Coccidiosis is a high-prevalence disease that annually entails huge costs for the poultry industry. Control of coccidiosis in poultry production is based on the use of coccidiostats and vaccines. However, along with the problem of drug resistance, there is a concern about food safety and drug residues in poultry products. The objective of this study was to evaluate the effect of sodium bisulfate (SBS) in comparison with monensin (M) and their combination (SBSM) effects on controlling coccidiosis in broilers. In a randomized design, 300 chickens (Ross 308) were divided into 5 treatments and 4 replications (15 birds per replicate). All birds, except the negative control (NC), were orally inoculated with 4 Eimeria species on 14 D of age. Treatments included were as follows: NC, an unsupplemented basal diet, nonchallenged; positive control, a basal diet unsupplemented, challenged with Eimeria spp; a basal diet supplemented with 5 g/kg of SBS; a basal diet supplemented with 1 g/kg of M; and a basal diet supplemented with 5 g/kg SBS and 1 g/kg M (SBSM). Oocyst shedding per gram (OPG) of the faecal sample from each experimental unit was counted on 5 to 14 D after inoculation. Two chicks from each experimental unit were euthanized to investigate intestinal lesions on day 5 after inoculation. The NC birds showed the highest BW gain and the lowest feed conversion ratio. The birds in the SBSM group had improved feed consumption compared with the M group in the prechallenge period (P < 0.05). All supplemented treatments resulted in a significant decrease in OPG. The M and SBSM treatments showed more efficacy than the SBS group (P < 0.05) in reducing OPG. There was a significant reduction in cecal lesions owing to supplementation with SBS, but the effect of SBS in the upper part of the intestine was lower than the M and SBSM groups (P < 0.05). Based on the results of this study, SBS has protective effects against coccidiosis in ceca, and the combination of M and SBS (SBSM) did not show any further improvement effect compared with M alone on the control of coccidiosis.

In experiment 1, 585 hatchlings were randomly placed in 15 replicate floor pens, (13 poults/pen) with recycled litter and provided feed and water ad libitum. Birds were randomly assigned to 3 dietary treatments from 1 to 12 wk: nonmedicated control, 0.15% functional oils, and 66-ppm monensin. From wk 13 to 20, each initial treatment group was further divided into 3 treatments-control (no additive), 0.15% of functional oils, or 20 ppm of virginiamycin to produce 9 different treatments, 5 replicate pens per treatment. Data on feed weights were collected weekly, and body weight bi-weekly. At termination (20 wk), birds were euthanized, and their meat was processed to determine mass of carcass sections and meat quality, while intestinal samples were collected for histology. In experiment 1, toms fed monensin or functional oils were 10.5 and 4.5% heavier (P < 0.05), respectively, than the controls at 12 wk. Birds fed monensin had a 4% improvement (P < 0.05) in feed conversion as compared to the other treatments. Neither virginiamycin nor the functional oils affected bird performance when fed from 13 to 20 wk. The jejunum villi surface area at 3 wk was most enhanced (P < 0.05) for the poults fed monensin. Supplementation with functional oils significantly reduced leg yield and thiobarbituric-acid reactive substances of white meat after 7 D of storage (P < 0.05). There were no effects on performance or carcass characteristics in experiment 2. While additional confirmatory studies are needed, functional oils in the diet of turkey toms may be a viable alternative to antibiotic growth promotants. The effects of a blend of encapsulated organic acids with essential oils (EOA) as an alternative to antimicrobial growth promoter (AGP) on growth performance and gut health of Eimeria spp./Clostridium perfringens (C. perfringens) in chickens infected with necrotic enteritis (NE) broilers was investigated. A total of 432 male Arbor Acres broilers (1-day-old) were randomly distributed into 6 treatment groups, namely noninfected negative control (A); NE-infected positive control (D); NE-infected broiler chickens fed a basal diet supplemented with 250 mg/kg bacitracin methylene disalicylate (BMD) plus 90 mg/kg monensin; and NE-infected broiler chicken fed 200; 500; and 800 mg/kg EOA (E, F, G, and H group). Feeding EOA at 200 and 500 mg/kg considerably improved the feed conversion ratio, reduced gut lesions, serum fluorescein isothiocyanate dextran level, and C. perfringens load in the caecum and liver of the NE-infected broiler chickens. This feed was similar to AGP. Furthermore, the increased villous height-to-crypt depth ratio and goblet cells counts, upregulated claudin-1, glucagon-like peptide-2 (GLP-2), insulin-like growth factor-2 (IGF-2) mRNA gene expression, downregulated occludin, zonula occludens-1 (ZO-1), toll-like receptor (TLR-4), interleukin (IL-1β), interferon γ (IFN-γ), TNF receptor-associated factor 6 (TRAF-6), tumor necrosis factor superfamily member 15 (TNFSF15), and Toll-interacting protein (Tollip) genes expression in the jejunum were observed in the NE-infected broiler chickens that received EOA at 200 and 500 mg/kg compared with those of the single NE-challenged groups without EOA supplementations (P < 0.05). The 16S analysis revealed that EOA supplemented with 200 or 500 mg/kg enriched relative abundance of Lactobacillus, unclassified_Lachnospiraceae, and Enterococcus, and carbohydrate metabolic pathways but suppressed unclassified_Erysipelotrichacease and organismal systems involved in the immune system (P < 0.05). Feeding EOA could alleviate NE-induced gut impairment and growth depression and modulate cecal microbiota composition, which has potential as antimicrobial alternatives.

Two experiments were conducted to evaluate the effects of dietary sophorolipids (SLs) supplementation as antibiotic alternatives on growth performance and gut health of chickens infected with Eimeria maxima. In experiment 1, 336 (zero-day-old) male broilers were used. The chickens were weighed and randomly allocated to the following 6 treatments groups with 7 chickens/cage and 8 cages/treatment: control group that received a basal diet (NC), positive control group that received a basal diet and was challenged with E. maxima (PC), PC+C18:1 lactonic diacetyled SL (SL1), PC+C18:1 deacetyled SL (SL2), PC+C18:1 monoacetyled SL (SL3), and PC+C18:1 diacetyled SL (SL4). Each SL (200 mg/kg feed) was added to the corresponding treatment group. In experiment 2, 588 (zero-day-old) male broilers were used. The chickens were randomly allocated to the following experimental groups with 10 or 11 chickens/cage and 8 cages/treatment: NC, PC, PC+ monensin at 90 mg/kg feed (MO), PC+SL1 at 200 mg/kg feed (SL1 200), PC+SL1 at 500 mg/kg feed (SL1 500), PC+SL4 at 200 mg/kg feed (SL4 200), and PC+SL4 at 500 mg/kg of feed (SL4 500). The chickens and feed were weighed at 0, 7, 14, 20, and 22 d to determine growth performance. In both experiments, all chickens except the NC group were orally infected with E. maxima (10,000 oocysts/chicken) at d 14. One chicken per cage was euthanized at d 20 to sample jejunal tissue to measure lesion scores, cytokines, and tight junction (TJ) proteins. Excreta samples were collected daily between d 20 and 22 to measure oocyst numbers. Data were analyzed using Mixed Model (PROC MIXED) in SAS. In experiment 1, SLs did not affect the growth of broiler chickens, but SL4 decreased (P0.05) the lesion score and oocyst number compared to PC chickens. In terms of cytokines and TJ protein gene expression, SLs increased (P0.05) IL-1β, IL-6, IL-17F, IL-4, IL-13, occludin, and ZO1 levels compared to PC chickens. In experiment 2, monensin increased (P0.05) body weight, and decreased (P0.05) the lesion score and oocyst number compared to the PC group. SL4 500 increased (P0.05) average daily gain and feed conversion ratio but decreased (P0.05) lesion score and fecal oocyst number. SL4 decreased (P0.05) IL-6, IL-17F, TNFSF-15, IL-2, and IL-10 levels but increased (P0.05) occludin and ZO-1 levels. Overall, dietary SL supplementation, especially SL4, improved growth and gastrointestinal functionality of young broiler chickens, demonstrating significant potential as an antibiotic alternative.

The aim of this study was to investigate the effect of dietary supplementation with nisin alone or in combination with salinomycin or monensin on broiler chickens in terms of growth performance, selected blood parameters, digestive enzyme activity, apparent nutrient digestibility, and tibiotarsus mineralization, as well as selected gastrointestinal tract (GIT) organ weights, intestinal length, and central immune organ weights. Two independent experiments, each including 400 one-day-old female Ross 308 chicks differing in ionophore coccidiostats, i.e., salinomycin and monensin supplementation, were conducted. The following treatments were applied: experiment 1: NA—no additives, SAL—salinomycin (60 mg/kg diet), NIS—nisin (2,700 IU/kg diet), SAL+NIS—salinomycin (60 mg/kg diet) and nisin (2,700 IU/kg diet); experiment 2: NA—no additives, MON—monensin (100 mg/kg diet), NIS—nisin (2,700 IU/kg diet) and MON+NIS—monensin (100 mg/kg diet) and nisin (2,700 IU/kg diet). The addition of nisin with or without ionophores to the birds’ diet improved broiler growth performance in terms of BWG and FCR (days 1 to 14) and BWG and FI (15 to 35 d; 1 to 35 d). Salinomycin showed effects similar to those of nisin influence on growth performance (1 to 35 d), while monensin supplementation resulted in lower BWG. Moreover, no additive effect between nisin and ionophores was observed. Nisin and salinomycin had no influence on the serum concentration of selected hormones and other blood biochemical parameters except glucose, which was reduced by nisin. A decrease in lipase activity was observed during nisin and salinomycin supplementation, while the apparent ileal digestibility of fat was not affected. However, the digestibility of crude protein increased with nisin administration. Additionally, the effects of nisin on decreasing the weight and length of GIT segments were observed. Supplementation with nisin and monensin was not associated with a negative impact on tibiotarsus mineralization and the immune organ index. This study suggests that nisin may be used in broiler nutrition as a growth promotor, with no negative influence on the bird's metabolism or immune status.

Poultry production faces several threats and challenges, one of the most important of which is avian coccidiosis which causes annual losses exceeding US$ 3 billion. Discovering new drugs or combinations of existing anticoccidials has become inevitable to overcome the emergence of coccidiosis resistance. This study evaluated a new combination of maduramicin and diclazuril in comparison to the well-known product Maxiban72 which consisted of narasin and nicarbazin, and the single effect of monensin as treatments for avian coccidiosis. A total of 750 1-day-old Indian River broiler chicks were allocated equally into 5 experimental groups with 6 replicates each as follows: 1) negative unchallenged control group (NC) fed the basal diet; 2) positive control group (PC) received the basal diet and inoculated with Eimeria; 3) PC + 100 mg monensin sodium (Atomonsin)/kg diet (MS); 4) PC + 5 mg maduramicin ammonium (Madramycin) + 2.5 mg diclazuril (Atozuril)/kg diet (MMD); and 5) PC + 40 mg narasin + 40 mg nicarbazin (MaxibanT72)/kg diet (NN). Anticoccidials improved (P < 0.01) growth performance, dressing (%) and carcass yield of inoculated birds compared to untreated-inoculated ones. Erythrogram and leukogram parameters were affected by Eimeria challenge. Total protein, globulin, cholesterol, triglycerides, superoxide dismutase and glutathione peroxidase levels in PC birds' serum were reduced (P < 0.05) while their values of liver enzymes, malondialdehyde and catalase were elevated (P < 0.01) when compared to NC ones. Serum immunoglobulin A, and jejunal gene expressions of interleukin-6 and interferon gamma were increased (P < 0.05) in PC group compared to NC group. Anticoccidial drugs restored values of the aforementioned biomarkers near to those of NC. Jejunal architecture in inoculated birds was improved by the anticoccidial treatments in MS, MMD, and NN. Fecal oocyst counts were significantly reduced in MMD, NN, and MS groups compared to PC group. Conclusively, although all examined anticoccidial drugs were effective in treating Eimeriosis, the anticoccidial combinations in MMD and NN groups were more effective than the single administration of MS in treating avian coccidiosis.

The turnover of intestinal epithelial cells is a dynamic process that includes adequate cell proliferation and maturation in the presence of microbiota and migration and seeding of immune cells in early gut development in chickens. We studied the effect of yeast-derived macromolecules (YDM) on performance, gut health, and immune system gene expression in the intestine of broiler chickens. One thousand eighty 1-d-old birds, with 60 birds per pen and 6 pens per treatment, were randomly assigned to 3 treatment diets; a diet containing monensin (control), control diet supplemented with bacitracin methylene disalycylate (BMD), and BMD diet supplemented with YDM. Feed intake, BW, mortality, ileum histomorphology, and gene expression of Toll-like receptors (TLR2b, TLR4, and TLR21), cytokines [interferon (IFN)-γ, IFN-β, IL-12p35, IL-1β, IL-6, IL-10, IL-8, IL-2, IL-4, and transforming growth factor (TGF)-β4], and cluster of differentiation (CD)40 in the ileum, cecal tonsil, bursa of Fabricius, and spleen were assessed. No significant overall difference in performance in terms of feed intake, BW gain, and G:F was observed among treatments (P > 0.05). The YDM diet resulted in significantly higher villi height and villi height:crypt depth ratio compared with BMD and control diets (P < 0.05). A significantly lower mortality was observed in the YDM treatment compared with both control and BMD treatments. Compared with the control, gene expression analysis in YDM treatment showed no major change in response in the ileum, whereas higher CD40, IFN-β, IL-β, IL-6, TGF-β4, IL-2, and IL-4 in the cecal tonsil; TLR2b, TLR4, TLR21, and TGF-β4 in the bursa of Fabricius; and TLR4, IL-12p35, IFN-γ, TGF-β4, and IL-4 in the spleen was observed (P < 0.05). In conclusion, supplementation of YDM supports pro- and anti-inflammatory cytokine production via T helper type 1 and 2 (Th1 and Th2) cell-associated pathways both locally and systemically with a stronger additive effect in the cecal tonsil in the presence of BMD in the diet of chickens.

A study was conducted to assess the effect of yeast-derived carbohydrates (YDC) on performance and innate immune responses of broiler chickens. In total, 1,080 one-day-old birds were randomly assigned to one of 3 dietary treatments (n = 360): a standard broiler diet containing monensin (control), control + bacitracin methylene disalycylate (BMD), and YDC treatment (control + YDC at 0.02%, 0.01%, and 0.005% for starter, grower, and finisher, respectively). Weekly BW, feed intake (FI), and feed conversion ratio (FCR) were recorded. Immune organ weights, gut morphology, gene expression, heterophil:lymphocyte (H:L), and serum IgG were determined at d 42. No significant difference in FCR, FI, and mortality was observed among treatments. However, BW gain in starter phase was higher in control and YDC treatments compared with BMD treatment. Ileal villi height, crypt depth, and their ratio were not significantly different among treatments, whereas villi width was lower in control and YDC treatments compared with BMD treatment. The number of goblet cells per unit area in the ileum was lower in BMD treatment compared with control and YDC treatments. Expression of TLR2b and IL-6 in the ileum and cecal tonsils was not significantly different among treatments (P > 0.05). Expression of TLR4 was downregulated in YDC treatment compared with control in the ileum. Expression of IL-12p35 and IFN-γ were downregulated in the YDC treatment only in the cecal tonsils. Compared with the control, the expression of IL-10 in both the ileum and the cecal tonsils was downregulated in YDC treatment. Serum IgG and H:L ratio were lower and higher, respectively, in the YDC treatment compared with control and BMD treatments. In conclusion, dietary inclusion of YDC affected intestinal cytokines anti-inflammatory profile on a gut location associated immune pathways manner, suggesting different immune pathways that require further studies in this field.

Aflatoxins (AF) are a major problem in broiler production and are significant economic and public health burdens worldwide. A commercial sodium bentonite (Na-B) adsorbent was used to prevent the effect of AF [50 µg of aflatoxin B₁ (AFB₁)/kg of feed] in broiler productivity, biochemical parameters, macroscopic and microscopic liver changes, and AFB₁ liver residues. The influence of Na-B (0.3%) and monensin (MON, 100 mg/kg), alone or in combination, was investigated in depth. The dietary treatments were as follows: treatment (T) 1: basal diet (B); T2: B + MON; T3: B + Na-B; T4: B + Na-B + MON; T5: B + AFB₁; T6: B + AFB₁ + Na-B + MON; T7: B + AFB₁ + MON; T8: B + AFB₁ + Na-B. Birds were fed dietary treatments for 28 d (d 18 to 46). No significant differences (P < 0.05) were observed among treatments with respect to broiler performance, biochemical parameters, or relative liver weights. With the exception of T8, all livers showed histopathological alterations, with accumulation of fat vacuoles. The normal appearance of livers from T8 showed the protective effect of Na-B against aflatoxicosis. The residual AFB₁ levels in livers from T5 to T8 ranged from 0.2 to 1.0 ng/g and were higher in livers from T6 (P < 0.05). Results of this study indicate a competition between AFB₁ and MON for adsorption sites on Na-B when feed contains low levels of the toxin, indicating a nonselective adsorption capacity of this particular Na-B. In addition, significant levels of AFB₁ in livers indicate that this determination is an important technique not only for diagnosis of aflatoxicosis in broilers, but also for quality control of avian products.

In July 1971, the polyether ionophorous antibiotic monensin was introduced in the United States for the control of coccidiosis in poultry. At that time, prospects for new anticoccidial agents were not good. Amprolium had enjoyed several years of use, but many other compounds had been abandoned as resistance to them developed. After the introduction of monensin, most commercial broilers were medicated with the drug and it is still widely used for this purpose today. Apart from in poultry, monensin is also used to control coccidiosis in game birds, sheep, and cattle. Indeed, more animals have been medicated with ionophores, such as monensin, for control of disease than any other medicinal agents in the history of veterinary medicine. In this review, we discuss the discovery, mode of action, and efficacy of monensin, together with matters of importance to the poultry industry such as commercial use, drug resistance, toxicity, pharmacology and residues, host immunity to coccidiosis, and effects in other avian species.

Tiamulin hydrogen fumarate is a semisynthetic derivative of the diterpene antibiotic pleuromutilin used in poultry medicine to treat mainly Mycoplasma- and Brachyspira-related diseases. Its use over 30 yr has not generally increased the development of resistance to these pathogens but occasionally resistant isolates are encountered. Tiamulin administered at therapeutic levels is relatively quickly absorbed, metabolized in the liver, and eliminated from the body of the bird after a withdrawal period of 72 h, and as a result, meat products can be safely consumed. A zero withdrawal period for eggs has been granted in several European Union states. When administered with different drugs, tiamulin has been shown to have an enhanced activity with the tetracyclines. There is a strong interaction, even death, with the ionophore anticoccidials monensin, narasin, and salinomycin when tiamulin is used at therapeutic levels, but this is dose-related and low doses do not interact. It is thought to be caused by the preferential metabolism of tiamulin in the liver resulting in a build up of the ionophore leading to clinical signs of overdosage. Tiamulin shows a milder interaction, such as temporary growth depression, with maduramicin and semduramicin but is compatible with lasalocid. Although tiamulin shows small benefits in improving performance in healthy animals, its main production benefit is in the face of infection, as a true therapeutic antibiotic.

Akşit, Hasan (Balikesir Author), Essential oil of oregano (OEO) has proven to be a potential candidate for controlling chicken coccidiosis. The aim of the current study is to determine whether OEO and an approved anticoccidial, monensin sodium (MON), as in-feed supplements could create a synergism when combined at low dosages. Day-old broiler chickens were separated into six equal groups with six replicate pens of 36 birds. One of the groups was given a basal diet and served as the control (CNT). The remaining groups received the basal diet supplemented with 100 mg/kg MON, 50 mg/kg MON, 24 mg/kg OEO, 12 mg/kg OEO, or 50 mg/kg MON + 12 mg/kg OEO. All of the chickens were challenged with field-type mixed Eimeria species at 12 d of age. Following the infection (i.e., d 13 to 42), the greatest growth gains and lowest feed conversion ratio values were recorded for the group of birds fed 100 mg/kg MON (P < 0.05), whereas results for the CNT treatment were inferior. Dietary OEO supplementations could not support growth to a level comparable with the MON (100 mg/kg). The MON programs were more efficacious in reducing fecal oocyst numbers compared to CNT and OEO treatments (P < 0.05). Serum malondialdehyde and nitric oxide concentrations were decreased (P < 0.01), whereas superoxide dismutase (P < 0.05) and total antioxidant status (P < 0.01) were increased in response to dietary medication with MON and OEO. All MON and OEO treatments conferred intestinal health benefits to chickens by improving their morphological development and enzymatic activities. The results suggest that OEO supported the intestinal absorptive capacity and antioxidant defense system during Eimeria infection; however, it displayed little direct activity on the reproductive capacity of Eimeria. This might be the reason for inferior compensatory growth potential of OEO compared to that MON following the challenge. This study was conducted to examine the effects of fructooligosaccharide (FOS) supplementation on growth performance, lymphoid organ weight, intestinal morphology, and immunological status in broilers (n=180) challenged with Salmonella Enteritidis lipopolysaccharides (LPS). Birds were randomly assigned into a 3×2 factorial arrangement that included 1) 3 dietary treatments from d one to 21: positive control (PC), wheat-corn-soybean meal based diet contained antibiotics (virginiamycin and monensin); negative control (NC), as PC without antibiotics; and NC+FOS, as NC supplemented with 0.5% FOS, and 2) 2 intraperitoneal injections: 2 mg/kg Salmonella Enteritidis LPS or sterile phosphate buffered saline (PBS) on d 21. Evaluation of digestive microbial ecology is necessary to understand effects of growth-promoting feed. In the current study, the dynamics of intestinal microbial communities (MC) were examined in broilers fed diets supplemented with a combination of antibiotic (bacitracin methylene disalicylate) and ionophore (Coban 60), and diets containing 1 of 2 essential oil (EO) blends, Crina Poultry (CP) and Crina Alternate (CA). Five treatments were analyzed: 1) unmedicated uninfected control; 2) unmedicated infected control; 3) feed additives monensin (bacitracin methylene disalicylate) + monensin (Coban 60; AI); 4) EO blend CP; and 5) EO blend CA. Additives were mixed into a basal feed mixture, and EO were adjusted to 100 ppm. Chicks were infected by oral gavage at 19 d of age with Eimeria acervulina, Eimeria maxima, and Eimeria tenella. Duodenal, ileal, and cecal samples were taken from 12 birds per treatment just before and 7 d after challenge; 2 samples each were pooled to give a final number of 6 samples total; and all pooled samples were frozen until used for DNA extraction. Denaturing gradient gel electrophoresis was used to examine PCR-amplified fragments of the bacterial 16S ribosomal DNA variable region. Results are presented as percentages of similarity coefficients (SC). Dendrograms of PCR amplicon or band patterns indicated MC differences due to intestinal location, feed additives, and cocci challenge. Essential oil blends CP and CA affected MC in all gut sections. Each EO had different effects over MC, and they differed in most instances from the AI group. The cocci challenge caused drastic MC population shifts in duodenal, ileal, and cecal sections (36.7, 55.4, and 36.2% SC, respectively). Diets supplemented with CP supported higher SC between pre- and postchallenge MC (89.9, 83.3, and 76.4%) than AI (81.8., 57.4, and 60.0%). We concluded that mixed coccidia challenge caused drastic shifts in MC. These EO blends modulated MC better than AI, avoiding drastic shifts after a mixed challenge.

Straight-run broiler chickens were raised either in floor pens or wire-floored cages (trial 1) or in floor pens only (trials 2, 3, and 4). Birds raised in floor pens had lower BW and feed intakes than those raised in cages. The administration of bacitracin in the feed increased feed intake from d 12 to d 35, decreased the feed conversion ratio during the same period in trial 2, and improved the weight gain of broilers from d 0 to 10 in trial 3. The concentrations of conjugated bile salts (taurocholic and taurochenodeoxycholic acids) were higher in the ileal contents of broilers administered the antimicrobials compared with untreated birds. Supplementation of the feed with monensin increased fat digestibility in the ileum of the birds. Although total numbers of bacteria in ileal contents were the same regardless of whether antimicrobials were administered or not, the bacterial community differed qualitatively. Populations of Lactobacillus salivarius were reduced in birds fed antimicrobials relative to untreated broilers. A representative ileal isolate of L. salivarius deconjugated bile salts in pure culture in the laboratory and in the ileal contents of ex-Lactobacillus-free chickens maintained in a protective environment and colonized by the Lactobacillus isolate. These observations provide a link between bile salt deconjugation in the ileum by L. salivarius and decreased weight gain of broilers. Lactobacillus salivarius populations could be targeted in future studies aimed at modification of the ileal bacterial community to achieve growth promotion of broilers without the administration of antimicrobial drugs.

The effect of sodium ions (Na+) on calcium (Ca2+)-mediated muscle damage in broiler chickens was investigated using an in vitro muscle preparation. Muscle Ca2+ accumulation was determined by 45Ca2+ uptake. Muscle damage was assessed by measurement of the efflux of the intracellular enzyme creatine kinase (CK) into the incubation medium. Loading muscle cells with Na+ by means of the sodium ionophore monensin led to concentration-dependent (25 to 200 microM) increases in 45Ca2+ uptakes and corresponding and proportional CK losses. The greatest responses occurred at 100 microM ionophore or greater, reflected in a 49% increase (P < 0.05) in 45Ca2+ uptake and an associated 140%-fold increase (P < 0.001) in CK efflux. Inhibition of muscle Na+/K+-ATPase activity with ouabain (2 mM) induced a 56% increase in 45Ca2+ uptake and a 60%-fold increase (P < 0.001) in total CK loss. The combined use of ionophore and ouabain resulted in 90 and 130%-fold elevations in 45Ca2+ uptake and CK loss, respectively. In monensin-treated muscles, inhibition of external Ca2+ influx from the incubation medium by chelation with 1,2 bis(2-aminophenoxy)ethane-N,N,N',N' tetracetic acid (5 mM) markedly reduced 45Ca2+ uptake (38%: P < 0.05) but increased CK release by 85% (P < 0.001). The results demonstrate that initial elevations in muscle Na+ can facilitate increases in muscle Ca2+ and lead to alterations in muscle cell membrane integrity and CK loss. The Na+-induced increases in myocellular Ca2+ may be mediated via direct extracellular Ca2+ entry or redistribution from internal Ca2+ stores. It is proposed that in order to reduce or prevent myopathies in poultry, exposure to conditions that may lead to elevations in muscle Na+ (e.g., increased muscle activity and stress or accidental ionophore toxicosis) should be avoided. The findings of this study have implications for management strategies of bird welfare, muscle pathology, and product quality.

During the period from January to June, combined-sex broiler chickens were inoculated with coccidia via drinking water at 14 d of age. In a completely randomized design (eight replicate pens; 88 chicks per pen) using built-up litter, experimental diets contained monensin plus 0.20% dietary sodium bicarbonate (SBC), which provided 0.054% sodium and 0.144% bicarbonate. Treatment with SBC significantly improved coccidial lesion score, 45-d body weight, and feed efficiency compared with monensin alone. In a 2 x 5 factorial trial using built-up litter pens (eight replicate pens; 88 chicks per pen) vs. each ionophore alone, 0.20% dietary SBC with monensin significantly improved body weight, uniformity, and feed efficiency; 0.20% SBC with halifuginone, lasalocid, monensin, or salinomycin significantly reduced mortality; and 0.20% SBC with lasalocid, monensin, or salinomycin significantly increased breast meat yield. In a 2x4 factorial trial (12 replicate pens; 88 chicks per pen) on built-up litter, corn-soy and corn-soy-meat diets (higher potassium, lower chloride) with monensin were evaluated using 0.054% sodium from SBC, NaCl, or sodium sulfate decahydrate (SSD). With both diet types, SBC (0.20%) or NaCl (0.139% extra) significantly improved weight uniformity, feed efficiency, mortality, and breast meat yield; however, the SSD results were closer to controls. In a 21-d battery brooder test using similar diets and design (2x4 factorial; 4 replicate pens; 10 chicks per pen), SBC and NaCl significantly reduced coccidial lesion scores; SSD produced a significant, but weaker effect. Extra NaCl significantly increased water intake (approximately 37%), water excretion (approximately 27%), and litter moisture (approximately 22%) with both diet types. The SSD did not affect water intake.

The efficacy of different concentrations of monensin in turkeys exposed to field isolates of Eimeria and the effect of the withdrawal of monensin from their diet were investigated. Results of a battery study indicated that 66 ppm monensin was effective against three species of Eimeria in the turkey. In floor pens in which poults were exposed to infection, use of 59.5, 79.4, and 99.2 ppm monensin resulted in reduced mortality and improved feed conversion at 3 wk of age compared with birds that had received no medication. Poults given 59.5 ppm monensin weighed more and had a better feed conversion at 3 wk than poults given 79.4 or 99.2 ppm monensin, but at 10 wk no differences in the body weight or feed conversion of poults given different concentrations of drug were apparent. At 14 wk (and from 10 to 14 wk), poults that had been given 99.2 ppm monensin had a lower feed intake and weighed less than birds that had been given 59.5 ppm of the drug but there were no significant differences in feed conversion. There was no significant difference in the weight gain or feed conversion of poults from 10 to 14 wk of age whether monensin was present in the feed or had been withdrawn. No evidence of compensatory growth was found in the present study.

Three experiments (Exp.) were conducted to determine the interactive effects of sodium bentonite (NaB) with the efficacy of monensin (MON) or salinomycin (SAL) in coccidiosis-infected chicks. Male broiler chicks 5 to 14 d of age were used, and each treatment was replicated with eight (Exp. 1) or four (Exp. 2 and 3) pens of five chicks each. In Exp. 1, MON (80 ppm), NaB (0.50%), or MON+NaB were fed to uninfected and coccidiosis-infected (5 x 10(5) sporulated Eimeria acervulina oocysts on Day 2 of the Exp.) chicks in a 2 x 2 x 2 factorial arrangement of treatments. Experiment 2 was identical to Exp. 1, but SAL (30 ppm) replaced MON as the anticoccidial additive. In Exp. 3, MON (55 ppm) or SAL (22 ppm) were added individually or with NaB (0.50%) to diets for uninfected or coccidiosis-infected chicks. Coccidial infection reduced (P0.01) gain, feed intake, gain:feed, plasma carotenoids, and percentage tibia ash in all experiments. The MON and SAL additions increased these response criteria in infected chicks (coccidiosis by anticoccidial, P0.07), except MON did not increase (P0.10) feed intake or tibia ash in Exp. 3. In Exp. 3, NaB partially reduced the positive effect of MON on daily gain (NaB by MON, P0.03), and of SAL on feed intake (NaB by SAL, P0.08). The NaB addition also increased gain:feed (P0.08), and the increase was greater in infected chicks (coccidiosis by NaB, P0.08). Also in Exp. 3, SAL increased feed intake more in chicks not fed NaB than in chicks fed NaB (SAL by NaB, P0.08). Dietary NaB (0.5%) may reduce the efficacy of MON and SAL in coccidiosis-infected chicks when these additives are added at less than recommended levels.

Three experiments were conducted to evaluate elevated dietary NaCl levels as a means of offsetting industry-observed reductions of growth, feed intake, and feed efficiency associated with early (35-d) coccidiostat withdrawal. In the first experiment, monensin (100 ppm) was withdrawn and dietary salt levels of 0.33, 0.48, 0.63, 0.78, or 0.93% provided from 35 to 42 d of age. Experiments 2 and 3 involved lasalocid (110 ppm) withdrawal and slat amounts of 0.33, 0.53, 0.73, or 0.93%. In all studies, a positive control of 0.33% salt and the coccidiostat was also given. Monensin withdrawal reduced body weight gain, which was not overcome by slat addition. Feed efficiency during the 1-wk period was improved to the level of the group receiving continued medication by salt amounts of 0.78% or above. In contrast to industry field observations, removal of lasalocid did not reduce body weight gain, feed intake, or water consumption, and elevation of salt levels resulted in no consistent improvements of weight gain, feed intake, or feed conversion. Water intake increased proportionally as salt concentration increased. Elevated salt levels do not appear to be a reliable means of offsetting reduced performance related to early coccidiostat withdrawal, nor were such performance problems demonstrable for lasalocid in these trials.

Three experiments (Exp.) were conducted to evaluate the interactive effects of dietary betaine (BET) and monensin (MON) in uninfected or Eimeria acervulina-infected chicks. The treatments were replicated with six (Exp. 1) or five (Exp. 2 and 3) pens of five chicks each. The experimental periods lasted 9 (Exp. 1 and 2) or 10 (Exp. 3) d each and the coccidiosis infections were established on Day 2 (Exp. 1 and 2) or Day 3 (Exp. 3) of the experiment. Average initial weight of the chicks was 101, 73, and 68 g in Exp. 1 to 3, respectively, and the initial age of the chicks was 5 (Exp. 1) or 4 (Exp. 2 and 3) d. A corn-soybean meal basal diet was used in each experiment. In Exp. 1, the effect of dietary BET (0, 0.1, or 0.5%) in uninfected or coccidiosis-infected (COC; 5 x 10(5) sporulated E. acervulina oocysts) chicks was investigated. In Exp. 2, the interactive effects of BET (0 or 0.1%) and MON (0 or 55 ppm) in uninfected or COC chicks were investigated in a 2 x 2 x 2 factorial arrangement of treatments. Experiment 3 was identical to Exp. 2, except the level of MON was 110 rather than 55 ppm. In Exp. 1, 2, and 3, COC reduced (P0.01) gain, feed intake (FI), feed efficiency (GF), and plasma carotenoid concentration (CAR) and increased (P0.01) lesion score (LS). In Exp. 1, gain and FI were decreased in uninfected chicks fed 0.1% BET but gain and FI were increased in COC chicks fed 0.1% BET (COC x BET quadratic, P0.01). Dietary BET linearly increased (P0.05) GF. In Exp. 2 and 3, MON increased (P0.01) gain, FI, GF, and CAR and decreased (P0.01) LS of COC chicks, but MON had no effect in uninfected chicks (COC x MON, P0.01). In Exp. 2, GF was increased more in chicks fed both MON and BET than in chicks fed MON (BET x MON, P0.06). In Exp. 3, BET increased GF of uninfected chicks fed MON and of COC chicks not fed MON (COC x BET x MON, P0.02). Betaine may have an effect on E. acervulina-infected chicks, but there is no conclusive evidence to indicate that the efficacy of MON is improved when fed in combination with BET.

The ability of selected anticoccidial drugs to inhibit the colonization of day-old male broiler chicks (Cornish Rocks) by Escherichia coli 0157:H7, strain 932 was examined. Chicks were challenged with 1.8 × 109E. coli 0157:H7 on Day 1, and fed diets supplemented with three selected anticoccidial drugs; monensin, nicarbazin, or robenidine. The cecal and colon fecal contents of the chicks were removed on Day 7, 14, and 21 postinoculation and examined for the concentration of E. coli 0157:H7 per gram of contents. Monensin effectively reduced cecal and colon colonization of the chicks by E. coli 0157:H7. By Day 7, there were significant reductions in the bacterial population of the cecal contents of chicks receiving the monensin-medicated feed, and by Day 21 no E. coli 0157:H7 was recovered from the cecal and colon contents. The bacterial counts in the colon contents of the nicarbazin- and robenidine-medicated and unmedicated chicks were significantly higher than the monensin-treated chicks. Bacterial populations in the colon contents were high only when there were high bacterial concentrations in the cecal contents.

The performance of broilers reared in floor pens and given monensin in the feed at 121 ppm was compared with that of birds given no drug. Feed intake and BW gain of medicated birds was significantly lower than that of unmedicated birds from 0 to 22 d of age. Feed intake and feed conversion of medicated birds was significantly reduced, compared with unmedicated birds, from 22 to 53 and 0 to 60 d of age. Total mortality, and mortality due to leg abnormalities from 22 to 53 and 0 to 60 d, was significantly lower in birds given monensin. There was no difference in the incidence of tibial dyschondroplasia (TD) by 60 d. No differences in mortality due to pulmonary hypertension syndrome (PHS) were observed for any age period. Birds removed from pens at 28 d that had received monensin had lower hematocrit and percentage saturation of hemoglobin with oxygen in the blood than unmedicated birds. No differences in these variables were found at 54 d. There were no differences in the right ventricle weight: total ventricular weight ratios or electrocardiogram lead II values at 28 or 54 d. The results indicate that PHS does not occur more frequently in broilers medicated with monensin.

Clay feed additives have been increasingly incorporated into animal diets to prevent aflatoxicosis. Due to the nonselective nature of the binding interaction, many important components of the diets could also be made unavailable because of these feed additives. The anticoccidial monensin (MON) could also be sequestered by these clays. The use of sodium bentonite (Na-B) from a mine in the province of Mendoza, Argentina, was investigated as a sequestering agent to prevent the effects of 100 µg/kg of dietary aflatoxin B(1) (AFB(1)). In vitro studies demonstrated that the above Na-B was a good candidate to prevent aflatoxicosis. They also showed that MON competes with AFB(1) for the adsorption sites on the clay surface and effectively displaces the toxin when it is in low concentration. Even though the levels of MON in diets, approximately 55 mg/kg, are high enough to not be significantly changed as a consequence of the adsorption, they can further affect the ability of the clays to bind low levels of AFB(1). An in vivo experiment carried out with poultry showed that 100 µg/kg of AFB(1) does not significantly change productive or biochemical parameters. However, liver histopathology not only confirmed the ability of this particular Na-B to prevent aflatoxicosis but also the decrease of this capacity in the presence of 55 mg/kg of MON. This is the first report stressing this fact and further research should be performed to check if this behavior is a characteristic of the assayed Na-B or of this type of clay. On the other hand, the presence of MON should also be taken into account when assaying the potential AFB(1) binding ability of a given bentonite.

A floor pen study was conducted to determine the effects of feed restriction with subsequent gorging and water restriction on 5- to 10-wk-old toms fed graded levels of monensin. Eight treatments were factorially arranged with four levels of dietary monensin (0, 60, 100, and 140 ppm) and two feeding-watering regimens (ad libitum and restricted). Restricted birds had access to feed daily from 0800 to 1200 h and had no access to water during this period. The ad libitum birds had full access to feed and water. Feed restriction decreased (P .10) on feed:gain ratio. The mortality rate of restricted birds was 82% less (P .10) influence on the incidence of mortality. No treatment-related abnormalities were observed during either interim or terminal necropsies. Neither clinical signs of ionophore toxicity nor pathologic findings attributed to monensin treatment were observed during the study. Under the conditions of this experiment, the gorging of monensin treated (60 to 140 ppm) feed without access to water will not adversely affect the health status, or increase the mortality rate, of 5- to 10-wk-old male turkeys.

Aflatoxins (AF) are a major problem in broiler production and are significant economic and public health burdens worldwide. A commercial sodium bentonite (Na-B) adsorbent was used to prevent the effect of AF [50 µg of aflatoxin B₁ (AFB₁)/kg of feed] in broiler productivity, biochemical parameters, macroscopic and microscopic liver changes, and AFB₁ liver residues. The influence of Na-B (0.3%) and monensin (MON, 100 mg/kg), alone or in combination, was investigated in depth. The dietary treatments were as follows: treatment (T) 1: basal diet (B); T2: B + MON; T3: B + Na-B; T4: B + Na-B + MON; T5: B + AFB₁; T6: B + AFB₁ + Na-B + MON; T7: B + AFB₁ + MON; T8: B + AFB₁ + Na-B. Birds were fed dietary treatments for 28 d (d 18 to 46). No significant differences (P0.05) were observed among treatments with respect to broiler performance, biochemical parameters, or relative liver weights. With the exception of T8, all livers showed histopathological alterations, with accumulation of fat vacuoles. The normal appearance of livers from T8 showed the protective effect of Na-B against aflatoxicosis. The residual AFB₁ levels in livers from T5 to T8 ranged from 0.2 to 1.0 ng/g and were higher in livers from T6 (P0.05). Results of this study indicate a competition between AFB₁ and MON for adsorption sites on Na-B when feed contains low levels of the toxin, indicating a nonselective adsorption capacity of this particular Na-B. In addition, significant levels of AFB₁ in livers indicate that this determination is an important technique not only for diagnosis of aflatoxicosis in broilers, but also for quality control of avian products.

Freshly excysted sporozoites (SZ) of the turkey coccidia Eimeria meleagrimitis and Eimeria adenoeides were incubated at 41 C in concentrations of monensin from .01 to 1.0 microgram/mL, washed free of the drug, and either processed for phase, fluorescence, and transmission electron microscopy or inoculated into cultures of turkey kidney cells. Phase microscopy indicated that after 1.5 h incubation in 1.0 micrograms/mL monensin, about 60% of the SZ of E. meleagrimitis had become notably rounded or displayed localized protrusions. These alterations were accompanied by ultrastructural abnormalities (in 90% of the SZ) including vacuoles in the cytoplasm, bulging and separation of plasma membrane layers, and dense bands in the refractile bodies that extended toward the periphery of the refractile body. Similar morphological and ultrastructural changes were observed in over half of the E. adenoeides SZ after 2 h incubation in 1.0 micrograms/mL monensin. Additionally, some specimens contained a pycnotic nucleus that was usually surrounded by a large vacuole. After 4 h incubation, almost all of the SZ displayed some degree of ultrastructural damage. Indirect fluorescent antibody labeling with parasite-specific monoclonal antibodies demonstrated clouds of antigen surrounding the monensin-treated but not the untreated SZ, suggesting an increase in permeability with incubation in monensin. With both E. meleagrimitis and E. adenoeides, the structural changes were reflected in a significant inhibition of cellular invasion. The inhibitory activity of monensin was concentration- and time-dependent in that the greatest inhibition of invasion was observed in SZ incubated for 4 h in 1.0 micrograms/mL of monensin; shorter incubation times or lower concentrations of monensin having less effect.

In vivo and in vitro experiments were conducted in an effort to elucidate the mechanism of suppression by halofuginone of skin strength in broilers. In the in vivo study, halofuginone was included at concentrations of 0, 1.5, 3, and 6 mg/kg of diet, corresponding to 0, 50, 100, and 200%, respectively, of the amount recommended for use as a coccidiostat. Each dietary treatment was given to 260 female broiler day-old chickens. Skin tearing was evaluated at the processing plant. Skin collagen and Kjeldahl-nitrogen were determined chemically. At the age of 7 wk, BW and feed efficiency were affected only in birds consuming the diet containing the highest concentration of the drug. Skin tearing increased but skin collagen concentration decreased in a dose-dependent manner. Fibroblasts were obtained by collagenase digestion from chicken skin and cultured. The cultured cells were incubated with various concentrations of halofuginone, monensin, and nicarbazin, and [3H]proline incorporation was evaluated in collagenase-digestible (representing mostly collagen) and nondigestible proteins exported by the cells into the medium. Halofuginone, at a concentration as low as 10(-11) M, inhibited incorporation of [3H]proline into collagenase-digestible proteins, but did not affect incorporation of [3H]proline into collagenase-nondigestible proteins. Even at concentrations as high as 10(-9) M, neither monensin nor nicarbazin affected collagenase-digestible proteins. The in vitro results suggest that halofuginone specifically inhibits collagen synthesis by skin fibroblasts. Results of both in vivo and in vitro trials suggest that the increase of skin tearing during processing, induced by halofuginone, is caused by direct suppression of skin collagen synthesis.
