- Abstract
Objective To examine the changes in the expression levels of Pleckstrin homology domain family A member 4(PLEKHA4) in glioma tissues and its influencing factors. Methods Fifty-two cases of glioma tissues from surgical resection and 52 cases of their corresponding paraneoplastic tissues were selected, of which 21 cases were WHO grade Ⅰ-Ⅱ and 31 cases were grade Ⅲ-Ⅳ, and 23 cases were wild-type and 29 cases were mutant type of p53 typing. Immunohistochemistry and protein immunoblotting were used to detect the expression level of PLEKHA4 in the paracancerous tissues and glioma tissues of patients with glioma. We observed the clinical characteristics and prognosis of glioma patients with different expression levels of PLEKHA4, and analyzed the factors affecting the expression level of PLEKHA4 in glioma tissues. Results PLEKHA4 was positively expressed in glioma tissues of 52 patients, and the sites of expression were mainly in the cytoplasm and cell membrane of glioma tissues; no positive expression of PLEKHA4 was seen in the paracancerous tissues. The relative expression levels of PLEKHA4 in the paracancerous tissues, grade Ⅰ-Ⅱ glioma, and grade Ⅲ-Ⅳ glioma were 0. 419 ± 0. 101,0. 968 ± 0. 061, and 1. 366 ± 0. 096, respectively. The relative expression of PLEKHA4 in the glioma tissues was higher than that in paracancerous tissues, and the relative expression of PLEKHA4 was higher in glioma tissues of grade Ⅲ-Ⅳ than that in glioma tissues of grade Ⅰ-Ⅱ, respectively (both P<0. 05) . Significant differences were found in the WHO classification and p53 typing of patients with different PLEKHA4 expression levels in glioma tissues (both P<0. 05) . Logistic regression analysis showed that WHO classification and p53 typing of gliomas were the independent influencing factors for PLEKHA4 expression (both P<0. 05) . At 2-year postoperative follow-up, the survival rate of patients with high PLEKHA4 expression was 28. 6%, the survival rate of patients with low PLEKHA4 expression was 70. 8%, and statistically significant difference was found in the survival rate between them (P<0. 05) . Conclusion PLEKHA4 was highly expressed in glioma tissues, WHO typing and p53 typing of patients were the influencing factors of PLEKHA4 expression level in glioma tissues, and the PLEKHA4 expression level was related to clinical prognosis of patients.
