Phthalates (PAEs) are ubiquitous in soils and food products and thus pose a high risk to human health. Herein, genome mining revealed a great diversity of bacteria with PAEs-degrading potential. Mining of the genome of Raoultella ornithinolytica XF201, a novel strain isolated from Dongxiang wild rice rhizosphere, revealed the presence of two silenced tandem genes pcdGH (encoding protocatechuate 3,4-dioxygenase, 3,4-PCD), key aromatic ring-cleaving genes in PAEs biodegradation. Ribosome engineering was successfully utilized to activate the expression of pcdGH genes to produce 3,4-PCD in the mutant XF201-G2U5. The mutant XF201-G2U5 showed high 3,4-PCD activity and could remove 94.5 % of di-n butyl phthalate (DBP) in 72 h. The degradation kinetics obeyed the first-order kinetic model. Strain XF201-G2U5 could also degrade the other PAEs and the main intermediate metabolites, ultimately leading to tricarboxylic acid cycle. Therefore, this strategy facilitates novel bacterial resources discovery for bioremediation of PAEs and other emerging contaminants.

With the rapid development of the nano-material and chemical industry, more and more microplastic (MP) and nano-material were discharged into the environment. In this study, a two-dimensional (2D) surface of Extended Darjaguin-Landau-Verwe-Overbeek (XDLVO) is proposed to quantitatively investigate the effect of polyamide (PA) on the transport of graphene oxide (GO) in porous media. The influences of mass fraction of PA, flow rate, GO concentration, ionic type and strength on the migration of GO in saturated porous media are investigated by column experiments and numerical models. The two-dimensional (2D) surfaces of XDLVO interaction energy between GO and GO, GO and QS, GO and PA, are firstly calculated to analyze the transport of GO in saturated porous media. Experimental results suggest the mobility of GO is enhanced when flow velocity and initial concentration of GO are increased. However, the mobility of GO is inhibited when the mass fraction of PA and ionic strength are increased. More important, the inhibitory effect of divalent cations on GO migration is stronger than that of monovalent cations. Simultaneously, XDLVO results suggest that ionic types and strengths are important factors affecting the mobility of GO in porous media, and the critical ionic strength is observed from the continuous variation of the secondary minimum trap of XDLVO interaction energy. Model results show that there is a linear relationship between the logarithm of the secondary minimum trap of XDLVO interaction energy and the parameters related to GO mobility, which suggests XDLVO energy surface has an important application significance in the accurate quantification of GO mobility in porous media. These findings contribute to GO transport affected by microplastic in porous media, thus laying a significant foundation for the environmental risk and contamination remediation.

The cultivation of crop cultivars with low pollutant accumulation is an important strategy to reduce the potential health risks of food produced from polluted soils. In this study, we identified three loose-leaf lettuce cultivars with low accumulation of perfluorooctanoic acid (PFOA), a highly toxic and persistent organic pollutant. PFOA concentrations in the shoots of low-PFOA cultivars were 3.7–5.5-fold lower than those of high-PFOA cultivars. The identification of low-PFOA cultivars could contribute to ensuring food safety despite cultivation in highly polluted soils (1 mg/kg) based on the tolerable daily PFOA intake (1.5 μg/kg/d). We detected lower desorbing fractions of PFOA in rhizosphere soil, lower bioconcentration factors, and higher distribution in the cell walls and organelles of roots in low-PFOA cultivars, all of which are key factors in limiting PFOA uptake and translocation from soil to shoots, than in high-PFOA cultivars. This study reveals the mechanism of PFOA uptake from soil to crop and lays a foundation for establishing a cost-effective strategy to plant crops in polluted soil and reduce exposure risk due to persistent organic pollutants in crops.

A pot experiment was conducted to investigate the effect of a series of β-cyclodextrin (β-CD) concentrations on bioremediation of soil co-contaminated with Cd and BDE-209 using amaranth and the white-rot fungus Phanerochaete chrysosporium, with BDE-209 degrading ability. Results showed that the white-rot fungus was beneficial to the growth of amaranth, Cd uptake and BDE-209 degradation. Addition of β-CD further increased biomass of both shoots and roots, shoot Cd concentrations and contents, chlorophyll concentrations and soil manganese peroxidase (MnP) activities. Furthermore, well-organized mesophyll cells were observed in β-CD treatments, implying that the combination of white-rot fungus and β-CD can alleviate the stresses of Cd and BDE-209 to mesophyll cells. The BDE-209 degradation rate was positively correlated to β-CD concentration and MnP activity in soil. Our results also revealed that RF+β0.8 treatment possessed the greatest Cd removal efficiency due to its well-configured mesophyll cells and the highest shoot biomass, chlorophyll concentration, and shoot Cd concentration. Considering simultaneous removal of Cd and BDE-209 from soil, using 0.8% β-CD to amaranth inoculated with white-rot fungus is a promising way forward for the phytoremediation of soil co-contaminated with Cd and BDE-209. A high percentage of mono-BDE was detected in inoculated amaranth, suggesting that BDE-209 was debrominated into low brominated PBDEs by the fungus in soil, which were then absorbed and further debrominated into mono-BDE in the plant.

Phthalate acid esters (PAEs) are environmentally ubiquitous and have aroused a worldwide concern due to their threats to environment and human health. Di-n-butyl phthalate (DBP) is one of the most frequently observed PAEs in the environment. In this study, a novel bacterium identified as Pseudomonas sp. YJB6 that isolated from PAEs-contaminated soil was determined to have strong DBP-degrading activity. A complete degradation of DBP in 200 mg/L was achieved within 3 days when YJB6 was cultivated at 31.4 °C with an initial inoculation size of 0.6 (OD600) in basic mineral salts liquid medium (MSM), pH 7.6. The degradation curves of DBP (50–2000 mg/L) fitted well the first-order kinetics model, with a half-life (t1/2) ranging from 0.86 to 1.88 d. The main degradation intermediates were identified as butyl-ethyl phthalate (BEP), mono-butyl phthalate (MBP), phthalic acid (PA) and benzoic acid (BA), indicating a new complex and complete biodegradation pathway presented by YJB6. DBP might be metabolized through de-esterification, β-oxidation, and hydrolysis, followed by entering the Krebs cycle of YJB6 as a final step. Strain YJB6 was successfully immobilized with sodium alginate (SA), polyvinyl alcohol (PVA), and SA-PVA. The immobilization significantly improved the stability and adaptability of the cells thus resulting in high volumetric DBP-degrading rates compared to that of the freely suspended cells. In addition, these immobilized cells can be reused for many cycles with well conserved in DBP-degrading activity. The ideal DBP degrading ability of the free and immobilized YJB6 cells suggests that strain YJB6, especially the SA-PVA+ YJB6 promises great potential to remove hazardous PAEs.

Sorption of organic contaminants by biochar greatly affects their bioavailability and fate in soils. Nevertheless, very little information is available regarding the effects of biochar on sorption and desorption of organic contaminants in different soil particle-size fractions. In this study, di-n-butyl phthalate (DBP), a prevalent organic contaminant in agricultural soils, was taken as a model contaminant. The effects of biochar on DBP sorption and desorption in six particle-size fractions (i.e., coarse sand, fine sand, coarse silt, fine silt, clay, and humic acid fractions) of paddy soil were investigated using batch sorption-desorption experiments. A straw-derived biochar with high specific surface area (116 m(2)/g) and high content of organic matter (OM) rich in aromatic carbon (67%) was prepared. Addition of this biochar (1% and 5%) significantly promoted the sorption and retention of DBP in all the paddy soil particle-size fractions at environmentally relevant DBP concentrations (2-12 mg/L) with 1.2-132-fold increase of the K-d values. With increasing addition rates of biochar, DBP retention by the biochar enhanced. The biochar's effectiveness was remarkably influenced by the physicochemical properties of the soil particle-size fractions, especially, the OM contents and pore size showed the most striking effects. A parameter (r(kd)) reflecting the biochar's effectiveness showed negative and positive correlations with OM contents and pore size of the soil particle-size fractions, respectively. Accordingly, strong effect of the biochar was found in the soil fractions with low OM contents and high pore size. The findings of this study gave insight into the effects and influencing factors of biochar on sorption and desorption of organic contaminants in soils at scale of various particle-size factions.
