Rationale: Conscious perception is thought to depend on global amplification of sensory input. In recent years, striatal dopamine has been proposed to be involved in gating information and conscious access, due to its modulatory influence on thalamocortical connectivity. Objectives: Since much of the evidence that implicates striatal dopamine is correlational, we conducted a double-blind crossover pharmacological study in which we administered cabergoline—a dopamine D2 agonist—and placebo to 30 healthy participants. Under both conditions, we subjected participants to several well-established experimental conscious-perception paradigms, such as backward masking and the attentional blink task. Results: We found no evidence in support of an effect of cabergoline on conscious perception: key behavioral and event-related potential (ERP) findings associated with each of these tasks were unaffected by cabergoline. Conclusions: Our results cast doubt on a causal role for dopamine in visual perception. Rationale: The specific role of neuromodulator systems in regulating rapid fluctuations of attention is still poorly understood. Objectives: In this study, we examined the effects of clonidine and scopolamine on multiple target detection in a rapid serial visual presentation task to assess the role of the central noradrenergic and cholinergic systems in temporal attention. Method: Eighteen healthy volunteers took part in a crossover double-dummy study in which they received clonidine (150/175 μg), scopolamine (1.2 mg), and placebo by mouth in counterbalanced order. A dual-target attentional blink task was administered at 120 min after scopolamine intake and 180 min after clonidine intake. The electroencephalogram was measured during task performance. Results: Clonidine and scopolamine both impaired detection of the first target (T1). For clonidine, this impairment was accompanied by decreased amplitudes of the P2 and P3 components of the event-related potential. The drugs did not impair second-target (T2) detection, except if T2 was presented immediately after T1. The attentional blink for T2 was not affected, in line with a previous study that found no effect of clonidine on the attentional blink. Conclusions: These and other results suggest that clonidine and scopolamine may impair temporal attention through a decrease in tonic alertness and that this decrease in alertness can be temporarily compensated by a phasic alerting response to a salient stimulus. The comparable behavioral effects of clonidine and scopolamine are consistent with animal studies indicating close interactions between the noradrenergic and cholinergic neuromodulator systems. Rationale: While it is well documented that substance users exhibit attentional bias toward addiction-related stimuli, the exact mechanism remains unclear. Objectives: To differentiate between distinct aspects of attentional allocation in the smoking-cue attentional bias observed in smokers. Methods: Active smokers (AS) and non-smoking controls completed spatial cueing tasks with pairs of smoking and neutral pictorial cues to measure attentional capture, and an attentional blink task with either a smoking or neutral image appearing behind the first target (T1) to measure aspects of attention separate from capture. In addition, we tested groups of sports enthusiasts, and non-enthusiasts in corresponding tasks replacing smoking images with sports-related images to address the possibility that effects found in the smoking study were due simply to greater stimulus familiarity. Results: Smoking cues reflexively capture smokers' attention, as AS showed a greater bias toward smoking cues in short stimulus-onset asynchrony (SOA; the time between the onset of two stimuli) trials, but not in trials with a longer SOA. These effects represent a facilitation of responding to smoking- versus neutral-cued targets, and were absent in the sports control task. The attentional blink effects were similar in the smoking- and sports-cue experiments: the special T1 resulted in better detection of the second target for the smokers and sports enthusiasts. Conclusions: Stimulus familiarity may contribute to some aspects of attentional bias in regular nicotine users, but selective quick capture of attention by smoking cues may be nicotine-habit specific. Recent theories posit an important role for the noradrenergic system in attentional selection in the temporal domain. In contrast, the spatially diffuse topographical projections of the noradrenergic system are inconsistent with a direct role in spatial selection. To test the hypotheses that pharmacological attenuation of central noradrenergic activity should (1) impair performance on the attentional blink task, a task requiring the selection of targets in a rapid serial visual stream of stimuli; and (2) leave intact the efficiency of the search for a target in a two-dimensional visuospatial stimulus array. Thirty-two healthy adult human subjects performed an attentional blink task and a visual search task in a double-blind, placebo-controlled, between-subject study investigating the effects of the α2 adrenoceptor agonist clonidine (150 μg, oral dose). No differential effects of clonidine vs placebo were found on the attentional blink performance. Clonidine slowed overall reaction times in the visual search task but did not impair the efficiency of the visual search. The attentional blink results are inconsistent with recent theories about the role of the noradrenergic system in temporal filtering and in mediating the attentional blink. This discrepancy between theory and data is discussed in detail. The visual search results, in combination with previous findings, suggest that the noradrenergic system is not directly involved in spatial attention processes but instead can modulate these processes in an indirect fashion.
