Non-destructive methods have been widely recognized for evaluating fruit quality traits of many horticultural crops and food processing industry. Destructive (analytical) test, and non-destructive evaluation of the quality traits were investigated and compared for ‘Red Rose’ tomato (Solanum lycopersicum L.) fruit grown under protected environment. Fresh tomato fruit at five distinctive maturity stages namely; breaker (BK), turning (TG), pink (PK), light-red (LR), and red (RD) were labeled and scanned using the handheld near infra-red (NIR) enhanced spectrometer at a wavelength range of 285–1200 nm. The labeled tomato samples were then measured analytically for flesh firmness, lycopene, β-carotene, total phenolic content (TPC) and total flavonoids content (TFC). The results revealed that quality traits could be estimated using NIR spectroscopy with a relatively high coefficient of determination (R2): 0.834 for total phenolic content, 0.864 for lycopene, 0.790 for total flavonoid content, 0.708 for β-carotene; and 0.679 for flesh firmness. The accumulation of Lyco and β-Car rapidly increased in tomatoes harvested between the TG and the LR maturity stages. Harvesting tomatoes at BK maturity stage resulted in significantly higher flesh firmness than harvesting at the later maturity stages. Tomato fruits had the lowest TPC and TFC contents at the earliest maturity stage (BK), while they had intermediate TPC and TFC levels at LR and RD maturity stages. NIR spectroscopic measurements of fruit firmness and lipophilic antioxidants in tomato fruit at various maturity stages were partially in accordance with those estimated by destructive (analytical) methods. Based on these findings, we recommend using non-destructive NIR spectroscopy as an effective tool for predicting tomato fruit quality during harvest stage and postharvest processing.

The use of synthetic antioxidants has been associated with serious concerns for human and environmental health. During ripening stages, tomato fruit is exposed to different abiotic stresses which not only influence its nutritional, mechanical, and functional properties at harvest, but also affect the quality and shelf life of the fruit during storage. This study investigated the pattern of changes in dietary antioxidants during various ripening stages of tomato fruit (cv. Red Rose) and their impact on storage behavior of the fruit during cold storage. Tomato fruits were harvested at mature green, breaker, turning, pink, light-red and red stages of maturity. Then, they were analysed for flesh firmness, soluble solids content, titratable acidity, total sugars, pH, dry matter content, lipophilic (lycopene, β-carotene, and total carotenoids), and hydrophilic (ascorbic acid, phenolic and flavonoids) antioxidants. Additional fruits were harvested at each maturity stage and divided into three equal lots, then were subjected to low-temperature (10 ± 1 °C) storage with 80 ± 5% RH, for 7, 14, and 21 days. Flesh firmness, and the levels of dietary antioxidants were analysed following the subsequent storage periods. The results revealed that the peak of hydrophilic antioxidants such as ascorbic acid, phenolic compounds, and flavonoids was between the ‘pink’ and the ‘light-red’ stages of fruit maturity. Whereas tomatoes harvested at the ‘red’ stage of maturity had the highest levels of lycopene and β-carotene. Both the stage of fruit maturity at harvest and duration of cold storage influenced flesh firmness, organoleptic and functional properties of ‘Red Rose’ tomato fruit. In conclusion, the results of the current investigation have practical implications in formulating foods with improved functional properties at processing industries.

Weeds and ornamental plants serve as a reservoir for geminiviruses and contribute to their dissemination, genome recombination and/or satellite capture. Ornamental sunflower (Helianthus spp.) plants exhibiting mild leaf curl symptoms were subjected to begomovirus and DNA-satellites isolation. The full-length genome of the isolated begomovirus clone (Od1-A) showed 96.8% nucleotide (nt) sequence identity with mesta yellow vein mosaic virus (MeYVMV; accession no. FR772081) whereas, alphasatellite (Od1-a) and betasatellite (Od1-b) clones showed their highest nt sequence identities at 97.4% and 98.2% with ageratum enation alphasatellite (AEA; accession no. FR772085) and papaya leaf curl betasatellite (PaLCuB; accession. no. LN878112), respectively. The evolutionary relationships, average evolutionary divergence and the recombination events were also inferred. The MeYVMV exhibited 9.5% average evolutionary divergence and its CP and Rep had 9.3% and 12.2%, concomitantly; the alphasatellite and the betasatellite had 8.3% and 5.2%, respectively. The nt substitution rates (site-1 year−1) were found to be 6.983 × 10-04 and 5.702 × 10-05 in the CP and Rep of MeYVMV, respectively. The dN/dS ratio and the Tajima D value of MeYVMV CP demonstrated its possible role in host switching. The absolute quantification of the begomovirus demonstrated that mild symptoms might have a correlation with low virus titer. This is the first identification of MeYVMV and associated DNA-satellites from ornamental sunflower in Pakistan. The role of sequence divergence, recombination and importance of MeYVMV along with DNA-satellites in extending its host range is discussed.

Superparasitism occurs in Cotesia glomerata L. (Hymenoptera: Braconidae), a gregarious endoparasitoid of Pieris spp. (Lepidoptera: Pieridae). The responses of Pieris brassicae L. larvae to superparasitism were examined in order to elucidate the ecological significance of this behaviour. Models of tritrophic interactions often imply that attraction of herbivore natural enemies by the plant constitutes a defence. Parasitoid attack on herbivores is assumed to result in a reduction in herbivory and or an increase in plant fitness. Coupled with the active involvement of the plant in producing signals, this can be seen as an indirect mediation of wound induced defence. The results show that superparasitism of P. brassicae by the parasitoid C. glomerata reduced survivorship but increased food consumption and weight growth in P. brassicae larvae. The duration of host larval development was found prolonged as the number of oviposition increased and superparasitized larvae (three to five time parasitized) grew slower than unparasitized larvae or larvae parasitized one or two times.

6. Morphological and physiological response of sour orange ( Citrus aurantium L.) seedlings to the inoculation of taxonomically characterized bacterial endophytes.

- Abstract
Entophytic bacteria (EBs) are very diverse and found in virtually all plant species studied. These natural EBs live insides the host plant and can be used to maximize crop and fruit yield by exploiting their potential. In this paper, EBs characterization from various citrus genotypes and their influence on the morphological and physiological functioning of sour orange ( Citrus aurantium ) seedlings are described. To assess the influence of 10 distinct EBs, three different techniques (injection, soil mix, and spray) were applied for single and mixed inoculation on sour orange ( C. aurantium ) seedlings. The selected strains were identified as firmicutes ( Enterococcus faecalis, Bacillus safensis, Bacillus cereus, Bacillus megaterium, Brevibacillus borstelensis & Staphylococcus haemolyticus), and gamma Proteobacteria ( Enterobacter hormachaei, Proteus mirabilis, Pseudomonas aeruginosa, & Pseudomonas sp.) by 16S rRNA gene sequencing. To investigate the influence of these EBs on host plant morphology, different parameters (morphometric) were recorded after five WOI (weeks of inoculation), including shoot/root length, shoot/root fresh and dry biomass, and biophysical analyses i.e., relative water content (RLWC). Physiological markers such as chlorophyll & carotenoid content, protein content, proline content, phenolics, and flavonoids were also analyzed to determine the influence of endophytes on sour orange seedlings. Five strains such as SM-34, SM-20, SM-36, SM-68, and SM-56 significantly improved the development and physiology of sour orange seedlings. Bacillus cereus and Pseudomonas aeruginosa produced the best outcomes in terms of plant growth. The relative quantification of bacterial inoculums was determined using real-time PCR. A rise in the number of bacterial cells in inoculated treatment suggests that bacterial strains survived and colonized successfully, and also shown their competitiveness with native bacterial community structure. As per the results of inoculation methods, soil mixing, and injection methods were determined to be effective for bacterial inoculation to plants but a variable trend was found for different parameters with test bacterial strains. Contamination of food chain by Polychlorinated biphenyls through use of pesticides, electric and industrial waste poses human health risk. In previous studies, PCB species were stated as endocrine disrupting pollutants and showed toxic health effects like cancerous and noncancerous in animals. The aim of this study was to investigate the levels of PCBs and its toxicity equivalence in food item from plant source to evaluate the health risk in Khanewal and Multan, Pakistan. Samples were collected and processed for further analysis of PCB species through GC/MS after extraction and clean up. The mean concentrations of PCBs ranged as 2.71-151.67 ng/g in beans and grains and 2.30-97.00 ng/g dry weight in vegetables and were lower than 200-3000 ng/g PCBs recommended by FDA tolerance level for all foods. The mean concentrations of two NDL-PCB species detected in all vegetables, beans and grains except S.indicum and T.aestivum were lower than maximum allowable concentration of non-dioxin like PCBs i.e. 40ngg
-1 reported by European Commission. Mean TEQ of sum of 14 PCB species ranged as 1.52-5.91 ng-WHO-TEQg-1 in vegetables and 1.46-10.04 ng-WHO-TEQg-1 in beans and grains. The present study concluded that the mean concentrations and mean TEQs of PCB species in most of the vegetables, beans and grains were found safe but due to higher consumption rate of some vegetables and grains, posed the moderate level of risk for human health. This study emphasizes on an implement of the strict rules regarding the use of restricted chemicals to diminish the effluence in food chains. 8. Potential health risks assessment cognate with selected heavy metals contents in some vegetables grown with four different irrigation sources near Lahore, Pakistan.

Carcinogenic and health hazard causing heavy metals have been increasing in our dietary stuffs due to large amount of industrial effluents being dumped in water bodies that are ultimately used for irrigation purposes. The study was aimed to assess and compare the mean concentrations of heavy metals (Cd, As and Pb) in soil and vegetables irrigated with four different sources (Ground water, river water, domestic sewage water and industrial untreated effluents and domestic waste water receiving drains) for the estimation of carcinogenic and non-carcinogenic health risk associated with them. Prepared samples were analyzed by through ICP-OES. Statistical analysis revealed that domestic sewage water and drains water usage for irrigation purposes leads to high values of Estimated Daily Intake (EDI) of metals through vegetation. To assess the carcinogenic effects values daily intakes, Total hazard quotients (THQs) and Health indexes (HI), while for carcinogenic effects, Total cancer risks (TCR) were determined. The results of present study revealed that the daily intakes of these metals are far less than that of permissible levels but their bio-accumulating behavior produce high risks to human health. The HI values revealed that waste water usage is producing the vegetables of high health risks. In adults, the HI of Phaseolus vulgaris , Spinacia oleracea , Brassica compestris , Raphnus sativus, Daucus carota and Solanum tuberosum assessed as 0.81, 1.52, 1.26, 0.12, 0.22, and 0.15 (ground water irrigation), 0.046, 0.75, 0.51, 0.68, 0.90 0.064 (River Ravi water irrigation), 1.23, 3.34, 4.81, 4.23, 1.41 and 3.43 (domestic sewage irrigation) and 3.04, 5.50, 6.08, 2.50, 5.34 and 5.13 (Drain waste water irrigation), respectively.
