We aimed to characterize scenarios where magnetic resonance elastography (MRE) of the liver was ordered and its impact on clinical course and management. 96 consecutive MRE examinations and subsequent encounters over 14 months were reviewed. Indication for MRE of the liver and subsequent management were abstracted from the medical record. In all cases, non-invasive assessment of liver fibrosis was the primary indication and at least one additional rationale was noted. There was a significant decrease in recommendations to undergo liver biopsy after MRE. Additionally, a greater percentage of those recommended to undergo biopsy completed the procedure after discussion of the results. Given the significant cost and rare but serious risks of liver biopsy, MRE of the liver provides an attractive, safer alternative that may have a comparable impact on management, or select cases where biopsy is essential to guide management. We demonstrate the versatility of MRE in real-world hepatology practice, including its utility as a non-invasive surrogate for liver biopsy.

Abstract Aim & background : Intraocular pressure (IOP) measured through the sclera has some advantages over IOP measured through cornea. This study aimed to determine the agreement between IOP as estimated by scleral Schiotz (SS) tonometer and Goldmann applanation tonometer (GAT). Methods: In this cross-sectional observational study, IOP was measured by GAT on the cornea and Schiotz tonometer on the temporal sclera. Axial length and central corneal thickness (CCT) were noted. IOP was then estimated from SS IOP as eGAT using a predictive formula. Agreement of IOP corrected for CCT (cGAT) with eGAT and SS IOP were assessed. The effects of age, axial length, refraction, CCT, gender, and ocular and systemic comorbidity on measured IOP were also assessed. Results: The study included 155 patients with a mean age of 54.27 ± 10.02 years. Mean IOP measured by GAT and scleral Schiotz were 16.58 ± 2.76 mm Hg and 16.10 ± 2.96 mm Hg, respectively. The mean IOP by cGAT and eGAT were 16.70 ± 2.80 mm Hg and 17.07 ± 3.10 mm Hg, respectively. Both eGAT & SS IOP had good agreement with cGAT on the Bland-Altmann plot (p = 0.96 & 0.51, respectively). Further, SS IOP showed good agreement even with GAT IOP (p = 0.78). Age, axial length, refraction, central corneal thickness, gender, and ocular and systemic comorbidity did not show significant correlation with IOP measurement. Conclusion: Estimated GAT IOP showed good agreement with measured GAT values. SS IOP also agreed well with GAT IOP. Clinical significance IOP measurement on the sclera using Schiotz can be used as an alternative to GAT.

Abstract In this work, we have focused on the preparation of copper hexacyanoferrate/boron doped rGO composites (abbreviated as CuHCF/B-rGO) by employing simple co-precipitation technique subsequently processed with ultrasonication method. The XRD spectra confirmed the existence of the cubic structure of copper hexacyanoferrate with high crystalline peaks. The prepared nanocomposite morphology was evaluated by scanning electron microscopy (SEM), and confirmed CuHCF nanoparticles formation with flake-like and wrinkled sheets. Pure CuHCF nanostructures revealed good OER action at 430 mV to obtain 10 mA/cm2. The obtained CuHCF product OER activity can be further upgraded by incorporating the electrically conductive boron doped reduced graphene oxide matrix into CuHCF nanostructures, for the reason that the overpotential of the CuHCF/B-rGO was reduced to 380 mV to attain 10 mA/cm2 with 88 mV/dec Tafel slope value. The doping of heteroatom considerably improves charge-transfer resistance of metal hexacyanoferrate, giving a small resistance value of 2.97 Ω, which was lower than that of CuHCF (5.57 Ω) and CuHCF/rGO (4.31 Ω). Furthermore, the catalytic activity of the CuHCF/B-rGO was stable at prolonged hours with a small decay of 12.5%. Therefore, this work offers new approach to stimulate the catalytic performance of metal hexcyanoferrate by highly conductive carbon-based materials for water splitting performance.

Abstract Appendicitis, an infection and inflammation of the appendix is a prevalent condition in children that requires immediate treatment. Rupture of the appendix may lead to several complications, such as peritonitis and sepsis. Appendicitis is medically diagnosed using urine, blood, and imaging tests. In recent times, Artificial Intelligence and machine learning have been a boon for medicine. Hence, several supervised learning techniques have been utilized in this research to diagnose appendicitis in pediatric patients. Six heterogeneous searching techniques have been used to perform hyperparameter tuning and optimize predictions. These are Bayesian Optimization, Hybrid Bat Algorithm, Hybrid Self-adaptive Bat Algorithm, Firefly Algorithm, Grid Search, and Randomized Search. Further, nine classification metrics were utilized in this study. The Hybrid Bat Algorithm technique performed the best among the above algorithms, with an accuracy of 94% for the customized APPSTACK model. Five explainable artificial intelligence techniques have been tested to interpret the results made by the classifiers. According to the explainers, length of stay, means vermiform appendix detected on ultrasonography, white blood cells, and appendix diameter were the most crucial markers in detecting appendicitis. The proposed system can be used in hospitals for an early/quick diagnosis and to validate the results obtained by other diagnostic modalities.

Abstract Cancer is a global issue and hence various efforts are being made. Iron oxide is considered a significant biochemical agent in the biomedical arena for cancer treatment. Marine macroalgae-mediated iron oxides especially, magnetite (Fe3O4) nanoparticles (NPs) are a prospective alternative to diagnose and treat cancer owing to their fluorescent and magnetic properties. We intend to appraise the usability of the aqueous extract of Rosenvingea intricata (R. intricata) in Fe3O4 NPs synthesis and to study their cytotoxic effects against human hepatocarcinoma (Hep3B) and pancreatic (PANC1) cancer cells. In the present study, R. intricata were collected from the coastal region of South Andaman, India. Aqueous extracts of R. intricata were utilized to synthesize Fe3O4 NPs via the co-precipitation method. Phycosynthesized Fe3O4 NPs exhibited wide peak at 400–600 nm from ultraviolet–visible diffused reflectance spectroscopic analysis which validated the formation of NPs. Band edge emission peak at 660 nm in fluorescent spectra confirmed the quantum confinement in Fe3O4 NPs. Fourier transform infrared spectroscopy confirmed the role of R. intricata as a capping and reducing agent with functional groups such as O–H, C–H, C=O, N=O, C=C, C–O, C–N, and C–S arising from amino acids, polysaccharides, aliphatic hydrocarbons, esters, amides, lignins, alkanes, aliphatic amines, and sulfates. Physicochemical properties such as crystallite size (14.36 nm), hydrodynamic size (84.6 nm), irregular morphology, elemental composition, particle size (125 nm), crystallinity, and saturation magnetization (0.90007 emu/g) were obtained from x-ray diffractometer, dynamic light scattering, scanning electron microscopy, energy dispersive x-ray spectrometer, high-resolution transmission electron microscopy, selected area electron diffraction and vibrating sample magnetometer techniques, respectively. The cell viability showed dose-dependent cytotoxic effects and enhanced the apoptosis against Hep3B and PANC1 cancer cells. R. intricata extract capped Fe3O4 NPs could be the most appropriate and effective nanomaterial for cancer treatment and management.

Abstract Presbyopia is an age-related physiological phenomenon in which eye gradually losses its ability to accommodate. It is one of the leading causes of visual impairment worldwide, especially in adults above the age of 40. If uncorrected, it can significantly impair a patient's quality of life. This study aims to evaluate the factors which affects patient’s need and willingness to accept presbyopic correction. This cross-sectional analytical study was carried out in a semi urban tertiary care hospital from Jan 2021 to June 2022 among patients aged 40 and above who presented to Outpatient department (OPD). Demographic details, medical history, presenting ocular complaints pertaining to presbyopia, spectacle use and decision regarding using near vision correction were noted. Ocular examination included refraction and ocular biometry. Factors that may have influenced complaints of presbyopia or willingness to accept presbyopic correction were analysed. Three hundred and forty two patients with a mean age of 48.55 ± 6.68 years were included. Of these, 262 (76.61%) patients presented with chief complaints related to presbyopia. Those with higher educational qualification (p = 0.031), hypermetropia (p = 0.021), shallower AC depth (p = 0.028) and on medications for systemic ailments (p = 0.01), were more likely to present with chief complaints attributable to presbyopia. Among them, those with higher educational qualifications (p = 0.02) and skilled workers were more likely to accept near vision glasses (p = 0.02), while those with lower Hb (p = 0.01) and myopia (p = 0.01) were less likely to accept correction for presbyopia. Among the 80 patients without chief complaints related to presbyopia, 35 (43.75%) were not willing to accept near vision glasses. Those with higher BMI (p = 0.04) and hypermetropes (p = 0.05) were more willing to accept presbyopic correction. Presbyopia constitutes a significant reason for patients above the age of 40 visiting eye care facility. Multiple socio-economic, systemic and ocular factors influenced both the chief complaints related to presbyopia and willingness to accept presbyopic correction.

Abstract Sperm transcriptomics provide insights into subtle differences in sperm fertilization competence. For predicting the success of complex traits like male fertility, identification of hub genes involved in various sperm functions are essential. The bulls from the transcriptome profiled samples (n = 21), were grouped into good and poor progressive motility (PM), acrosome integrity (AI), functional membrane integrity (FMI) and fertility rate (FR) groups. The up-regulated genes identified in each group were 87, 470, 1715 and 36, respectively. Gene networks were constructed using up- and down-regulated genes from each group. The top clusters from the upregulated gene networks of the PM, AI, FMI and FR groups were involved in tyrosine kinase (FDR = 1.61E−11), apoptosis (FDR = 1.65E−8), translation (FDR = 2.2E−16) and ribosomal pathway (FDR = 1.98E−21), respectively. From the clusters, the hub genes were identified and validated in a fresh set of semen samples (n = 12) using RT-qPCR. Abstract Theoretically, it is more challenging to anticipate the conversion and selectivity of a photochemical experiment compared to thermally generated reactivity. This is due to the interaction of light with a photoreactive substrate. Photochemical reactions do not yet receive the same level of broad analytical study. Here, we close this research gap by presenting a methodology for statistically forecasting the time-dependent progression of photoreactions using widely available LEDs. This study uses NiS/ZnO in perovskite (MAPbI3) solar cells as an additive (5 volume %). The effect of monolithic perovskite solar cells (mPSCs) on forecasting the wavelength of LEDs has been carefully investigated using various characterization methods, including X-ray diffraction (XRD) and Transmission electron microscopy (TEM). The photocatalytic activity was analyzed by measuring the voltage produced. Various factors like selectivity, stability and sensitivity were also examined. This work provides a new perspective to validate NiS/ZnO photocatalysts for predicting the wavelength of different light sources and to apply in photolithography.

Abstract The paper examines the influence of carbon coating on iron nanopowder used as a sintering aid for water-atomized iron powder. Iron nanopowder without such a coating was used as a reference sintering aid to isolate the influence of the carbon coating. Both nanopowder variants were characterised using XPS and HRTEM. The results showed a core–shell structure for both variants. The iron nanopowder is covered by a 3–4 nm thick iron oxide layer, while the carbon-coated iron nanopowder is encapsulated with several nanometric carbon layers. Thermogravimetry conducted in a pure hydrogen environment shows a multipeak behaviour for the carbon-coated iron nanopowder, while a single peak behaviour is observed for the iron nanopowder. Two types of micro/nanobimodal powders were obtained by mixing the nanopowder with water-atomized iron powder. Improved linear shrinkage was observed during sintering when the carbon-coated iron nanopowder was added. This can be explained by the reduction in surface diffusion in the nanopowder caused by the carbon coating, which allows the nanopowder to sinter at higher temperatures and improves densification. Carbon and oxygen analysis, density measurements, optical microscopy and JMatPro calculations were also performed.

Abstract Scutellaria baicalensis is a well-studied medicinal plant belonging to the Lamiaceae family, prized for the unique 4′-deoxyflavones produced in its roots. In this study, three native species to the Americas, S. lateriflora, S. arenicola, and S. integrifolia were identified by DNA barcoding, and phylogenetic relationships were established with other economically important Lamiaceae members. Furthermore, flavone profiles of native species were explored. 4′-deoxyflavones including baicalein, baicalin, wogonin, wogonoside, chrysin and 4′-hydroxyflavones, scutellarein, scutellarin, and apigenin, were quantified from leaves, stems, and roots. Qualitative, and quantitative differences were identified in their flavone profiles along with characteristic tissue-specific accumulation. 4′-deoxyflavones accumulated in relatively high concentrations in root tissues compared to aerial tissues in all species except S. lateriflora. Baicalin, the most abundant 4′-deoxyflavone detected, was localized in the roots of S. baicalensis and leaves of S. lateriflora, indicating differential accumulation patterns between the species. S. arenicola and S. integrifolia are phylogenetically closely related with similar flavone profiles and distribution patterns. Additionally, the S. arenicola leaf flavone profile was dominated by two major unknown peaks, identified using LC–MS/MS to most likely be luteolin-7-O-glucuronide and 5,7,2′-trihydroxy-6-methoxyflavone 7-O-glucuronide. Collectively, results presented in this study suggest an evolutionary divergence of flavonoid metabolic pathway in the Scutellaria genus of Lamiaceae.

Abstract Immunotherapy is a promising new treatment approach for hepatocellular carcinoma (HCC), but there are numerous barriers to immunotherapy in HCC, including an immunosuppressive microenvironment and the “immunotolerance” of the liver. Hyperthermia treatment modalities are standard of care for early stage HCC, and hyperthermia is known to have immunomodulatory effects. We have developed a molecularly targeted photothermal ablation (MTPA) technology that provides thermally tunable, tumor-specific heat generation. The purpose of this study was to evaluate the morphologic and immunologic effects of MTPA in an immunotherapy-resistant syngeneic mouse model of HCC in a background of toxin-induced cirrhosis. We found that the anatomic, cellular, and molecular features of this model recapitulate the characteristics of advanced human HCC. MTPA as a monotherapy and in combination with immune checkpoint therapy significantly increased intratumoral CD3+ and activated CD8+ T cells while decreasing regulatory T cells relative to control or immune checkpoint therapy alone based on immunohistochemistry, flow cytometry, and single cell RNA sequencing data. Furthermore, we identified evidence of MTPA’s influence on systemic tumor immunity, with suppression of remote tumor growth following treatment of orthotopic tumors. The results of this study suggest that tumor-specific hyperthermia may help overcome resistance mechanisms to immunotherapy in advanced HCC.

Abstract Cancer cell heterogeneity (CCH) is crucial in understanding cancer progression and metastasis. The CCH is one of the stumbling blocks in modern medicine's therapeutics and diagnostics . An in-vitro model of co-culture systems of MCF-7, HeLa, HEK-293, with THP-1 cells showed the occurrence of EpCAM positive (EpCAM+) and EpCAM negative (EpCAM−) heterogenetic cancer cell types labeled with the Quantum Dot antibody conjugates (QDAb). This in-vitro model study could provide insights into the role of rare cancer cells manifestation and their heterogeneity in metastatic progression and risk for severe infections in these patients. We successfully report the presence of CCH based on the fluorescence ratios of the co-cultured cancer cells when treated with the QDAb. These short-term mimic co-cultures give a compelling and quite associated model for assessing early treatment responses in various cancers.

Abstract Erianthus arundinaceus [Retzius] Jeswiet, a wild relative of sugarcane has a high biomass production potential and a reservoir of many genes for superior agronomic traits and tolerance to biotic and abiotic stresses. A comparative physiological, anatomical and root transcriptome analysis were carried out to identify the salt-responsive genes and metabolic pathways associated with salt-tolerant E. arundinaceus genotype IND99-907 and salinity-sensitive sugarcane genotype Co 97010. IND99-907 recorded growth of young leaves, higher proline content, higher relative water content, intact root anatomical structures and lower Na+/K+, Ca2+/K+ and Mg2+/K+ ratio as compared to the sugarcane genotype Co 97010. Abstract Predicting language therapy outcomes in bilinguals with aphasia (BWA) remains challenging due to the multiple pre- and poststroke factors that determine the deficits and recovery of their two languages. Computational models that simulate language impairment and treatment outcomes in BWA can help predict therapy response and identify the optimal language for treatment. Here we used the BiLex computational model to simulate the behavioral profile of language deficits and treatment response of a retrospective sample of 13 Spanish-English BWA who received therapy in one of their languages. Specifically, we simulated their prestroke naming ability and poststroke naming impairment in each language, and their treatment response in the treated and the untreated language. BiLex predicted treatment effects accurately and robustly in the treated language and captured different degrees of cross-language generalization in the untreated language in BWA. Our cross-validation approach further demonstrated that BiLex generalizes to predict treatment response for patients whose data were not used in model training. These findings support the potential of BiLex to predict therapy outcomes for BWA and suggest that computational modeling may be helpful to guide individually tailored rehabilitation plans for this population.

Abstract Fall Armyworm (FAW), Spodoptera frugiperda, is a polyphagous pest capable of feeding over 80 plant species and was indigenous to the Western Hemisphere. Within a span of 4 years, FAW has established itself throughout most of the regions in Africa and Asia causing significant losses in maize production. Owing to its revamped distribution range, it would be prudent to analyze the ensuing genetic changes and study the emerging phylogeographic patterns across the world. In this regard, we would like to provide a current snapshot of genetic diversity of FAW in India 2 years after the initial introduction and compare it with the worldwide diversity in order to trace the origins and evolutionary trajectories of FAW in India. We have investigated around 190 FAW samples from different regions in India for strain identity and polymorphism analysis on the basis of partial mitochondrial cytochrome oxidase I (COI) gene sequences. Apart from the ancestral rice and corn strain haplotype, our study demonstrates the presence of 14 more haplotypes unique to India at a haplotype diversity of 0.356. We were also able to record inter-strain hybrid haplotypes of rice and corn strains in India. Regional heterogeneity within Indian populations seems to be quite low representative of extensive migration of FAW within India. Distribution analysis of pairwise differences and rejection of neutrality tests suggest that the FAW population in India might be undergoing expansion. Our data is consistent with the findings suggesting a recent and common origin for invasive FAW populations in Asia and Africa, and does not indicate multiple introductions to India. This study reports the highest genetic diversity for Indian FAW populations to date and will be useful to track the subsequent evolution of FAW in India. The findings would have important ramifications for FAW behavior and composition throughout the world.

Abstract Language outcomes after speech and language therapy in post-stroke aphasia are challenging to predict. This study examines behavioral language measures and resting state fMRI (rsfMRI) as predictors of treatment outcome. Fifty-seven patients with chronic aphasia were recruited and treated for one of three aphasia impairments: anomia, agrammatism, or dysgraphia. Treatment effect was measured by performance on a treatment-specific language measure, assessed before and after three months of language therapy. Each patient also underwent an additional 27 language assessments and a rsfMRI scan at baseline. Patient scans were decomposed into 20 components by group independent component analysis, and the fractional amplitude of low-frequency fluctuations (fALFF) was calculated for each component time series. Post-treatment performance was modelled with elastic net regression, using pre-treatment performance and either behavioral language measures or fALFF imaging predictors. Analysis showed strong performance for behavioral measures in anomia (R2 = 0.948, n = 28) and for fALFF predictors in agrammatism (R2 = 0.876, n = 11) and dysgraphia (R2 = 0.822, n = 18). Models of language outcomes after treatment trained using rsfMRI features may outperform models trained using behavioral language measures in some patient populations. This suggests that rsfMRI may have prognostic value for aphasia therapy outcomes.

Diagnosis and treatment of vocal fold lesions has been a long-evolving science for the otolaryngologist. Contemporary practice requires biopsy of a glottal lesion in the operating room under general anesthesia for diagnosis. Current in-office technology is limited to visualizing the surface of the vocal folds with fiber-optic or rigid endoscopy and using stroboscopic or high-speed video to infer information about submucosal processes. Previous efforts using optical coherence tomography (OCT) have been limited by small working distances and imaging ranges. Here we report the first full field, high-speed, and long-range OCT images of awake patients' vocal folds as well as cross-sectional video and Doppler analysis of their vocal fold motions during phonation. These vertical-cavity surface-emitting laser source (VCSEL) OCT images offer depth resolved, high-resolution, high-speed, and panoramic images of both the true and false vocal folds. This technology has the potential to revolutionize in-office imaging of the larynx.

Abstract This report discusses the first demonstration of electrophoresis assisted time-of-flow mass spectrometry using ‘U’ shaped hollow nanomechanical resonators (HNR). Capillary electrophoresis was coupled with the HNR based mass detection to overcome low ionic conductivity of channels embedded in the HNR preventing direct in-situ electrophoretic separation. The flow of analytes through the HNR was achieved by balancing the hydrodynamic pressure to override the electromotive force and inhibit the motion of analytes towards the anode for capillary electrophoresis. The resonance frequency shifts of the HNR vibrating around 1.5 MHz were correlated with the time of the passage of the protein bands to construct the mass spectrum. The proposed concept was demonstrated by constructing a mass spectrum of egg white proteins in the molecular weight range of 14–250 kDa. When compared to regular polyacrylamide gel electrophoresis, our method not only provides a precise and fast readout but also avoids the use of chemical staining. This study paves a new route for low-cost and on-chip mass spectrometers with ultra-miniaturized dimensions.

Abstract Sterility mosaic disease (SMD) is one of the serious production constraints that may lead to complete yield loss in pigeonpea. Three mapping populations including two recombinant inbred lines and one F2, were used for phenotyping for SMD resistance at two locations in three different years. Genotyping-by-sequencing approach was used for simultaneous identification and genotyping of SNPs on above mentioned populations. In total, 212,464, 89,699 and 64,798 SNPs were identified in ICPL 20096 × ICPL 332 (PRIL_B), ICPL 20097 × ICP 8863 (PRIL_C) and ICP 8863 × ICPL 87119 (F2) respectively. By using high-quality SNPs, genetic maps were developed for PRIL_B (1,101 SNPs; 921.21 cM), PRIL_C (484 SNPs; 798.25 cM) and F2 (996 SNPs; 1,597.30 cM) populations. The average inter marker distance on these maps varied from 0.84 cM to 1.65 cM, which was lowest in all genetic mapping studies in pigeonpea. Composite interval mapping based QTL analysis identified a total of 10 QTLs including three major QTLs across the three populations. The phenotypic variance of the identified QTLs ranged from 3.6 to 34.3%. One candidate genomic region identified on CcLG11 seems to be promising QTL for molecular breeding in developing superior lines with enhanced resistance to SMD.

Abstract Fusarium wilt (FW) is one of the most important biotic stresses causing yield losses in pigeonpea. Genetic improvement of pigeonpea through genomics-assisted breeding (GAB) is an economically feasible option for the development of high yielding FW resistant genotypes. In this context, two recombinant inbred lines (RILs) (ICPB 2049 × ICPL 99050 designated as PRIL_A and ICPL 20096 × ICPL 332 designated as PRIL_B) and one F2 (ICPL 85063 × ICPL 87119) populations were used for the development of high density genetic maps. Genotyping-by-sequencing (GBS) approach was used to identify and genotype SNPs in three mapping populations. As a result, three high density genetic maps with 964, 1101 and 557 SNPs with an average marker distance of 1.16, 0.84 and 2.60 cM were developed in PRIL_A, PRIL_B and F2, respectively. Based on the multi-location and multi-year phenotypic data of FW resistance a total of 14 quantitative trait loci (QTLs) including six major QTLs explaining >10% phenotypic variance explained (PVE) were identified. Comparative analysis across the populations has revealed three important QTLs (qFW11.1, qFW11.2 and qFW11.3) with upto 56.45% PVE for FW resistance. This is the first report of QTL mapping for FW resistance in pigeonpea and identified genomic region could be utilized in GAB.
