Abstract During mammalian spermatogenesis, the ubiquitin proteasome system maintains protein homoeostasis (proteastasis) and spermatogenic cellular functions. DCAF17 is a substrate receptor in the ubiquitin CRL4 E3 Ligase complex, absence of which causes oligoasthenoteratozoospermia in mice resulting in male infertility. To determine the molecular phenomenon underlying the infertility phenotype caused by disrupting Dcaf17, we performed RNA-sequencing-based gene expression profiling of 3-weeks and 8-weeks old Dcaf17 wild type and Dcaf17 disrupted mutant mice testes. At three weeks, 44% and 56% differentially expressed genes (DEGs) were up- and down-regulated, respectively, with 32% and 68% DEGs were up- and down-regulated, respectively at 8 weeks. DEGs include protein coding genes and lncRNAs distributed across all autosomes and the X chromosome. Gene ontology analysis revealed major biological processes including proteolysis, regulation of transcription and chromatin remodelling are affected due to Dcaf17 disruption. We found that Dcaf17 disruption up-regulated several somatic genes, while germline-associated genes were down-regulated. Up to 10% of upregulated, and 12% of downregulated, genes were implicated in male reproductive phenotypes. Moreover, a large proportion of the up-regulated genes were highly expressed in spermatogonia and spermatocytes, while the majority of downregulated genes were predominantly expressed in round spermatids. Collectively, these data show that the Dcaf17 disruption affects directly or indirectly testicular proteastasis and transcriptional signature in mouse.

Abstract Low fresh gas flow rates are recommended because of their benefits, however, its use is limited due to associated risks. The main purpose of this study was to investigate whether 300 mL of fresh gas flow that practised with automated gas control mode is applicable and safe. The second aim is to show that automated mode can provide economic benefits. Sixty hepatectomy cases who suitable criterias were included to cohort study in three groups as prospective, sequential, observational. An operating room were allocated only for this study. 300 mL fresh gas flow with automated mode (groupA3), 600 mL fresh gas flow with automated mode (groupA6) and, 600 mL fresh gas flow with manually (groupM6) was applied. Abstract Machine learning (ML) holds great promise in transforming healthcare. While published studies have shown the utility of ML models in interpreting medical imaging examinations, these are often evaluated under laboratory settings. The importance of real world evaluation is best illustrated by case studies that have documented successes and failures in the translation of these models into clinical environments. A key prerequisite for the clinical adoption of these technologies is demonstrating generalizable ML model performance under real world circumstances. The purpose of this study was to demonstrate that ML model generalizability is achievable in medical imaging with the detection of intracranial hemorrhage (ICH) on non-contrast computed tomography (CT) scans serving as the use case. An ML model was trained using 21,784 scans from the RSNA Intracranial Hemorrhage CT dataset while generalizability was evaluated using an external validation dataset obtained from our busy trauma and neurosurgical center. This real world external validation dataset consisted of every unenhanced head CT scan (n = 5965) performed in our emergency department in 2019 without exclusion. The model demonstrated an AUC of 98.4%, sensitivity of 98.8%, and specificity of 98.0%, on the test dataset. On external validation, the model demonstrated an AUC of 95.4%, sensitivity of 91.3%, and specificity of 94.1%. Evaluating the ML model using a real world external validation dataset that is temporally and geographically distinct from the training dataset indicates that ML generalizability is achievable in medical imaging applications.

Abstract The liver is a unique organ that has a phenomenal capacity to regenerate after injury. Different surgical procedures, including partial hepatectomy (PH), intraoperative portal vein ligation (PVL), and associated liver partition and portal vein ligation for staged hepatectomy (ALPPS) show clinically distinct recovery patterns and regeneration. The observable clinical differences likely mirror some underlying variations in the patterns of gene activation and regeneration pathways. In this study, we provided a comprehensive comparative transcriptomic analysis of gene regulation in regenerating rat livers temporally spaced at 24 h and 96 h after PH, PVL, and ALPPS. The time-dependent factors appear to be the most important determinant of post-injury alterations of gene expression in liver regeneration. Gene expression profile after ALPPS showed more similar expression pattern to the PH than the PVL at the early phase of the regeneration. Early transcriptomic changes and predicted upstream regulators that were found in all three procedures included cell cycle associated genes (E2F1, CCND1, FOXM1, TP53, and RB1), transcription factors (Myc, E2F1, TBX2, FOXM1), DNA replication regulators (CDKN1A, EZH2, RRM2), G1/S-transition regulators (CCNB1, CCND1, RABL6), cytokines and growth factors (CSF2, IL-6, TNF, HGF, VEGF, and EGF), ATM and p53 signaling pathways. The functional pathway, upstream, and network analyses revealed both unique and overlapping molecular mechanisms and pathways for each surgical procedure. Identification of molecular signatures and regenerative signaling pathways for each surgical procedure further our understanding of key regulators of liver regeneration as well as patient populations that are likely to benefit from each procedure.

Abstract Although biopsies and tumor resection are prognostically beneficial for glioblastomas (GBM), potential negative effects have also been suggested. Here, using retrospective study of patients and intravital imaging of mice, we identify some of these negative aspects, including stimulation of proliferation and migration of non-resected tumor cells, and provide a strategy to prevent these adverse effects. By repeated high-resolution intravital microscopy, we show that biopsy-like injury in GBM induces migration and proliferation of tumor cells through chemokine (C-C motif) ligand 2 (CCL-2)-dependent recruitment of macrophages. Blocking macrophage recruitment or administrating dexamethasone, a commonly used glucocorticoid to prevent brain edema in GBM patients, suppressed the observed inflammatory response and subsequent tumor growth upon biopsy both in mice and in multifocal GBM patients. Taken together, our study suggests that inhibiting CCL-2-dependent recruitment of macrophages may further increase the clinical benefits from surgical and biopsy procedures.

Abstract Hybrid energy systems (HESs) are the most important sources of energy demand-supply, have developed significantly around the world. Microgrids, renewable energy sources, remote telecommunications stations, greenhouses, etc., are being considered as HESs applications. Optimal sizing of these systems is considered as one of the important issues related to energy management. In this paper, the Response Surface Methodology (RSM) is proposed for the optimal sizing of a Photovoltaic (PV) system in a HESs. The suggested procedure solves the optimization problem by considering the factors affecting PV output power about the environmental conditions of the HESs. Providing a mathematical model for each of the input parameters and the ability to assessment the sensitivity of each of the input variables are the most important advantages of the proposed technique. In this paper, the RSM provides the most optimal sizing related to the PV system by considering climatic and geographical factors in the study site, and technical and economic issues related to the HESs. The optimal model obtained is evaluated by the Analysis of Variance (ANOVA) evaluation method, which is one of the important techniques of statistical evaluation. It should be noted that the RSM technique can be utilized to optimize all components of any HES.

Abstract Power quality is a crucial determinant for integrating wind energy into the electrical grid. This integration necessitates compliance with certain standards and levels. This study presents cascadedfuzzy power control (CFPC) for a variable-speed multi-rotor wind turbine (MRWT) system. Fuzzy logic is a type of smart control system already recognized for its robustness, making it highly suited and reliable for generating electrical energy from the wind. Therefore, the CFPC technique is proposed in this work to control the doubly-fed induction generator (DFIG)-based MRWT system. This proposed strategy is applied to the rotor side converter of a DFIG to improve the current/power quality. The proposed control has the advantage of being model-independent, as it relies on empirical knowledge rather than the specific characteristics of the DFIG or turbine. Moreover, the proposed control system is characterized by its simplicity, high performance, robustness, and ease of application. The implementation of CFPC management for 1.5 MW DFIG-MRWT was carried out in MATLAB environment considering a variable wind speed. The obtained results were compared with the direct power control (DPC) technique based on proportional-integral (PI) controllers (DPC-PI), highlighting that the CFPC technique reduced total harmonic distortion by high ratios in the three tests performed (25%, 30.18%, and 47.22%). The proposed CFPC technique reduced the response time of reactive power in all tests by ratios estimated at 83.76%, 65.02%, and 91.42% compared to the DPC-PI strategy. Also, the active power ripples were reduced by satisfactory proportions (37.50%, 32.20%, and 38.46%) compared to the DPC-PI strategy. The steady-state error value of reactive power in the tests was low when using the CFPC technique by 86.60%, 57.33%, and 72.26%, which indicates the effectiveness and efficiency of the proposed CFPC technique in improving the characteristics of the system. Thus this control can be relied upon in the future.

Abstract Cybersecurity is critical in today’s digitally linked and networked society. There is no way to overestimate the importance of cyber security as technology develops and becomes more pervasive in our daily lives. Cybersecurity is essential to people’s protection. One type of cyberattack known as “credential stuffing” involves using previously acquired usernames and passwords by attackers to access user accounts on several websites without authorization. This is feasible as a lot of people use the same passwords and usernames on several different websites. Maintaining the security of online accounts requires defence against credential-stuffing attacks. The problems of credential stuffing attacks, failure detection, and prediction can be handled by the suggested EWOA-ANN model. Here, a novel optimization approach known as Enhanced Whale Optimization Algorithm (EWOA) is put on to train the neural network. The effectiveness of the suggested attack identification model has been demonstrated, and an empirical comparison will be carried out with respect to specific security analysis.

Abstract The paper proposes a nonlinear controller called dual super-twisting sliding mode command (DSTSMC) for controlling and regulating the rotor side converter (RSC) of multi-rotor wind power systems that use doubly-fed induction generators. It was proposed that this controller be developed as an alternative to the direct power control (DPC), which makes use of a pulse width modulation (PWM) strategy to regulate the RSC's functioning. Overcoming the power/current quality issue with the proposed technique (DPC-DSTSMC-PWM) is characterized by great robustness and excellent performance. The designed strategy was contrasted with the standard method of control and other methods already in use. So, the unique proposed control strategy’s robustness, performance, efficiency, and efficacy in enhancing system characteristics were tested and validated in Matlab/Simulink. In both tests, the proposed method resulted in significant improvements, reducing active power ripples by 83.33%, 57.14%, and 48.57% in the proposed tests. When compared with the traditional regulation method, the reduction rates of reactive power ripples are 64.06%, 52.47%, and 68.7% in the tests. However, in contrast to the conventional method, the proposed tests showed a decrease of between 72.46%, 50%, and 76.22% in the value of total harmonic distortion (THD) of the provided currents. These ratios show how effective the proposed plan is in ameliorating and enhancing aspects of the energy system.

Abstract This work develops a hybrid active power filter (HAPF) in this article to operate in conjunction with the energy storage system (ESS), wind power generation system (WPGS), and solar energy system (SES). It employs three level shunt voltage source converters (VSC) connected to the DC-bus. Optimization of the gain values of the fractional-order proportional integral derivative controller (FOPIDC) and parameter values of the HAPF is achieved using the Jaya grey wolf hybrid algorithm (GWJA). The primary objectives of this study, aimed at enhancing power quality (PQ), include: (1) ensuring swift stabilization of DC link capacitor voltage (DCLCV); (2) reducing harmonics and improving power factor (PF); (3) maintaining satisfactory performance under different combinations of loads like EV charging load, non linear load and solar irradiation conditions. The proposed controller's performance is evaluated through three test scenarios featuring different load configurations and irradiation levels. Additionally, the HAPF is subjected to design using other optimization algorithms such as genetic algorithm (GA), particle swarm optimization (PSO), and ant colony optimization (ACO) to assess their respective contributions to PQ improvement.

Abstract Placenta accreta spectrum (PAS) presents a significant obstetric challenge, associated with considerable maternal and fetal-neonatal morbidity and mortality. Nevertheless, it is imperative to acknowledge that a noteworthy subset of PAS cases remains undetected until the time of delivery, thereby contributing to an augmented incidence of morbidity among the affected individuals. The delayed identification of PAS not only hinders timely intervention but also exacerbates the associated health risks for both the maternal and fetal outcomes. This underscores the urgency to innovate strategies for early PAS diagnosis. In this study, we aimed to explore plasma proteins as potential diagnostic biomarkers for PAS. Integrated transcriptome and proteomic analyses were conducted to establish a novel diagnostic approach. A cohort of 15 pregnant women diagnosed with PAS and delivering at Inonu University Faculty of Medicine between 01/04/2021 and 01/01/2023, along with a matched control group of 15 pregnant women without PAS complications, were enrolled. Abstract It is noted that the traditional direct filed-oriented control (DFOC) is widely used in the field of electric power generation from wind due to its fast response dynamic, ease of implementation and simplicity, but this strategy is characterized by the presence of large ripples at the level of both active and reactive powers. This work presents a new algorithm for DFOC strategy of an asynchronous generator (AG) in a wind power (WP) system, which is based on the use of a new nonlinear controller called fractional-order synergetic control–fractional-order proportional-integral (FOSC–FOPI) controller, where the proposed technique parameters are calculated using the particle swarm optimization (PSO) strategy. It has been observed that the DFOC–FOSC–FOPI–PSO strategy is robust and works well in case of changing generator parameters. Three tests were performed to study the behavior of the designed technique under different working conditions, where the behavior of the DFOC–FOSC–FOPI–PSO strategy was compared with the behavior of the traditional DFOC technique in terms of power ripple ratio, overshoot, steady-state error, response time, tracking reference, and current quality. The simulation of the designed technique based on the FOSC–FOPI–PSO strategy of the AG–WP system is carried out using Matlab software, where the simulation results showed that the suggested technique is better than the classical technique (with PI controller) in terms of improving response time of active power (33.33%) and reactive power (10%) in second test, reduction of the steady-state error of reactive power (96.95%) and active power (97.14) in first test, minimization of harmonic distortion of current (96.57%) in third test and significant minimization of ripples of active power (99.67%, 44.69%, and 98.95%) and reactive power (99.25%, 53.65%, and 70.50%) in the three tests. The effectiveness of the DFOC–FOSC–FOPI–PSO strategy is very high, so it can be a reliable solution for controlling various generators.

Abstract This paper studies the possibility of connecting Wind Farms (WF) to the electric grid with the use of finite space model predictive command (FS-MPC) to manage wind farms to improve the quality of the current output from the doubly-fed induction generator (DFIG) with considering fault ride-through technique. This proposed system can generate active power and enhance the power factor. Furthermore, the reduction of harmonics resulting from the connection of non-linear loads to the electrical grid is achieved through the self-active filtering mechanism in DFIGs-WF, facilitated by the now algorithm proposed. FS-MPC technique has the ability to improve system characteristics and greatly reduce active power ripples. Therefore, MATLAB software is used to implement and verify the safety, performance, and effectiveness of this designed technique compared to the conventional strategy. The results obtained demonstrated the effectiveness of the proposed algorithm in handling the four operational modes (Maximum power point tracking, Delta, Fault, and Filtering). Additionally, the suggested technique exhibited flexibility, robustness, high accuracy, and fast dynamic response when compared to conventional strategies and some recently published scientific works. On the other hand, the THD value of the current was significantly reduced, obtaining at one test time the values 56.87% and 0.32% before and after filtering, respectively 27.50% and 0.26% at another time of testing, resulting in an estimated THD reduction percentage of 99.43% and 99.05%, respectively. These high percentages prove that the quality of the stream is excellent after applying the proposed strategy.

Abstract Cardiovascular diseases (CVDs) are a serious public health issue that affects and is responsible for numerous fatalities and impairments. Ischemic heart disease (IHD) is one of the most prevalent and deadliest types of CVDs and is responsible for 45% of all CVD-related fatalities. IHD occurs when the blood supply to the heart is reduced due to narrowed or blocked arteries, which causes angina pectoris (AP) chest pain. AP is a common symptom of IHD and can indicate a higher risk of heart attack or sudden cardiac death. Therefore, it is important to diagnose and treat AP promptly and effectively. To forecast AP in women, we constructed a novel artificial intelligence (AI) method employing the tree-based algorithm known as an Explainable Boosting Machine (EBM). EBM is a machine learning (ML) technique that combines the interpretability of linear models with the flexibility and accuracy of gradient boosting. We applied EBM to a dataset of 200 female patients, 100 with AP and 100 without AP, and extracted the most relevant features for AP prediction. We then evaluated the performance of EBM against other AI methods, such as Logistic Regression (LR), Categorical Boosting (CatBoost), eXtreme Gradient Boosting (XGBoost), Adaptive Boosting (AdaBoost), and Light Gradient Boosting Machine (LightGBM). We found that EBM was the most accurate and well-balanced technique for forecasting AP, with accuracy (0.925) and Youden's index (0.960). We also looked at the global and local explanations provided by EBM to better understand how each feature affected the prediction and how each patient was classified. Our research showed that EBM is a useful AI method for predicting AP in women and identifying the risk factors related to it. This can help clinicians to provide personalized and evidence-based care for female patients with AP.

Abstract This work designs a powerful new nonlinear control technique using synergetic control (SC), proportional-integral (PI) controller, and genetic algorithm (GA) for multi-rotor wind energy (MRWE) conversion systems, whereby an asynchronous generator (AG) is used to achieve optimal energy extraction. The direct power control (DPC) technique is used based on the proposed SC-PI-GA (SPI-GA) technique to control the AG-based MRWE system, where this new nonlinear control technique is used to achieve stable control characteristics under random changes in wind speed and to provide great robustness against modeling uncertainties. Moreover, the pulse width modulation (PWM) technique is used to control the AG inverter due to its simplicity and ease of implementation. In this proposed DPC-SPI-GA technique, we need to measure current and voltage to estimate the active power and the reactive power. Also, inner loops are not used in this proposed DPC-SPI-GA technique as is the case in the field-oriented control (FOC) technique, where the proposed system in this work is characterized by an integrated structure. Three different tests are proposed to study and verify the behavior of the designed DPC-SPI-GA strategy compared to the traditional DPC technique.
