Abstract The mechanical properties of rocks in cold regions undergo significant changes as a result of decades of freeze‒thaw cycles with seasonal variations, which can lead to a series of geological disasters, such as collapse. This study investigates the evolution of the mechanical characteristics and internal progressive damage characteristics of mixed granite under freeze‒thaw cycling and axial loading. By measuring the mass, wave velocity, and uniaxial compressive strength of rock samples and combining these metrics with acoustic emission (AE) characteristics, the physical and mechanical properties and microfracture development of mixed granite after different numbers of freeze‒thaw cycles were investigated. The results indicate that as the number of freeze‒thaw cycles increases, the longitudinal wave velocity, uniaxial compressive strength, and elastic modulus of the mixed granite decrease nonlinearly, while the peak strain gradually increases. Combined with the stress‒strain curve, the AE characteristics can be divided into four stages. As the number of freeze‒thaw cycles increases, the AE cumulative count decreases, and the AE counts of the four stages are different. The low-frequency-high-amplitude signals first increases and then tends to stabilize, and they only appeared in the third and fourth stages. At the same time, the proportion of the low-frequency ratio gradually increases, and the proportion of the high-frequency ratio decreases. In addition, based on the rise time/amplitude (RA) and average frequency (AF) characteristics and failure modes, it was found that the internal crack types of mixed granite transition from shear cracks to tensile cracks, among which tensile cracks play a crucial role in rock failure.

Abstract In practical engineering, the development of surface cracks is one of the most important reasons for the destruction of the rock mass, and the development of complex morphology fractures on the rock mass surface significantly influences rock mass mechanics. This paper addresses the freeze–thaw damage issue in rock mass containing surface fractures in cold regions. Tuffs and a control group are selected as research samples, with the control group being prefabricated surface jointed specimens with an inclination angle of 70° and a fracture depth ratio d (crack depth) /t (sample width) of 0.26. The study analyzes the mass, wave velocity loss, macro-microcosmic fracture damage morphology, and mechanical properties of the two specimen groups through laboratory freeze–thaw cycle tests, uniaxial compression tests, and scanning electron microscopy examinations. The results show an overall decrease in mass, wave velocity, and uniaxial compressive strength as the cycle number increases, with the prefabricated jointed group samples showing more significant changes. However, the two specimen groups exhibit different macroscopic failure fracture states. In addition, scanning electron microscopy images illustrate that after freeze–thaw cycles, the large rock mass particles break into smaller fragments, resulting in looser particle arrangements and a transition from initial surface cementation to point contact., which weakens the compressive strength of the rock mass. The paper also explains the mechanism of the diminishing impact of freeze–thaw cycles on the strength of the rock mass after a certain number of cycles. The research outcomes hold significant reference value for engineering construction in cold regions.

Abstract Endophytes have been shown to promote plant growth and health. In the present study, a Bacillus velezensis CH1 (CH1) strain was isolated and identified from high-quality oats, which was capable of producing indole-3-acetic acid (IAA) and strong biofilms, and capabilities in the nitrogen-fixing and iron carriers. CH1 has a 3920 kb chromosome with 47.3% GC content and 3776 code genes. Compared genome analysis showed that the largest proportion of the COG database was metabolism-related (44.79%), and 1135 out of 1508 genes were associated with the function “biosynthesis, transport, and catabolism of secondary metabolites.” Furthermore, thirteen gene clusters had been identified in CH1, which were responsible for the synthesis of fifteen secondary metabolites that exhibit antifungal and antibacterial properties. Additionally, the strain harbors genes involved in plant growth promotion, such as seven putative genes for IAA production, spermidine and polyamine synthase genes, along with multiple membrane-associated genes. The enrichment of these functions was strong evidence of the antimicrobial properties of strain CH1, which has the potential to be a biofertilizer for promoting oat growth and disease resistance.

Abstract A set of nozzle equipment for proton therapy is currently under development at China Institute of Atomic Energy (CIAE). To facilitate the off-line commissioning of the whole equipment, a set of ionization chamber signal generation system, known as the test electronics, was designed. The results showed that the system can simulate the beam position, beam fluence (which exhibits a positive correlation with the dose), and other related analog signals generated by the proton beam when it traverses the ionization chamber. Moreover, the accuracy of the simulated beam position is within ± 0.33 mm, and the accuracy of the simulated beam fluence signal is within ± 1%. The test electronics can output analog signals representing environmental parameters. The test electronics meets the design requirements, which can be used for the commissioning of the nozzle system as well as the treatment control system without the presence of the proton beam.

Abstract The optimal design of groundwater circulation wells (GCWs) is challenging. The key to purifying groundwater using this technique is its proficiency and productivity. However, traditional numerical simulation methods are limited by long modeling times, random optimization schemes, and optimization results that are not comprehensive. To address these issues, this study introduced an innovative approach for the optimal design of a GCW using machine learning methods. The FloPy package was used to create and implement the MODFLOW and MODPATH models. Subsequently, the formulated models were employed to calculate the characteristic indicators of the effectiveness of the GCW operation, including the radius of influence (R) and the ratio of particle recovery (Pr). A detailed collection of 3000 datasets, including measures of operational efficiency and key elements in machine learning, was meticulously compiled into documents through model execution. The optimization models were trained and evaluated using multiple linear regression (MLR), artificial neural networks (ANN), and support vector machines (SVM). The models produced by the three approaches exhibited notable correlations between anticipated outcomes and datasets. For the optimal design of circulating well parameters, machine learning methods not only improve the optimization speed, but also expand the scope of parameter optimization. Consequently, these models were applied to optimize the configuration of the GCW at a site in Xi’an. The optimal scheme for R (Q = 293.17 m3/d, a = 6.09 m, L = 7.28 m) and optimal scheme for Pr (Q = 300 m3/d, a = 3.64 m, L = 1 m) were obtained. The combination of numerical simulations and machine learning is an effective tool for optimizing and predicting the GCW remediation effect.

Abstract Syndecan-binding protein (SDCBP) was reported to stimulate the advancement of esophageal squamous cell carcinoma (ESCC) and could potentially be a target for ESCC treatment. There is a growing corpus of research on the anti-tumor effects of iron chelators; however, very few studies have addressed the involvement of dexrazoxane in cancer. In this study, structure-based virtual screening was employed to select drugs targeting SDCBP from the Food and Drug Administration (FDA)-approved drug databases. The sepharose 4B beads pull-down assay revealed that dexrazoxane targeted SDCBP by interacting with its PDZ1 domain. Additionally, dexrazoxane inhibited ESCC cell proliferation and anchorage-independent colony formation via SDCBP. ESCC cell apoptosis and G2 phase arrest were induced as measured by the flow cytometry assay. Subsequent research revealed that dexrazoxane attenuated the binding ability between SDCBP and EGFR in an immunoprecipitation assay. Furthermore, dexrazoxane impaired EGFR membrane localization and inactivated the EGFR/PI3K/Akt pathway. In vivo, xenograft mouse experiments indicated that dexrazoxane suppressed ESCC tumor growth. These data indicate that dexrazoxane might be established as a potential anti-cancer agent in ESCC by targeting SDCBP.

Abstract The digital economy provides new impetus for the high-quality development of manufacturing industry. Through the DEA-Malmquist model and panel regression model, this paper confirmed that there is a positive and significant relationship between the development of digital economy and the green total factor productivity (GTFP) of manufacturing industry. The research result is as follows: (1) the development of digital economy can enhance the overall GTFP of manufacturing industry. (2) The green technology progress brought by the development of digital economy is the main path to promote the GTFP of manufacturing industry. (3) The heterogeneity analysis shows that the impact of digital economy on GTFP of high pollution manufacturing industry is significantly positive, the impact of labor-intensive manufacturing industry is significantly negative, and the impact of technology intensive manufacturing industry is not obvious. The contributions of this study are as follow. In terms of theory, this study theoretically continues Solow’s classical theory, demonstrating the scientific nature of digital technology progress in promoting GTFP growth. In empirical analysis, this study build a new digital economy development level evaluation index system based on the perspective of manufacturing industry. In addition, this study also add a labor-technology-pollution perspective for the development of relevant policies.

Abstract Multiple myeloma (MM) is a prevalent hematological malignancy that poses significant challenges for treatment. Dysregulated cholesterol metabolism has been linked to tumorigenesis, disease progression, and therapy resistance. However, the correlation between cholesterol metabolism-related genes (CMGs) and the prognosis of MM remains unclear. Univariate Cox regression analysis and LASSO Cox regression analysis were applied to construct an overall survival-related signature based on the Gene Expression Omnibus database. The signature was validated using three external datasets. Enrichment analysis and immune analysis were performed between two risk groups. Furthermore, an optimal nomogram was established for clinical application, and its performance was assessed by the calibration curve and C-index. A total of 6 CMGs were selected to establish the prognostic signature, including ANXA2, CHKA, NSDHL, PMVK, SCAP and SQLE. The prognostic signature demonstrated good prognostic performance and correlated with several important clinical parameters, including number of transplants, International Staging System, albumin, beta2-Microglobulin and lactate dehydrogenase levels. The function analysis and immune analysis revealed that the metabolic pathways and immunologic status were associated with risk score. The nomogram incorporating the signature along with other clinical characteristics was constructed and the discrimination was verified by the calibration curve and C-index. Our findings indicated the potential prognostic connotation of cholesterol metabolism in MM. The development and validation of the prognostic signature is expected to aid in predicting prognosis and guiding precision treatment for MM.

Abstract With the aging of the population, an increasing number of elderly patients are opting for artificial joint replacement, leading to the exploration of various rapid rehabilitation programs in the perioperative period. In this study, we aimed to investigate the effectiveness of combining recombinant human erythropoietin and iron sucrose in altering the range and trend of postoperative hemoglobin in patients undergoing arthroplasty. Specifically, we will examine whether this combination can effectively alter the rise and fall of postoperative haemoglobin, identify the inflection point of haemoglobin change or recovery after arthroplasty, and assess the effect of treatment on serum iron in postoperative blood. We conducted a retrospective study of 138 patients who underwent unilateral total joint arthroplasty by the same surgeon in the same hospital before July 2022. The results of this study may provide valuable insights for the development of effective rehabilitation programs for patients undergoing arthroplasty.

Abstract The YOLOv4 approach has gained significant popularity in industrial object detection due to its impressive real-time processing speed and relatively favorable accuracy. However, it has been observed that YOLOv4 faces challenges in accurately detecting small objects. Its bounding box regression strategy is rigid and fails to effectively leverage the asymmetric characteristics of objects, limiting its ability to enhance object detection accuracy. This paper proposes an enhanced version of YOLOv4 called KR–AL–YOLO (keypoint regression strategy and angle loss based YOLOv4). The KR–AL–YOLO approach introduces two customized modules: an keypoint regression strategy and an angle-loss function. These modules contribute to improving the algorithm’s detection accuracy by enabling more precise localization of objects. Additionally, KR–AL–YOLO adopts an improved feature fusion technique, which facilitates enhanced information flow within the network, thereby further enhancing accuracy performance. Experimental evaluations conducted on the COCO2017 dataset demonstrate the effectiveness of the proposed method. KR–AL–YOLO achieves an average precision of 45.6%, surpassing both YOLOv4 and certain previously developed one-stage detectors. The utilization of keypoint regression strategy and the incorporation of robust feature fusion contribute to superior object detection accuracy in KR–AL–YOLO compared to YOLOv4.

Abstract Acute myeloid leukemia (AML) is characterized by an unfavorable prognosis due to the presence of self-renewing leukemic stem cells (LSCs). The presence of T-cell immunoglobulin mucin-3 (TIM-3) on the surface of LSCs has been observed in various types of human AML, exerting an impact on the prognostic outcome. Exploring the hub genes associated with varying levels of TIM-3 expression offers a valuable approach to enhance our understanding of the underlying mechanisms involving TIM-3 and to identify potential prognostic indicators in AML. Nevertheless, to date, no research studies have reported a prognostic model that relies on the level of TIM-3 expression. In our study, we screen the hub-genes based on different expression level of TIM-3 through WGCNA. The prognostic risk-scoring model was constructed based on hub-genes. The results show the risk prognostic model has extraordinary ability to predict prognosis in both the training and validation sets. The high-risk group present poor prognosis with mutation of NPM1, TP53 (Multiple Hit) and FLT3(multiple hit), while IDH2 (Missense Mutation), MUC16 (Multiple Hit/Missense Mutation) occur mutation in low-risk group presenting favorite prognosis than high-risk group. Leukocyte cell–cell adhesion, regulation of T cell activation and I-κB kinase/NF-κB signaling enriched in high-risk group, involving in HSCs or LSCs anchoring to BM, which implicated in LSCs survival and chemotherapy resistance. B7-H3 (CD276) and CD276 would be the potential immune targets in high-risk group. The risk score model may help in distinguishing immune and molecular characteristics, predicting patient outcomes.

Abstract Optical tweezers exert a strong trapping force on cells, making it crucial to analyze the movement of trapped cells. The rotation of cells plays a significant role in their swimming patterns, such as in sperm cells. We proposed a fast deep-learning-based method that can automatically determine the projection orientation of ellipsoidal-like cells without additional optical design. This method was utilized for analyzing the planar rotation of trapped sperm cells using an optical tweezer, demonstrating its feasibility in extracting the rotation of the cell head. Furthermore, we employed this method to investigate sperm cell activity by examining variations in sperm rotation rates under different conditions, including temperature and laser output power. Our findings provide evidence for the effectiveness of this method and the rotation analysis method developed may have clinical potential for sperm quality evaluation.

Abstract Fuchs endothelial corneal dystrophy (FECD) is a slowly evolving, bilateral disease of the corneal endothelium, characterized by an abnormal accumulation of extracellular matrix (ECM) in the basement membrane (Descemet’s membrane, DM). This results in the formation of small round excrescences, called guttae, and a progressive disappearance of endothelial cells. In the intermediate stage, the numerous guttae create significant optical aberrations, and in the late stage, the loss of endothelial function leads to permanent corneal edema. The molecular components of guttae have not been fully elucidated. In the current study, we conducted shotgun proteomics of the DMs, including guttae, obtained from patients with FECD and revealed that 32 proteins were expressed only in the FECD-DMs but not in the DMs of control subjects. Subsequent enrichment analyses identified associations with multiple ECM-related pathways. Immunostaining of flat-mounted DMs confirmed that 4 of the top 5 identified proteins (hemoglobin α, SRPX2, tenascin-C, and hemoglobin γδεβ) were expressed in FECD-DMs but not in non-FECD-DMs. Fibrinogen α was strongly expressed in FECD-DMs, but weakly expressed in non-FECD-DMs. We also demonstrated that matrix-assisted laser desorption ionization imaging mass spectrometry (MALDI-IMS) can display the in situ spatial distribution of biomolecules expressed in the DM, including the guttae.

Abstract Risky decision-making is critical to survival and development, which has been compromised in elderly populations. However, the neural substrates of altered financial risk-taking behavior in aging are still under-investigated. Here we examined the intrinsic putamen network in modulating risk-taking behaviors of Balloon Analogue Risk Task in healthy young and older adults using resting-state fMRI. Compared with the young group, the elderly group showed significantly different task performance. Based on the task performance, older adults were further subdivided into two subgroups, showing young-like and over-conservative risk behaviors, regardless of cognitive decline. Compared with young adults, the intrinsic pattern of putamen connectivity was significantly different in over-conservative older adults, but not in young-like older adults. Notably, age-effects on risk behaviors were mediated via the putamen functional connectivity. In addition, the putamen gray matter volume showed significantly different relationships with risk behaviors and functional connectivity in over-conservative older adults. Our findings suggest that reward-based risky behaviors might be a sensitive indicator of brain aging, highlighting the critical role of the putamen network in maintaining optimal risky decision-making in age-related cognitive decline.

Abstract Organic field-effect transistors (OFETs) with polymer charge-trapping dielectric, which exhibit many advantages over Si-based memory devices such as low cost, light weight, and flexibility, still suffer challenges in practical application due to the unsatisfied endurance characteristics and even the lack of fundamental of behind mechanism. Here, we revealed that the degradation of endurance characteristics of pentacene OFET with poly(2-vinyl naphthalene) (PVN) as charge-storage layer is dominated by the deep hole-traps in PVN by using the photo-stimulated charge de-trapping technique with the fiber-coupled monochromatic-light probes. The depth distribution of hole-traps in PVN film of pentacene OFET is also provided.

Abstract The serological diagnostic criteria for the immune-tolerant (IT) phase have not been strictly defined and it is hard to determine an accurate rate for significant histologic changes among IT patients. The aim of this study was to establish a baseline rate of significant histologic changes and to determine the main characteristics of IT patients. We systematically searched PubMed, Embase, and Web of Science. Studies reporting liver biopsy results (inflammation grade or fibrosis stage) for adults with chronic hepatitis B virus (HBV) infection in the IT phase diagnosed by serological criterion were included to pool the rate of significant histologic changes. Studies that enrolled subjects with confirmed chronic HBV infection in the IT phase diagnosed by serological and liver biopsy criteria (dual criteria) were included to pool the mean values of main characteristics among IT patients. Of 319 studies screened, 15 were eventually included in the meta-analysis. The pooled rates of significant liver fibrosis and inflammatory activity for 10 studies were 10% (95% confidence interval [CI] 0.06–0.18) and 16% (95% CI 0.07–0.31), respectively. The pooled mean values of age, alanine aminotransferase level, HBV DNA level, and HBsAg level for another 5 studies with IT patients diagnosed by dual criteria were 30.7 years (95% CI 27.31–34.09), 26.64 IU/mL (95% CI 24.45–28.83), 8.41 log10 cp/mL (95% CI 7.59–9.23), and 4.24 log10 IU/mL (95% CI 3.67–4.82), respectively. Significant histologic changes were not rare events among IT patients. Strictly defined serological diagnostic criteria for the IT phase are warranted.

Abstract Amino acid-mediated metabolism is one of the key catabolic and anabolic processes involved in diverse cellular functions. However, the role of the semi-essential amino acid arginine in normal and malignant hematopoietic cell development is poorly understood. Here we report that a continuous supply of exogenous arginine is required for the maintenance/function of normal hematopoietic stem cells (HSCs). Surprisingly, knockout of Slc7a3 (CAT3), a major L-arginine transporter, does not affect HSCs in steady-state or under stress. Although Slc7a3 is highly expressed in naïve and activated CD8 T cells, neither T cell development nor activation/proliferation is impacted by Slc7a3 depletion. Furthermore, the Slc7a3 deletion does not attenuate leukemia development driven by Pten loss or the oncogenic Ptpn11 E76K mutation. Arginine uptake assays reveal that L-arginine uptake is not disrupted in Slc7a3 knockout cells. These data suggest that extracellular arginine is critically important for HSCs, but CAT3 is dispensable for normal hematopoiesis and leukemogenesis.

Abstract To better understand the molecular mechanisms of intracranial aneurysm (IA) pathogenesis, we used gene coexpression networks to identify hub genes and functional pathways associated with IA onset. Two Gene Expression Omnibus (GEO) datasets encompassing intracranial aneurysm tissue samples and cerebral artery control samples were included. To discover functional pathways and potential biomarkers, weighted gene coexpression network analysis was employed. Next, single-gene gene set enrichment analysis was employed to investigate the putative biological roles of the chosen genes. We also used receiver operating characteristic analysis to confirm the diagnostic results. Finally, we used a rat model to confirm the hub genes in the module of interest. The module of interest, which was designated the green module and included 115 hub genes, was the key module that was most strongly and negatively associated with IA formation. According to gene set variation analysis results, 15 immune-related pathways were significantly activated in the IA group, whereas 7 metabolic pathways were suppressed. In two GEO datasets, SLC2A12 could distinguish IAs from control samples. Twenty-nine hub genes in the green module might be biomarkers for the occurrence of cerebral aneurysms. SLC2A12 expression was significantly downregulated in both human and rat IA tissue. In the present study, we identified 115 hub genes related to the pathogenesis of IA onset and deduced their potential roles in various molecular pathways; this new information may contribute to the diagnosis and treatment of IAs. By external validation, the SLC2A12 gene may play an important role. The molecular function of SLC2A12 in the process of IA occurrence can be further studied in a rat model.

Abstract To study and prevent the water hazards of deep coal mines roof in the Inner Mongolia–Shaanxi (IM–S) mining area, it is essential to correctly evaluate the water-richness distributions of water-bearing aquifers in roof. This paper puts forward a sediment control method for water-richness law and zoning in the roof aquifers of deep Jurassic coals. To determine the vertical distance of direct water-bearing aquifers, the height of fractured water-conducting zone was detected by an underground network parallel electrical method. The plane and lateral spatial distribution patterns of the water-bearing aquifers and the control of the water-richness distribution was analyzed with the sediment control method. An evaluation system that consisted of four indicators, i.e., sedimentary environmental impact index, interlayer ratio of sandstone and mudstone, sandstone thickness, geophysical water-richness anomaly index was constructed. Furthermore, an Analytic Hierarchy Process (AHP) was introduced to establish the comprehensive zoning map. Finally, through the example analysis of Muduchaideng coal mine, the zoning evaluation results of water-richness were verified by the mine inflow. The findings of this study provide scientific guidance for prevention and control of mine water hazards in the IM–S mining area.

Abstract The co-pyrolysis of oily sludge with biomass to prepare carbon materials is not only an effective way to mitigate oily sludge pollution, but it is also a method of obtaining carbon materials. In this study, a carbon material (OS-CS AC) was obtained by the direct co-pyrolysis of oily sludge (OS) and corn stalks (CS) and then applied to Cr(VI) removal. According to the hydroxy and carboxy masking experiments and the characterization of OS-CS AC by FT-IR, SEM, XPS, XRD, and N2 physical adsorption–desorption, Cr(VI) can be adsorbed efficiently through pore filling, the surface oxygen-containing functional groups can promote the reduction of Cr(VI) to Cr(III) through electron donors, and the greater the electrostatic attraction between the electron-donating functional groups of OS-CS AC and the Cr(VI) is, the stronger the ability to remove Cr(VI). In addition, the removal process was discussed, and the results indicated that the McKay kinetic model, Langmuir isotherm model and Van't Hoff thermodynamic model were the most suitable models for removal. The main factors affecting the removal of Cr(VI) were discussed, and the removal of Cr(VI) reached 99.14%, which gives a comprehensive utilization way of oily sludge and corn stalks.

Abstract With global climate change and the rapid urbanization, urban flood and drought disasters are frequent and urban water supply systems are facing a sea of serious challenges. It is crucial to assess the resilience of urban water supply systems and develop corresponding disaster mitigation and improvement strategies. Urban water supply systems include many subsystems, but existing researches generally focus on a single subsystem. Therefore, this paper proposes a correlation analysis method and a factor analysis method for the resilience evaluation index system of urban water supply systems by combining each subsystem and applying grey system theory. The method can reflect the four dimensions of the water supply process (water source, water plant, supply and distribution network and users) and the five dimensions of the urban management system (society, natural environment, economy, physics and organization). Taking Qingdao as an example, a multi-level integrated evaluation model based on a cloud model is applied to simulate and analyze the resilience of Qingdao's water supply system. As a result, decision support is provided for planning and building resilience systems for urban water systems in the short and long term, based on four main factors.

Abstract The initial in situ stress field influences underground engineering design and construction. Since the limited measured data, it is necessary to obtain an optimized stress field. Although the present stress field can be obtained by valley evolution simulation, the accuracy of the ancient stress field has a remarkable influence. This paper proposed a method using the generative adversarial network (GAN) to obtain optimized lateral stress coefficients of the ancient stress field. A numerical model with flat ancient terrain surfaces is established. Utilizing the nonlinear relationship between measured stress components and present burial depth, lateral stress coefficients of ancient times are estimated to obtain the approximate ancient stress field. Uniform designed numerical tests are carried out to simulate the valley evolution by excavation. Coordinates, present burial depth, present lateral stress coefficients and ancient regression factors of lateral stress coefficients are input to GAN as real samples for training, and optimized ancient regression factors can be predicted. The present stress field is obtained by excavating strata layers. Numerical results show the magnitude and distribution law of the present stress field match well with measured points, thus the proposed method for the stress field inversion is effective.

Abstract The purpose is to explore the brain’s structural difference in local morphology and between-region networks between two types of peripheral neuropathic pain (PNP): postherpetic neuralgia (PHN) and lower back pain (LBP). A total of 54 participants including 38 LBP and 16 PHN patients were enrolled. The average pain scores were 7.6 and 7.5 for LBP and PHN. High-resolution structural T1 weighted images were obtained. Both grey matter volume (GMV) and morphological connectivity (MC) were extracted. An independent two-sample t-test with false discovery rate (FDR) correction was used to identify the brain regions where LBP and PHN patients showed significant GMV difference. Next, we explored the differences of MC network between LBP and PHN patients and detected the group differences in network properties by using the two-sample t-test and FDR correction. Abstract Bemisia tabaci is one of the most notorious agricultural pests in the world. A vicious circle among insect resistance, dose increased, environment and human body impaired as the overuse of synthetic pesticides are becoming increasingly evident. Entomopathogenic Beauveria sp. is known as an effective natural enemy to control B. tabaci. Therefore, this study aimed to purify and identify the biological compounds from Beauveria sp. LY2 via extensive chromatographic techniques, NMR and MS and evaluated for their insecticidal activities against B. tabaci via contact and feeding assay. The outcome identified that one new cerebroside, cerebroside F (1), nine known compounds, cerebroside B (2), bassiatin (3), methyl 1,4-dihydro-4-oxo-2-quinolinecarboxylate (4), cerevisterol (5), 9-hydroxycerevisterol (6), 6-dehydrocerevisterol (7), (22E,24R)-ergosta-8(14),22-diene-3β,5α,6β,7α-tetrol (8), melithasterol B (9) and ergosterol peroxide (10) were isolated. Among the known compounds, methyl 1,4-dihydro-4-oxo- 2-quinolinecarboxylate (4) was isolated from natural origin for the first time. It is demonstrable from the results that compounds 3, 4 and 7 strongly featured insecticidal activities against B. tabaci, being the LC50 value as 10.59, 19.05, 26.59 μg/mL respectively in contact as well as 11.42, 5.66, 5.65 μg/mL respectively in feeding experiment. Moreover, no adverse effect on plant growth/height or phytotoxicity was observed on pepper, cucumber, tomato and cotton. The data from the current study has provided the foundation for the use of newly purified compounds against Bemisia tabaci as an alternative to synthetic chemical compounds.

Abstract Aberrant mechanical factor is one of the etiologies of the intervertebral disc (IVD) degeneration (IVDD). However, the exact molecular mechanism of spinal mechanical loading stress-induced IVDD has yet to be elucidated due to a lack of an ideal and stable IVDD animal model. The present study aimed to establish a stable IVDD mouse model and evaluated the effect of aberrant spinal mechanical loading on the pathogenesis of IVDD. Eight-week-old male mice were treated with lumbar spine instability (LSI) surgery to induce IVDD. The progression of IVDD was evaluated by μCT and Safranin O/Fast green staining analysis. The metabolism of extracellular matrix, ingrowth of sensory nerves, pyroptosis in IVDs tissues were determined by immunohistological or real-time PCR analysis. The apoptosis of IVD cells was tested by TUNEL assay. IVDD modeling was successfully produced by LSI surgery, with substantial reductions in IVD height, BS/TV, Tb.N. and lower IVD score. LSI administration led to the histologic change of disc degeneration, disruption of the matrix metabolism, promotion of apoptosis of IVD cells and invasion of sensory nerves into annulus fibrosus, as well as induction of pyroptosis. Moreover, LSI surgery activated Wnt signaling in IVD tissues. Mechanical instability caused by LSI surgery accelerates the disc matrix degradation, nerve invasion, pyroptosis, and eventually lead to IVDD, which provided an alternative mouse IVDD model.
