The intestinal microbiota has attracted tremendous attention in the field of the poultry industry due to its critical role in the modulation of nutrient utilization, immune system, and consequently the improvement of the host health and production performance. Accumulating evidence implies intestinal microbiota of laying hens is a potential mediator to improve the prevalent issues in terms of egg quality decline in the late phase of laying production. However, the regulatory effect of intestinal microbiota on egg quality in laying hens remains elusive, which requires consideration of microbial baseline composition and succession during their long lifespans. Notable, although Firmicutes, Bacteroidetes, and Proteobacteria form the vast majority of intestinal microbiota in layer hens, dynamic intestinal microbiota succession occurs throughout all laying periods. In addition to the direct effects on egg safety, intestinal microbiota and its metabolites such as short-chain fatty acids, bile acids, and tryptophan derivatives, are suggested to indirectly modulate egg quality through the microbiota-gut-liver/brain-reproductive tract axis. These findings can extend our understanding of the crosstalk between intestinal microbiota and the host to improve egg quality and safety. This paper reviews the compositions of intestinal microbiota in different physiological stages of laying hens and their effects on egg quality and proposes that intestinal microbiota may become a potential target for modulating egg quality and safety by nutritional strategies in the future.

The current study was performed to explore the effects of dietary supplementation of Saccharomyces cerevisiae hydrolysate (SCH) on growth performance, immune function, and intestinal health in broiler chicken. A total of 300 Ross 308 male broilers (1-day-old) were randomly assigned to 2 dietary treatments including a basal diet (control group), and a basal diet supplemented with SCH feed additive (500 mg/kg in starter and grower phase, and 250 mg/kg in finisher phase). Each treatment had 6 replicates with 25 birds each. The results showed that the addition of SCH promoted growth during d 15 to 28 (P0.05). Although the addition of SCH had no significant effect on the intestinal relative indexes, it significantly increased the jejunum villus height (VH) and the ratio of villus height to crypt depth (VCR) of jejunum, and decreased the crypt depth (CD) of ileum (P0.05). Furthermore, SCH addition significantly downregulated the mRNA expression of immunomodulatory genes (TNF-α, IL-1β, and IL-6), and upregulated the tight junction genes (ZO-1 and Claudin-1) (P0.05). High throughput sequencing analysis of bacterial 16S rRNA revealed that dietary SCH supplementation altered cecum microbiota. Alpha diversity analysis showed that a higher bacterial richness in cecum of broilers fed with SCH. The composition of cecum microbiota regulated by SCH addition was characterized by an increased abundance of Firmicutes and a reduced abundance of Bacteroidetes. At the genus level, dietary SCH resulted in a decrease of Bacteroides and an increase of short-chain fatty acids (SCFA) -producing bacteria including Lactobacillus and Faecalibacterium. Taken together, dietary SCH supplementation can stimulate the growth of broilers by regulating the intestinal immunity and barrier function, and improving the intestinal morphology, which may be related to the enhancement of bacterial diversity and the changes of intestinal microbial composition.

The present study determined the effects of in ovo feeding (IOF) of N-acetyl-L-glutamate (NAG) on early intestinal development and growth performance of broilers. A total of 702 fertile broiler eggs were randomly divided into 3 treatments: 1) non-punctured control group, 2) saline-injected control group, and 3) NAG solution–injected group (1.5 mg/egg). At 17.5 D of incubation, 300 μL of each solution was injected into each egg of injected groups. Results indicated that the hatchability and healthy chicken rate were not affected by NAG injection (P > 0.05). The objective of this study was to evaluate the effect of dietary chloride (Cl) levels on performance, eggshell mechanical quality, and ultrastructure in layers based on the substitution of sodium chloride (NaCl) by sodium sulfate (Na2SO4). Three hundred sixty Jing Brown laying hens aged 43 wk were randomly divided into 5 groups and fed with corn–soybean meal diets containing 0.06, 0.10, 0.15, 0.20, and 0.25% total Cl inclusion. Every group had 8 replicates of 9 birds each. The feeding trial lasted for 12 wk. The results showed dietary 0.06% Cl due to complete substitution of NaCl by Na2SO4, depressed performance (P < 0.05) from 45 to 54 wk of age, increased serum creatinine level (P < 0.05), and caused visible renal tubular atrophy. Dietary Cl levels quadratically affected breaking strength, thickness, and weight of eggshell (P < 0.05). Better eggshell quality could be obtained when NaCl was partly replaced by Na2SO4 in laying hen diets maintaining Cl level at 0.10 or 0.15%. Moreover, the eggshell ash content was affected by Cl levels in a quadratic (P < 0.001) manner, with higher values observed in the 0.10 and 0.15% Cl groups (P < 0.05). Besides, the eggshell ultrastructural data showed that the total thickness and effective thickness significantly increased (P < 0.05) and mammillary thickness decreased (P < 0.05) in the group of dietary 0.15% Cl compared with the groups of 0.06 and 0.25% Cl. In conclusion, the complete substitution of dietary NaCl by Na2SO4 may induce Cl deficiency and depress laying performance and eggshell quality. Na2SO4 could partly replace NaCl in diets for laying hens (43–54 wk of age) without adverse effects on performance at the dietary Cl level from 0.10 to 0.25%. Better eggshell quality could be obtained when NaCl was partly replaced by Na2SO4 in laying hen diets maintaining Cl level at ∼0.15%.

This study was conducted to determine the standardized ileal digestible amino acids (SID AA) and nitrogen-corrected apparent metabolizable energy (AMEn) contents of 6 wheats from different origins in China and incidentally to investigate the effects of exogenous xylanase addition on SID AA and AMEn determination in broiler chicks. A total of 480 chicks were divided into 48 cages of 10 birds each balanced for body weight and fed 8 types of diets in a completely randomized design (6 replicated cages per diet) from 21 to 26 d of age. The individual wheat constituted the only source of crude protein in a semi-purified experimental diet. A nitrogen-free diet was designed to estimate basal endogenous AA loss and determine the SID AA. This study was conducted to investigate the comparative effects of dietary supplementation with microalgae oil or fish oil on fatty acid composition, sensory quality, and overall acceptability of table eggs. A total of six hundred thirty, 30-week-old, Hy-Line Brown laying hens were allocated to 7 dietary treatments. Layers were fed with a control diet or the control diet supplemented with graded levels of docosahexaenoic acid (DHA; 1.25, 2.50, and 5.00 mg/g feed) derived from microalgae oil or fish oil. The feeding trial lasted 10 wk. Enrichment of eggs with DHA by dietary supplementation with microalgae oil or fish oil enhanced yolk DHA and total n-3 polyunsaturated fatty acid (PUFA) enrichment and reduced n-6-to-n-3 ratio in a dose-response manner, whereas the efficiency decreased (P 0.05). This study was designed to evaluate the effect of dietary Ser on performance, egg quality, serum indices, and ileal mucosal immunity in laying hens fed low crude protein (LCP), essential amino acids (EAA) balanced diets. A total of 480 Hy-Line Brown layers at 24 wk of age were randomly assigned into 5 dietary treatments with 8 replicates of 12 birds each. Treatments included a control diet (16.49% CP), and 4 LCP, EAA balanced diets (14.05% CP) supplemented with 0, 0.114%, 0.306%, 0.498% L-Ser, respectively. Dietary Ser supplementation linearly increased hen-day egg production (HDEP; P < 0.05) and decreased feed-to-egg ratio (P < 0.05) among LCP groups from wk 6 to 10, and the optimal HDEP of layers occurred at Ser level of 0.498%. At the end of wk 10, birds in the control had higher albumen height and thick white proportion than those fed the LCP diet without Ser addition (P < 0.05), and presented a lower yolk color score than all LCP groups (P < 0.05). Among LCP groups, serum total protein and globulin contents were significantly increased by dietary Ser addition at the levels of 0.306% and 0.498% (P < 0.05), and had a linear response to the supplemental Ser levels (P < 0.05). Furthermore, dietary 0.498% Ser supplementation significantly increased serum immunoglobulin G and immunoglobulin M contents (P < 0.05) and up-regulated the expression of mucin 2, secretory immunoglobulin A, and relevant glycosyltransferases of O-glycosylation in ileal mucosa (P < 0.05). The increased expression of proinflammatory cytokines IFN-γ and IL-1β induced by LCP diets (P < 0.05) was reversed following 0.498% Ser addition (P < 0.05). Collectively, dietary CP reduction by 2.44% could maintain the productive performance of layers when it was fortified with certain EAA, though poor albumen quality, and ileal inflammation were occurred. The addition of Ser to LCP diets improved performance probably through enhancing humoral and ileal mucosal immunity and attenuating the ileal inflammation of layers.

This study investigated the effects of dietary chloride (Cl) reduction on laying performance and eggshell quality by substitution of sodium bicarbonate (NaHCO3) or sodium sulfate (Na2SO4) for part of dietary sodium chloride (NaCl), and further explored its mechanism for improving eggshell quality. A total of 360 29-wk-old Hy-line Brown laying hens were randomly allocated to 5 dietary treatments, including a basal diet contained 0.33% NaCl (control group, 0.27% dietary Cl), and 4 experimental diets that contained 0.21% and 0.15% dietary Cl by substituting Na2SO4 or NaHCO3 for part of NaCl in the basal diet. No significant differences were observed in blood Na+, Cl−, K+ and Ca2+ levels and pH value as well as serum creatinine and uric acid contents among 5 treatments (P > 0.05). Dietary Cl reduction increased egg production and ADFI during wk 33 to 36, 37 to 40 and 29 to 40 of age and decreased feed conversion ratio during wk 37 to 40 of age (P < 0.05). The hens fed with diets containing 0.15% Cl increased eggshell breaking strength, thickness and weight ratio in wk 40 of age (P 0.05). Overall, dietary Cl reductions could improve laying performance and eggshell quality by substitution of NaHCO3 or Na2SO4 for part of NaCl, and there were no differences in the improvements between these two substitutes. The improved eggshell quality may be attributed to improved eggshell ultrastructure and increased supply of eggshell Ca2CO3.

Eggshell color is an important shell quality trait that influences consumer preference. It is also of particular importance with respect to sexual signaling and the physiological and mechanical properties of shell pigment. Pigments include protoporphyrin IX, biliverdin, and traces of biliverdin zinc chelates, with brown eggs being notably rich in protoporphyrin IX, the synthesis of which has a marked effect on the intensity of brown eggshell color. This pigment is initially synthesized in the eggshell gland within the oviduct of laying hens and is subsequently deposited throughout the cuticular and calcareous layers of brown eggshell. In this review, we describe the factors affecting brown eggshell color and potential targets for the regulation of pigment synthesis. Protoporphyrin IX synthesis might be compromised by synthetase-mediated pigment synthesis, the redox status of the female birds, and regulation of the nuclear transcription factors associated with δ-aminolevulinic acid synthetase1. We believe that this review will provide a valuable reference for those engaged in studying eggshell depigmentation.

The objective of this study was to evaluate the effects of dietary Bacillus subtilis supplementation and calcium (Ca) levels on performance, eggshell quality, intestinal morphology, and relative calbindin-D28k (CALB1) mRNA level of laying hens in the late phase of production. An experiment employing a 2 × 3 factorial arrangement of 3 levels of Ca (3.5, 4.0, and 4.5%) and the absence or presence of B. subtilis was carried out with a total of 576 Hy-Line Brown laying hens aged 72 to 79 wk. Every group had 8 replicates of 12 birds each. The results showed that 4.0 and 4.5% Ca levels improved (P < 0.05) apparent retention and serum Ca content of aged laying hens. Compared with the 3.5% Ca level, the 4.0% Ca level in diets increased (P < 0.05) thickness, eggshell weight, shell ratio, and eggshell Ca content of aged laying hens. Moreover, breaking strength, thickness, eggshell weight, shell ratio, eggshell Ca content, apparent retention of Ca in g/day, apparent retention of Ca in percent, villus height, villus height/crypt depth, serum Ca level, and relative CALB1 mRNA level of aged laying hens were all increased (P < 0.05) by B. subtilis supplementation in diets. The supplemental B. subtilis decreased feed conversion ratio (P = 0.001) significantly. In addition, there was an interaction effect between increased Ca levels from 3.5 to 4.5% and B. subtilis supplementation on crypt depth in the duodenum (P < 0.05). In conclusion, we found that both the increase in dietary Ca level from 3.5 to 4.5% and B. subtilis supplementation could enhance intestinal Ca absorption and improve eggshell quality of laying hens in the late phase of production (72-79 wk of age). Dietary supplementation of B. subtilis accompanying the 4.0% Ca level was appropriate in enhancement of eggshell quality.

We evaluated the effect of photoperiod on ovarian morphology, reproductive hormone secretion, and hormone receptor mRNA expression in layer ducks during the pullet phase. A total of 480 71-d-old Jinding layer ducks were randomly divided into 5 groups that received 6L (hours of light):18D (hours of darkness), 8L:16D, 10L:14D, 12L:12D, or 14L:10D, respectively. Each group had 6 replicates with 16 birds each. The photoperiod feeding trial lasted 80 d until 150 d of age. The age at first egg (AFE), the total number, and weight of eggs increased linearly with increasing photoperiods (P0.05); lower values of AFE occurred with photoperiods ≥8 h, whereas a higher total number and weight of eggs occurred with photoperiods ≥10 h, compared with 6L:18D (P0.05). Oviduct weight, ovary percentage, and initial and bare stroma (weight and percentage) increased quadratically with increasing photoperiods (P0.05), and 10.24, 10.01, and 10.10 h were the optimal photoperiods for oviduct weight, bare stroma (follicles ≥2 mm in diameter removed) weight, and bare stroma percentage, respectively, as calculated from reliable regression equations (R2 ≥ 0.5791). Compared with 6L:18D, 10L:14D had a higher total large white follicle weight, small yellow follicle number, and weight (P0.05). In addition, higher serum levels of follicle-stimulating hormone, luteinizing hormone, and progesterone were observed with ≥10-h photoperiods (P0.05), as were levels of hormone receptor mRNA expression in ovarian follicles (P0.05), with the highest values for both measures at 10L:14D. In the hypothalamus, mRNA expression of gonadotropin-releasing hormone increased in ≥8-h photoperiods, with the highest value at 10L:14D. In contrast, gonadotropin-inhibitory hormone increased in photoperiods ≥12 h (P0.05). In conclusion, an appropriate photoperiod led to early sexual maturity and improved the development of reproductive organs and ovarian follicles through effects on reproductive hormones and their receptors; 10 to 10.24 h is an adequate photoperiod for layer ducks during the pullet phase.

We investigated the effect of photoperiod on performance, ovarian morphology, reproductive hormones levels, and their receptors mRNA expressions in laying ducks. After adaption, 300 252-day-old Jinding laying ducks were randomly allocated to 5 groups, receiving 12L:12D, 14L:10D, 16L:8D, 18L:6D, or 20L:4D, respectively. Each treatment had 6 replicates of 10 birds each. The feeding trial lasted 8 wk. Egg production, egg mass, and ADFI increased linearly and quadratically with increasing photoperiods (P < 0.05), and the higher values of them occurred in photoperiods ≥ 16 h, compared with 12L:12D (P > 0.05). Initial and bare stroma weight increased quadratically, while total large white follicle (LWF) number and weight increased linearly and quadratically, with increasing photoperiods (P < 0.05). The higher values of them occurred in 16L:8D and 18L:6D treatments as well as the higher total LWF weight also occurred in 20L:4D, compared with 12L:12D (P > 0.05). Besides, 16.93 and 16.93 h were the optimal photoperiods for bare stroma (follicles ≥ 2 mm in diameter removed) weight and total LWF weight, respectively, calculated from reliable regression equations (R2 ≥ 0.5071). Compared with 12L:12D, the higher levels of estradiol, progesterone, follicle-stimulating hormone (FSH) as well as the higher expressions of estrogen, luteinizing hormone (LH) and progesterone receptors were observed in ≥16 h photoperiods (P < 0.05), while the higher LH level and FSH receptor expression only occurred in 16L:8D and 18L:6D (P < 0.05). In the hypothalamus, higher mRNA expression of gonadotropin-releasing hormone occurred in 16L:8D and 18L:6D groups (P < 0.05). Meanwhile, gonadotropin-inhibitory hormone and prolactin increased in 20-hour photoperiod (P < 0.05), and the latter may be due to theup-regulation of vasoactive intestinal peptide expression (P < 0.05). To sum up, an appropriate photoperiod could improve the performance and reproductive organ and ovarian follicles development through reproductive hormones and their receptors, and 16.56 to 10.93 h is an adequate photoperiod for laying ducks.

The objective of the current study was to investigate the effect of dietary β-alanine supplementation on growth performance, meat quality, antioxidant ability, carnosine content, and gene expression of carnosine-related enzymes in broiler chicks. We randomly assigned 540 1-day-old Arbor Acres broilers to 5 dietary treatments supplemented with 0 (control group), 250, 500, 1,000, or 2,000 mg/kg of β-alanine (mg β-alanine per kg feed). Each treatment included 6 replicates of 18 birds. The feeding trial lasted for 42 d. Dietary β-alanine supplementation linearly and quadratically increased the average daily gain (ADG) during the starting period (d 1 to 21, P = 0.02 and P = 0.002). The feed conversion ratio (FCR) decreased quadratically in response to dietary β-alanine supplementation during the starting and entire periods (P0.001 and P = 0.003, respectively). For the entire period, the predicted best FCR would be achieved when β-alanine was fed at a level of 1,100 mg/kg from quadratic regression. The concentrations of carnosine and β-alanine in breast muscle increased quadratically with dietary β-alanine supplementation (d 42, P0.001 and P = 0.001, respectively). The predicted dietary β-alanine level for highest breast carnosine content was 1,196 mg/kg. Dietary supplementation with β-alanine reduced the taurine concentrations in plasma (d 42, linear and quadratic, P 0.001). Breast muscle yield increased linearly and quadratically in response to dietary β-alanine addition (d 21, P = 0.017 and P = 0.007). Dietary supplementation with β-alanine quadratically reduced the shear force (P = 0.003), whereas a*45 min and a*24 h values increased quadratically in response to dietary β-alanine supplementation (d 42, P = 0.020 and P = 0.021, respectively). Dietary β-alanine addition quadratically enhanced the expression of carnosine synthase and taurine transporter mRNAs (P0.05). Overall, dietary β-alanine supplementation improved growth performance and carnosine content, ameliorated antioxidant capacity and meat quality, and upregulated the gene expression of carnosine synthesis-related enzymes in broiler chicks.

We investigated the effect of photoperiod on eggshell quality, bone quality characteristics and bone metabolism related enzymes and factors in laying ducks. After adaption, 300 Jinding laying ducks (252-day-old) were randomly divided into 5 treatments, receiving 12L (hours of light):12D (hours of darkness), 14L:10D, 16L:8D, 18L:6D or 20L:4D, respectively. Each group had 6 replicates of 10 birds each. The feeding experiment lasted 8 wk. Compared with 12L:12D, the higher values of eggshell breaking strength occurred in ≥18 h photoperiods at the end of 6 wk, and in ≥16 h photoperiods at the end of 4 wk, with the common highest values in 18 h photoperiod (P ˂ 0.05). Besides, 18L:6D had higher values of ultimate load Fu and cortical cross-sectional area A in tibia, femur, and ulna (P ˂ 0.05), compared with 12L:12D. The higher values of proximal bone mineral content (BMC; tibia), distal BMC (ulna), total Ca (tibia), and cortical volumetric bone mineral density (vBMC; tibia and ulna) were observed in 16L:8D and 18L:6D treatments (P ˂ 0.05). Meanwhile, 18 h photoperiod group had the higher proximal BMC (femur) and total Ca in ulna (P ˂ 0.05). In serum, compared with 12L:12D group, the higher ALP activity occurred in ≥16 h photoperiods (0:00 and 18:00), with the highest values in 18L:6D treatment (P ˂ 0.05); the higher values of TGF-β (6:00) and OC (6:00 and 18:00) were simultaneously observed in 18 h photoperiod (P ˂ 0.05). Moreover, values of trACP activity, TNF-α and IL-6 contents decreased in ≥18 h photoperiods at 0:00 (P ˂ 0.05), compared with 12L:12D group. To sum up, an appropriate photoperiod could improve eggshell quality, bone strength and mineral content through increasing osteogenesis during the light time and decreasing resorption activity during the dark, and 18 h is an adequate photoperiod for the eggshell and bone quality of laying ducks.

This study aimed to evaluate the effect of photoperiod on growth performance and bone development in layer ducks during the pullet phase. A total of 480 11-wk-old Jinding layer ducks were randomly divided into 5 groups, receiving 6L (light): 18D (dark), 8L: 16D, 10L: 14D, 12L: 12D, 14L: 10D, respectively. Each group had 6 replicates of 16 birds each. The feeding trial lasted 80 d until 150 d of age. The results showed that 8L: 16D increased body weight (BW) and average daily gain (ADG; P0.05) compared with 6L:18D, whereas average daily feed intake, feed conversion ratio, and mortality rate had no difference among all treatments (P0.05). The midpoint perimeter (tibia and femur) and work to fracture (Wu; tibia) increased quadratically (P0.05), in response to photoperiod increment. Compared with 6L:18D, 8L:16D had higher tibia midpoint perimeter, ultimate load (Fu), and cortical cross-sectional area (P0.05), accompanied with higher content of total Ca, distal bone mineral content (BMC), and cortical volumetric BMC (vBMC; P0.05). A linear increase was observed in trabecular volumetric bone mineral density (vBMD) and vBMC (tibia and femur), elastic energy (femur), work to fracture Wu and young modulus of elasticity E (tibia) in response to photoperiod increase (P0.05), and treatment with 14L:10D had higher ultimate load Fu and total Ca of tibia (P0.05). Besides, estradiol and testosterone levels (serum), estrogen receptor gene mRNA expression (bone tissue), alkaline phosphatase and tartrate-resistant acid phosphatase activities, transforming growth factor-β, osteocalcin and tumor necrosis factor-α contents increased in ≥12 h photoperiods (P0.05). In summary, appropriate photoperiod increased BW, ADG, cortical bone generation, and bone mineralization, whereas only medullary bone formation increased with photoperiods. These results suggest that 8L:16D could be an adequate photoperiod for layer ducks' growth and bone development during the pullet phase.

The objective of this study consisting of 2 trials was to investigate the antioxidant role of conjugated linoleic acid (CLA) isomers (c9, t11-CLA and t10, c12-CLA) and the underlying mechanism by which they act in modulating redox status in a primary laying hen hepatocyte culture. In trial 1, the cytotoxicity of CLA isomers or linoleic acid (LA) (0, 25, 50, 100, 200, 400, 800 μmol/L) was evaluated by 3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyltetrazolium bromide (MTT) assay. The concentration of CLA isomers or LA (25, 50, 100 μmol/L) for proper antioxidant activity was evaluated by measuring the antioxidant enzyme activity. In trial 2, there were 5 groups: control group, cells were untreated; H2O2 group, cells were exposed to 4 mmol/L H2O2 for 2 h; c9, t11 or t10, c12 or LA group, cells were treated with c9, t11-CLA or t10, c12-CLA or LA for 24 h and then exposed to 4 mmol/L H2O2 for 2 h. Trial 1 showed that the non-toxic dose range for CLA isomers was 0 to 200 μmol/L. The optimum concentration of c9, t11-CLA and t10, c12-CLA for trial 2 was 100 μmol/L. In trial 2, pretreatment with t10, c12-CLA but not c9, t11-CLA attenuated the increase in reactive oxygen species (ROS) compared to hydrogen peroxide (H2O2) group (P < 0.05). t10, c12-CLA elevated the superoxide dismutase (SOD) and catalase (CAT) activities compared with the H2O2 group (P < 0.05). In addition, t10, c12-CLA up-regulated the mRNA expression of nuclear factor E2-related factor-2 (Nrf2) as well as its target genes, Cu-Zn superoxide dismutase (SOD1) and CAT (P < 0.05). Pretreatment with t10, c12-CLA but not c9, t11-CLA decreased Nrf2 protein expression in the cytoplasm and increased Nrf2 protein expression in the nucleus compared with the H2O2 group (P < 0.05). The results indicate that t10, c12-CLA exhibits a stronger antioxidant capacity than c9, t11-CLA in primary cultured laying hen hepatocytes. t10, c12-CLA increases the activity and mRNA expression of antioxidant enzymes via facilitating nuclear translocation of Nrf2.

The objective of this study was to evaluate the effects of dietary supplementation with Moringa oleifera leaf (MOL) on performance, carcass characteristics, meat quality, and oxidative stability of breast muscle in broilers. A total of 720 1-d-old male Arbor Acres birds were randomly divided into 6 dietary groups, which were fed a basal diet supplemented with 0, 1, 2, 5, 10, and 15% MOL, respectively. Each group had 6 replicates of 20 birds each. The feeding trial lasted for 42 d. The results showed dietary MOL supplementation linearly and quadratically decreased body weight and average daily gain (P0.01), and increased feed conversion ratio (P0.001). Abdominal fat decreased linearly and quadratically in response to the supplementation of MOL in diets, both on d 21 and 42 (P0.001). In breast muscle, dietary supplementation with MOL quadratically increased the contents of C18:2, C18:3n-3, C20:4, polyunsaturated fatty acids (PUFA), n-3 PUFA, n-6 PUFA (P0.01), and decreased thrombogenic index (TI; P = 0.019). Dietary inclusion of MOL improved meat color, evidenced by quadratically reduced b* (yellowness) values (45 min postmortem, P = 0.001; 24 h postmortem, P = 0.018) and increased a* (redness) values (24 h postmortem, P0.001). Besides, diets supplemented with MOL quadratically decreased malondialdehyde (MDA) levels in breast muscle during storage (P0.001). Plasma total anti-oxidative capacity, total superoxide dismutase, glutathione peroxidase activities increased quadratically (P0.01), whereas MDA decreased quadratically (P0.001), in response to dietary MOL supplementation. In summary, MOL could be used as a feed ingredient for broilers to improve PUFA contents, oxidative stability, color of breast muscle, and abdominal fat without adverse effects on growth performance, with an inclusion of 1.56% in the diets.

Beta-hydroxy-β-methylbutyrate (HMB), the metabolite of leucine, plays an important role in muscle protein metabolism. To investigate the effect of dietary HMB calcium (HMB-Ca) on growth performance, breast muscle development, and serum parameters in broiler chickens, a total of two hundred seventy 1-d-old Arbor Acres male broiler chicks were randomly allotted into 3 dietary treatments supplemented with 0, 0.05%, or 0.1% HMB-Ca during the starter (1 to 21 d) and grower (22 to 42 d) period. The results showed that broilers fed 0.1% HMB-Ca diet had higher ADG during the starter or the whole period, and gain 148 g more BW than the chicks fed the control diet at 42 d of age (P < 0.05). At 21 d of age, birds receiving 0.1% HMB-Ca had more breast muscle yield, less abdominal fat than the control, and more dressing percentage than birds fed the control or 0.05% HMB-Ca diet (P < 0.05). At 42 d of age, 0.1% HMB-Ca increased breast muscle yield than the control and decreased abdominal fat compared with the control or 0.05% HMB-Ca group (P < 0.05). In comparison with the control, feeding 0.1% HMB-Ca increased the triiodothyronine, thyroxine, triiodothyronine/thyroxine ratio and decreased the serum uric acid level at d 21 (P < 0.05). At 42 d of age, serum thyroxine level was elevated in the 0.05% HMB-Ca treatment, and the uric acid concentration was significantly decreased by the 0.1% HMB-Ca-supplemented diet (P < 0.05). Dietary HMB-Ca did not affect the growth hormone or insulin content. This study suggested that dietary supplementation of HMB-Ca improved growth performance, stimulated the breast muscle development, and decreased the abdominal fat deposition in broiler chickens, and the favorable effects were more pronounced in the starter phase. The growth promotion effect of HMB-Ca may be partly related to the increased serum thyroid hormones in broiler chickens.

The aim of this study was to compare the effects of stunning methods [electrical stunning (ES) vs. gas stunning (GS)] on lipid oxidation in broiler meat and to investigate possible mechanisms of lipid oxidation by measuring plasma variables, muscle reactive oxygen species (ROS), and TBA reactive substance (TBARS) concentrations, muscle fiber ratios, and mRNA levels of avian uncoupling protein (avUCP), avian adenine nucleotide translocator, and avian peroxisome proliferator-activated receptor-γ coactivator-1α (avPGC-1α). Arbor Acres broilers (n = 36) were not stunned (control) or were exposed to the following stunning treatments: 40% CO(2) + 21% O(2) + N(2); 60% CO(2) + 21% O(2) + N(2); 35 V, 47 mA, 400 Hz; 50 V, 67 mA, 160 Hz; and 65 V, 86 mA, 1,000 Hz. The ROS level in tibialis anterior (TA; P0.05) and the TBARS concentration in pectoralis major (PM; P0.01) were decreased in the GS groups compared with the ES groups at 45 min postmortem. However, the TBARS concentrations at 24 h postmortem in the PM and TA groups were not affected by stunning method (ES or GS). Compared with ES, GS caused greater expression of avUCP mRNA (1.47-fold in PM, and 2.41-fold in TA) and avPGC-1α mRNA (1.42-fold in PM, and 2.08-fold in TA). In conclusion, the upregulation of avUCP and avPGC-1α reduced ROS accumulation and lipid oxidation at 45 min postmortem in the skeletal muscles of broilers stunned with hypercapnic moderate oxygenation GS. However, these changes were not sufficient to cause a difference in meat lipid oxidation at 24 h postmortem between broilers stunned with hypercapnic moderate oxygenation GS and those stunned with low-current, high-frequency ES.

We studied the effects of dietary oxidized oils on serum lipid metabolic indices, estradiol level, and the gene expression of apolipoprotein B-100 and apolipoprotein VLDL-II in laying hens. Hy-Line Grey hens (280 ± 10 d old; average egg production, 90.0 ± 2.5%) were allotted to 1 of 4 dietary treatments, which were supplemented with 0 (control group), 1% (low oxidized group), 2% (moderately oxidized group), or 4% (highly oxidized group) thermally oxidized soybean oil. Each treatment contained 6 replicates, with 12 birds each. The feeding trial lasted for 30 d. Laying performance data were recorded weekly. Other indices were measured on d 0, 2, 6, 14, and 30 of the feeding trial. Hens in the moderately and highly oxidized groups had significantly lowered feed conversion ratios (P < 0.05). Those in the highly oxidized group also had decreased concentrations of serum very low density lipoprotein cholesterol on d 30 (P < 0.05) compared with the very low density lipoprotein cholesterol of hens in the moderately oxidized group. Hens in the moderately oxidized group had significantly increased expression of apolipoprotein B-100 (P < 0.05) from d 6 to 30. Consequently, hepatic triglyceride increased in this group on d 30 (P < 0.05). Serum triglyceride decreased in the moderately oxidized group on d 30 (P < 0.05), which may have been caused by the activation of peroxisome proliferator-activating receptor α. Serum estradiol levels were not significantly affected by oxidized oils at any time of measurement, but were significantly different between d 0 and 30 within the moderately oxidized group. This fact indicated that the effect of oxidized oils on apolipoprotein B-100 might partially be a cumulative result of the increasing secretion of estradiol. The results suggested that oxidized oil may affect the performance of laying hens through the regulation of apolipoproteins and estradiol.

The aim of this study was to compare the effects of gas stunning (GS) and electrical stunning (ES) on energy metabolism in Arbor Acres broilers. Thirty-six birds were slaughtered without stunning (control) or after stunning with the following treatments: 40% CO(2) + 21% O(2) + N(2) (G40%); 60% CO(2) + 21% O(2) + N(2) (G60%); 35 V, 47 mA, 400 Hz (E35V); 50 V, 67 mA, 160 Hz (E50V); and 65 V, 86 mA, 1,000 Hz (E65V). Muscle samples were obtained from the pectoralis major (breast) and tibialis anterior (leg) muscles in ambient temperature within 45 min postmortem and stored at -80°C. Blood pH decreased consistently with GS (G40% and G60%) compared with ES and the control (P0.01). No consistent differences were observed between GS and ES in the plasma variables, glycolytic potential, adenosine phosphates, or fiber intensities. Plasma lactate increased with G40% and E35V (P0.05), whereas plasma uric acid and urea nitrogen increased with E35V (P0.05) compared with the control. Compared with the control, the intensity of type IIB fibers decreased in broilers stunned with E35V and E50V (P0.05) and glycolytic potential increased (P0.01) with G60% in the breast muscle and decreased (P0.01) in the leg muscle with all the stunning treatments except for E50V. Energy decreased (lower adenosine triphosphate, higher adenosine monophosphate, and adenosine monophosphate:adenosine triphosphate ratio, P0.05) in breast muscle with G40% compared with ES at high currents (E50V and E65V). However, the adenosine phosphates with GS were not significantly different (P0.05) from ES at low current (E35V) in either breast or leg muscle. In conclusion, no essential difference in energy metabolism was found in broilers stunned with ES and GS when ES was based on low current and high frequency and GS was based on hypercapnic moderate oxygenation. This study indicated that G40% was potentially a superior stunning variable.

To investigate the effects of transport stress on blood metabolism, glycolytic potential, and meat quality in male Chinese Lingnan meat-type yellow-feathered chickens, three hundred sixty 72-d-old birds were randomly assigned to 1 of 5 treatments: unstressed control, 45-min (short-term) transport with 45-min (short-term) recovery, 45-min transport with 3-h (long-term) recovery, 3-h (long-term) transport with 45-min recovery, and 3-h transport with 3-h recovery. Each treatment consisted of 6 replicates with 12 birds each. All birds (except control group) were transported according to a designed protocol. Long-term transport decreased the plasma glucose level (P < 0.001), glycogen concentration in both breast (P = 0.023) and thigh (P = 0.012) muscles, and affected the size of IIb fibers in the tibialis anterior by decreasing the area (P = 0.031) and increasing the density (P = 0.046), drip loss (P = 0.045), and the a* (redness; P = 0.040) value in breast muscle. Long-term recovery after transport also decreased the plasma glucose level (P = 0.018) and showed a trend toward decreased breast glycogen concentration (P = 0.064). These results suggested that transport did not cause any noticeable changes in overall meat quality characteristics, although breast meat drip loss and meat color were affected. A long-term recovery from a period of feed and water deprivation also caused hypoglycemia and did not improve meat quality in Lingnan meat-type yellow-feathered chickens.

To evaluate the effect of flavin-containing monooxygenase 3 (FMO3) genotype and dietary choline supplementation on trimethylamine (TMA) metabolism in HyLine Brown laying hens, a 3 × 2 two-factorial arrangement was employed with FMO3 genotypes (AA, AT, and TT) and dietary choline supplemental levels (370 and 2,960 mg/kg of diet) as main effects. At 46 wk of age, 108 hens of AT genotype and 108 hens of TT genotype were randomly allotted to one of the 2 dietary treatments, and each dietary treatment consisted of 6 replicates with 9 birds each. A total of 24 hens with AA genotype was allotted to one of the 2 dietary treatments that consisted of 6 replicates with 2 hens. Hens were fed the diet with 370 mg/kg of choline supplementation for 1 wk of adaptation followed by a 6-wk trial period. Yolk TMA concentration was increased by dietary supplemental choline at 2,960 mg/kg (P0.05), and TT hens showed a higher TMA content in egg yolks than that in AA and AT hens (P0.05). Dietary supplementation of choline at 2,960 mg/kg increased the TMA concentration of cecal chyme (P0.05) and serum (P0.05). Hepatic FMO3 mRNA levels in hens were reduced by higher choline added to the diet (P0.05). The TMA and methimazole oxidation rate in AA hens was higher than those in AT and TT hens (P0.05). A higher choline diet decreased hepatic FMO3 activity by 33.99% (P0.05) and 61.39% (P0.05) toward TMA and methimazole, respectively. These results suggest that lower hepatic FMO3 activity caused by the mutation may be responsible for the genotype difference in the TMA metabolism. Exposure to a high dosage of dietary choline increased TMA synthesis in the cecum, suppressed activity of FMO3 in liver, and consequently aggravated the burden of TMA metabolism, especially in TT hens.

This study was designed to determine the effect of electrical stunning variables (low currents and high frequencies) on meat quality, glycolytic potential, and blood parameters in broilers. A total of 54 broilers were stunned with 9 electrical stunning methods for 18 s using sinusoidal alternating currents combining 3 current levels (35 V, 47 mA; 50 V, 67 mA; and 65 V, 86 mA) with 3 frequencies (160, 400, and 1,000 Hz). Samples for meat quality were obtained from the pectoralis major (PM) and musculus iliofibularis (MI), and samples for glycogen metabolism were taken from the PM and tibialis anterior muscle at 45 min postmortem. The use of high frequency reduced the shear value in PM (400 and 1,000 Hz vs. 160 Hz; P0.01) and cooking loss in MI (1,000 Hz vs. 160 and 400 Hz; P0.01). The shear value of PM decreased at high frequency (400 and 1,000 Hz) when current was high (50 V, 67 mA and 65 V, 86 mA; P0.01) but increased at high frequency (1,000 Hz) when current was low (35 V, 47 mA). Stunning with 1,000 Hz (vs. 160 Hz) caused low glycogen and glycolytic potential in PM (P0.05). Plasma corticosterone decreased (P0.05) at high currents (≥50 V, 67 mA) but was not affected by changes in frequency. Electrical current interacted with frequency in plasma glucose, redness 24 h postmortem, shear value (PM), pH 24 h postmortem (MI), and glycolytic potential (tibialis anterior; P0.05). This study indicated that high stunning frequencies (400 and 1,000 Hz) may improve meat quality without aggregating stress when the current was not too low (50 V, 67 mA).

This study aimed to investigate the effects of different CO(2) concentrations on blood variables, glycolytic potential (GP), and meat quality of hot-boned muscles in broilers. Thirty broilers were exposed to one of the following 5 gas mixtures for 90 s: 40% CO(2) + 30% O(2) + N(2) (control), 30% CO(2) + 21% O(2) + N(2) (G30%), 40% CO(2) + 21% O(2) + N(2) (G40%), 50% CO(2) + 21% O(2) + N(2) (G50%), and 60% CO(2) + 21% O(2) + N(2) (G60%). Samples were taken from the pectoralis major (PM), musculus iliofibularis (MI), and tibialis anterior muscles 45 min postmortem. The ultimate pH in both the PM (vs. G30% and G40%) and MI (vs. G40%) was decreased with G60% (P0.05), whereas drip loss in the PM (vs. G30%, P = 0.01) was increased with G60%. Drip loss in the MI (vs. control and G30%, P0.01) was increased with G50%. Lightness after 24 h in PM (vs. G30% and G40%, P0.01) was increased with G50%. In MI, lightness after 24 h was slightly decreased with G40% compared with the control (P0.10). The GP value in the PM was lower in the G30% and G40% than in G60% (P0.05), and the GP value in the tibialis anterior was the lowest in G30% (P0.01). Plasma corticosterone, plasma glucose, and meat quality (pH, lightness, redness, yellowness) 45 min postmortem were not affected by CO(2) levels (P0.05). In conclusion, stunning broilers with low CO(2) levels (30 and 40%) improved meat quality but had no advantage in animal welfare compared with high CO(2) levels (50 and 60%).

The effect of transport stress on superoxide production and adenosine phosphate concentration in addition to avian uncoupling protein (avUCP), avian adenine nucleotide translocator, and avian peroxisome proliferator-activated receptor-gamma coactivator-1alpha mRNA levels of skeletal muscles in broilers was investigated. Arbor Acres chicks (n = 360, 46 d old, males) were randomly allotted to 1 of 5 treatments: unstressed control, 45-min (short-term) transport with 45-min (short-term) recovery, 45-min transport with 3-h (long-term) recovery, 3-h (long-term) transport with 45-min recovery, and 3-h transport with 3-h recovery. Each treatment consisted of 6 replicates with 12 birds each. All birds (except control group) were transported according to a designed protocol. Transport time affected reactive oxygen species production in the thigh muscle (P < 0.05), adenosine triphosphate (ATP) content and energy charge (EC) in both breast and thigh muscles (P < 0.05 for all 4 comparisons), ATP:adenosine diphosphate (ADP) ratio in the breast muscle (P < 0.05), and avUCP mRNA levels in the thigh muscle (P < 0.05). Long-term transport increased (P < 0.05) reactive oxygen species production, ATP content, ATP:ADP ratio, and EC in the thigh muscle, but it decreased ATP content, ATP:ADP ratio, and EC in the breast muscle. Long-term transport increased avUCP mRNA in the thigh muscle (P < 0.05). Long-term recovery increased the ATP (P < 0.05) and ADP (P < 0.05) concentrations, avian adenine nucleotide translocator mRNA (P < 0.05), and avian peroxisome proliferator-activated receptor-gamma coactivator-1alpha mRNA (P < 0.05) in the thigh muscle, whereas EC decreased (P < 0.05) in the breast muscle. There were interactions between transport and recovery time on ATP (P < 0.05), EC (P < 0.05), and avUCP mRNA level (P < 0.05) in the thigh muscle. This study suggests that long-term transport accelerates muscle energy metabolism and lipid peroxidation. A long-term recovery may help alleviate cellular damage and maintain meat quality by reducing the rate of energy metabolism and scavenging of free radicals formed.
