The purposes of this study were to determine the intervals when placental corticotrophic-releasing hormone (CRH) was most responsive to maternal cortisol. A sample of 203 women each were evaluated at 15, 19, 25 and 31 weeks gestation and followed to term. Placental CRH and maternal adrenocorticotropin hormone (ACTH), B-endorphin and cortisol were determined from plasma. CRH levels increased faster and were higher in women who delivered preterm compared with women who delivered at term (F3,603 = 5.73, p < .001). Simple effects indicated that CRH levels only at 31 weeks predicted preterm birth (F1,201 = 5.53, p = .02). Levels of cortisol were higher in women who delivered preterm at 15 weeks gestation (F1,201 = 4.45, p = .03) with a similar trend at 19 weeks gestation. Hierarchical regression suggested that the influence on birth outcome of maternal cortisol early in pregnancy was mediated by its influence on placental CRH at 31 weeks. Elevated cortisol at 15 weeks predicted the surge in placental CRH at 31 weeks (R = .49, d.f. = 1,199, Fchange = 61.78, p < .0001). Every unit of change in cortisol (microg/dl) at 15 weeks was associated with a 34 unit change of CRH (pg/ml) at 31 weeks. These findings suggested that early detection of stress signals by the placenta stimulated the subsequent release of CRH and resulted in increased risk for preterm delivery.

Proopiomelanocortin (POMC) contains several interesting, behaviorally active peptides. Release patterns of these fragments have been related to bizarre episodes of self-injurious behavior (SIB) among autistic individuals. Moreover, elevation in beta-endorphin (betaE) but not ACTH levels was associated with a positive response to an acutely administered, centrally acting opioid blocker among autistic individuals exhibiting SIB. In the present study, POMC fragments were measured in 12 self-injurious patients before and after long term (3 month) treatment with an opiate blocker naltrexone (NTX). POMC fragments were sampled from blood collected at the beginning of the baseline and placebo-controlled treatment phases of the study. Results indicated that the co-release (coupling) of POMC fragments were stable over time and the profile of POMC fragments in plasma predicted the effectiveness of a CNS acting drug in autistic subjects who self-injure.

Twelve rats received peripheral injections (20 micrograms/rat) of alpha-MSH, beta-endorphin or the vehicle solution and were subsequently tested for the motor and cardiac responses to repeated presentations of intense acoustic stimuli. Each subject received all treatments in a counterbalanced order with 3-day periods between each session. beta-Endorphin tended to decrease the amplitude of the habituated motor startle reflex, while alpha-MSH produced a slight increase in basal heart rate during the habituation session. Neither peptide had any effect on the cardiac response to intense acoustic stimulation. The effects of the two peptides were not directly antagonistic but they are consistent with the hypothesis that complex attentional processes were facilitated by MSH/ACTH fragments and inhibited by the endorphins.

Three oral doses (5, 10 and 20 mg) of an analog of ACTH 4-9 were compared with a vehicle control and d-amphetamine (10 mg). In a double-blind procedure, five men and five women were tested at weekly intervals with each treatment. In each session, four visual event-related potentials (ERPs) were recorded at hourly intervals. Visual ERPs were averaged from the electroencephalogram recorded from the left and right hemisphere. Dosage, time after administration, hemisphere of the brain and sex of the subject influenced the ERP. The ACTH 4-9 analog decreased amplitude of P100 but increased integrated activity of the ERP. This effect peaked at 60 minutes then "recovered." The effects of the peptide were more pronounced with doses of 5 and 10 mg, in the right hemisphere of men and in the left hemisphere of women. The findings indicated that the ACTH 4-9 analog influenced components of ERP commonly related to the perceptual/attentional state of the organism in a sexually dimorphic manner.

Adrenocorticotropic Hormone (ACTH), Melanocyte-Stimulating Hormone (MSH), and related peptides have been shown to have several neurogenic effects: alteration of cerebral protein synthesis, RNA synthesis, protein phosphorylation, and neurotransmitter turnover. Furthermore, there appears to be an ACTH containing circuit in the CNS which originates in the arcuate nucleus. Changes in concentration of the peptides in this family have been shown to alter electrophysiology, neuromuscular function, and behavior (e.g., grooming, learning) in infrahuman subjects. These findings suggest that the neuropeptides MSH and ACTH influence the capacity of an organism to efficiently evaluate information and influence the affective functioning of humans.

In two experiments the effects of the pituitary peptide α-MSH, the hypothalamic tripeptide MIF-I (P-L-G-NH2) and the pineal hormone melatonin were investigated on the attenuation of morphine analgesia measured by a tail flick test. In Experiment 1, α-MSH had minimal effect on morphine analgesia, whereas, MIF-I and melatonin clearly delayed the onset of morphine analgesia, and melatonin also shortened the duration of analgesia. Experiment 2 was designed to investigate the possible synergistic effect of MIF-I and melatonin. The combined treatment of MIF-I and melatonin significantly delayed the onset of morphine analgesia, and melatonin alone shortened the duration of analgesia. The relationshps among the pituitary, hypothalamus and the pineal for the modulation of pain and response to morphine were discussed.

In two experiments the effects of melatonin were investigated on the motor, cardiac and emotional components of the startle reflex induced by an intense acoustic stimulus. Melatonin inhibited the motor movements and significantly decreased the HR responses of rats pretested with these procedures. Melatonin decreased the HR responses to stimulation whereas the HR responses of the rats treated with the vehicle solution increased. Melatonin treatment also inhibited the startle-induced defecation compared with the vehicle injection. These findings suggest that melatonin exerts inhibitory effects on the CNS, on sensory reflex and on stress responses.

Learning and attention were examined in rats after injections of one of several molecules related to adrenocorticotropic hormone (ACTH) and melanocyte-stimulating hormone (MSH). The initial phase of the learning process was linearly related to the length of the peptide with the smallest fragment (MSH/ACTH 4-10) improving learning the most and the largest molecule (ACTH 1-24) exerting no effect. Later stages of the learning problem, which were sensitive to the attentional state of the organism, resulted in U-shaped relations with the length of the same peptides. Enhancement of attention was significant only for the MSH compounds. These data indicate that some behaviors may be influenced as a function of the redundant information contained in the molecule while other behaviors may be discretely related to the specific conformation of the molecule.
