Objective To examine the changes in the expression levels of Pleckstrin homology domain family A member 4（PLEKHA4) in glioma tissues and its influencing factors. Methods Fifty-two cases of glioma tissues from surgical resection and 52 cases of their corresponding paraneoplastic tissues were selected, of which 21 cases were WHO grade Ⅰ-Ⅱ and 31 cases were grade Ⅲ-Ⅳ, and 23 cases were wild-type and 29 cases were mutant type of p53 typing. Immunohistochemistry and protein immunoblotting were used to detect the expression level of PLEKHA4 in the paracancerous tissues and glioma tissues of patients with glioma. We observed the clinical characteristics and prognosis of glioma patients with different expression levels of PLEKHA4, and analyzed the factors affecting the expression level of PLEKHA4 in glioma tissues. Results PLEKHA4 was positively expressed in glioma tissues of 52 patients, and the sites of expression were mainly in the cytoplasm and cell membrane of glioma tissues； no positive expression of PLEKHA4 was seen in the paracancerous tissues. The relative expression levels of PLEKHA4 in the paracancerous tissues, grade Ⅰ-Ⅱ glioma, and grade Ⅲ-Ⅳ glioma were 0. 419 ± 0. 101，0. 968 ± 0. 061, and 1. 366 ± 0. 096, respectively. The relative expression of PLEKHA4 in the glioma tissues was higher than that in paracancerous tissues, and the relative expression of PLEKHA4 was higher in glioma tissues of grade Ⅲ-Ⅳ than that in glioma tissues of grade Ⅰ-Ⅱ, respectively (both P<0. 05) . Significant differences were found in the WHO classification and p53 typing of patients with different PLEKHA4 expression levels in glioma tissues (both P<0. 05) . Logistic regression analysis showed that WHO classification and p53 typing of gliomas were the independent influencing factors for PLEKHA4 expression (both P<0. 05) . At 2-year postoperative follow-up, the survival rate of patients with high PLEKHA4 expression was 28. 6%, the survival rate of patients with low PLEKHA4 expression was 70. 8%, and statistically significant difference was found in the survival rate between them (P<0. 05) . Conclusion PLEKHA4 was highly expressed in glioma tissues, WHO typing and p53 typing of patients were the influencing factors of PLEKHA4 expression level in glioma tissues, and the PLEKHA4 expression level was related to clinical prognosis of patients. 目的比较后路椎弓根螺钉内固定联合颗粒骨打压椎间植骨与自体髂骨块+颗粒骨椎间植骨治疗退行 性下腰椎不稳的临床疗效。方法选取行经后路减压+椎间植骨+椎弓根螺钉内固定治疗退行性下腰椎不稳患 者40例，按融合方式不同分为两组。A组行椎间颗粒骨打压植骨18例，B组行自体髂骨(2枚椎间融合器大小)+ 颗粒骨填充打压22例;观察两组患者术前、术后1周及术后12个月JOA评分、椎间隙高度，比较两组临床疗效。 结果所有患者术后腰背部疼痛及下肢根性症状明显改善，A、B组患者术后1周及术后12个月JOA评分较术前 差异有统计学意义(P均<0.05)，两组间比较差异无统计学意义(P>0. 05)。术后12个月B组的融合率为 93.5%，A组为90.9%，差异无统计学意义(P >0.05);术后12个月椎间隙高度B组高于A组(P <0.05);A、B两 组术后1周和术后12个月椎间隙高度均明显高于术前(P均<0.05)。结论椎弓根螺钉内固定联合自体髂骨块 +颗粒骨打压充填椎间植骨融合治疗退行性下腰椎不稳在术后椎间隙高度维持方面较单纯椎间打压植骨更有 优势。 [ABSTRACT FROM AUTHOR]

目的 探讨磷脂酰肌醇激酶3(PIK3C3)小干扰RNA(siRNA)-pool对大鼠PC12细胞PIK3C3及P62/SQSTM1表达的影响.方法 将处于对数生长期的PC12细胞随机分为正常组、载体组、阴性对照组及siRNA 25、50、100 nmol/L组.正常组常规培养,其余组均给予载体Lipofectamine2000转染;在转染的同时,阴性对照组加入200 pmol/L FAM-siRNA;siRNA 25、50、100 nmol/L组采用siRNA-pool法分别加入含50、100、200 nmol/L siRNA.各组转染24 h时,采用MTT法检测细胞存活率,采用RT-PCR法检测PIK3C3 mRNA相对表达量;转染48 h时,采用免疫细胞化学法检测P62/SQSTM1表达.结果 转染24 h时,正常组、载体组、阴性对照组及siRNA 25、50、100 nmol/L组细胞存活率分别为100%、(98.4±1.7)%、(99.7±2.4)%、(104.3±2.8)%、(101.6±5.1)%、(96.9±1.9)%,组间比较P均>0.05;PIK3C3 mRNA相对表达量分别为1、1.01±0.11、0.93±0.07、0.85±0.06、0.78±0.03、0.65±0.04,siRNA 50 nmol/L组PIK3C3 mRNA相对表达量均低于正常组及载体组,siRNA 100 nmol/L组均低于其他各组,组间比较P<0.05或<0.01. 正常组及siRNA 100 nmol/L组P62/SQSTM1表达光密度值分别为0.038±0.001、0.054±0.004,组间比较P<0.01.结论 PIK3C3 siRNA-pool可抑制PC12细胞PIK3C3表达,促进P62/SQSTM1蓄积 [ABSTRACT FROM AUTHOR]

目的：对比观察不同剂量米力农静脉泵入治疗肺动脉高压（ PAH）的有效性及安全性。方法 PAH患者120例，根据米力农用药剂量分为A、B、C三组，各40例。 A、B、C组在常规用地高辛、安体舒通、卡托普利等治疗基础上，分别以0．25、0．5、0．8μg／（kg· min）的米力农微量泵持续泵入，连续5 d。米力农治疗5 d后进行疗效评价。记录各组6分钟步行试验距离、Borg呼吸困难指数、肺动脉压功能分级、平均肺动脉压（PAP）、氧合指数（OI）、心输出量（CO）、心脏指数（ CI ）、肝肾功能相关指标变化。观察治疗过程中出现的不良事件及不良反应。结果各组治疗后6分钟步行试验距离增加，且B、C组与A组相比，P均＜0．05。各组治疗后Brog呼吸困难指数和肺动脉压功能分级指数、PAP、OI、CO、CI较治疗前改善（P均＜0．05），且C组治疗后CI高于A、B组（P均＜0．05）。三组治疗前后肝肾功能指标差异无统计学意义。 A组发生头痛2例、无力2例、心率加快1例、低血压1例、血小板减少0例，不良反应发生率为15％；B组分别为4、3、1、0、0、例和20％；C组分别为3、5、1、0、0例和22．5％；三组不良反应发生率相比，P均＞0．05。结论0．25、0．5、0．8μg／（kg· min）的米力农治疗PAH均有效，不良反应较少，以0．8μg／（kg· min）剂量效果最佳。 [ABSTRACT FROM AUTHOR]

Objective To investigate the application value of blood-oxygen functional imaging system (BOFIS) in diagnosis of breast neoplasms. Methods Totally 201 patients with histopathologically verified breast lesions were selected, and the histology revealed there were 140 cases of benign lesions and 61 cases of malignancies. BOFIS, ultrasound and mammography were respectively used on these patients. The sensitivity, specificity and accuracy of BOFIS, ultrasound, mammography and combined criteria were analyzed. Results The sensitivities of BOFIS, ultrasound and mammography were 77. 14%, 94. 29% and 90. 71%. The specificities were 54. 10%, 86. 89% and 83. 61%, respectively. The accuracies were 70. 15%, 92. 04% and 88. 56%, respectively. Significant difference was found between them (P<0. 05). The combination with BOFIS can improve the diagnostic sensitivity of ultrasound/mammography alone. Conclusions BOFIS provides a new way for the diagnosis of breast disease. After combining with the conventional examination (ultrasound or mammography), BOFIS improves the diagnostic sensitivity, and can be used as a beneficial supplement to the imageological examination of breast.
