Abstract Premature ovarian insufficiency (POI), a major cause of female infertility, is defined as follicular atresia and a rapid loss of germ cells in women of reproductive age due to ovarian failure. Recently, findings from several studies have indicated that human umbilical cord mesenchymal stem cells (hUMSCs) can alleviate ovarian dysfunction resulting from POI. However, the mechanisms underlying this effect require further clarification. In this study, a mouse model of POI was established as achieved with an intraperitoneal injection of cyclophosphamide (CTX) into female C57BL/6J mice in vivo. These POI mice received a 1-week intervention of hUMACs. In addition, an in vitro POI model was also included. The cultured supernatants of hUMSCs and glycogen synthase kinase 3 beta (GSK3β) inhibitor (SB216763) were used to treat theca cells (TCs) exposed to CTX. Hematoxylin and Eosin (H&E) staining and Enzyme-linked immunosorbent assay (ELISA) were used to assess ovarian structure and morphology, as well as endocrine function in these POI mice. Based on results from the ELISA and JC-1 labeling, CTX exerted significant detrimental effects on testosterone levels and the mitochondrial membrane potential in TCs. Subsequently, Western Blot, Immunofluorescence staining (IF), and Quantitative real-time polymerase chain reaction (qRT-PCR) were used to evaluate various indicators of testosterone synthesis function and mitochondrial dynamics in ovaries and TCs of POI mice. In vivo, dysfunctions in ovarian structure and function in the POI mouse model were effectively restored following hUMSCs treatment, and abnormalities in hormone synthesis were significantly reduced. Furthermore, when the stem cell supernatants of hUMSCs were applied to TCs in vitro we found that GSK3β expression was reduced, the imbalance of mitochondrial dynamics was alleviated, and the ability of mitochondrial testosterone synthesis was increased. Taken together, our results indicate that hUMSCs treatment can restore the imbalance of mitochondrial dynamics and restart testosterone synthesis of TCs by suppressing GSK3β expression, ultimately alleviating POI damage.

Abstract Coal mining may lead to ground subsidence in a long term and is widely distributed, which can cause environmental damage and other disasters. Predicting the dynamic process of ground subsidence in real time is very important for offering theoretical or technical guidance to deal with the consequences of mining. In this study, we developed a prediction method for dynamic ground subsidence using a time function model that considers two stages of surface subsidence and reflects the law of surface subsidence in goaf. We applied the model to the Barapukuria mine, and our simulation shows that the prediction results are in good agreement with the monitoring data. Our results suggest that the dynamic development of the ground subsidence basin may be an effective measure to assess the loss of ground and provide early warning of oncoming hazards.

Abstract The process of blasting stress wave propagation and crack propagation is directly affected by the physical properties of the rock mass and internal joints in the rock. In soft and hard rock layers, the blasting process is more complicated since the blasting stress wave needs to penetrate two kinds of rocks with different physical properties and the interface between soft rock and hard rock. In this study, the modal transformation of stress waves at the interface of layered composite rock was analyzed, and the process was reproduced by finite element analysis. Furthermore, the development law of cracks was explored. The research results demonstrated that in the single blasting-hole model, a triangular crack area caused by reflected stress waves appeared at the rock interface of rock medium I near the blast hole. In rock medium II, the tensile crack generated by the interface wave appeared on the side away from the blast hole. Besides, the development of the tensile crack was associated with the incident mode of the blast stress wave and the incident angle. In the deep hole blasting model, the incidence of the detonation wave front from hard rock to soft rock promoted the fragmentation of the hard rock.

Abstract This study determined the Water Production Function (WPF) and Optimal Irrigation Schedule (OIS) for rice (Oryza sativa L.) cultivated with drip irrigation under plastic film. Six different field capacity levels were established, 100% (W1), 90% (W2), 80% (W3), 70% (W4) and 60% (W5). The results showed that, the rice growth and yields and quality were significantly affected by the different irrigation treatments. The rice height and yield decreased from W1to W4, the W2 is the highest yield. The lower the amount of irrigation water applied was, the higher the Irrigation Water Use Efficiency values were. A WPF model was established for this cropping system, and the water sensitivity indices calculated by the mathematical model showed that the crop water sensitivity decreased in the order booting stage > flowering stage > tillering stage > filling stage. Based on this result, the OIS determined by the dynamic solution of several models was as follows: the optimal irrigation levels were 750 m3 ha−1 in the tillering stage, 2125 m3 ha−1 in the jointing-booting stage, 1050 m3 ha−1 in the heading-flowering stage and 325 m3 ha−1 in the milk stage. The WPF and OIS developed in this study provide a theoretical basis for the implementation of rice cultivation with drip irrigation under plastic film in arid regions of China.

Abstract There is no study exploring the association between triglyceride-glucose (TyG) index and skeletal muscle mass in Chinese adolescents. Therefore, the objective of this study is to explore the association between TyG index and appendicular lean mass (ALM) in Chinese adolescents. In this study, 1336 adolescents (805 boys, 60.25%) aged 12–18 years in China were randomly selected through a stratified cluster sampling. According to the tertiles of TyG index, we separated all participants into three groups, and LM was measured by Bioelectrical Impedance Analysis. Abstract This paper investigated the preparation method and the dispersion behaviour of Modified Carbon Nanotube-fiber Reinforcements (MCNF), the change laws and the effect mechanisms of dynamic compressive strength of MCNF concretes. Electrophoresis method was used to prepare MCNF and its interfacial shear performance was tested by interfacial shear strength (IFSS) test. In addition, the dispersion behavior of MCNF in simulated concrete solution was verified by turbidity method. Split Hopkinson Pressure Bar (SHPB), Scanning Electron Microscope (SEM) and Mercury Intrusion Porosimetry (MIP) tests were carried on concrete samples with different volume fractions (0%, 0.1%, 0.2%, 0.3%, 0.4%) of MCNF. The results show that carbon nanotubes are easier to deposit to the negative electrode, and the higher the content of polycarboxylate superplasticizer, the more obvious the dispersity of MCNF in alkaline environment. The dynamic compressive strength of MCNF concrete was 14.0–35.5% higher than that of untreated concrete, and reached the maximum when the MCNF content was about 0.3%. The MCNF was wrapped in concrete matrix and promoted hydration reaction of interface between cement and MCNF from microscopic observation. The addition of MCNF could increase the porosity. The volume percentage of ≥ 100 nm pore decreased first and then increased. Reasons for the improvement strength of MCNF concrete is that the bridging effect is stronger with the increase of MCNF content (≤ 0.3%) and limited when the MCNF content is equal to 0.4%. MCNF concrete could be used in actual engineering with high requirements for dynamic load.

Abstract Lean NAFLD is a special phenotypic closely correlated with metabolic syndrome (MS). The aim of this study is to investigate the MS development and the gender differences in lean NAFLD population. Participants were divided into 4 groups by BMI and NAFLD status. Descriptive analysis was performed to characterize baseline information. A total of 18,395 subjects were participated, and 1524 incident cases of MS were documented. Then, Kaplan–Meier curves were used to present the MS outcomes in different groups, and the NAFLD was found to be a riskier factor than obesity for MS. Subgroup analysis showed significantly higher MS incidence in female than male among lean NAFLD group, which is different from other groups. Although with higher prevalence in male, lean NAFLD seems to be a more harmful phenotype for females according to the TG, ALT and GGT levels. The logistic regressive analysis was performed to show the impact of NAFLD status and BMI changes on MS risk. Lean non-NAFLD subjects merely developed to NAFLD with no BMI status changes exhibited highest MS risk (ORs = 1.879, 95% CI 1.610–2.292) than that with both BMI increase and NAFLD development (ORs = 1.669, 95% CI 1.325–2.104). It also suggests the metabolic specificity of this population.

Abstract Acoustic emission (AE) characterization is an effective technique to indirectly capture the failure process of quasi brittle rock. In previous studies, both experiments and numerical simulations were adopted to investigate the AE characteristics of rocks. However, as the most popular numerical model, the moment tensor model (MTM) cannot be constrained by the experimental result because there is a gap between MTM and experiments in principle, signal processing and energy analysis. In this paper, we developed a particle-velocity-based model (PVBM) that enabled direct monitoring and analysis of the particle velocity in the numerical model and had good robustness. The PVBM imitated the actual experiment and could fill in gaps between the experiment and MTM. AE experiments of marine shale under uniaxial compression were carried out, and the results were simulated by MTM. In general, the variation trend of the experimental result could be presented by MTM. Nevertheless, the magnitudes of AE parameters by MTM presented notable differences of more than several orders of magnitude compared with those by the experiment. We sequentially used PVBM as a proxy to analyse these discrepancies and systematically evaluate the AE characterization of rocks from the experiment to numerical simulation, considering the influence of wave reflection, energy geometrical diffusion, viscous attenuation, particle size and progressive deterioration of rock material. The combination of MTM and PVBM could reasonably and accurately acquire AE characteristics of the actual AE experiment of rocks by making full use of their respective advantages.

Abstract The relationship between visceral adiposity index (VAI) and unhealthy metabolic phenotype remained unclear in children and adolescents. This study aimed to investigate their association and compared the ability of VAI and traditional adiposity indicators (body mass index, waist circumference and waist-to-height ratio) to predict metabolically unhealthy phenotype among normal-weight, overweight and obese children and adolescents. In this cross-sectional study, 1722 children and adolescents aged 12–18 years were selected by cluster random sampling, underwent a questionnaire survey, physical examination and biochemical tests. Participants were divided into four phenotypes according to the combination of the weight status determined by body mass index (BMI) and metabolic syndrome components. Receiver operating characteristic (ROC) analysis and multivariate logistic regression were used to compare the predictive capacity between VAI and traditional adiposity indicators and their relationship with metabolically unhealthy phenotype. Abstract Amomum tsao-ko, as an edible and medicinal variety, has been cultivated for more than 600 years in China. Recently, two cultivars, A. tsao-ko and Amomum paratsao-ko, were found in A. tsao-ko planting area. The two cultivars are often confused because of the similar phenotype and difficult to distinguish through sensory judgment. In this study, the non-targeted gas chromatography-mass spectrometry (GC–MS) metabolomics combined with near-infrared spectroscopy (NIRS) were used for dissecting the two cultivars with phenotypic differences. According to principal component analysis (PCA) loading diagram and orthogonal partial least squares discriminant analysis (OPLS-DA) S-plot of the metabolites, the accumulation of major components including 1,8-cineole, α-phellandrene, (E)-2-decenal, (−)-β-pinene, (E)-2-octenal, 1-octanal, D-limonene, and decanal, were present differences between the two cultivars. Seven metabolites potential differentiated biomarkers as β-selinene, decamethylcyclopentasiloxane, (E,Z)-2,6-dodecadienal, (E)-2-hexenal, (E)-2-decenal, isogeranial, 1,8-cineole and β-cubebene were determined. Although A. tsao-ko and A. paratsao-ko belong to the same genera and are similar in plant and fruit morphology, the composition and content of the main components were exposed significant discrepancy, so it is necessary to distinguish them. In this study, the discriminant model established by GC–MS or NIRS combined with multivariate analysis has achieved a good classification effect. NIRS has the advantages of simple, fast and nondestructive and can be used for rapid identification of varieties and fruit tissues.

Abstract The recent COVID-19 pandemic poses a serious threat to global public health, thus there is an urgent need to define the molecular mechanisms involved in SARS-CoV-2 spike (S) protein-mediated virus entry that is essential for preventing and/or treating this emerging infectious disease. In this study, we examined the blocking activity of human COVID-19 convalescent plasma by cell–cell fusion assays using SARS-CoV-2-S-transfected 293 T as effector cells and ACE2-expressing 293 T as target cells. We demonstrate that the SARS-CoV-2 S protein exhibits a very high capacity for membrane fusion and is efficient in mediating virus fusion and entry into target cells. Importantly, we find that COVID-19 convalescent plasma with high titers of IgG neutralizing antibodies can block cell–cell fusion and virus entry by interfering with the SARS-CoV-2-S/ACE2 or SARS-CoV-S/ACE2 interactions. These findings suggest that COVID-19 convalescent plasma may not only inhibit SARS-CoV-2-S but also cross-neutralize SARS-CoV-S-mediated membrane fusion and virus entry, supporting its potential as a preventive and/or therapeutic agent against SARS-CoV-2 as well as other SARS-CoV infections.

Abstract Utilizing methods such as scanning electron microscopy observation and mercury intrusion porosimetry, this paper investigates the basic microstructure and pore structure properties of polymer-cement composite joint sealants for pavements, and analyzes the effects and rules of various material types, ratio parameters and processing conditions. Further, the fractal characteristics and variation rules of pore size distribution are investigated for the joint sealants by introducing the fractal theory. The results show that changes in material type, ratio parameter and processing condition produce insignificant effects on the basic microstructure properties and configuration of joint sealants, with effects reflected primarily in the change of sealant pore structure. Measures like increasing the powder-liquid ratio and cement ratio, blending with sulphoaluminate cement or mica powder, adding latex powder or coupling agent, cold drawing and hot pressing, as well as ultraviolet irradiation treatment are all capable of reducing the total pore volume of joint sealants and refining their pore structure. In contrast, opposite effects are yielded when low-grade cement is used, styrene-acrylic emulsion is blended, or plasticizer is added. Additionally, after blending with talc powder or adding carbon fiber additive, the total pore volume of joint sealants remains basically unchanged or reduced, despite the coarsened pore structure. The total pore volume of joint sealants increases after wet–dry cycling treatment, while no obvious change in the pore size distribution is observed. Pore size distribution of the studied joint sealants presents distinct fractal characteristics, and the corresponding fractal dimension of pore surface area ranges between 2.6 and 2.8.

Abstract HOXA transcript antisense RNA myeloid-specific 1 (HOTAIRM1) is a long non-coding RNA (lncRNA) that plays a pivotal role in regulating myeloid cell development via targeting HOXA1 gene expression. We and others have previously shown that myeloid-derived suppressor cells (MDSCs), a heterogeneous population of immature myeloid cells, expand during chronic viral (HCV, HIV) infections. However, the role of HOTAIRM1 in the development and suppression of MDSCs during viral infection remains unknown. In this study, we demonstrate that the expressions of HOTAIRM1 and its target HOXA1 are substantially upregulated to promote the expressions of immunosuppressive molecules, including arginase 1, inducible nitric oxide synthase, signal transducer and activator of transcription 3, and reactive oxygen species, in CD33+ myeloid cells derived from hepatitis C virus (HCV)-infected patients. We show that HCV-associated exosomes (HCV-Exo) can modulate HOTAIRM1, HOXA1, and miR124 expressions to regulate MDSC development. Importantly, overexpression of HOTAIRM1 or HOXA1 in healthy CD33+ myeloid cells promoted the MDSC differentiation and suppressive functions; conversely, silencing of HOTAIRM1 or HOXA1 expression in MDSCs from HCV patients significantly reduced the MDSC frequency and their suppressive functions. In essence, these results indicate that the HOTAIRM1-HOXA1-miR124 axis enhances the differentiation and suppressive functions of MDSCs and may be a potential target for immunomodulation in conjunction with antiviral therapy during chronic viral infection.

Abstract Macrodomains are conserved protein folds associated with ADP-ribose binding and turnover. ADP-ribosylation is a posttranslational modification catalyzed primarily by ARTD (aka PARP) enzymes in cells. ARTDs transfer either single or multiple ADP-ribose units to substrates, resulting in mono- or poly-ADP-ribosylation. TARG1/C6orf130 is a macrodomain protein that hydrolyzes mono-ADP-ribosylation and interacts with poly-ADP-ribose chains. Interactome analyses revealed that TARG1 binds strongly to ribosomes and proteins associated with rRNA processing and ribosomal assembly factors. TARG1 localized to transcriptionally active nucleoli, which occurred independently of ADP-ribose binding. TARG1 shuttled continuously between nucleoli and nucleoplasm. In response to DNA damage, which activates ARTD1/2 (PARP1/2) and promotes synthesis of poly-ADP-ribose chains, TARG1 re-localized to the nucleoplasm. This was dependent on the ability of TARG1 to bind to poly-ADP-ribose. These findings are consistent with the observed ability of TARG1 to competitively interact with RNA and PAR chains. We propose a nucleolar role of TARG1 in ribosome assembly or quality control that is stalled when TARG1 is re-located to sites of DNA damage.

Abstract Congenital cataract is the most frequent inherited ocular disorder and the most leading cause of lifelong visual loss. The screening of pathogenic mutations can be very challenging in some cases, for congenital cataracts are clinically and genetically heterogeneous diseases. The aim of this study is to investigate the mutation spectrum and frequency of 54 cartaract-associated genes in 27 Chinese families with congenital cataracts. Variants in 54 cataract-associated genes were screened by targeted next-generation sequencing (NGS) and then validated by Sanger sequencing. We identified pathogenic variants in 62.96% (17/27) of families, and over 52.94% (9/17) of these variants were novel. Among them, three are splicing site mutations, four are nonsense mutations, seven are missense mutations, two are frame shift mutations and one is intronic mutation. This included identification of: complex ocular phenotypes due to two novel PAX6 mutations; progressive cortical cataract and lamellar cataract with lens subluxation due to two novel CRYGS mutations. Mutations were also found in rarely reported genes including CRYBA4, CRYBA2, BFSP1, VIM, HSF4, and EZR. Our study expands the mutation spectrum and frequency of genes responsible for congenital cataracts. Targeted next-generation sequencing in inherited congenital cataract patients provided significant diagnostic information.
