Detection of subsurface hydrodynamic anomalies plays a significant role in groundwater resource management and environmental monitoring. In this paper, based on data from the groundwater level, atmospheric pressure, and precipitation in the Chengdu area of China, a method for detecting outliers considering the factors affecting groundwater levels is proposed. By analyzing the factors affecting groundwater levels in the monitoring site and eliminating them, simplified groundwater data is obtained. Applying sl-Pauta (self-learning-based Pauta), iForest (Isolated Forest), OCSVM (One-Class SVM), and KNN to synthetic data with known outliers, testing and evaluating the effectiveness of 4 technologies. Finally, the four methods are applied to the detection of outliers in simplified groundwater levels. The results show that in the detection of outliers in synthesized data, the OCSVM method has the best detection performance, with a precision rate of 88.89%, a recall rate of 91.43%, an F1 score of 90.14%, and an AUC value of 95.66%. In the detection of outliers in simplified groundwater levels, a qualitative analysis of the displacement data within the field of view indicates that the outlier detection performance of iForest and OCSVM is better than that of KNN. The instability of rock slope is still a very frequent geological disaster, which seriously affects people's life and production activities. Previous studies have mainly focused on deformation mechanism, prediction, and control of hard rock with single lithology, while there are limited studies on the theoretic computational method of the stability for soft–hard interbedded anti-inclined rock strata. In this study, a geomechanical model for the toppling failure of soft–hard-interbedded anti-inclined rock slope is established. The modes of failure for soft and hard rock strata are analyzed, the computational formula of the downward thrust for each anti-inclined rock stratum is derived, and the stability safety factor of each rock stratum is defined. A theoretical computational method for determining the potentially most dangerous failure surface of soft–hard-interbedded anti-inclined rock slope is proposed. By comparing with the existing research results, the theoretical solving method proposed in this study can well solve the location of the potentially most dangerous failure surface of soft–hard-interbedded anti-inclined rock slope. The potentially most dangerous failure surface of this kind of slope is approximately planar, and the angle between it and the normal plane of the rock strata is an acute angle within 30°. Anoikis is a unique form of apoptosis associated with vascularization and distant metastasis in cancer. Eliminating anoikis resistance in tumor cells could be a promising target for improving the prognosis of terminal cancer patients. However, current studies have not elaborated on the prognosis effect of anoikis-related long non-coding RNAs (lncRNAs) in cutaneous melanoma. Pre-processed data, including RNA sequences and clinical information, were retrieved from TCGA and GTEx databases. After a series of statistical analyses, anoikis-related lncRNAs with prognostic significance were identified, and a unique risk signature was constructed. Risk scores were further analyzed in relation to the tumor microenvironment, tumor immune dysfunction and exclusion, immune checkpoint genes, and RNA methylation genes. The indicators were also used to predict the potentially sensitive anti-cancer drugs. An anoikis-related lncRNAs risk signature consisting of LINC01711, POLH-AS1, MIR205HG, and LINC02416 was successfully established in cutaneous melanoma. Overall survival and progression-free survival of patients were strongly linked with the risk score, independently of other clinical factors. The low-risk group exhibited a more beneficial immunological profile, was less affected by RNA methylation, and was more sensitive to the majority of anti-cancer drugs, all of which indicated a better prognostic outcome. We investigate the quantum synchronization phenomena of two mechanical oscillators of different frequencies in two optomechanical systems under periodically modulating cavity detunings or driving amplitudes, which can interact mutually through an optical fiber or a phonon tunneling. The cavities are filled with Kerr-type nonlinear medium. It is found that, no matter which the coupling and periodically modulation we choose, both of the quantum synchronization of nonlinear optomechanical system are more appealing than the linear optomechanical system. It is easier to observe greatly enhanced quantum synchronization with Kerr nonlinearity. The effect of crystal plane orientation on tribochemical removal of monocrystalline silicon was investigated using an atomic force microscope. Experimental results indicated that the tribochemical removal of silicon by SiO2 microsphere presented strong crystallography-induced anisotropy. Further analysis suggested that such anisotropic tribochemical removal of silicon was not dependent on the crystallography-dependent surface mechanical properties (i.e., hardness and elastic modulus), but was mainly attributed to various atomic planar density and interplanar spacing in different crystal planes. Phenomenological results speculated that higher density of silicon atom could promote the formation of Si-O-Si bonds between the SiO2 microsphere and silicon substrate, resulting in more severe tribochemical material removal. Larger interplanar spacing with smaller energy barrier facilitated the rupture of the Si-Si network with the help of mechanical shearing stress, which caused more serious wear of the silicon surface. In this study, we investigated the dosage effect of gemcitabine, an inhibitor of ribonucleotide reductase (RR), on cellular levels of ribonucleotides and deoxyribonucleotides using high performance liquid chromatography-electrospray ionization tandem mass spectrometric method. As anticipated, after 4-h incubation of non-small cell lung cancer (A549) cells with gemcitabine at 0.5 and 2 μM, there were consistent reductions in levels of deoxyribonucleoside diphosphates (dNDP) and their corresponding deoxyribonucleoside triphosphates (dNTP). However, after 24-h exposure to 0.5 μM gemcitabine, the amounts of dNTP were increased by about 3 fold, whereas cells after 24-h 2 μM gemcitabine treatment exhibited deoxycytidine diphosphate (dCDP), deoxyadenosine diphosphate (dADP) and deoxyguanosine diphosphate (dGDP) levels less than 50% of control values, with deoxycytidine triphosphate (dCTP) and deoxyguanosine triphosphate (dGTP) returning to the control level. Using cell cycle analysis, we found that 24-h incubation at 0.5 μM gemcitabine resulted in a significant increase in S phase arrest, while 2 μM treatment increased G0/G1 population. Our data demonstrated the correlation between the level of RR and the increased levels of dNTPs in the group of 0.5 μM treatment for 24-h with a markedly reduced level of dFdCTP. Accordingly, we proposed that the dosage of dFdC could determine the arrested phase of cell cycle, in turn affecting the recovery of dNTPs pools. [ABSTRACT FROM AUTHOR]

A tribochemistry-induced selective etching approach is proposed for the first time to produce silicon nanostructures without lattice damage. With a ~1 nm thick SiOx film as etching mask grown on Si(100) surface (Si(100)/SiOx) by wet-oxidation technique, nano-trenches can be produced through the removal of local SiOx mask by a SiO2 tip in humid air and the post-etching of the exposed Si in potassium hydroxide (KOH) solution. The material removal of SiOx mask and Si under low load is dominated by the tribochemical reaction at the interface between SiO2 tip and Si/SiOx sample, where the contact pressure is much lower than the critical pressure for initial yield of Si. High resolution transmission electron microscope (HRTEM) observation indicates that neither the material removal induced by tribochemical reaction nor the wet etching in KOH solution leads to lattice damage of the fabricated nanostructures. Chronic hepatitis B virus (HBV) infection is a major global health burden. Functional exhaustion and numerical reduction of HBV-specific cytotoxic T lymphocytes (CTLs) in the liver and peripheral blood limit anti-HBV CTL activity in patients with chronic HBV infection (CHB). However, the ongoing anti-HBV CD8+ T cell responses in the lymphoid organs are largely unknown due to the infeasibility of obtaining lymphoid organs from CHB patients. Here we demonstrate that the percentage of HBV-specific CD8+ T cells is higher in the spleen of CHB patients than that from peripheral blood and liver. Although they do respond to TCR stimulation and produce IFNγ, the cells proliferate poorly. Furthermore, miR-720 expression is upregulated in HBV-specific CD8+ T cells. Overexpression of miR-720 in primary human CD8+ T cells inhibits TCR stimulation-induced proliferation. We also demonstrate that TGFβ sustains miR-720 upregulation after TCR stimulation, and blood TGFβ levels are associated with the outcome of type I interferon treatment of CHB patients. Akt and nuclear factor kappa B (NF-κB) signaling pathways are involved in germ cell apoptosis and inflammation after testicular heat stress (THS). We observed that after THS induced by the exposure of rat testes to 43 °C for 20 min, their weight decreased, the fraction of apoptotic testicular germ cells significantly increased, and the proliferation of germ cells was inhibited. In addition, THS lowered serum testosterone (T) level, whereas the levels of follicle stimulating hormone and luteinizing hormone were not significantly changed. The ultrastructure of the seminiferous tubules became abnormal after THS, the structure of the blood-testis barrier (BTB) became loose, and the Sertoli cells showed a trend of differentiation. The level of phosphorylated Akt was reduced, whereas the amount of phosphorylated NF-κB p65 was augmented by THS. Wuzi-Yanzong (WZYZ), a classic Chinese medicine prescription for the treatment of male reproductive dysfunctions, alleviated the changes induced by THS. In order to determine the mechanism of action of WZYZ, we investigated how this preparation modulated the levels of T, androgen receptor (AR), erythropoietin (EPO), EPO receptor, and Tyro-3, Axl, and Mer (TAM) family of tyrosine kinase receptors. We found that WZYZ activated the Akt pathway, inhibited the Toll-like receptor/MyD88/NF-κB pathway, and repaired the structure of BTB by regulating the levels of T, AR, TAM receptors, and EPO. In conclusion, these results suggest that WZYZ activates the Akt pathway and inhibits the NF-κB pathway by acting on the upstream regulators, thereby improving spermatogenesis deficit induced by THS. Genetic factors play a role in the risk of idiopathic pulmonary fibrosis (IPF). Specifically, MUC5B rs35705950 non-risk alleles and immunologic aberrations were associated with the IPF's progression. However, rare genetic variants have not been systematically investigated in Chinese IPF patients. In this study, we aimed to improve understanding of the genetic architecture of IPF in the Chinese population and to assess whether rare protein-coding variants in the immunity pathway genes are enriched in the IPF patients with non-risk alleles at rs35705950. A case–control exome-wide study including 110 IPF patients and 60 matched healthy controls was conducted. rs35705950 was genotyped by Sanger sequencing. To identify genes enriched in IPF, gene-based association analyses were performed. Identified genes were included for further pathway analyses using gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Associations between rs35705950 and genes enriched in the immunity pathway were also tested. 226 genes that were enriched with deleterious variants were identified in IPF patients. Out of them, 36 genes were significantly enriched in GO and KEGG pathways in the IPF. Pathway analyses implicated that these genes were involved in the immune response and cell adhesion. Rare protein-altering variants in genes related to the immunity pathway did not significantly differ between patients with a MUC5B risk allele and individuals without risk allele. We drafted a comprehensive mutational landscape of rare protein-coding variants in the Chinese IPF and identified genes related to immune response and cell adhesion. The ability to propagate via small diaspores is crucial for the invasion of a clone plant that does not reproduce sexually in its introduced range. We investigated the effects of node and internode adjacent mode, fragment type, burial orientation and position of the node in relation to the soil surface on the sprouting and growth of alligator weed (Alternanthera philoxeroides (Martius) Griseb.). All the factors had effects and interaction effects on the sprouting rate and growth. As a whole fragment in all treatments, the fragments with basal node buried upward on the soil surface, exhibited the best above-ground growth and root growth. The one-node fragment with basal node buried downward above the soil surface and upward under the soil surface significantly decreased the above-ground growth and root growth compared to that of the two-node fragment. Therefore, the one-node fragments were more affected by environmental conditions than the two-node fragments. The results indicated that reducing the number of nodes of a fragment and burying the node under the soil or orienting it downward above the soil surface could be applied to control the invasion of alligator weed. We investigate the formation of discrete breathers (DBs) and the dynamics of the mixture of two-species Bose-Einstein condensates (BECs) in open boundary optical lattices using the discrete nonlinear Schrödinger equations. The results show that the coupling of intra- and interspecies interaction can lead to the existence of pure single-species DBs and symbiotic DBs (i.e., two-species DBs). Furthermore, we find that there is a selective distillation phenomenon in the dynamics of the mixture of two-species BECs. One can selectively distil one species from the mixture of two-species BECs and can even control dominant species fraction by adjusting the intra- and interspecies interaction in optical lattices.
