of the experimental autoimmune dementia rats. These decreases are specific to the anti-cholinergic high molecular weight neurofilament subunit immune response of the experimental autoimmune dementia rats and are not observed in control rats which were immunized with chemically heterogeneous high molecular weight neurofilament subunit. The decrease in density of forebrain cholinergic neurons in experimental autoimmune dementia rats may mimic pathogenic processes in Alzheimer's disease and supports a role for anti-cholinergic high molecular weight neurofilament subunit antibodies in the degeneration of cholinergic neurons in the disease.

Experimental autoimmune dementia is a rat model designed to examine the potential role of anti-cholinergic neurons antibodies in neuronal degeneration in dementia and Alzheimer's disease. We have previously shown that sera of patients with Alzheimer's disease contain antibodies which bind specifically to the high molecular weight neurofilament protein of the purely cholinergic electromotor neurons of Torpedo. Production of such antibodies in experimental autoimmune dementia rats by prolonged immunization with the Torpedo cholinergic high molecular weight neurofilament subunit results in accumulation of antibodies in the septum and hippocampus of the immunized rats, in a marked decrease in the density of forebrain cholinergic neurons, and in memory deficits. In the present study we characterized the open-field behavior of experimental autoimmune dementia rats, and examined whether, like in dementia, the spatiotemporal organization of their behavior is impaired. The results obtained revealed that experimental autoimmune dementia rats travel shorter distances; explore a smaller part of the open-field; and perform less round-trips to the key location--the home base--in reference to which their behavior is normally organized. The shrinkage of the explored space and the reduced number of round trips are independent of the amount of locomotion and represent a deterioration in the organization of behavior in time and space. These behavioral changes are specific to the anti-cholinergic immune response of experimental autoimmune dementia rats as they are not observed in rats which were immunized with chemically heterogeneous high molecular weight neurofilament subunit.(ABSTRACT TRUNCATED AT 250 WORDS)

We have previously shown that neurofilaments in enteric neurons are immunologically distinct from those found in the central nervous system. In particular, one monoclonal antibody to the medium molecular weight neurofilament subunit, called NN18, stained the perikarya of enteric neurons very weakly, if at all, although other medium molecular weight neurofilament subunit antibodies clearly showed the presence of significant amounts of medium molecular weight neurofilament subunit. We have since located the epitope for NN18 in a peptide sequence at the carboxy terminal tail of medium molecular weight neurofilament subunit and have now looked for further neurofilament antibodies that stain the same region. We found two monoclonal antibodies, RMO1 and RMO59, that recognize the same peptide. These antibodies also show much reduced staining in myenteric neurons compared to those in the central nervous system, suggesting that this region of the medium molecular weight neurofilament subunit is specifically modified in certain cells. In the developing enteric nervous system, we found that all the antibodies show strong staining of enteric neurons in the neonatal rat, but that with NN18, RMO1, and RMO59, the staining intensity decreases during further development, and by postnatal day 21 all three demonstrate decreased or absent staining identical to the adult. These results indicate that myenteric neurons, in contrast to the brain, have a developmentally regulated modification in a specific region of the medium molecular weight neurofilament subunit, which may reflect adaptation to structural stress by myenteric neurons.

It has been reported that injection of the antitumoral drug cis-dichlorodiammineplatinum at 10 days of life affects cerebellar development in rats. After a single dose of 5 micrograms/g of body weight, the formation of granule cells is decreased and the maturation of postmitotic neurons is slowed down. A substantial time after treatment, reduced cell packing density of the internal granule layer and atrophy of the molecular layer can be observed. In addition, there is degeneration of some Purkinje cells and Golgi neurons. In spite of all these alterations, the regular architecture of the cerebellar folia is retained in many places. In the present study, we used immunocytochemistry with an immune serum raised against glutamic acid decarboxylase to further characterize the cis-dichlorodiammineplatinum-induced alterations of GABAergic neurons. The aim was to examine cerebellar development and to test for factors controlling the settling of GABAergic circuits. At all post-treatment intervals, most of the Purkinje and Golgi neurons and molecular layer interneurons showed stronger anti-glutamic acid decarboxylase immunoreactivity than in controls; this may have been due to altered fixation because of cis-dichlorodiammineplatinum-induced damages to the blood vessels; but could also reflect cellular retention of the enzyme, maybe due to cis-dichlorodiammineplatinum-induced damage of the microtubular apparatus. After seven days, large roundish immunoreactive varicosities were present in the molecular layer adjacent to the Purkinje cell dendritic poles. These varicosities, which were not observed in control animals, may be terminals of Purkinje cell axon recurrent collaterals contributing to the supraganglionic plexus, whose abnormal development would compensate for the reduced inhibitory inputs from inhibitory interneurons and/or Purkinje cells, which degenerated at early post-treatment intervals. At later post-treatment intervals (15 and 21 days), there were also alterations in the pericellular basket at the Purkinje cell axon hillock, which was poorly developed in or absent from the majority of cells. The finding was confirmed by morphological observation of basket cells in Golgi-Cox preparation and immunocytochemistry with an antibody raised against 200,000 mol. wt phosphorylated neurofilaments. It is concluded that early changes in anti-glutamic acid decarboxylase immunoreactivity of neurons may be due to a direct interference of the drug with the cellular metabolic pathways. The late anomalies in the anti-glutamic acid decarboxylase immunoreactivity appear to be secondary to changes in the tissue cytoarchitecture rather than being primary cis-dichlorodiammineplatinum-induced lesions of the cells.

The studies described have sought to determine what, if any, relationship exists between axons and the senile plaque, a hallmark histopathological feature of Alzheimer's disease. A double stain was performed on both early and late Alzheimer frontal cortex tissues in order to examine the interaction between axons stained with antibodies against the 200,000 mol. wt neurofilament subunit (NFP-200) of the axon cytoskeleton and Thioflavin-S, a fluorescent dye that stains plaques. Serial photomicrographs of plaques were taken and axon and plaque profiles were three-dimensionally reconstructed. Analysis of computer-processed images revealed that there were fewer axons within plaques than in regions lying one and two plaque distances away. When axons were observed passing through plaques, swelling and disruption of normal morphology was frequently present. Statistical analyses of axon counts within and around placques showed a gradient of axon density, with increased numbers occurring at progressive distances from the placque. Similar patterns were seen for early and late stages of the disease. The results of this study indicate that disruption of the axonal cytoskeleton may occur within the regions occupied by plaques.
