In this study, a new type of multi-arch tunnel construction method is proposed. This effort is undertaken due to the many disadvantages of the traditional multi-arch tunnel construction method. Furthermore, this method omits the construction of a middle pilot tunnel, and it has the advantages of safety, high efficiency, and being economical. When using the method of continuous arch tunneling without a middle pilot tunnel, the blasting of the first tunnel and the following tunnel has a greater impact on the surrounding rock damage, as well as on the supporting structure of the same cross-section. Therefore, this study uses LS-DYNA finite element software to construct a three-dimensional numerical model. In addition, the perimeter rock damage law and mechanical response characteristics of the supporting structure in the same cross-section of the first tunnel, as well as the following tunnel after blasting without a middle pilot tunnel, are analyzed. At the same time, the results of the study are based on optimizing the blasting program, and these are then applied to the field. Through the results, it is found that, after blasting a continuous arch tunnel without a medial pilot tunnel, the surrounding rock damage in the arch cross-region of the double-arch tunnel (where the arch top and the arch shoulder are more significant) and the effective stress of the supporting structure exceed the strength design value. In addition, the maximum adequate pressure is distributed in the medial diaphragm wall. With the optimized blasting scheme, the range of the peripheral rock damage is reduced by a maximum of 67%, and the effective stress in the supporting structure is reduced by 25.9–64.8%. The research results are of great significance in terms of improving construction safety, economic efficiency, and project quality, as well as in promoting the research and development of new work methods for double-arch tunnels. Based on the macroscopic structure control theory, The slate with a significant bedding plane is a composite rock mass composed of rock blocks containing microscopic defects, joint surface closure elements, and shear deformation elements. Considering the coupling damage effect of water erosion and triaxial compressive load on bedding structure plane, the transversely isotropic damage constitutive model of slate under triaxial compressive load is derived with the dip angle of bedding and confining pressure as the variable. Firstly, based on the statistical theory of continuous damage mechanics and the maximum tensile strain criterion, the transversely isotropic deformation constitutive model of rock block with micro-defects is given; Secondly, based on the phenomenological theory of closed deformation and shear-slip deformation mechanism of layered structural plane under the coupling action of water erosion and triaxial compression load, the calculation formula of axial deformation of layered structural plane under the coupling action is given; Finally, to verify the accuracy of the established constitutive model, triaxial compression tests are carried out to study the influence of dip angle and confining pressure on the macroscopic mechanical properties and mechanism of slate. The results show that: the established triaxial compression damage constitutive model of bedding slate can accurately describe the stress–strain relationship of bedding slate after water environment erosion. With the increase of bedding dip angle, the strength and deformation capacity of the bedding slate first decreases and then increases, showing a U-shaped distribution as a whole. The CO2 distribution in the atmosphere remains unclear for the complexity of the long-range vertical transport process and other influencing factors. In this work, regression analysis was used to verify the accuracy of CO2 concentrations datasets. Geostatistical analyses were used to investigate the spatiotemporal distributions of CO2 at 7 levels from near the surface to the mid-troposphere (0~5 km). Spatial correlation and time series analyses were used to further determine the diffusion characteristics of the CO2 concentration based on the horizontal wind (NCEP R2), which is one of the main driving factors. The results showed that the horizontal, not vertical, diffusion of CO2 becomes increasingly more prominent with the decrease in atmospheric pressure to the mid-troposphere, whereas many regions, such as the Rocky Mountains and Qinghai-Tibet Plateau, have constant low values throughout the year due to the influence of high topography (up to 10.756 ppmv lower than that near the surface). These areas form low CO2 concentration 'windows' keeping letting thermal infrared energy out into space. This study is the first to question the existing view of the closure of the 'greenhouse effect'. Cryptosporidium spp., Giardia duodenalis, and Blastocystis sp. are common intestinal protozoans that infect humans and animals worldwide. A survey that assessed the prevalence, molecular characteristics, and zoonotic potential of these pathogens was conducted on a variety of dogs in Guangzhou, southern China. A total of 651 canine stool samples from household (n = 199), shelter (n = 149), breeding (n = 237), and pet market dogs (n = 66) were collected from eight districts in Guangzhou. Cryptosporidium spp., Giardia duodenalis, and Blastocystis sp. were detected by PCR amplification of the SSU rRNA gene. Giardia duodenalis-positive specimens were further assigned into assemblages using the glutamate dehydrogenase gene. Cryptosporidium spp., G. duodenalis, and Blastocystis sp. were found in 21 (3.2%), 20 (3.1%), and 35 (5.4%) samples, respectively. The overall prevalence of shelter dogs (40.28%, 60/149) was significantly higher than that of household (3.0%, 6/199), breeding (2.1%, 5/237), and pet market dogs (7.5%, 5/66) (χ2 = 154.72, df = 3, P < 0.001). Deworming in the past 12 months had a strong protective effect on the risk of contracting parasite infections (P < 0.001). No significant differences were detected between age or sex groups (P > 0.05). Dog-specific C. canis (n = 19) and zoonotic C. parvum (n = 2) were the only two Cryptosporidium species. Sequence analysis revealed the presence of three G. duodenalis assemblages: dog-specific assemblages D (n = 14) and C (n = 5), and cat-specific F (n = 1). Zoonotic Blastocystis ST3 (n = 28) was the dominant subtype, followed by ST1 (n = 6) and ST10 (n = 1). To our knowledge, this is the first large-scale investigation on the occurrence and molecular characteristics of Blastocystis sp. in dogs in China. Our results indicated that the dogs seemed to play a negligible role as reservoirs for Cryptosporidium spp. and G. duodenalis transmission to humans, but they are potential novel suitable hosts of Blastocystis sp. A strict sentinel surveillance system of dogs should be established to minimise the zoonotic risk of spreading blastocystosis among humans and dogs. [ABSTRACT FROM AUTHOR]

The effect of ultra-narrowband light absorption enhancement is presented by using metamaterials with symmetry-broken square silicon patches (SSPs). The symmetry of the SSP can be broken by introducing a narrow slit deviating from its center. By breaking the symmetry of the SSPs, slit resonance mode with standing wave patterns can be excited, and the locations of the absorption peaks can be well estimated by using the Fabry-Pérot (F-P) cavity model. Although there is no excitation of surface plasmon resonance, ultra-narrowband light absorption can be achieved by minimizing the reflectance through perfect impedance matching and simultaneously eliminating the transmittance by the metallic substrate. Good ultra-narrowband absorption features can be maintained as the parameters of the buffer layer and the SSPs are altered. When this type of symmetry-broken SSPs-based metamaterial is used in refractive-index sensors, it shows excellent sensing properties due to its stable ultra-narrowband absorption enhancement. Similar land-use types usually have similar soil properties, and, most likely, similar microbial communities. Here, we assessed whether land-use types or soil chemical properties are the primary drivers of soil microbial community composition, and how changes in one part of the ecosystem affect another. We applied Ion Torrent sequencing to the bacterial and fungal communities of five different land-use (vegetation) types in the Loess Plateau of China. We found that the overall trend of soil quality was natural forest > plantation > bare land. Dominant bacterial phyla consisted of Proteobacteria (42.35%), Actinobacteria (15.61%), Acidobacteria (13.32%), Bacteroidetes (8.43%), and Gemmatimonadetes (6.0%). The dominant fungi phyla were Ascomycota (40.39%), Basidiomycota (38.01%), and Zygomycota (16.86%). The results of Canonical Correspondence Analysis (CCA) and Redundancy Analysis (RDA) based on land-use types displayed groups according to the land-use types. Furthermore, the bacterial communities were mainly organized by soil organic carbon (SOC). The fungal communities were mainly related to available phosphorus (P). The results suggested that the changes of land use type generated changes in soil chemical properties, controlling the composition of microbial community in the semiarid Loess Plateau region. Our comparative genomic analysis showed that the numbers of plant cell wall (PCW)- and fungal cell wall (FCW)-degradation-associated carbohydrate-active enzymes (CAZymes) in necrotrophic and hemibiotrophic fungi are significantly larger than that in most biotrophic fungi. However, our transcriptional analyses of CAZyme-encoding genes in Melampsora larici-populina, Puccinia graminis and Sclerotinia sclerotiorum showed that many genes encoding PCW- and FCW-degradation-associated CAZymes were significantly up-regulated during the infection of both necrotrophic fungi and biotrophic fungi, indicating an existence of a universal mechanism underlying PCW degradation and FCW reorganization or modification, which are both intimately involved in necrotrophic and biotrophic fungal infection. Furthermore, our results showed that the FCW reorganization or modification was also related to the fungal development. Additionally, our transcriptional analysis of the secretome of S. sclerotiorum showed that many secreted protein-encoding genes were dramatically induced during infection. Among them, a small, cysteine-rich protein SsCVNH was experimentally confirmed to be essential for the virulence and sclerotial development, indicating that the small secreted proteins might also play crucial roles as potential effectors in host-non-specific necrotrophic fungi. Carbon fixation pathway of plants (CFPP) in photosynthesis converts solar energy to biomass, bio-products and biofuel. Intriguingly, a large number of heterotrophic fungi also possess enzymes functionally associated with CFPP, raising the questions about their roles in fungal development and in evolution. Here, we report on the presence of 17 CFPP associated enzymes (ten in Calvin-Benson-Basham reductive pentose phosphate pathway and seven in C4-dicarboxylic acid cycle) in the genome of Sclerotinia sclerotiorum, a heterotrophic phytopathogenic fungus, and only two unique enzymes: ribulose-1, 5-bisphosphate carboxylase-oxygenase (Rubisco) and phosphoribulokinase (PRK) were absent. This data suggested an incomplete CFPP-like pathway (CLP) in fungi. Functional profile analysis demonstrated that the activity of the incomplete CLP was dramatically regulated during different developmental stages of S. sclerotiorum. Subsequent experiments confirmed that many of them were essential to the virulence and/or sclerotial formation. Most of the CLP associated genes are conserved in fungi. Phylogenetic analysis showed that many of them have undergone gene duplication, gene acquisition or loss and functional diversification in evolutionary history. These findings showed an evolutionary links in the carbon fixation processes of autotrophs and heterotrophs and implicated the functions of related genes were in course of continuous change in different organisms in evolution. Virus-vector relationships can be complex and diverse as a result of long-term coevolution. Understanding these interactions is crucial for disease and vector management. Rice stripe virus (RSV) is known to be transovarially transmitted within its vector, Laodelphax striatellus, and causes serious rice stripe disease. In RSV-infected L. striatellus, we found contrasting changes in vector fecundity, physiology, primary endosymbionts (i.e. yeast-like symbionts, YLS) and feeding behavior that can interact to affect the spread of RSV. RSV-infected L. striatellus exhibited a significant decrease in fecundity that could lead a reduction of viruliferous individuals in populations. As a potential response to this loss, RSV infection also significantly shortened nymphal stage duration, which can strengthen RSV vertical circulation in L. striatellus populations and promote RSV spreading by adult migration and dispersal. Down-regulated JHAMT and up-regulated CYP307A1 in the juvenile hormone and ecdysteroid pathways, respectively, were linked to accelerated development. RSV-infected adults were also found to have higher body weight in conjunction with increased YLS abundance. Furthermore, prolonged host plant phloem exposure to salivation by RSV-infected adults should further enhance RSV horizontal transmission. Our study highlights potential strategies of RSV in enhancing its transmission, and provides new insights into the complexity of virus-vector interactions. [ABSTRACT FROM AUTHOR]
