This study introduces a deep learning approach to predicting demographic features from meibography images. A total of 689 meibography images with corresponding subject demographic data were used to develop a deep learning model for predicting gland morphology and demographics from images. The model achieved on average 77%, 76%, and 86% accuracies for predicting Meibomian gland morphological features, subject age, and ethnicity, respectively. The model was further analyzed to identify the most highly weighted gland morphological features used by the algorithm to predict demographic characteristics. The two most important gland morphological features for predicting age were the percent area of gland atrophy and the percentage of ghost glands. The two most important morphological features for predicting ethnicity were gland density and the percentage of ghost glands. The approach offers an alternative to traditional associative modeling to identify relationships between Meibomian gland morphological features and subject demographic characteristics. This deep learning methodology can currently predict demographic features from de-identified meibography images with better than 75% accuracy, a number which is highly likely to improve in future models using larger training datasets, which has significant implications for patient privacy in biomedical imaging.

These results suggest that tear film evaporation initiates a pathway that results in the onset of ocular discomfort and the stimulus to blinking. The presence of a subset of subjects with no or minimal OSC who nevertheless have a short MIBP indicates that evaporative cooling is not the only mechanism responsible for the onset of ocular discomfort.

Asprosin, an adipokine, was recently discovered in 2016. Here, the correlation between asprosin and metabolic-associated fatty liver disease (MAFLD) was examined by quantitatively assessing hepatic steatosis using transient elastography and controlled attenuation parameter (CAP). According to body mass index (BMI), 1276 adult participants were enrolled and categorized into three groups: normal, overweight, and obese. The study collected and evaluated serum asprosin levels, general biochemical indices, liver stiffness measure, and CAP via statistical analysis. In both overweight and obese groups, serum asprosin and CAP were greater than in the normal group (p < 0.01). Each group showed a positive correlation of CAP with asprosin (p < 0.01). The normal group demonstrated a significant and independent positive relationship of CAP with BMI, low-density lipoprotein cholesterol (LDL-C), asprosin, waist circumference (WC), and triglycerides (TG; p < 0.05). CAP showed an independent positive association (p < 0.05) with BMI, WC, asprosin, fasting blood glucose (FBG), and TG in the overweight group, and with high-density lipoprotein cholesterol (HDL-C) showed an independent negative link (p < 0.01). CAP showed an independent positive relationship (p < 0.05) with BMI, WC, asprosin, TG, LDL-C, FBG, glycated hemoglobin A1c (HbA1c), and alanine transferase in the obese group. CAP also showed an independent positive link (p < 0.01) with BMI, WC, asprosin, TG, LDL-C, and FBG in all participants while independently and negatively correlated (p < 0.01) with HDL-C. Acute myocardial infarction (AMI) commonly precedes ventricular remodeling, heart failure. Few dynamic molecular signatures have gained widespread acceptance in mainstream clinical testing despite the discovery of many potential candidates. These unmet needs with respect to biomarker and drug discovery of AMI necessitate a prioritization. We enrolled patients with AMI aged between 30 and 70. RNA-seq analysis was performed on the peripheral blood mononuclear cells collected from the patients at three time points: 1 day, 7 days, and 3 months after AMI. PLC/LC-MS analysis was conducted on the peripheral blood plasma collected from these patients at the same three time points. Differential genes and metabolites between groups were screened by bio-informatics methods to understand the dynamic changes of AMI in different periods. We obtained 15 transcriptional and 95 metabolite expression profiles at three time points after AMI through high-throughput sequencing. AMI-1d: enrichment analysis revealed the biological features of 1 day after AMI primarily included acute inflammatory response, elevated glycerophospholipid metabolism, and decreased protein synthesis capacity. Phosphatidylcholine (PC) and phosphatidylethanolamine (PE) might stand promising biomarkers to differentiate post-AMI stage. Anti-inflammatory therapy during the acute phase is an important direction for preventing related pathology. AMI-7d: the biological features of this stage primarily involved the initiation of cardiac fibrosis response and activation of platelet adhesion pathways. Accompanied by upregulated TGF-beta signaling pathway and ECM receptor interaction, GP5 help assess platelet activation, a potential therapeutic target to improve haemostasis. AMI-3m: the biological features of 3 months after AMI primarily showed a vascular regeneration response with VEGF signaling pathway, NOS3 and SHC2 widely activated, which holds promise for providing new therapeutic approaches for AMI. Our analysis highlights transcriptional and metabolomics signatures at different time points after MI, which deepens our understanding of the dynamic biological responses and associated molecular mechanisms that occur during cardiac repair., (© 2024. In the field of the Internet of Things, image acquisition equipment is the very important equipment, which will generate lots of invalid data during real-time monitoring. Analyzing the data collected directly from the terminal by edge calculation, we can remove invalid frames and improve the accuracy of system detection. SSD algorithm has a relatively light and fast detection speed. However, SSD algorithm do not take full advantage of both shallow and deep information of data. So a multiscale feature fusion attention mechanism structure based on SSD algorithm has been proposed in this paper, which combines multiscale feature fusion and attention mechanism. The adjacent feature layers for each detection layer are fused to improve the feature information expression ability. Then, the attention mechanism is added to increase the attention of the feature map channels. In previous studies, the numerical modeling and analyzing methods onto industrial or vehicle airbags dynamics were revealed to have high accuracy regarding their actual dynamic properties, but there are scarcely airbag stiffness modeling and comfortableness investigations of nursing cushion or mattress airbags. This study constructs a numerical model illustrating the association between the stiffness property and the internal gas mass of the wedge-shaped airbag of nursing appliance, and then the airbag stiffness variation discipline is described based on various inflation volumes. To start with, based on an averaged pressure prerequisite, a dynamic simulation model of the wedge-shaped airbag is established by the fluid cavity approach. For this modeling, the elastic mechanical behaviors of airbag material are determined according to a material constitutive model built by the quasi-static uniaxial tensile test. Besides, verification experiments clarify that the presented modeling method is accurate for airbag stiffness behavior prediction, and then can be effectively applied into design and optimization phases of wedge-shaped airbags. Ultimately, based on the simulation and experimental results, it is found that the wedge-shaped airbag stiffness exhibits a three stages characteristic evolution with the gas mass increase. Then the mathematical relationship between the airbag stiffness and gas mass is obtained by numerical fitting, which provides a vital basis for structural optimization and differentiated control of nursing equipment airbags., (© 2023. The recent release of large-scale healthcare datasets has greatly propelled the research of data-driven deep learning models for healthcare applications. However, due to the nature of such deep black-boxed models, concerns about interpretability, fairness, and biases in healthcare scenarios where human lives are at stake call for a careful and thorough examination of both datasets and models. In this work, we focus on MIMIC-IV (Medical Information Mart for Intensive Care, version IV), the largest publicly available healthcare dataset, and conduct comprehensive analyses of interpretability as well as dataset representation bias and prediction fairness of deep learning models for in-hospital mortality prediction. First, we analyze the interpretability of deep learning mortality prediction models and observe that (1) the best-performing interpretability method successfully identifies critical features for mortality prediction on various prediction models as well as recognizing new important features that domain knowledge does not consider; (2) prediction models rely on demographic features, raising concerns in fairness. Therefore, we then evaluate the fairness of models and do observe the unfairness: (1) there exists disparate treatment in prescribing mechanical ventilation among patient groups across ethnicity, gender and age; (2) models often rely on racial attributes unequally across subgroups to generate their predictions. We further draw concrete connections between interpretability methods and fairness metrics by showing how feature importance from interpretability methods can be beneficial in quantifying potential disparities in mortality predictors. Our analysis demonstrates that the prediction performance is not the only factor to consider when evaluating models for healthcare applications, since high prediction performance might be the result of unfair utilization of demographic features. Our findings suggest that future research in AI models for healthcare applications can benefit from utilizing the analysis workflow of interpretability and fairness as well as verifying if models achieve superior performance at the cost of introducing bias., (© 2022. Understanding molecular mechanisms of intervertebral disc degeneration (IDD) and providing a novel target for the treatment of IDD have important implications. We sought to explore a new promising gene target for the treatment of IDD. This study integrated 19,678 genes of 38 IDD patients from two gene datasets. Differentially Expressed Genes (DEGs) of annulus fibrosus were analyzed in groups with mild disc degeneration (MDD) and severe disc degeneration (SDD). We screened the hub gene through biological information technology (bioinformatic) methods. Then, we further validated the hub gene using annulus fibrosus and nucleus pulposus tissues from 12 patients with qRT-PCR. In addition, we explored its underlying molecular mechanism with GO, KEGG and GSEA. Through multiple screening bioinformatics methods, the hub gene CD63 was identified. The qRT-PCR explored that CD63 decreased significantly in SDD group compared to that in MDD group (P < 0.001). The GO, KEGG and GSEA of CD63 explored significant enrichment of the molecular features (P < 0.001), including the cellular component (Extracellular matrix, P < 0.001), the molecular function (collagen binding, P < 0.001), the biological processes (protein targeting, collagen fibril organization and platelet degranulation, P < 0.001) and the signaling pathways. Plasmonic metasurfaces, representing arrays of gap-surface plasmon (GSP) resonators and consisting of arrays of metal nanobricks atop thin dielectric layers supported by thick metal films, constitute an important subclass of optical metasurfaces operating in reflection and enabling the realization of numerous, diverse and multiple, functionalities. The available phase variation range is however limited to being [Formula: see text], a circumstance that complicates the metasurface design for functionalities requiring slowly varying phases over the whole range of [Formula: see text], e.g., in holographic applications. The available phase range also determines the wavelength bandwidth of metasurfaces operating with linearly polarized fields due to the propagation (size-dependent) nature of the reflection phase. We suggest an approach to extend the phase range and bandwidth limitations in the GSP-based metasurfaces by incorporating a pair of detuned GSP resonators into a metasurface elementary unit cell. With detailed simulations related to those for conventional single-resonator metasurfaces and proof-of-concept experiments, we demonstrate that the detuned-resonator GSP metasurfaces designed for beam steering at [Formula: see text] wavelength exhibit the extended reflection phase and operation bandwidth. We believe that the considered detuned-resonator GSP metasurfaces can advantageously be exploited in applications requiring the design of arbitrary phase gradients and/or broadband operation with linearly polarized fields.

A rising proportion of the world population suffers from food-related allergies, including incompatibilities to apples. Although several allergenic proteins have been found in apples, the most important proteins that cause allergic reactions to apples in Central-Northern Europe, and North America are the Mal d 1 proteins, which are homologues of the birch pollen allergen Bet v 1. As the demand for hypoallergenic fruits is constantly increasing, we selected apple genotypes with a low total content of Mal d 1 by enzyme-linked immunosorbent assay analysis from segregating populations and tested the tolerability of these fruits through a human provocation study. This tiered approach, which exploited the natural diversity of apples, led to the identification of fruits, which were tolerated by allergic patients. In addition, we found a significant correlation (coefficient >0.76) between the total Mal d 1 content and flavan-3-ol amount and show that the isoform composition of the Mal d 1 proteins, which was determined by LC-MS/MS has a decisive effect on the tolerability of apple genotypes. The approach presented can be applied to other types of fruit and to other allergenic proteins. Therefore, the strategy can be used to reduce the allergen content of other plant foods, thereby improving food safety for allergy subjects.

Fengyun-4A is the new generation of Chinese geostationary meteorological satellites. Land surface albedo, land surface emissivity and land surface temperature are key states for land surface modelling. In this paper, the land surface albedo, land surface emissivity and land surface temperature data from Fengyun-4A were assimilated into the Integrated Urban land Model. The Fengyun-4A data are one of the data sources for the land data assimilation system which devoted to produce the high spatial and temporal resolution, multiple parameters near real-time land data sets. The Moderate-Resolution Imaging Spectroradiometer (MODIS) LSA and LSE data, the Institute of Atmospheric Physics, China Academy of Sciences (IAP) 325 m tower observation data and the observed 5 cm and 10 cm soil temperature data in more than 100 sites are used for validation. The results indicate the MODIS land surface albedo is much smaller than the Fengyun-4A and is superior to the Fengyun-4A for the Institute of Atmospheric Physics, China Academy of Sciences 325 m tower site. The Moderate-Resolution Imaging Spectroradiometer land surface emissivity is smaller than the Fengyun-4A in barren land surface and the differences is relatively small for other land use and land cover categories. In most regions of the research area, the Fengyun-4A land surface albedo and land surface emissivity are larger than those of the simulations. After the land surface albedo assimilation, in most regions the simulated net radiation was decreased. After the land surface emissivity assimilation, in most regions the simulated net radiation was increased. After the land surface temperature assimilation, the biases of the land surface temperature were decreased apparently; the biases of the daily average 5 cm and 10 cm soil temperature were decreased.

We show that through the wave energy conserved and reversible process of coherent interactions of scalar waves in a multi-channel system joint by a common junction, it is possible to generate outgoing waves only in certain channels by controlling the incoming waves. We refer to such processes as coherent perfect channeling (CPC). As two particular examples, we report experimental and theoretical investigations of CPC with two incoming coherent waves in three and four-channel waveguides that are completely channeled into one or two other waveguides mediated by a deep subwavelength dimension scatterer at the common junction. Two such scatterers are discovered, one confirmed by experiments and the other predicted by theory, and their scattering matrices are constructed. Scatterers with other CPC scattering matrices are explored, and preliminary investigations of their properties are conducted. The scattering matrix formulism makes it possible to extend the domain of CPC to other scalar waves, such as electromagnetic waves and quantum wavefunctions.

Synchronous gastric tumors that consist of both gastrointestinal stromal tumor (GIST) and adenocarcinoma are rare. We studied the clinicopathological and molecular characteristics of six cases containing both gastric adenocarcinoma and GIST. By means of immunohistochemical analysis, all GIST cells expressed CD117, CD34 and Dog1 in all six synchronous gastric adenocarcinomas with GIST, and in GIST alone. Sequencing analysis demonstrated that exon 11 c-kit mutations were present in two of six synchronous tumors and four of five GISTs. One of the two exon 11 c-kit mutations in synchronous adenocarcinomas with GISTs was an uncommon mutation of CTT > CCA at amino acid 576, and the other was a GTT deletion at amino acid 560. The mutation was a homozygous A > G mutation in exon 12 (amino acid 567) of PDGFR-α. We concluded that the exon 11 mutations were the most important in both cases of synchronous gastric adenocarcinoma with GIST and GIST alone. The mutation rate was higher in GIST alone than in synchronous adenocarcinoma with GIST.

Elevated ground-level ozone (O 3 ), which is an important aspect of air quality related to public health, has been causing increasing concern. This study investigated the spatiotemporal distribution of ground-level O 3 concentrations in China using a dataset from the Chinese national air quality monitoring network during 2013-2015. This research analyzed the diurnal, monthly and yearly variation of O 3 concentrations in both sparsely and densely populated regions. In particular, 6 major Chinese cities were selected to allow a discussion of variations in O 3 levels in detail, Beijing, Chengdu, Guangzhou, Lanzhou, Shanghai, and Urumchi, located on both sides of the Heihe-Tengchong line. Data showed that the nationwide 3-year MDA8 of ground-level O 3 was 80.26 μg/m 3 . Ground-level O 3 concentrations exhibited monthly variability peaking in summer and reaching the lowest levels in winter. The diurnal cycle reached a minimum in morning and peaked in the afternoon. Yearly average O 3 MDA8 concentrations in Beijing, Chengdu, Lanzhou, and Shanghai in 2015 increased 12%, 25%, 34%, 22%, respectively, when compared with those in 2013. Compared with World Health Organization O 3 guidelines, Beijing, Chengdu, Guangzhou, and Shanghai suffered O 3 pollution in excess of the 8-hour O 3 standard for more than 30% of the days in 2013 to 2015.

We report the experimental realization of acoustic coherent perfect absorption (CPA) of four symmetric scatterers of very different structures. The only conditions necessary for these scatterers to exhibit CPA are that both the reflection and transmission amplitudes of the scatterers are 0.5 under one incident wave, and there are two collinear and counter-propagating incident waves with appropriate relative amplitude and phase. Nearly 1000 times in the modulation of output power has been demonstrated by changing the relative phase of the incident waves over 180°. We further demonstrate that these scatterers could potentially be sensitive devices to detect the small differences between two nearly equal incident waves. A 27% change in the strength of the scattering wave has been demonstrated for every degree of phase deviation from the optimum condition between the incident waves.

Surface wettability is of importance for electrochemical reactions. Herein, its role in electrochemical hydrogen evolution reactions is investigated using light-sensitive nanotubular TiO 2 supported Pt as hydrogen evolution electrodes (HEEs). The HEEs are fabricated by photocatalytic deposition of Pt particles on TiO 2 nanotubes followed by hydrophobization with vaporized octadecyltrimethoxysilane (OTS) molecules. The surface wettability of HEEs is subsequently regulated in situ from hydrophobicity to hydrophilicity by photocatalytic decomposition of OTS molecules using ultraviolet light. It is found that hydrophilic HEEs demonstrate a larger electrochemical active area of Pt and a lower adhesion force to a gas bubble when compared with hydrophobic ones. The former allows more protons to react on the electrode surface at small overpotential so that a larger current is produced. The latter leads to a quick release of hydrogen gas bubbles from the electrode surface at large overpotential, which ensures the contact between catalysts and electrolyte. These two characteristics make hydrophilic HEEs generate a much high current density for HERs. Dual-phase (Fe 83 Ga 17 ) 100-x Tb x alloys with 0 ≤ x ≤ 1 were synthesized by arc melting and homogenization treatment. The microstructures and the corresponding mechanical properties were systematically investigated. The chemical composition of the body centered cubic matrix is Fe 83 Ga 17 . The monoclinic second phase was composed of meltable precipitates with approximate composition Fe 57 Ga 33 Tb 10 . The nano-hardness of matrix and precipitates were 2.55 ± 0.17 GPa and 6.81 ± 1.03 GPa, respectively. Both the ultimate tensile strength (UTS) and fracture strain (ε) of the alloys were improved by the precipitates for x ≤ 0.2 alloys, but the strain decreases significantly at higher values of x. As potential structural-functional materials, the best mechanical properties obtained were a UTS of 595 ± 10 MPa and an ε of 3.5 ± 0.1%, four-fold and seven-fold improvements compared with the un-doped alloy. The mechanism for these anomalous changes of mechanical properties was attributed to the dispersed precipitates and semi-coherent interfaces, which serve as strong obstacles to dislocation motion and reduce the stress concentration at the grain boundaries. A sizeable improvement of magnetostriction induced by the precipitates in the range 0 ≤ x ≤ 0.2 was discovered and an optimal value of 150 ± 5 ppm is found, over three times higher than that of the un-doped alloy.

Much greater surface-to-volume ratio of hierarchical nanostructures renders them attract considerable interest as prototypical gas sensors. In this work, a novel resistive gas sensor based on TiO2/Ag0.35V2O5 branched nanoheterostructures is fabricated by a facile one-step synthetic process and the ethanol sensing performance of this device is characterized systematically, which shows faster response/recovery behavior, better selectivity, and higher sensitivity of about 9 times as compared to the pure TiO2 nanofibers. The enhanced sensitivity of the TiO2/Ag0.35V2O5 branched nanoheterostructures should be attributed to the extraordinary branched hierarchical structures and TiO2/Ag0.35V2O5 heterojunctions, which can eventually result in an obvious change of resistance upon ethanol exposure. This study not only indicates the gas sensing mechanism for performance enhancement of branched nanoheterostructures, but also proposes a rational approach to design nanostructure based chemical sensors with desirable performance.

Inherited aortopathy, which is characterized by a high risk of fatal aortic aneurysms/dissections, can occur secondarily to several syndromes. To identify genetic mutations and help make a precise diagnosis, we designed a gene panel containing 15 genes responsible for inherited aortopathy and tested 248 probands with aortic disease or Marfan syndrome. The results showed that 92 individuals (37.1%) tested positive for a (likely) pathogenic mutation, most of which were FBN1 mutations. We found that patients with a FBN1 truncating or splicing mutation were more prone to developing severe aortic disease or valvular disease. To date, this is the largest reported cohort of Chinese patients with aortic disease who have undergone genetic testing. Therefore, it can serve as a considerable dataset of next generation sequencing data analysis of Chinese population with inherited aortopathy. Additionally, according to the accumulated data, we optimized the analysis pipeline by adding quality control steps and lowering the false positive rate.

Prevalence of atopic dermatitis (AD) is increasing worldwide. Up to date, there has been no face-to-face nation-wide study in China. We aim to explore the prevalence of clinical diagnosed AD in children aged 1-7 ys in China. Twelve metropolises were chosen from different areas of China. In each region, we selected 4-10 kindergartens and 2-5 vaccination clinics randomly. A complete history-taking and skin examination were performed by dermatologists. The definite diagnosis of AD and the severity were determined by two or three dermatologists. All criteria concerned in UK diagnosis criteria, characteristic presentation of AD and atypical manifestations were recorded in detail. A total of 13998 children from 84 kindergartens and 40 vaccination clinics were included. The prevalence of AD was 12.94% by clinical diagnosis of dermatologists overall, with 74.6% of mild AD. Comparatively, prevalence of AD based on UK diagnostic criteria was 4.76%. This is the first face-to-face nation-wide study in Chinese children aged 1-7 ys, revealing that the prevalence of AD in children is closer to that of wealthier nations.

White spot syndrome virus (WSSV) is a lethal pathogen of shrimp and many other crustaceans, including crayfish. However, the molecular mechanism underlying its cellular entry remains elusive due to the lack of shrimp cell lines for viral propagation. Crayfish hematopoietic tissue (Hpt) cell culture was recently established as a good model for WSSV infection study. Here, we showed that multiple endocytic routes, including clathrin-mediated endocytosis (CME), macropinocytosis and caveolae-mediated endocytosis, were indispensably employed for the viral entry into Hpt cell of the crayfish Cherax quadricarinatus. Intriguingly, cellular autophagic activity was positively correlated with efficient viral entry, in which a key autophagy-related protein, γ-aminobutyric acid receptor-associated protein (Cq-GABARAP), that not only localized but also co-localized with WSSV on the Hpt cell membrane, strongly facilitated WSSV entry by binding to the viral envelope VP28 in a CME-dependent manner that was negatively regulated by Cq-Rac1. Furthermore, cytoskeletal components, including Cq-β-tubulin and Cq-β-actin, bound to both recombinant rCq-GABARAP and WSSV envelope proteins, which likely led to viral entry promotion via cooperation with rCq-GABARAP. Even under conditions that promoted viral entry, rCq-GABARAP significantly reduced viral replication at an early stage of infection, which was probably caused by the formation of WSSV aggregates in the cytoplasm.

IL-6 (interleukin 6) plays an important role in the development and growth of hepatocellular carcinoma (HCC) via both classic signaling and trans-signaling pathways. Soluble gp130 (sgp130) is known to be a natural inhibitor of the trans-signaling pathway. In the present study, our goal was to investigate whether recombinant sgp130 could suppress the initiation and progression of HCC in mouse models. Our results demonstrate that sgp130 induced an apoptosis of HepG2 cells and inhibited the clonogenicity of HepG2 in vitro. Moreover, the IL-6 trans-signaling pathway is significantly suppressed by sgp130 as reflected by the decrease in the level of STAT3 phosphorylation and other inflammatory factors both in vitro and in vivo. In the DEN-induced HCC mouse model, intravenous injection of sgp130 attenuated hepatic fibrosis at 16 weeks and reduced the initiation and progression of primary HCC at 36 weeks. Furthermore, our results also demonstrate that intravenous administration of sgp130 significantly suppressed the growth and metastasis of xenograft human HCC in NOD/SCID mice.

Urban heat island (UHI) is one of the most focuses in urban climate study. The parameterization of the anthropogenic heat (AH) is crucial important in UHI study, but universal method to parameterize the spatial pattern of the AH is lacking now. This paper uses the NOAA DMSP/OLS nighttime light data to parameterize the spatial pattern of the AH. Two experiments were designed and performed to quantify the influences of the AH to land surface temperature (LST) in eastern China and 24 big cities. The annual mean heating caused by AH is up to 1 K in eastern China. This paper uses the relative LST differences rather than the absolute LST differences between the control run and contrast run of common land model (CoLM) to find the drivers. The heating effect of the anthropogenic footprint has less influence on relatively warm and wet cities.

Spontaneous imbibition happens in many natural and chemical engineering processes in which the mean advancing front usually follows Lucas-Washburn's law. However it has been found that the scaling law does not apply in many cases. There have been few criteria to determine under what conditions the Washburn law works. The effect of gravity on spontaneous imbibition in porous media was investigated both theoretically and experimentally. The mathematical model derived analytically was used to calculate the imbibition rates in porous media with different permeabilities. The results demonstrated that the effect of gravity on spontaneous imbibition was governed by the hydraulic conductivity of the porous media (permeability of the imbibition systems). The criteria for applying the Lucas-Washburn law have been proposed. The effect of gravity becomes more apparent with the increase in permeability or with the decrease in CGR number (the ratio of capillary pressure to gravity forces) and may be ignored when the CGR number is less than a specific value N(*)(cg) ≅ 3.0. The effect of gravity on imbibition in porous media can be modeled theoretically. It may not be necessary to conduct spontaneous imbibition experiments horizontally in order to exclude the effect of gravity, as has been done previously.

Radioresistance has been an important factor in restricting efficacy of radiotherapy for non-small cell lung cancer (NSCLC) patients and new approaches to inhibit cancer growth and sensitize irradiation were warranted. Despite the important role of ubiquitin/proteasome system (UPS) during cancer progression and treatment, the expression and biological role of ubiquitin (Ub) in human NSCLC has not been characterized. In this study, we found that ubiquitin was significantly overexpressed in 75 NSCLC tissues, compared to their respective benign tissues by immunohistochemistry (P < 0.0001). Knock-down of ubiquitin by mixed shRNAs targeting its coding genes ubiquitin B (UBB) and ubiquitin C (UBC) suppressed the growth and increased the radiosensitivity in NSCLC H1299 cells. Apoptosis and γ H2AX foci induced by X-ray irradiation were enhanced by knock-down of ubiquitin. Western blot and immunostaining showed that knock-down of ubiquitin decreased the expression and translocation of NF-κB to the nucleus by reduced phospho-IκBα after irradiation. Suppression of ubiquitin decreased the proliferation and radioresistance of H1299 transplanted xenografts in nude mice by promoting apoptosis. Taken together, our results demonstrate the critical role of ubiquitin in NSCLC proliferation and radiosensitivity. Targeting ubiquitin may serve as a potentially important and novel approach for NSCLC prevention and therapy.

CYP3A4 metabolizes more than 50% of the drugs on the market. The large inter-individual differences of CYP3A4 expression may contribute to the variability of human drug responses. Post-transcriptional regulation of CYP3A4 is poorly understood, whereas transcriptional regulation has been studied much more thoroughly. In this study, we used multiple software programs to predict miRNAs that might bind to CYP3A4 and identified 112 potentially functional miRNAs. Then a luciferase reporter system was used to assess the effect of the overexpression of each potentially functional miRNA in HEK 293T cells. Fourteen miRNAs that significantly decreased reporter activity were measured in human liver samples (N = 27) as candidate miRNAs. To establish a more effective way to analyze in vivo data for miRNA candidates, the relationship between functional miRNA and target mRNA was modeled mathematically. Taking advantage of this model, we found that hsa-miR-577, hsa-miR-1, hsa-miR-532-3p and hsa-miR-627 could significantly downregulate the translation efficiency of CYP3A4 mRNA in liver. This study used in silico, in vitro and in vivo methods to progressively screen functional miRNAs for CYP3A4 and to enhance our understanding of molecular events underlying the large inter-individual differences of CYP3A4 expression in human populations.
