Ambulatory recording of the preejection period (PEP) can be used to measure changes in cardiac sympathetic nervous system (SNS) activity under naturalistic conditions. Here, we test the ECG T-wave amplitude (TWA) as an alternative measure, using 24-h ambulatory monitoring of PEP and TWA in a sample of 564 healthy adults. The TWA showed a decrease in response to mental stress and a monotonic decrease from nighttime sleep to daytime sitting and more physically active behaviors. Within-participant changes in TWA were correlated with changes in the PEP across the standardized stressors (r = .42) and the unstandardized naturalistic conditions (mean r = .35). Partialling out changes in heart rate and vagal effects attenuated these correlations, but they remained significant. Ambulatory TWA cannot replace PEP, but simultaneous recording of TWA and PEP provides a more comprehensive picture of changes in cardiac SNS activity in real-life settings.

This study estimated the heritability of 24-h heart rate variability (HRV) measures, while considering ceiling effects on HRV at low heart rates during the night. HRV was indexed by the standard deviation of all valid interbeat intervals (SDNN), the root mean square of differences between valid, successive interbeat intervals (RMSSD), and peak-valley respiratory sinus arrhythmia (pvRSA). Sleep and waking levels of cardiac vagal control were assessed in 1,003 twins and 285 of their non-twin siblings. Comparable heritability estimates were found for SDNN (46%‐53%), RMSSD (49%‐ 54%), and pvRSA (48%‐57%) during the day and night.Anighttime ceiling effect was revealed in 10.7% of participants by a quadratic relationship between mean pvRSA and the interbeat interval. Excluding these participants did not change the heritability estimates.The genetic factors influencing ambulatory pvRSA, RMSSD, and SDNN largely overlap.These results suggest that gene-finding studies may pool the different cardiac vagal indices and that exclusion of participants with low heart rates is not required. Metrics of heart period variability are widely used in the behavioral and biomedical sciences, although somewhat confusingly labeled as heart rate variability (HRV). Despite their wide use, HRV metrics are usually analyzed and interpreted without reference to prevailing levels of cardiac chronotropic state (i.e., mean heart rate or mean heart period). This isolated treatment of HRV metrics is nontrivial. All HRV metrics routinely used in the literature exhibit a known and positive relationship with the mean duration of the interval between two beats (heart period): as the heart period increases, so does its variability. This raises the question of whether HRV metrics should be “corrected” for the mean heart period (or its inverse, the heart rate). Here, we outline biological, quantitative, and interpretive issues engendered by this question. We provide arguments that HRV is neither uniformly nor simply a surrogate for heart period. We also identify knowledge gaps that remain to be satisfactorily addressed with respect to assumptions underlying existing HRV correction approaches. In doing so, we aim to stimulate further progress toward the rigorous use and disciplined interpretation of HRV. We close with provisional guidance on HRV reporting that acknowledges the complex interplay between the mean and variability of the heart period.

We tested for a genetic influence on magnetoencephalogram (MEG)-recorded somatosensory evoked fields (SEFs) in 20 monozygotic (MZ) and 14 dizygotic (DZ) twin pairs. Previous electroencephalogram (EEG) studies that demonstrated a genetic contribution to evoked responses generally focused on characteristics of representative brain potentials. Here we demonstrate significantly smaller amplitude differences within MZ compared to DZ twin pairs for the complete SEF time series (across left and right hand SEFs: 0.37 vs. 0.60 pT(2) and 0.28 vs. 0.39 pT(2) for primary [SI] and secondary [SII] sensory cortex activation) and higher MZ than DZ wave shape correlations (.71 vs. .44 and .52 vs. .35 for SI and SII activation). Our findings indicate a genetic influence on MEG-recorded evoked brain activity and also confirm our recent conclusion (van 't Ent, van Soelen, Stam, De Geus, & Boomsma, 2009) that higher MZ resemblance for EEG amplitudes is not trivially reflecting greater MZ concordance in intervening biological tissues.

We investigated the relationship between three electrophysiological indices of response anticipation in a spatial delayed response task with a low and high memory load manipulation: a slow cortical potential (SCP), theta desynchronization, and upper alpha synchronization. Individual differences in these three measures were examined in 531 adult twins and siblings. Heritability of the SCP at occipital-parietal leads varied from 30% to 43%. Heritability of upper alpha synchronization (35% to 65%) and theta desynchronization (31% to 50%) was significant at all leads. Theta desynchronization and upper alpha synchronization were significantly correlated (r � 43%), but SCP was not correlated with either. The effect of working memory load on all three measures was not heritable. Response anticipation reliably evokes an SCP, upper alpha synchronization and theta desynchronization, but variation in these measures reflects different (genetic) sources. It has been hypothesized that the ratio of heart rate variability in the low- (LF) and high- (HF) frequency bands may capture variation in cardiac sympathetic control. Here we tested the temporal stability of the LF/HF ratio in 24-h ambulatory recordings and compared this ratio to the preejection period (PEP), an established measure of cardiac sympathetic control. Good temporal stability was found across a period of 3.3 years (.46

The extent to which various measures of ambulatory respiratory sinus arrhythmia (RSA) capture the same information across conditions in different subjects remains unclear. In this study the root mean square of successive differences (RMSSD), peak valley RSA (pvRSA), and high frequency power (HF power) were assessed during ambulatory recording in 84 subjects, of which 64 were retested after about 3 years. We used covariance structure modeling to test the equality of the correlations among three RSA measures over two test days and three conditions (daytime sitting or walking and nighttime sleep) and in groups with low, medium, and high mean heart rate (HR), or low, medium, and high mean respiration rate (RR). Results showed that ambulatory RMSSD, pvRSA, and HF power are highly correlated and that their correlation is stable across time, ambulatory conditions, and a wide range of resting HR and RR values. The extent to which variation in posture and respiration can confound pre-ejection period and respiratory sinus arrhythmia (RSA) as indices of cardiac sympatho-vagal activity was examined. Within-subjects changes in these measures were assessed in 36 subjects during different postures and (paced) respiratory frequencies. Changes from supine to sitting to standing led to reduced RSA values and longer pre-ejection periods, reflecting the known decrease in vagal but not the increase of sympathetic activity. Multilevel path analysis showed that within-subjects changes in sympatho-vagal balance were faithfully reflected by changes in interbeat interval, but imperfectly by changes in RSA and pre-ejection period. It was concluded that pre-ejection period should be stratified for posture and RSA for respiratory frequency to reliably index changes in sympatho-vagal balance when these factors are prone to change (e.g., during 24-h ambulatory recording). This study examines the effects of competition and competitiveness on hemodynamics. Cardiovascular activity was measured in 27 men at resting baseline and during a car racing game, which comprised a solo race against time and three races against an experimenter. To assess hematocrit, blood was collected at rest and after the final race. Trait competitiveness was assessed by questionnaire. Competition elicited increases in hematocrit, blood pressure, heart rate, and total peripheral resistance, as well as decreases in preejection period and heart rate variability. The final race was rated as more competitive than the solo race. Compared to intrapersonal solo racing, the final interpersonal race was associated with shorter preejection periods and faster heart rates, markers of beta-adrenergic activation. Although trait competitiveness was not associated with beta-adrenergic activation, variations in state competitiveness were.

This study tested various sources of changes in respiratory sinus arrhythmia (RSA). Twenty-two healthy participants participated in three experimental conditions (mental stress, relaxation, and mild physical exercise) that each consisted of three breathing parts (normal breathing, breathing compressed room air, and breathing compressed 5% CO2-enriched air). Independent contributions to changes in RSA were found for changes in tonic vagal modulation of heart rate, central respiratory drive (i.e., PaCO2), respiratory depth, and respiratory frequency. The relative contributions to changes in RSA differed for mental stress and physical exercise. It is concluded that uncorrected RSA will suffice to index within-subject changes in tonic vagal modulation of heart rate in most situations. However, if the central respiratory drive is expected to change, RSA should ideally be corrected for changes in PaCO2, respiratory depth, and respiratory frequency.

We examined differences between migraine patients and matched healthy controls in anticipatory processes preceding a warning stimulus and preceding a response stimulus during a forewarned choice reaction time task. We manipulated stimulus preceding negativity (SPN) by inserting full response information either at the instant of the warning stimulus (cue) or at the instant of the response stimulus. In contrast to control subjects, migraineurs with aura show low anticipation towards an informative cue and high anticipation towards a noninformative cue. Migraineurs without aura showed a cortical hypoactivation during motor preparation prior to the response stimulus. We propose a functional deficiency within frontal structures or the anterior cingulate cortex in migraine. This might explain the reduced anticipation, as well as the slow responses during selective attention that we previously reported in these patients.

Progress in molecular genetic psychophysiology, on display in this special issue, shows that the affective and cognitive processes tagged by psychophysiological endophenotypes are highly polygenic. Identifying overlap in the genetic variants that influence these endophenotypes as well as behavioral traits can help us understand where in the brain, in which stage, and during what type of information processing these variants play a role in normal and abnormal behavior. The Minnesota Twin Family Study has demonstrated that the assessment of genetic markers and extensive psychophysiological experimentation can be done at a genetic epidemiological scale, that is, in thousands of subjects. A genome-wide association meta-analysis consortium consisting of psychophysiological research teams following the Minnesotan example is the obvious next step.

This study examined the acute immunological effects of two laboratory stressors, expected to evoke distinct patterns of cardiac autonomic activity; namely an “active coping” time-paced memory test, and a “passive coping” stressful video showing surgical operations. We measured salivary S-IgA, IgA-subclasses ~IgA1, IgA2!, and secretory component ~SC!. SC is responsible for the transport of S-IgA across the epithelium, and thus a rate-determining step in S-IgA secretion. Thirty-two male undergraduates were subjected to both stressors and a control video ~a didactic television program!. The memory test induced a typical “fight-flight” response, characterized by increases in heart rate and blood pressure in association with a decrease in cardiac preejection period ~PEP! and vagal tone. The surgical video produced a “conservation-withdrawal”-like response, characterized by an enhanced vagal tone, a decrease in heart rate, and a moderate sympathetic coactivation ~as indicated by a shortened PEP and an increased systolic pressure!. The memory test induced an increase in the concentration and, to a lesser extent, in the output of S-IgA, IgA1, and SC. The output of IgA2 was not significantly affected. For the surgical video, a different pattern emerged: During stressor exposure S-IgA remained unaffected, against the background of a small increase in SC output. However, 10 min after the surgical video S-IgA levels had decreased. This decrease in S-IgA was paralleled by a decrease in IgA1, but not IgA2. We conclude that acute stress can have both enhancing and suppressive effects on secretory immunity, the IgA1 subclass in particular. The mechanisms that underlie these divergent responses may include stressor-specific patterns of autonomic activation. The observation that aerobically fit persons react to physical load with a smaller sympathetic response than do less fit subjects suggests that their response to psychological stress might also be reduced. The evidence for this, however, is far from consistent. It was argued that this inconsistency might be due to the incomplete measurement of the response system involved. In the present experiment two groups, which differed strongly in maximal aerobic power, were compared with respect to their cardiovascular response to a laboratory stressor. In addition to the traditional heart rate and blood pressure measurements, pre-ejection period, cardiac output, and peripheral resistance were assessed. Since only the part of the stress response that is not accounted for by metabolic needs might have pathological significance, the so-called "additional" responses were also measured. Fitness was shown to be associated with smaller sympathetic effects on both heart and vessels. The decrement in pre-ejection period and the heart rate response were smaller in the high fit group. The total peripheral resistance and diastolic blood pressure responses pointed to a much stronger vascular reactivity in the low fit group. Unexpectedly cardiac output did not increase during stress. The strong heart rate response in the low fit group was compensated by a reduction in stroke volume, which probably originated in an increased peripheral resistance. The largest discrepancy between the effects of sympathetic activation during stress and exercise occurred in the vessels, as demonstrated by the large "additional" response in total peripheral resistance during stress. The data point to the often neglected role of vascular processes in experiments of this kind. Furthermore, it seems necessary to take the effect of fitness into account when comparing subjects with respect to their cardiovascular stress response.
