The co-evaporation approach was used to examine the host-guest interaction and to explore the cytotoxic and antibacterial properties of an important anti-cancer medication, 6-mercaptopurine monohydrate (6-MP) with β-cyclodextrin (β-CD). The UV-Vis investigation confirmed the inclusion complex's (IC) 1 : 1 stoichiometry and was also utilized to oversee the viability of this inclusion process. FTIR, NMR, and XRD, among other spectrometric techniques, revealed the mechanism of molecular interactions between β-CD and 6-MP which was further hypothesized by DFT to verify tentative outcomes. TGA and DSC studies revealed that 6-MP's thermal stability increased after encapsulation. Because of the protection of drug 6-MP by β-CD, the formed IC was found to have higher photostability. Green gold nanoparticles (GNPs) were prepared from black tea extract (BTE) and used to examine the chemosensitivity of doxorubicin in colon cancer cell line HCT116.

Ligands like alkanethiol (e.g. dodecanethiol, hexadecanethiol, etc.) and polymers (e.g. poly(vinyl pyrrolidone), polyethylene glycol-thiol) capped to the gold nanoparticles (AuNPs) are widely used in biomedical field as drug carriers and as promising materials for probing and manipulating cellular processes. Ligand functionalised AuNPs are known to interact with the pulmonary surfactant (PS) monolayer once reaching the alveolar region. Therefore, it is crucial to understand the interaction between AuNPs and PS monolayers. Using coarse-grained molecular dynamics simulations, the effect of ligand density, and ligand length have been studied for two classes of ligands on a PS model monolayer consisting of DPPC, POPG, cholesterol and SP-B (mini-peptide). The ligands considered in this study are alkanethiol and polyethylene glycol (PEG) thiol as examples of hydrophobic and hydrophilic ligands, respectively. It was observed that the interaction between AuNPs and PS changes the biophysical properties of PS monolayer in compressed and expanded states. The AuNPs with hydrophilic ligand, can penetrate through the monolayer more easily, while the AuNPs with hydrophobic ligand are embedded in the monolayer and participated in deforming the monolayer structure particularly the monolayer in the compressed state. The bare AuNPs hinder to lower the monolayer surface tension value at the interface, however introducing ligand to the bare AuNPs or increasing the ligand length and density have an impact of lowering of monolayer surface tension to a minor extent. The simulation results guide the design of ligand protected NPs as drug carriers and can identify the nanoparticles' potential side effects on lung surfactant.

Adsorption mechanism of corticosteroid drug hydrocortisone in the lung surfactant monolayer.

Andrographis paniculata (Burm. f.) Nees and Andrographis nallamalayana J.L.Ellis have traditionally been used to treat various ailments such as mouth ulcers, intermittent fever, inflammation, snake bite. This study compares the comparative in vitro cytotoxic activity, and phytochemical profiling of methanol extract of A. nallamalayana (ANM) and A. paniculata (APM). UPLC-ESI-QTOF-MS/MS analysis has been performed. The cytotoxic activity of crude methanol extracts were evaluated against three different cancer cell lines (HCT 116, HepG2, and A549 cell line). Both plants' extract exhibited significant cytotoxic activity against tested cell lines in a dose-dependent manner. IC50 of ANM and APM in HCT 116 cell was 11.71 ± 2.48 μg ml−1 and 45.32 ± 0.86 μg ml−1 and in HepG2 cell line was 15.65 ± 2.25 μg ml−1 and 60.32 ± 1.05 μg ml−1 respectively. Cytotoxicity of these two extracts was comparatively similar in A549 cells. ANM induced cytotoxicity involved programmed cell death, externalisation of phosphatidylserine, ROS generation, up-regulation and down-regulation of major apoptotic markers. HRMS analysis of ANM and APM resulted in the identification of 59 and 42 compounds, respectively. Further, using the MS/MS fragmentation approach, 20 compounds, of which 18 compounds were identified for the first time from ANM, which belongs to phenolic acids, flavonoids, and their glycosides. Three known compounds, echioidinin, skullcapflavone I and 5,2′,6′-trihydroxy-7-methoxyflavone 2′-O-β-D-glucopyranoside, were isolated from A. nallamalayana and their crystal structures were reported for the first time. Subsequently, seven major compounds were identified in A. nallamalayana by direct comparison (retention time and UV-spectra) with authentic commercial standards and isolated compounds using HPLC-UV analysis. The cytotoxicity of phytochemicals from both the plants using in silico tools also justify their in vitro cytotoxic activity. It is the first report on the comparative characterisation of phytochemicals present in the methanolic extract of both the species of Andrographis, along with the cytotoxic activity of A. nallamalayana.

Fixation of atmospheric dinitrogen in plants by [Mo-Fe] cofactor of nitrogenase enzyme takes place efficiently under atmospheric pressure and normal temperature. In search for an alternative methodology for the highly energy intensive Haber-Bosch process, design and synthesis of highly efficient inorganic and organometallic complexes by mimicking the structure and function of [Mo-Fe] cofactor system is highly desirable for ammonia synthesis from dinitrogen. An ideal catalyst for ammonia synthesis should effectively catalyse the reduction of dinitrogen in the presence of a proton source under mild to moderate conditions, and thereby, significantly reducing the cost of ammonia production and increasing the energy efficacy of the process. In the light of current research, it is evident that there is a plenty of scope for the development and enhanced performance of the inorganic and organometallic catalysts for ammonia synthesis under ambient temperature and pressure. The review furnishes a comprehensive outlook of numerous organometallic catalysts used in the synthesis of ammonia from dinitrogen in the past few decades.

In this work, core–shell supramolecular assembly polymeric nano-architectures containing hydrophilic and hydrophobic segments were synthesized via reversible addition fragmentation chain transfer (RAFT) polymerization. Herein, polyethylene glycol methyl ether methacrylate (PEGMA), and stearic acid were used to synthesize the poly(PEGMA) homopolymer and stearyl ethyl methacrylate (SEMA), respectively. Then, PEGMA and SEMA were polymerized through controlled RAFT polymerization to obtain the final diblock copolymer, poly(PEGMA-co-SEMA) (BCP). Model anticancer drug, doxorubicin (DOX) was loaded on BCPs. Interestingly, efficient DOX release was observed at acidic pH, similar to the cancerous environment pH level. Significant cellular uptake of DOX loaded BCP50 (BCP50-DOX) was observed in MDA-MB-231 triple negative breast cancer cells and resulted in a 35 fold increase in anticancer activity against MDA MB-231 cells compared to free DOX. Scanning electron microscopy (SEM) imaging confirmed the apoptosis mediated cellular death. These core–shell supramolecular assembly polymeric nano-architectures may be an efficient anti-cancer drug delivery system in the future.

Biodegradable natural polymers and macromolecules for transient electronics have great potential to reduce the environmental footprint and provide opportunities to create emerging and environmentally sustainable technologies. Creating complex electronic devices from biodegradable materials requires exploring their chemical design pathways to use them as substrates, dielectric insulators, conductors, and semiconductors. While most research exploration has been conducted using natural polymers as substrates for electronic devices, a very few natural polymers have been explored as dielectric insulators, but they possess high dielectric constants. Herein, for the first time, we have demonstrated a natural polyphenol-based nanomaterial, derived from tannic acid as a low-κ dielectric material by introducing a highly nanoporous framework with a silsesquioxane core structure. Utilizing natural tannic acid, porous “raspberry-like” nanoparticles (TA-NPs) are prepared by a sol–gel polymerization method, starting from reactive silane unit-functionalized tannic acid. Particle composition, thermal stability, porosity distribution, and morphology are analyzed, confirming the mesoporous nature of the nanoparticles with an average pore diameter ranging from 19 to 23 nm, pore volume of 0.032 cm3 g−1 and thermal stability up to 350 °C. The dielectric properties of the TA-NPs, silane functionalized tannic acid precursor, and tannic acid are evaluated and compared by fabricating thin film capacitors under ambient conditions. The dielectric constants (κ) are found to be 2.98, 2.84, and 2.69 (±0.02) for tannic acid, tannic acid-silane, and TA-NPs, respectively. The unique chemical design approach developed in this work provides us with a path to create low-κ biodegradable nanomaterials from natural polyphenols by weakening their polarizability and introducing high mesoporosity into the structure.

The present work reported is a simple and selective method for the colorimetrical detection of L-cysteine in Lens culinaris (or lentils) using Au–Ag core–shell (Au core Ag shell) composite nanoparticles as a chemical probe. The phenomenon is based on the color change of composite nanoparticles from yellowish brown to light blue, followed by a shift of the localized surface plasmon resonance (LSPR) absorption band in the UV-visible region (i.e., 200–800 nm) with the addition of L-cysteine into the solution of bimetallic nanoparticles. The mechanism for the detection of L-cysteine is based on the electrostatic interaction of the metal ion with the thiol group of the amino acid, which causes the red shift of the LSPR band at 685 nm. The size distribution, morphology, composition and optical properties of the Au–Ag core–shell composite nanoparticles were characterized by transmission electron microscopy (TEM), dynamic light scattering (DLS), energy dispersive X-ray diffraction (EDX), UV-visible spectrophotometer and Fourier transform infrared spectroscopy (FTIR) techniques. An excellent linearity range for the present method was observed in the range of 20–140 μg mL−1 with a limit of detection at 1.95 μg mL−1 and correlation coefficient (R2) of 0.986. A good% recovery of 4.0% showed the selectivity of the method for L-cysteine determination from sample matrices. The advantageous features of the present method are being simple, rapid, low cost and selectivity towards the determination of L-cysteine in lentils.

Inorganic double halide perovskites have a wide range of applications in low-cost photovoltaic and optoelectronic devices. In this manuscript, we have studied their structural, electronic, mechanical and optical properties using density functional theory (DFT) simulations. In this work, hydrostatic pressure is induced from 0 to 50 GPa. Disordered Ag and Bi atoms have a large impact on band gap energy; in this case, the indirect band gap is transferred towards a direct band gap. We have seen that pressure-driven samples have transformed a band energy semiconductor into a metallic one. Under the induced hydrostatic pressure, the covalent bond is transformed into a metallic bond and the bond lengths are reduced. Meanwhile, pressure-induced samples enhance symmetry breaking in [AgBr6]5− and [BiBr6]3− octahedra, which reduces the density of states of the Fermi surface and lowers the total energy. The mechanical behaviors demonstrated that the studied materials are mechanically stable as well as ductile and their ductile nature is enhanced by the driving pressure. The absorption peak is shifted towards the low energy region with increased hydrostatic pressure. The absorptivity and dielectric constant values are also increased with driving pressure. Phase transformed double halide perovskites triggered by outside stimuli produce several outstanding materials properties, giving great scope for a broad range of applications. This type of pristine and disordered double halide perovskite with pressure-driven semiconductor-to-metal phase transition samples may have potential applications in optoelectronic and photovoltaic devices.

We report herein the development of a new pyridine-pyrazole based bis-bidentate asymmetric chemosensor that shows excellent turn-on chelation-enhanced Al3+-responsive fluorescence. The presence of two ‘hard’ phenolic hydroxyl groups plays a pivotal role in switching-on the sensing through coordination to the ‘hard’ Al3+ ion, while the mechanism can be interpreted by the chelation-enhanced fluorescence (CHEF) process. The X-ray single structure show a planar conjugated structure of the ligand, which was further stabilized by extensive H-bonding and π–π stacking. The photophysical studies related to the sensing behavior of the titular ligand toward aluminum was investigated in detail using various spectroscopic techniques like UV-Vis, photoluminescence, fluorescence and time-correlated single-photon count (TCSPC) and time-resolved NMR. The spectroscopic methods also confirm the selective detection of Al3+ ion in the presence of other metal ions. The theoretical calculations using Density Functional Theory (DFT) and the Time Dependent Density Functional Theory (TD-DFT) provide further insight on the mechanistic aspects of the turn-on sensing behavior including the electronic spectra of both the ligand and the complex. Interestingly, the as-synthesized H2DPC–Al complex can also be utilized as a fluorescence-based sensor for various nitroaromatics including picric acid, for which an INHIBIT logic gate can also be constructed. The as synthesized complex was subsequently used as a fluorescent probe for imaging of human breast adenocarcinoma (MCF7) cells using live cell confocal microscopic techniques.

The present study demonstrates binding interactions and Forster resonance energy transfer (FRET) between bovine serum albumin (BSA) and a series of structurally and electronically diverse phenothiazine (PTZ) and anthracene (ANT) dyes. Upon selective excitation of tryptophan (Trp) residues of BSA, radiationless energy transfer to a dye takes place, resulting in fluorescence quenching of the former. Fluorescence quenching mechanisms, FRET parameters, possible locations, and binding constants of dyes with the BSA have been examined to deduce a structure–property relationship. The mechanism of quenching is apparently static in nature. PTZ dyes with heteroatoms and a pentyl tail (C5-PTZ) attached to them were found to have a stronger binding affinity with BSA as compared to ANT dyes. Stronger binding affinities of C5-PTZ dyes with BSA result in greater energy transfer efficiencies (ET). A dye with a strong electron-withdrawing group present in it has shown better energy accepting capability. A FRET study with dicyanoaniline (DCA) analogs of PTZ and ANT dyes (C5-PTZDCA and ANTDCA, respectively) revealed that ET depends on electronic and structural factors of molecules. An almost orthogonal geometry between ANT and DCA moieties (∼79°) in ANTDCA induces the greater extent of electron transfer from ANT to DCA, showing a higher ET for this dye as compared to C5-PTZDCA in which the torsion angle is only ∼38°. Further, the observed facts have been validated by experimentally determined bandgaps (using cyclic voltammetry experiments) for all the dyes. Thus, the hydrophobic character and the presence of interactive substituents along with the electron-accepting abilities majorly control the FRET for such dyes with BSA.

Nanofabrication techniques that can generate large and complex 3D structures with nanoscale features are becoming increasingly important in the fields of biomedicine, micro-optics, and microfluidics. Direct laser writing via two-photon polymerization (DLW-TPP) is one such technique that relies on nonlinear absorption of light to form nanoscale 3D features. Although DLW-TPP provides the required nanoscale resolution, its built height is often limited to less than a millimetre. This height limitation is driven by the need to tightly focus the laser beam at arbitrary depths within the photopolymer. This requirement necessitates matching the photopolymer's refractive index to specific values but the required techniques have not been disseminated widely in the open scientific literature. To address this knowledge gap, we test two universal, different approaches to generate refractive index-matched polymeric and preceramic resins and demonstrate their performance by printing of fine submicron features in 3D structures as tall as 2.5 mm. Specifically, we achieve index-matching by mixing commercially-available resins or covalent modification of functional monomers. This work investigates the relationship of voxel shape to RI mismatch, and presents tuning of RI through mixing and covalent modification to a nonconventional material system of preceramic resin which has never been demonstrated before. We demonstrate the material flexibility by generating 3D silicon oxycarbide structures from preceramic resists while simultaneously eliminating the part-height limitation of conventional DLW-TPP.

A novel multi-metal oxide nanocomposite, Ag2O·SrO·CaO, was synthesized by a facile co-precipitation method followed by calcinations. The synthesized nanocomposite was characterized by XRD, FESEM, EDS, TEM, FTIR spectroscopy and photoluminescence (PL) spectroscopy. The composite showed enhanced photocatalytic activity under visible light irradiation and excellent anti-bacterial performance against both Gram-positive and Gram-negative bacteria. Here, the synthesized Ag2O·SrO·CaO nanomaterials were deposited on a glassy carbon electrode (GCE) in the form of a thin film to fabricate the desired electrochemical sensor and subjected to I–V analysis of 3-chlorophenol (3-CP) in a phosphate buffer solution (PBS). A calibration curve was plotted from the linear relation of current versus concentration and used to calculate the sensitivity (8.9684 μA μM−1 cm−2), linear dynamic range (LDR, 0.1 nM to 0.01 mM) and lower limit of detection (DL, 97.12 ± 4.86 pM). The analytical parameters of the sensor such as response time, reproducibility and long-term stability in the detection of 3-CP were reliable. Finally, it was used to analyze real samples collected from various environmental sources and found to be acceptable.

Ocular diseases have a significant effect on vision and quality of life. Drug delivery to ocular tissues is a challenge to formulation scientists. The major barriers to delivering drugs to the anterior and posterior segments include physiological barriers (nasolacrimal drainage, blinking), anatomical barriers (static and dynamic), efflux pumps and metabolic barriers. The static barriers comprise the different layers of the cornea, sclera, and blood-aqueous barriers whereas dynamic barriers involve conjunctival blood flow, lymphatic clearance and tear drainage. The tight junctions of the blood-retinal barrier (BRB) restrict systemically administered drugs from entering the retina. Nanocarriers have been found to be effective at overcoming the issues associated with conventional ophthalmic dosage forms. Various nanocarriers, including nanodispersion systems, nanomicelles, lipidic nanocarriers, polymeric nanoparticles, liposomes, niosomes, and dendrimers, have been investigated for improved permeation and effective targeted drug delivery to various ophthalmic sites. In this review, various nanomedicines and their application for ophthalmic delivery of therapeutics are discussed. Additionally, scale-up and clinical status are also addressed to understand the current scenario for ophthalmic drug delivery.

A series of naphthalimide-chalcone/pyrazoline conjugates was prepared and evaluated for their anti-breast cancer potential against estrogen responsive, i.e. MCF-7 (ER+), and triple-negative, i.e. MDA-MB-231 (ER−), cell lines. The structure-activity-relationship (SAR) was deduced based on the influence of linker length, substituents on the phenyl ring and the generated functionalities, on anti-proliferative activities. Docking simulations further delineate the type of interactions of the designed molecules with the selected targets. This report discloses the scope of triazole tethered naphthalimide-chalcone/pyrazoline conjugates as anti breast cancer agents.

In this work, a tri-metal based nanocomposite was synthesized and characterized. A detailed investigation of the photocatalytic dye degradation efficiency of the nanocomposite under visible light showed promising results in a wide pH range, both acidic and basic medium. Studies on anti-bacterial activity against seven pathogenic bacteria, including both Gram positive and Gram negative species, were conducted in the presence and absence of light and compared with the standard antibiotic gentamicin. The minimum inhibitory concentration (MIC) values of Ag·NiMn2O4 against multidrug-resistant (MDR) pathogens ranged from 0.008 to 0.65 μg μL−1, while the minimum bactericidal concentration (MBC) was found to be 0.0016 μg μL−1. The nanomaterial, Ag·NiMn2O4 was deposited onto the surface of a glassy carbon electrode (GCE; 0.0316 cm2) as a thin film to fabricate the chemical sensor probe. The proposed sensor showed linear current (vs. concentration) response to m-THyd (m-tolyl hydrazine) from 1.0 pM to 0.01 mM, which is denoted as the linear dynamic range (LDR). The estimated sensitivity and detection limit of the m-THyd sensor were found to be 47.275 μA μM−1 cm−2 and 0.97 ± 0.05 pM, respectively. As a potential sensor, it is reliable due to its good reproducibility, rapid response, higher sensitivity, working stability for long duration and efficiency in the analysis of real environmental samples.

This review contains up-to-date knowledge and recent advancements on the essentiality, sources, and toxicological profile of nickel and its different compounds. Nickel is a recognized essential element for several important biological processes like the healthy growth of plants, animals, and soil/water microbes; though an excess amount of nickel intoxicates flora and fauna. Nickel is found to affect the photosynthetic function of higher plants; it can severely degrade soil fertility and causes many chronic diseases in humans. Due to the huge growth in the nickel industry and consumption of nickel-containing products, environmental pollution has become inevitable by the element nickel and also varieties of its by-products through all the phases of making, utilization and dumping. We have focused on the importance of agenda 2030 (UN 17 SDGs) during the preparation of the write-up and have highlighted goals 3, 6, 8, 9, 11, 12, 13, 14, and 15 by elaborately discussing associated points. The plausible molecular mechanism of nickel toxicity is presented in simple diagrams. The article elaborates on possible methods for remediation of nickel toxicity and the treatment of nickel dermatitis and nickel cancer. Recent advancements in the understanding of the dual aspects of nickel as beneficial and a carcinogen are the key subject of this article.

The development of drug carriers based on nanomaterials that can selectively carry chemotherapeutic agents to cancer cells has become a major focus in biomedical research. A novel pH-sensitive multifunctional envelope-type mesoporous silica nanoparticle (SBA-15) was fabricated for targeted drug delivery to human colorectal carcinoma cells (HCT-116). SBA-15 was functionalized with folic acid (FA), and the material was loaded with the water-insoluble flavonoid, quercetin (QN). Additionally, acid-labile magnetite Fe3O4 nanoparticles were embedded over the FA-functionalized QN-loaded monodisperse SBA-15 to prepare the highly orchestrated material FA-FE-SBA15QN. The in vitro and in vivo anti-carcinogenic efficacy of FA-FE-SBA15QN was carried out to explore the pH-sensitive QN release with putative mechanistic aspects. FA-FE-SBA15QN caused a marked tumor suppression, and triggered mitochondrial-dependent apoptosis through a redox-regulated cellular signaling system. Furthermore, FA-IO-SBA-15-QN initiated the c-Jun N-terminal Kinase (JNK)-guided H2AX phosphorylation, which relayed the downstream apoptotic signal to the phosphorylate tumor suppressor protein, p53. On the other hand, the selective inhibition of heat shock protein-27 (HSP-27) by FA-FE-SBA15QN augmented the apoptotic fate through JNK/H2AX/p53 axis. The in vitro and in vivo magnetic resonance imaging (MRI) studies have indicated the theranostic perspective of the composite. Thus, the result suggested that the newly synthesized FA-FE-SBA15QN could be used as a promising chemo theranostic material for the management of carcinoma.

Naturally extracted glycosaminoglycan chondroitin sulphate is the reactive product of N-acetylgalactosamine and D-glucuronic acid. Chondroitin sulfate (CS) extracted from Scophthalmus maximus, H. scabra, E. fraudatrix, M. magnum, and H. mexicana has shown remarkable anticoagulant, articular cartilage repair, corneal lesion healing, antidiabetic, and antiproliferative effects. Also, platinum and strontium nanoparticles of chondroitin sulfate are effective in osteoarthritis and exert anti-HSV2 and anti-angiogenic properties. A combination of chondroitin sulfate and RNA lipolexes demonstrates gene silencing effects in liver fibrosis. Chondroitin sulfate has also been used as a carrier for loxoprofen hydrogel preparation. Oligosaccharides of chondroitin sulfate showed effective inhibition of bovine testicular hyaluronidase enzyme as an antibacterial agent during pregnancy. Monoclonal antibody-recognized chondroitin sulfate A was effectively used to treat ameloblastoma. Selenium-chondroitin sulfate nanoparticles demonstrated positive effects in therapy of Kashin–Beck disease (KBD) and osteoarthritis.
