Overexpressing a heat shock factor gene ( TaHsfA6b T) from wheat provides thermotolerance in barley by constitutive expression of heat and other abiotic stress-response genes. Temperature is one of the most crucial abiotic factors defining the yield potential of temperate cereal crops, such as barley. The regulators of heat shock response (HSR), heat stress transcription factors (Hsfs), modulate the transcription level of heat-responsive genes to protect the plants from heat stress. In this study, an Hsf from wheat (TaHsfA6b) is overexpressed in barley for providing thermotolerance. Transgenic barley lines overexpressing TaHsfA6b showed improvement in thermotolerance. The constitutive overexpression of a TaHsfA6b gene upregulated the expression of major heat shock proteins and other abiotic stress-responsive genes. RNA-seq and qRT-PCR analysis confirmed the upregulation of Hsps, chaperonins, DNAJ, LEA protein genes, and genes related to anti-oxidative enzymes in transgenic lines. Excessive generation and accumulation of reactive oxygen species (ROS) occurred in wild-type (WT) plants during heat stress; however, the transgenic lines reflected improved ROS homeostasis mechanisms, showing lesser ROS accumulation under high temperature. No negative phenotypic changes were observed in overexpression lines. These results suggest that TaHsfA6b is a regulator of HSR and its overexpression altered the expression patterns of some main stress-related genes and enhanced the thermotolerance of this cereal crop.

Five RALF (rapid alkalinization factor)-like genes, named ScRALF1 to 5, were isolated from fertilized ovule and ovary cDNA libraries of Solanum chacoense. They showed high sequence similarities with the RALF protein sequence from Nicotiana tabacum, and exhibited the characteristic architecture of RALF polypeptides. All ScRALFs were moderately to highly expressed at some stage of fruit maturation. ScRALF1 and ScRALF3 were predominantly expressed in ovaries and larger fruits, while ScRALF2, ScRALF4, and ScRALF5 were also expressed in other tissues, indicating that while some RALFs may be involved in fruit maturation, others could be involved in other developmental processes. Wounding or treatment of plants with growth regulators involved in plant defense responses had no significant impact on the mRNA level of any of these genes. These results suggest and support previous data showing that RALF peptides are more likely to act as a small peptide involved in plant development than in defense responses.

The mRNA expression of the Solanum chacoense Ovule Receptor Kinase 17 (ScORK17), a receptor kinase of the LRR-VI subfamily, is highly specific to the female reproductive tissues. No LRR-VI subfamily members in any plant species have yet been attributed a function. A phylogenetic tree inferred using the kinase domain of LRR-VI subfamily members separated the family into two clades: one containing an average of 8.2 LRR per protein and a second clade containing an average of 2.7. In situ hybridization analyses showed that the ScORK17 signal was mainly detected in the single ovule integument and in the endothelium. Transient expression analysis also revealed that ScORK17 was N-glycosylated in planta. Overexpression of ScORK17 in S. chacoense did not produce plants with an altered phenotype. However, when heterologous transformation was performed with a full-length ScORK17 clone in A. thaliana, the resulting transgenic plants showed reduced seed set, mainly due to aberrant embryo sac development, thus supporting a developmental role for ScORK17 in ovule and seed development.
