Myeloproliferative neoplasms (MPNs) are a type of chronic hematological tumor accompanied by bone marrow failure or leukemia. A nanoparticle-assisted digital loop-mediated isothermal amplification (nano-dLAMP) platform was established for the analysis of MPNs in this study. Microarray chips with four physical partitions were fabricated for the simultaneous detection of calreticulin type 1 (CALR-1), calreticulin type 2 (CALR-2), and janus kinase 2 V617F (JAK2 V617F) mutations and an internal reference gene. Polymerase chain reaction (PCR) additives and nanoparticles were used to make the traditional loop mediated isothermal amplification (LAMP) suitable for nanoliter-scale amplification. The results suggested that nanoparticles could improve the amplification performance of nanoliter LAMP. Quantitative detection of the main MPN molecular markers could be performed simultaneously in one four-partition microarray chip within 1 h. The detection sensitivity values for CALR-1, CALR-2, and JAK2 V617F were 0.5 %, 0.1 %, and 0.5 % mutation burden, respectively. The agreement between the developed platform and the commercial Quantstudio 3D was high at 99 % (280/281). Simultaneous analyzing multiple protein and nucleic acid biomarkers in one clinical sample can provide clinically valuable results for the diagnosis, monitoring, and management of diseases. However, current diagnostic platforms primarily rely on independent detection of proteins/nucleic acids, which is time-consuming and labor-intensive. Here, we develop a Highly-Parallelized and Low-Sample-Size (HPLSS) chip for simultaneously detecting protein and nucleic acid biomarker in hepatocellular carcinoma (HCC). We assess the limit of detections for five different markers (0.14 ng mL-1 for alpha fetoprotein (AFP), 0.09 ng mL-1 for protein induced by vitamin K absence or antagonist-II (PIVKA II), 7.94 copies μL-1 for microRNA-21, 4.88 copies μL-1 for microRNA-122, and 5.83 copies μL-1 for microRNA-223). We also evaluate the performance using clinical samples and compared it with commercial kits. Our method exhibited a small sample volume required (∼20 μL), multiplexed ability and adjustable throughput (five targets in each of six samples in one assay). Importantly, the combined evaluation of multiple biomarkers of different dimensions can improve the specificity and sensitivity of diagnosis. A fiber-optic humidity sensor based on excessively tilted fiber grating (Ex-TFG) wrapped with tin dioxide (SnO 2)/polyvinyl alcohol (PVA) hybrid nanofibers is presented. Relative humidity (RH) rise causes the changes both in refractive index (RI) and geometry size of the film. The transmitted spectrum of the Ex-TFG changes as a result, owing to the strong interaction of the excited forward propagating cladding modes to the surface coating. Experimental results demonstrate the maximum intensity sensitivity of 0.43 dB/%RH in RH range of 35%−75%. Compared with pure PVA film coated ones, the RH sensitivity of the SnO 2 /PVA wrapped grating is vastly enhanced by one order of magnitude. The reticular morphology of electrospun SnO 2 /PVA film around the Ex-TFG promises the unimpeded permeation of water molecules, and then the sensor exhibits an ultrafast response of ∼67 ms and recovery times of ∼83 ms, by monitoring the human breathing with different rhythms. Lastly, the negligible spectral deformation to temperature perturbations verifies the temperature de-correlation of the designed RH sensor. Abstract Blood clotting time monitoring is of great importance in predicting the risk of haemorrhage and cardiovascular diseases. In this study, a new P-Lamb wave sensor was developed to measure activated partial thromboplastic time (aPTT) with enhanced sensitivity. The sensor utilizes a piezoelectric and hydrophobic material, Parylene-C, as a waveguide for the P-Lamb wave sensor in order to monitor the aPTT in whole blood samples. With a smooth surface, the P-Lamb wave sensor reduced energy loss and prevented leaky modes, with a decrease of 33% ± 1.25% in the surface roughness and an increase of 2.75% ± 0.15% in the signal. The experimental results demonstrated that, compared to a quartz crystal microbalance (QCM), a P-Lamb sensor with high sensitivity (the frequency shift of a P-Lamb sensor is approximately 200 times greater than that of a QCM) improved stability and enhanced repeatability. Parylene-C worked as a phonon coupling layer, which improved the stability and sensitivity of the Lamb sensor. The relative standards deviation (RSD) of the aPTT measurement was 2.11%, which was 1.22 times lower than reported values. The P-Lamb wave sensor was also tested for its sensitivity to different concentrations of heparin and was found to exhibit an increase in coagulation time with an increasing heparin concentration. Compared to the SYSMEX CS 5100 haematology analyser, the clinical coefficient index (R2) was 0.99467. Considering its compact size, low cost, and mass production of the chip unit, the developed P-Lamb wave sensor is a promising device for point-of-care diagnosis of haemostasis and personal health monitoring. Development of a simple, sensitive and specific method for HIV-1 assay is demanded. In some emergency situations, a portable reverse transcription loop-mediated isothermal amplification (RT-LAMP) platform is necessary. A novel hydrogen ion-selective RT-LAMP (H+-RT-LAMP) sensor was developed for highly sensitive HIV-1 using hydrogen ion concentration as a signal producing compound. The H+-RT-LAMP sensor was based on screen-printed with carbon-doped graphene conductive layer, carbon nanotubes (CNTs) solid electrolyte layer, and reference electrode KCl functional layer, which provided superior electrical signals, high sensitivity and stability for HIV-1 detection. The rate of hydrogen ion production was related to the viral load (VL) during the amplification of HIV-1. The sensitive HIV-1 RNA detection range was achieved from 0 copies/μL to 105 copies/μL. The proposed H+-RT-LAMP sensor was applied to detect HIV-1 RNA in real human serum. 57 samples were detected and satisfactory results were obtained. Compared with commercial instruments, the detection results of H+-RT-LAMP sensor demonstrated a perfect concordance rate of 96.5 %, and the limit of detection (LOD) for the clinical samples was 10 copies per tube. Time to get the result is from 10 min to 25 min. Therefore, the proposed H+-RT-LAMP sensor was particularly suitable as a point-of-care device for the on-site detection of HIV-1.
