First principle calculations were performed to investigate the structural, mechanical, electronic properties, and thermodynamic properties of three binary Mg–B compounds under pressure, by using the first principle method. The results implied that the structural parameters and the mechanical properties of the Mg–B compounds without pressure are well matched with the obtainable theoretically simulated values and experimental data. The obtained pressure–volume and energy–volume revealed that the three Mg–B compounds were mechanically stable, and the volume variation decreases with an increase in the boron content. The shear and volume deformation resistance indicated that the elastic constant Cij and bulk modulus B increased when the pressure increased up to 40 GPa, and that MgB7 had the strongest capacity to resist shear and volume deformation at zero pressure, which indicated the highest hardness. Meanwhile, MgB4 exhibited a ductility transformation behaviour at 30 GPa, and MgB2 and MgB7 displayed a brittle nature under all the considered pressure conditions. The anisotropy of the three Mg–B compounds under pressure were arranged as follows: MgB4 > MgB2 > MgB7. Moreover, the total density of states varied slightly and decreased with an increase in the pressure. The Debye temperature ΘD of the Mg–B compounds gradually increased with an increase in the pressure and the boron content. The temperature and pressure dependence of the heat capacity and the thermal expansion coefficient α were both obtained on the basis of Debye model under increased pressure from 0 to 40 GPa and increased temperatures. This paper brings a convenient understanding of the magnesium–boron alloys.

The remediation of heavy metal-contaminated soils has attracted increased attention worldwide. The immobilization of metals to prevent their uptake by plants is an efficient way to remediate contaminated soils. This work aimed to seek the immobilization of cadmium in contaminated soils via a combination method. Flask experiments were performed to investigate the effects of hydroxyapatite (HAP) and the Cupriavidus sp. strain ZSK on soil pH and DTPA-extractable cadmium. Pot experiments were carried out to study the effects of the combined amendment on three plant species. The results showed that HAP has no obvious influence on the growth of the strain. With increasing concentrations of HAP, the soil pH increased, and the DTPA-extractable Cd decreased. Via the combined amendment of the strain and HAP (SH), the DTPA-extractable Cd in the soil decreased by 58.2%. With the combined amendment of the SH, the cadmium accumulation in ramie, dandelion, and daisy decreased by 44.9%, 51.0%, and 38.7%, respectively. Moreover, the combined amendment somewhat benefitted the growth of the three plant species and significantly decreased the biosorption of cadmium. These results suggest that the immobilization by the SH combination is a potential method to decrease the available cadmium in the soil and the cadmium accumulation in plants.

Medical image segmentation is a fundamental step in medical analysis and diagnosis. In recent years, deep learning networks have been used for precise segmentation. Numerous improved encoder–decoder structures have been proposed for various segmentation tasks. However, high-level features have gained more research attention than the abundant low-level features in the early stages of segmentation. Consequently, the learning of edge feature maps has been limited, which can lead to ambiguous boundaries of the predicted results. Inspired by the encoder–decoder network and attention mechanism, this study investigates a novel multilayer edge attention network (MEA-Net) to fully utilize the edge information in the encoding stages. MEA-Net comprises three major components: a feature encoder module, a feature decoder module, and an edge module. An edge feature extraction module in the edge module is designed to produce edge feature maps by a sequence of convolution operations so as to integrate the inconsistent edge information from different encoding stages. A multilayer attention guidance module is designed to use each attention feature map to filter edge information and select important and useful features. Through experiments, MEA-Net is evaluated on four medical image datasets, including tongue images, retinal vessel images, lung images, and clinical images. The evaluation values of the Accuracy of four medical image datasets are 0.9957, 0.9736, 0.9942, and 0.9993, respectively. The values of the Dice coefficient are 0.9902, 0.8377, 0.9885, and 0.9704, respectively. Experimental results demonstrate that the network being studied outperforms current state-of-the-art methods in terms of the five commonly used evaluation metrics. The proposed MEA-Net can be used for the early diagnosis of relevant diseases. In addition, clinicians can obtain more accurate clinical information from segmented medical images.

CX-5461 is a G-quadruplex (G4) ligand currently in trials with initial indications of clinical activity in cancers with defects in homologous recombination repair. To identify more genetic defects that could sensitize tumors to CX-5461, we tested synthetic lethality for 480 DNA repair and genome maintenance genes to CX-5461, pyridostatin (PDS), a structurally unrelated G4-specific stabilizer, and BMH-21, which binds GC-rich DNA but not G4 structures. We identified multiple members of HRD, Fanconi Anemia pathways, and POLQ, a polymerase with a helicase domain important for G4 structure resolution. Significant synthetic lethality was observed with UBE2N and RNF168, key members of the DNA damage response associated ubiquitin signaling pathway. Loss-of-function of RNF168 and UBE2N resulted in significantly lower cell survival in the presence of CX-5461 and PDS but not BMH-21. RNF168 recruitment and histone ubiquitination increased with CX-5461 treatment, and nuclear ubiquitination response frequently co-localized with G4 structures. Pharmacological inhibition of UBE2N acted synergistically with CX-5461. In conclusion, we have uncovered novel genetic vulnerabilities to CX-5461 with potential significance for patient selection in future clinical trials.

Lubricating technologies are essential for saving energy and identifying effective, environmentally friendly layered materials that meet the demands of solid lubricants is an important direction. Herein, we probed the relationships between load-carrying capacity and crystal structure. The results showed that increasing the sliding resistance of interlayers using corrugated layers increased the load-carrying capacity of layered solid lubricants. This finding expands on the traditional lubrication mechanism of layered materials. Following the rules, rapid selection of layered potassium magnesium and calcium phosphates (K-LMP and K-LCP) as effective solid lubricants was achieved using crystallographic data and strict filtering criteria. In order to prepare materials suitable for lubrication, the synthesis of K-LMP and K-LCP was optimised. These materials could be utilised in the food, textile or marine machinery industries in the future. The developed method could successfully guide the synthesis of application-oriented materials.

Antibiotic resistance is a major global health threat. High prevalences of colonization and infection with multi-drug resistance organisms (MDROs) have been reported in patients undergoing dialysis. It is unknown if this finding extends to patients with mild and moderate/severe kidney disease. An observational study included all adult incident patients hospitalized with a discharge diagnosis of infection in four hospitals from Guangzhou, China. Inclusion criteria: Serum creatinine measurement at admission together with microbial culture confirmed infections. Exclusion criterion: Undergoing renal replacement therapy. Four categories of Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) estimated glomerular filtration rate (eGFR) were compared: eGFR ≥ 105, 60–104 (reference), 30–59, and 2. The odds ratio of MDROs, defined as specific pathogens (Staphylococcus aureus, Enterococcus spp., Enterobacteriaceae, Pseudomonas aeruginosa and Acinetobacter spp.) resistant to three or more antibiotic classes, were calculated using a multivariable logistic regression model across eGFR strata. Of 94,445 total microbial culture records, 7,288 first positive cultures matched to infection diagnosis were selected. Among them, 5,028 (68.9%) were potential MDROs. The odds of infections by MDROs was 19% and 41% higher in those with eGFR between 30–59 ml/min/1.73 m2 (Adjusted odds ratio, AOR): 1.19, 95% CI:1.02–1.38, P = 0.022) and eGFR 2 (AOR: 1.41, 95% CI:1.12–1.78, P = 0.004), respectively. Patients with impaired renal function have a higher risk of infections by MDROs. Kidney dysfunction at admission may be an indicator for need of closer attention to microbial culture results requiring subsequent change of antibiotics.

Each year from April to May, high mortality rates are reported in red swamp crayfish (Procambarus clarkii) cultured in Jiangsu and other regions, in China, and this phenomenon has come to be known as “Black May” disease (BMD). Therefore, in order to investigate the possible causes of this disease, this study gathered BMD-affected P. clarkii samples and performed transcriptome analysis on hepatopancreas, gill, and muscle tissues. A total of 19,995,164, 149,212,804, and 222,053,848 clean reads were respectively obtained from the gills, muscle, and hepatopancreas of BMD-affected P. clarkii, and 114,024 unigenes were identified. The number of differentially expressed genes (DEGs) in gill, muscle, and hepatopancreas was 1703, 964, and 476, respectively. GO and KEGG enrichment analyses of the DEGs were then conducted. Based on KEGG pathway enrichment analysis, the most significantly differentially expressed pathways were mainly those involved with metabolism, human disease, and cellular processes. Further analysis of the significantly DEGs revealed that they were mainly related to the mitochondrial-mediated apoptosis pathway and that the expression of these DEGs was mostly down-regulated. Moreover, the expression of genes related to immune and metabolism-related pathways was also significantly down-regulated, and these significantly-inhibited pathways were the likely causes of P. clarkii death. Therefore, our results provide a basis for the identification of BMD causes.

X-ray dynamic tomographic microscopy offers new opportunities in the volumetric investigation of dynamic processes. Due to data complexity and their sheer amount, extraction of comprehensive quantitative information remains challenging due to the intensive manual interaction required. Particularly for dynamic investigations, these intensive manual requirements significantly extend the total data post-processing time, limiting possible dynamic analysis realistically to a few samples and time steps, hindering full exploitation of the new capabilities offered at dedicated time-resolved X-ray tomographic stations. In this paper, a fully automatized iterative tomographic reconstruction pipeline (rSIRT-PWC-DIFF) designed to reconstruct and segment dynamic processes within a static matrix is presented. The proposed algorithm includes automatic dynamic feature separation through difference sinograms, a virtual sinogram step for interior tomography datasets, time-regularization extended to small sub-regions for increased robustness and an automatic stopping criterion. We demonstrate the advantages of our approach on dynamic fuel cell data, for which the current data post-processing pipeline heavily relies on manual labor. The proposed approach reduces the post-processing time by at least a factor of 4 on limited computational resources. Full independence from manual interaction additionally allows straightforward up-scaling to efficiently process larger data, extensively boosting the possibilities in future dynamic X-ray tomographic investigations. © 2020, The Author(s)., Scientific Reports, 10 (1), ISSN:2045-2322

As a new plant biostimulant, poly-γ-glutamic acid (γ-PGA) may be an effective anti-drought agent that can efficiently alleviate the damage to plants under drought stress. In this study, the effects of γ-PGA on the physiological responses of oilseed rape (Brassica napus L.) seedlings under drought stress were investigated using hydroponics. Growth and development of the rape seedlings were significantly inhibited in a polyethylene glycol-simulated drought environment. However, 12 d after application of γ-PGA under drought stress, the fresh weight, chlorophyll content, and relative water content of rape seedlings all markedly increased. Moreover, proline content and antioxidant enzyme activity were all markedly enhanced, and the malondialdehyde content was significantly reduced in rape seedlings treated with γ-PGA. Furthermore, the content of the important anti-drought response hormone, abscisic acid (ABA), as well as the expression levels of the ABA metabolism regulation genes BnNCED3, BnZEP, and BnAAO4, significantly increased. These results indicate that γ-PGA may induce elements of a tolerance system to drought stress by promoting ABA accumulation in B. Napus.

Here, we bridge the gap between carbon mineralization and laundry detergent builder with sodium oxalate. Daily laundry can help mineralize carbon dioxide. First, we screen an environment-friendly process to produce sodium oxalate, using CO2 as a raw material. Then, we evaluate the properties of sodium oxalate as a detergent builder and prove the formation of calcium oxalate under laundry conditions. Our data suggest that sodium oxalate has excellent calcium-removing properties. Detergents based on sodium oxalate have good detergency. Furthermore, solid calcium oxalates (calcium oxalate monohydrate or calcium oxalate dihydrate, which has good stability in water and thermal stability), is obtained from washing waters. These results demonstrate the possibility of using sodium oxalate as detergent builder. The whole process can transform the greenhouse gas CO2 into commodity chemicals and can mineralize carbon.

A series of novel triazine-containing pore-tunable carbon materials (NT-POP@800-1-6), which was synthesized via pyrolysis of porous organic polymers (POPs) without any templates. NT-POP@800-1-6 possess moderate BET surface areas of 475–736 m2 g−1, have permanent porosity and plenty of nitrogen units in the skeletons as effective sorption sites, and display relatively rapid guest uptake of 56–192 wt% in iodine vapour in the first 4 h. In addition, all the samples exhibit the outstanding CO2 adsorption capacity of 2.83–3.96 mmol g−1 at 273 K and 1.05 bar. Furthermore, NT-POP@800-1-6 show good selectivity ratios of 21.2–36.9 and 3.3–7.5 for CO2/N2 or CH4/N2, respectively. We believe that our new building block design provides a general strategy for the construction of triazine-containing carbon materials from various extended building blocks, thereby greatly expanding the range of applicable molecules.

Straw is one of the most abundant stock of renewable biomass from crop production. However, its utilization efficiency is still very low. Although co-cultivation of fungi increases the degrading rate, the co-cultivation condition needs to be optimized. To optimize the co-culture condition of Phanerochaete chrysosporium and Trichoderma viride degrading rice straw, we first tested the antagonistic characteristic between the fungi. The results showed that the best co-culture pattern was to first inoculate P. chrysosporium and culture for 4 days, then inoculate T. viride, and co-culture the two fungi for 4 days. The optimum fermentation condition was 14% (w/v) of inoculum concentration, the equivalent inoculation of the fungi, culture temperature at 30 °C, and 1:1.4 for solid-liquid ratio. Under the optimum condition, the degradation ratios of lignin and cellulose were 26.38% and 33.29%, respectively; the soluble carbon content in the culture product was 23.07% (w/v). The results would provide important reference information for the efficient utilization of rice straw to produce more accessible energy resources, such as ethanol and glucose.

We herein report the construction of a novel azo-linked conjugated microporous polymers (Ag@AzoTPE-CMP), which possesses permanent porous structure and Ag+ loading up of 7.62% in the skeleton as effective sorption sites. Ag@AzoTPE-CMP shows considerable adsorption capacity of iodine of 202 wt% in iodine vapor at 350 K. In addition, Ag@AzoTPE-CMP can effectively remove heavy ions from ethanol-water solution.

A series of conjugated microporous polymers containing thiophene-moieties (SCMP-COOH@1-3) was obtained by a homo-coupling polymerization reaction. Then the SCMP-COOH@1-3 were directly pyrolyzed without any templates to synthesize the porous carbon networks, named as SCMP-600@1, 2 and 3. SCMP-600@1-3 possess moderate BET surface area of 362–642 m2 g−1, have a permanent porous structure and plenty of sulfur and oxygen units in the skeletons as effective sorption sites, and display a high absorption performance for iodine vapour with an uptake up to 204 wt.%. In addition, SCMP-COOH@1-3 polymers can be used to effectively detect mercury ion from ethanol-water solution. Interestingly, under the same concentration of Hg2+ conditions, the detection ability of mercury ion of porous materials increased with the increase of the pore volumes and the specific surface.

Silicosis is a common occupational disease and represents a significant contributor to respiratory morbidity and mortality worldwide. Lipid-laden macrophages, or foam cells, are observed in the lungs of patients with silicosis but the mechanisms mediating their formation remain poorly understood. In this study, we sought to elucidate the mechanisms by which silica promotes foam cell formation in the lung, and to determine whether uptake of lipids alone is sufficient to drive TGF-β production by alveolar macrophages. Consistent with previous reports, we found that foam cells were markedly increased in the lungs of patients with silicosis and that these findings associated with both higher levels of intracellular lipid levels (oxidized LDL, ox-LDL) and elevated transcript levels for the lipid scavenger receptor CD36 and the nuclear receptor PPARγ. Employing a rat alveolar macrophage cell line, we found that exposure to silica dust or ox-LDL alone had a modest effect on the induction of foam cell formation and only silica was capable of inducing the production of TGF-β. In contrast, foam cell formation and TGF-β production were both dramatically increased when cells were exposed to a combination of silica dust and ox-LDL. Moreover, we found that these endpoints were markedly attenuated by either blocking CD36 or inhibiting the activity of PPARγ. Altogether, our findings suggest that foam cell formation and TGF-β production are driven by the simultaneous uptake of silica and lipids in alveolar macrophages and that strategies aimed at blocking lipid uptake by alveolar macrophages might be effective in ameliorating fibrotic responses to silica in the lung.

Recent studies have reported intrinsic metabolic reprogramming in Pkd1 knock-out cells, implicating dysregulated cellular metabolism in the pathogenesis of polycystic kidney disease. However, the exact nature of the metabolic changes and their underlying cause remains controversial. We show herein that Pkd1 k o /ko renal epithelial cells have impaired fatty acid utilization, abnormal mitochondrial morphology and function, and that mitochondria in kidneys of ADPKD patients have morphological alterations. We further show that a C-terminal cleavage product of polycystin-1 (CTT) translocates to the mitochondria matrix and that expression of CTT in Pkd1 ko/ko cells rescues some of the mitochondrial phenotypes. Using Drosophila to model in vivo effects, we find that transgenic expression of mouse CTT results in decreased viability and exercise endurance but increased CO2 production, consistent with altered mitochondrial function. Our results suggest that PC1 may play a direct role in regulating mitochondrial function and cellular metabolism and provide a framework to understand how impaired mitochondrial function could be linked to the regulation of tubular diameter in both physiological and pathological conditions.

Ethanol has extensive effects on sleep and daytime alertness, causing premature disability and death. Adenosine, as a potent sleep-promoting substance, is involved in many cellular and behavioral responses to ethanol. However, the mechanisms of hypnotic effects of ethanol remain unclear. In this study, we investigated the role of adenosine in ethanol-induced sleep using C57BL/6Slac mice, adenosine A2A receptor (A2AR) knockout mice, and their wild-type littermates. The results showed that intraperitoneal injection of ethanol (3.0 g/kg) at 21:00 decreased the latency to non-rapid eye movement (NREM) sleep and increased the duration of NREM sleep for 5 h. Ethanol dose-dependently increased NREM sleep, which was consistent with decreases in wakefulness in C57BL/6Slac mice compared with their own control. Caffeine (5, 10, or 15 mg/kg), a nonspecific adenosine receptor antagonist, dose-dependently and at high doses completely blocked ethanol-induced NREM sleep when administered 30 min prior to (but not after) ethanol injection. Moreover, ethanol-induced NREM sleep was completely abolished in A2AR knockout mice compared with wild-type mice. These findings strongly indicate that A2AR is a key receptor for the hypnotic effects of ethanol, and pretreatment of caffeine might be a strategy to counter the hypnotic effects of ethanol.

Predominantly based on studies from high-income countries, reduced estimated glomerular filtration rate (eGFR) has been associated with increased risk of infections and infection-related hospitalizations (IRHs). We here explore in-hospital outcomes of IRHs in patients with different kidney function. A total of 6,283 adults, not on renal replacement therapy, with a discharge diagnosis of infection, and with an eGFR 1–12 months before index hospitalization, were included from four hospitals in China. We compared in-hospital outcomes (death, intensive care unit (ICU) admission, length of hospital stay (LOHS) and medical expenses), between patients with and without chronic kidney disease (CKD, defined as eGFR ≤ 60 ml/min per 1.73 m2 of body surface area) by mixed-effects logistic regression model or generalized linear model. The odds for in-hospital mortality (adjusted odds ratios (OR) = 1.41; 95% CI 1.02–1.96) and ICU admission (OR = 2.18; 95% CI 1.64–2.91) were higher among patients with CKD. Sperm motility is the most important indicator in evaluating roosters’ fecundity. However, the genetic mechanisms underlying chicken sperm motility is not yet clear. Long non-coding RNA (lncRNA) play epigenetic roles in reproduction. In this study, RNA sequencing was employed to profile the testis transcriptome (lncRNA and mRNA) of six Beijing-you cocks divergent in sperm motility. In total, 2,597 lncRNAs were identified in the chicken testis, including 1,267 lincRNAs, 975 anti-sense lncRNAs, and 355 intronic lncRNAs. They shared similar features with previous studies. Of these lncRNAs, 124 were differentially expressed. Among 17,690 mRNAs detected in this study, 544 were differentially expressed, including a bunch of genes with known functions on sperm motility. GO annotation analysis revealed these genes were involved in ATP binding, cilium assembly, and oxidation-reduction processes. Integrating analysis of lncRNA and mRNA profiles predicted 10 lncRNA-gene pairs, including 8 co-regulated and 2 inversely-regulated pairs. To the best of our knowledge, this is the first genome-wide investigation of the lncRNAs in the chicken testis associated with sperm motility. Our results provided a catalog of chicken testis lncRNAs and genes worthy of further studies to understand their roles in cocks’ reproductive performance regulation.

In recent years, Microcystis aeruginosa blooms have occurred throughout the world, causing huge economic losses and destroying aquatic ecosystems. It is necessary to develop effective and ecofriendly methods to control M. aeruginosa blooms. Here, we report a high algicidal activity of prodigiosin (PG) against M. aeruginosa as well as the algicidal mechanism. PG showed high algicidal activity against M. aeruginosa, with a 50% lethal dose (LD50) of 5.87 μg/mL in 72 h. A combination of methods, including propidium iodide and Annexin V-fluorescein staining assays and light and electron microscopy indicated the existence of two modes of cell death with features similar to those in eukaryotic programmed cell death: necrotic-like and apoptotic-like. Biochemical and physiological analyses showed that PG generates reactive oxygen species (ROS), which induce lipid peroxidation, damage the membrane system and destroy the function of the photosystem. A proteomics analysis revealed that many proteins were differentially expressed in response to PG stress and that most of these proteins were involved in important metabolic processes, which may trigger necrotic-like or apoptotic-like cell death. The present study sheds light on the multiple toxicity mechanisms of PG on M. aeruginosa and its potential for controlling the occurrence of M. aeruginosa blooms in lakes.

Lysine acetylation and succinylation are major types of protein acylation that are important in many cellular processes including gene transcription, cellular metabolism, DNA damage response. Malfunctions in these post-translational modifications are associated with genome instability and disease in higher organisms. In this study, we used high-resolution nano liquid chromatography-tandem mass spectrometry combined with affinity purification to quantify the dynamic changes of protein acetylation and succinylation in response to ultraviolet (UV)-induced cell stress. A total of 3345 acetylation sites in 1440 proteins and 567 succinylation sites in 246 proteins were identified, many of which have not been reported previously. Bioinformatics analysis revealed that these proteins are involved in many important biological processes, including cell signalling transduction, protein localization and cell metabolism. Crosstalk analysis between these two modifications indicated that modification switches might regulate protein function in response to UV-induced DNA damage. We further illustrated that FEN1 acetylation at different sites could lead to different cellular phenotypes, suggesting the multiple function involvement of FEN1 acetylation under DNA damage stress. These systematic analyses provided valuable resources and new insight into the potential role of lysine acetylation and succinylation under physiological and pathological conditions.

Alginate, a natural acidic polysaccharide extracted from marine brown seaweeds, is composed of different blocks of β-(1, 4)-D-mannuronate (M) and its C-5 epimer α-(1, 4)-L-guluronate (G). Alginate-derived guluronate oligosaccharide (GOS) readily activates macrophages. However, to understand its role in immune responses, further studies are needed to characterize GOS transport and signalling. Our results show that GOS is recognized by and upregulates Toll-like receptor 4 (TLR4) on RAW264.7 macrophages, followed by its endocytosis via TLR4. Increased expression of TLR4 and myeloid differentiation protein 2 (MD2) results in Akt phosphorylation and subsequent activation of both nuclear factor-κB (NF-κB) and mechanistic target of rapamycin (mTOR). Moreover, GOS stimulates mitogen-activated protein kinases (MAPKs); notably, c-Jun N-terminal kinase (JNK) phosphorylation depends on TLR4 initiation. All these events contribute to the production of inflammatory mediators, either together or separately. Our findings also reveal that GOS induces cytoskeleton remodelling in RAW264.7 cells and promotes macrophage proliferation in mice ascites, both of which improve innate immunity. Conclusively, our investigation demonstrates that GOS, which is dependent on TLR4, is taken up by macrophages and stimulates TLR4/Akt/NF-κB, TLR4/Akt/mTOR and MAPK signalling pathways and exerts impressive immuno-stimulatory activity.

A series of conjugated microporous polymers (CMPs) based on 1,3,6,8-tetrabromocarbazole (N4CMP-1–5) is synthesized via Suzuki cross-coupling or Sonogashira polycondensation. The porosity properties and surface area of these polymer networks can be finely tuned by using a linker with different geometries or strut length. These polymers show the Brunauer-Emmett-Tellerthe (BET) surface areas ranging from 592 to 1426 m2 g−1. The dominant pore sizes of the polymers on the basis of the different linker are located between 0.36 and 0.61 nm. Gas uptake increases with BET surface area and micropore volume, N4CMP-3 polymer can capture CO2 with a capacity of 3.62 mmol g−1 (1.05 bar and 273 K) among the obtained polymers. All of the polymers show high isosteric heats of CO2 adsorption (25.5–35.1 kJ mol−1), and from single component adsorption isotherms, IAST-derived ideal CO2/N2 (28.7–53.8), CO2/CH4 (4.6–5.2) and CH4/N2 (5.7–10.5) selectivity. Furthermore, N4CMPs exhibit the high CO2 adsorption capacity of 542–800 mg g−1 at 318 K and 50 bar pressure. These data indicate that these materials are a promising potential for clean energy application and environmental field.

Poly-γ-glutamic acid (γ-PGA) is a microbe-secreted isopeptide that has been shown to promote growth and enhance stress tolerance in crops. However, its site of action and downstream signaling pathways are still unknown. In this study, we investigated γ-PGA-induced tolerance to salt and cold stresses in Brassica napus L. seedlings. Fluorescent labeling of γ-PGA was used to locate the site of its activity in root protoplasts. The relationship between γ-PGA-induced stress tolerance and two signal molecules, H2O2 and Ca2+, as well as the γ-PGA-elicited signaling pathway at the whole plant level, were explored. Fluorescent labeling showed that γ-PGA did not enter the cytoplasm but instead attached to the surface of root protoplasm. Here, it triggered a burst of H2O2 in roots by enhancing the transcription of RbohD and RbohF, and the elicited H2O2 further activated an influx of Ca2+ into root cells. Ca2+ signaling was transmitted via the stem from roots to leaves, where it elicited a fresh burst of H2O2, thus promoting plant growth and enhancing stress tolerance. On the basis of these observation, we propose that γ-PGA mediates stress tolerance in Brassica napus seedlings by activating an H2O2 burst and subsequent crosstalk between H2O2 and Ca2+ signaling.

To evaluate the diagnostic performance of a new two-dimensional shear wave speed (SWS) imaging (i.e. Toshiba shear wave elastography, T-SWE) in differential diagnosis of breast lesions. 225 pathologically confirmed breast lesions in 218 patients were subject to conventional ultrasound and T-SWE examinations. The mean, standard deviation and ratio of SWS values (m/s) and elastic modulus (KPa) on T-SWE were computed. Besides, the 2D elastic images were classified into four color patterns. The area under the receiver operating characteristic (AUROC) curve analysis was performed to evaluate the diagnostic performance of T-SWE in differentiation of breast lesions. Compared with other quantitative T-SWE parameters, mean value expressed in KPa had the highest AUROC value (AUROC = 0.943), with corresponding cut-off value of 36.1 KPa, sensitivity of 85.1%, specificity of 96.6%, accuracy of 94.2%, PPV of 87.0%, and NPV of 96.1%. The AUROC of qualitative color patterns in this study obtained the best performance (AUROC = 0.957), while the differences were not significant except for that of Eratio expressed in m/s (AUROC = 0.863) (P = 0.03). In summary, qualitative color patterns of T-SWE obtained the best performance in all parameters, while mean stiffness (36.05 KPa) provided the best diagnostic performance in the quantitative parameters.

The translocation and assembly module (TAM) in bacteria consists of TamA and TamB that form a complex to control the transport and secretion of outer membrane proteins. Herein, we demonstrated that the DR_1462-DR_1461-DR_1460 gene loci on chromosome 1 of Deinococcus radiodurans, which lacks tamA homologs, is a tamB homolog (DR_146T) with two tamB motifs and a DUF490 motif. Mutation of DR_146T resulted in cell envelope peeling and a decrease in resistance to shear stress and osmotic pressure, as well as an increase in oxidative stress resistance, consistent with the phenotype of a surface layer (S-layer) protein SlpA (DR_2577) mutant, demonstrating the involvement of DR_146T in maintenance of cell envelope integrity. The 123 kDa SlpA was absent and only its fragments were present in the cell envelope of DR_146T mutant, suggesting that DR_146T might be involved in maintenance of the S-layer. A mutant lacking the DUF490 motif displayed only a slight alteration in phenotype compared with the wild type, suggesting DUF490 is less important than tamB motif for the function of DR_146T. These findings enhance our understanding of the properties of the multilayered envelope in extremophilic D. radiodurans, as well as the diversity and functions of TAMs in bacteria.

Lamb-dip spectroscopy of the C−N stretching band of methylamine has been systematically extended to P-, Q-, and R-branch by using microwave sidebands of a large number of CO2 laser lines as frequency-tunable infrared sources in a sub-Doppler spectrometer. Lamb-dip signals of more than 150 spectral lines have been observed with a resolution of 0.4 MHz and their frequencies have been precisely measured with an accuracy of ±0.1 MHz. More than 30 closed combination loops have been formed, which unambiguously confirm the assignments. For over 150 vibrational excited levels in 27 substates, refined term values have been determined and expanded in J(J + 1) power-series to determine the substate origins and the effective rotational constants. For transitions with Aa torsion-inversion symmetry in torsional state υt = 0, 57 K-doublet lines displaying asymmetry splittings have been observed and the splitting constants for levels with K = 1, 2, and 3 in the excited states have been determined. Our results provide accurate experimental information for spectroscopic studies of the interesting vibrational perturbations and intermode interactions related to the C−N stretching mode, directly support astronomical surveys, and are very relevant in practice to identification and frequency determination of the CO2-laser-pumped far-infrared laser lines of methylamine.

This study tested whether activation of adrenoreceptors in chondrocytes has roles in degenerative remodelling of temporomandibular joint (TMJ) and to determine associated mechanisms. Unilateral anterior crossbite (UAC) was established to induce TMJ degeneration in rats. Saline vehicle, α2- and β-adrenoreceptor antagonists or agonists were injected locally into the TMJ area of UAC rats. Cartilage degeneration, subchondral bone microarchitecture and the expression of adrenoreceptors, aggrecans, matrix metalloproteinases (MMPs) and RANKL by chondrocytes were evaluated. Chondrocytes were stimulated by norepinephrine to investigate signal transduction of adrenoreceptors. Increased α2A-adrenoreceptor expression was observed in condylar cartilage of UAC rats, together with cartilage degeneration and subchondral bone loss. Norepinephrine depresses aggrecans expression but stimulates MMP-3, MMP-13 and RANKL production by chondrocytes through ERK1/2 and PKA pathway; these effects were abolished by an α2A-adrenoreceptor antagonist. Furthermore, inhibition of α2A-adrenoreceptor attenuated degenerative remodelling in the condylar cartilage and subchondral bone, as revealed by increased cartilage thickness, proteoglycans and aggrecan expression and decreased MMP-3, MMP-13 and RANKL expressions in cartilage, increased BMD, BV/TV and decreased Tb.Sp in subchondral bone. Conversely, activation of α2A-adrenoreceptor intensified aforementioned degenerative changes in UAC rats. It is concluded that activation of α2A-adrenergic signal in chondrocytes promotes TMJ degenerative remodelling by chondrocyte-mediated pro-catabolic activities.

Lysine acetylation and succinylation are major types of protein acylation that are important in many cellular processes including gene transcription, cellular metabolism, DNA damage response. Malfunctions in these post-translational modifications are associated with genome instability and disease in higher organisms. In this study, we used high-resolution nano liquid chromatography-tandem mass spectrometry combined with affinity purification to quantify the dynamic changes of protein acetylation and succinylation in response to ultraviolet (UV)-induced cell stress. A total of 3345 acetylation sites in 1440 proteins and 567 succinylation sites in 246 proteins were identified, many of which have not been reported previously. Bioinformatics analysis revealed that these proteins are involved in many important biological processes, including cell signalling transduction, protein localization and cell metabolism. Crosstalk analysis between these two modifications indicated that modification switches might regulate protein function in response to UV-induced DNA damage. We further illustrated that FEN1 acetylation at different sites could lead to different cellular phenotypes, suggesting the multiple function involvement of FEN1 acetylation under DNA damage stress. These systematic analyses provided valuable resources and new insight into the potential role of lysine acetylation and succinylation under physiological and pathological conditions.
