Abstract India produces huge quantities of agricultural residues and stubbles and mainly disposed by burning on site causing air pollution. The organic matter present in the residues and stubble may be utilized by anaerobic digestion as a source of renewable energy subsequently reducing emission of greenhouse gases caused by burning. In the present study, solid state anaerobic digestion (SSAD) of pearl millet straw was investigated at mesophilic and thermophilic temperature with four different total solid (TS) content (15, 20, 25 and 30%). Results showed that 20 and 25% TS generated maximum methane (124.1 ± 7 and 162.4 ± 9L/kg VS) at mesophilic and thermophilic temperature respectively. However, increasing TS content beyond 25% did not show significant increment on methane yield. Analytical analysis showed correlation between the reduction of volatile solids and methane yield as well as VFA (volatile fatty acid) accumulation at high TS content. Also, VlseKriterijuska Optimizacija Komoromisno Resenje (VIKOR) and Technique for Order Preference by Similarity to Ideal Solution (TOPSIS) methods as MultiCriteria Decision Making modelling (MCDM) applied to select best possible alternative for SSAD of pearl millet. MCDM analysis showed that VIKOR method endorsed the experimental results.

Abstract Naumure Multipurpose Project (NMP) featuring a 169 m high Concrete Face Rock Filled Dam (CFRD) is the proposed reservoir project in the West Rapti River with an installed capacity of 218.34 MW. Most of the rivers in Nepal carry significant sediment loads that will consequently catalyze reservoir sedimentation. This phenomenon prevails as the primary factor in reducing reservoirs useful life, making ineffective for both flood control and hydroelectricity generation. Ultimately, such process of sedimentation has adverse impacts on projects economic feasibility and long-term sustainability. Therefore, the objective of this research was to examine the expected sediment deposition pattern in the NMP reservoir throughout its operational period by employing 1D HEC-RAS model to simulate the sedimentation process. The model was simulated for 10, 20, 30, 40 and 50 years. Yang’s equation as a sediment transport function, Active layer as a bed sorting method and Toffaleti as a fall velocity method were best suited for the river reach. The delta deposition was formed between 11 km and 22 km upstream of the dam region in the Jhimruk river, with the sediment deposition depth reaching peaks of about 23 m, 38 m, 39 m, 41 m and 49 m in 10, 20, 30, 40 and 50 years, respectively. Similarly, the delta deposition was formed between 13 km and 33 km upstream of dam region in the Madi river, with the sediment deposition depth reaching peaks of about 47 m, 62 m, 60 m, 68 m and 75 m in 10, 20, 30, 40 and 50 years respectively. Headcutting of delta deposition occurred between 20 and 30 year due to high flood during low stage of reservoir. Furthermore, the study revealed that about 6.22%, 11.61%, 15.94%, 22.96% and 25.65% of the storage capacity of NMP reservoir will be depleted in 10, 20, 30, 40 and 50 years respectively.

As a means to understand human neuropsychiatric disorders from human brain samples, we compared the transcription patterns and histological features of postmortem brain to fresh human neocortex isolated immediately following surgical removal. Compared to a number of neuropsychiatric disease-associated postmortem transcriptomes, the fresh human brain transcriptome had an entirely unique transcriptional pattern. To understand this difference, we measured genome-wide transcription as a function of time after fresh tissue removal to mimic the postmortem interval. Within a few hours, a selective reduction in the number of neuronal activity-dependent transcripts occurred with relative preservation of housekeeping genes commonly used as a reference for RNA normalization. Gene clustering indicated a rapid reduction in neuronal gene expression with a reciprocal time-dependent increase in astroglial and microglial gene expression that continued to increase for at least 24 h after tissue resection. Predicted transcriptional changes were confirmed histologically on the same tissue demonstrating that while neurons were degenerating, glial cells underwent an outgrowth of their processes. The rapid loss of neuronal genes and reciprocal expression of glial genes highlights highly dynamic transcriptional and cellular changes that occur during the postmortem interval. Understanding these time-dependent changes in gene expression in post mortem brain samples is critical for the interpretation of research studies on human brain disorders.

Abstract This work presents a study on the surface tension of liquid Aluminum–Nickel (Al–Ni) alloys. Obtaining adequate values of surface tension for this system is not a simple task as these alloys present the formation of atomic clusters with short-range order at certain compositions, which dramatically influences surface tension. The Compound Forming Model predicts the influence of these clusters on surface tension, but experimental limitations have obstructed its validation due to deficient thermodynamic data. This work attempts to overcome some of these limitations by using Molecular Dynamics (MD). By comparing the obtained results from MD simulations with those of an equivalent system without clusters, it was possible to infer the role of the atomic clusters on Al–Ni surface tension. It was found that these clusters increase surface tension by decreasing the Al content at the surface. They achieve this reduction in Al content at the surface by trapping Al atoms and hindering their travel to the surface.

Abstract There is a complete lack of highly sensitive and specific biomarkers for early pancreatic ductal adenocarcinoma (PDAC) diagnosis, limiting multi-modal therapeutic options. Mitochondrial DNA (mtDNA) is an excellent resource for biomarker discovery because of its high copy number and increased mutational frequency in cancer cells. We examined if mtDNA mutations can be detected in circulating extracellular vesicles (EVs) of PDAC patients and used for discerning between cancer and non-cancer subjects. A greater yield of circulating EVs (~ 1.4 fold; p = 0.002) was obtained in PDAC patients (n = 20) than non-cancer (NC) individuals (n = 10). PDAC-EVs contained a higher quantity of total DNA (~ 5.5 folds; p = 0.0001) than NC-EVs and had greater enrichment of mtDNA (~ 14.02-fold; p = 0.0001). PDAC-EVs also had higher levels of cardiolipin (a mitochondrial inner-membrane phospholipid), suggestive of their mitochondrial origin. All mtDNA mutations in PDAC-EVs were unique and frequency was remarkably higher. Most mtDNA mutations (41.5%) in PDAC-EVs were in the respiratory complex-I (RCI) (ND1-ND6), followed by the RCIII gene (CYTB; 11.2%). Among the non-coding genes, D-Loop and RNR2 exhibited the most mutations (15.2% each). Altogether, our study establishes, for the first time, that mtDNA mutations can be detected in circulating EVs and potentially serve as a tool for reliable PDAC diagnosis.

Noble metal nanoclusters are ultrasmall nanomaterials with tunable properties and huge application potential; however, retaining their enhanced functionality is difficult as they readily lose their properties without stabilization. Here, we demonstrate a facile synthesis of highly photostable silver nanoclusters in a polymer thin film using visible light photoreduction. Furthermore, the different stages of the nanocluster formation are investigated in detail using absorption and fluorescence spectroscopy, fluorescence microscopy, and atomic force microscopy. A cost-effective fabrication of photostable micron-sized fluorescent silver nanocluster barcode is demonstrated in silver-impregnated polymer films using a low-power continuous-wave laser diode. It is shown that a laser power of as low as 0.75 mW is enough to write fluorescent structures, corresponding to the specifications of a commercially available laser pointer. The as-formed nanocluster-containing microstructures can be useful in direct labeling applications such as authenticity marking and fluorescent labeling.
