A highly regioselective aerobic bromination of aromatic compounds has been achieved using Fe3O4@SiO2/CuO nanocatalyst and O2 as a green oxidant. In this procedure, the reactions gave high yields and para-selectivity for the various aromatic compounds. Mechanistic studies are described, and the possible mechanisms are proposed. Recyclability of the Fe3O4@SiO2/CuO nanocatalyst has also been explored upon aerobic bromination of aromatic compounds. 2,4-Diphenyl thiazole is an important organic intermediate and its derivatives contain multiple biological properties. In the present study, we reported a new protocol to synthesize 2,4-diphenyl thiazole analogs, which involved the bromination of ethyl benzoylacetates with NBS in the presence of 2-hydroxypropyl-β-cyclodextrin, followed by a direct cyclization with thiobenzamides in water. The photocatalytic bromination of 2-arylimidazo[1,2-a]pyridines is described in this paper. This reaction uses the readily accessible and shelf-stable CBr4 as a bromine source. The α-bromination of carbonyl compounds is one of the most important transformations and also important precursors in synthetic organic chemistry. A simple and efficient bromination of aniline, aniline derivatives, and analogs have been developed. Forty three examples were given and the highest yield reached was 98%. Different substrates including substituted aniline, pyridin-amine, N-substituted aniline, N,N-disubstituted aniline, N-phenyl-amide, N-phenyl-sulfonamide, and nitrogen-containing heterocycles were all reactive and selectively generated desired bromo-products. An efficient method has been developed for the synthesis of 1-(arylimino)naphthalen-2(1H)-ones through the cascade reaction of anilines and 2-naphthols promoted by NaBr/K2S2O8/Ce(NH4)2(NO3)6. Using this protocol, a series of 1-(arylimino)naphthalen-2(1H)-ones was obtained in good to excellent yields (17 examples, 70–92% yields). The reactions may proceed through the following steps: bromination of 2-naphthols by in-situ-generated bromine from NaBr and K2S2O8 to afford 1-bromonaphthalen-2-ols, coupling of 1-bromonaphthalen-2-ols with anilines to afford the corresponding amines, and subsequent oxidation of the amines into the products by Ce(NH4)2(NO3)6. Practical and economical synthesis of synthetically valuable 3,3,3-triethoxypropyne, ketal protected phenyl and methyl substituents prop-2-ynones is described. Bromination and subsequent 18-crown-6 catalyzed elimination of triethylorthoacrylate and ketal protected terminal alkenes with methyl and phenyl substituent which are inturn readily available from triethylorthopropionate, 3-chlorobutan-2-one and propiophenone afforded multigram quantities (>10 g) of corresponding functionalized terminal alkynes. Exploration of the synthetic utility of these alkynes is also demonstrated by the acetylenic substitution of the phenylalaninol derived 1,2-cyclic sulfamidate to deliver chiral alkynylated amines. α -Bromoacetals of ketones were prepared from various secondary alcohols with 1,3-dibromo-5,5-dimethylhydantoin (DBDMH) and ethylene glycol through oxidation, bromination, and acetalization in one pot without the use of other catalysts under mild conditions. Expedient and highly efficient synthesis of previously unknown 2-methyl-5-nitrobenzo[b]thiophene has been developed, furnishing the target product in four steps with 87% overall yield. A simple method of electrochemical bromination of a series of cyclic and acyclic enes (styrene and substituted styrenes, stilbene, indene, and cyclooctene) in a biphasic water–chloroform mixture mediated by bromide/bromine redox system is reported. Aqueous 25% NaBr/H2SO4is used as the electrolyte. Regio- and stereoselective dibromination of enes is achieved. In the present study, 20 new tetracyclic 1,4-benzothiazines (4a–4 t) were conveniently synthesized in good yields and characterized by different spectral and physical techniques. The lipid peroxidation of fatty acids leads to secondary products, among which several carbonyl compounds are of concern in food toxicology. The detection of these reactive aldehydes for identification and evaluation is required. Derivatization is necessary to improve their stability and detection in liquid chromatography/high-resolution mass spectrometry (LC/HRMS) trace analyses. Therefore, a brominatedO-benzylhydroxylamine, namely 1-((ammoniooxy)methyl)-2-bromobenzene chloride, was selected as a new probe for the mild and selective derivatization of carbonyl compounds. New oxime ethers were thus synthesized under mild reaction and workup conditions, with full analytical characterization. The relevance of the chemical reaction was assessed with nine aldehydes, especially conjugated and deuterium-labeled aldehydes, and two ketones. This article presents an efficient method for the synthesis of core-brominated naphthalene diimides (NDI) from naphthalene dianhydride (NDA). A procedure for monitoring the NDA bromination reaction by1H NMR spectroscopy is described, allowing for optimization and greater consistency of this reaction. Furthermore, the subsequent bis-imidization reaction of the brominated NDA product has been significantly enhanced using microwave-assisted conditions, with recovery of pure product via simple filtration in excess of 90% of theoretical yield. Diethyl azodicarboxylate (DEAD) is a well-known coupling reagent that can be readily synthesized from diethylhydrazodicarboxylate (DEHD). The bromination of commercially available ethyl allophanate (1) in CHCl3, followed by the Hofmann-type rearrangement reaction of the resultingN-brominated species2and3in C2H5OH in the presence of 1,8-diazabicyclo[5.4.0]-7-undecene (DBU), gave DEHD in good yield from a one-pot process. Interestingly, however, the bromination and Hofmann-type rearrangement reactions did not occur in the presence of N(C2H5)3. These results therefore suggest that this reaction is reliant upon a high level of reactivity during the bromination reaction to give2and3, and that theseN-brominated species require the presence of a strong and nonnucleophilic base to undergo the Hofmann-type rearrangement to give DEHD. A novel procedure for the preparation of organic ammonium tribromides (OATBs) is described from their corresponding bromides. Quaternary ammonium bromides (QABs) and a N,N′-heterocyclic dibromide are efficiently oxidized to their corresponding monotribromides and bistribromide bym-chloroperbenzoic acid (MCPBA) in the presence of 2 and 4 equiv of KBr, respectively. The reactions are carried out in an aqueous medium without the use of any mineral acid or metal catalyst/promoters. A variety of tribromides are synthesized in very good yields including a hitherto unknown reagent, 1,10-(ethane-1, 2-diyl)phenanthrolinediium bistribromide (EPDBT). EPDBT is investigated as brominating agent and found to be highly effective for selective bromination of active methylene groups of a variety of 1,3-diketones and β-ketoesters. N,N-Dibromo-p-toluenesulfonamide (TsNBr2) has been found to be a new reagent for bromination of aromatic compounds. The reaction is extremely fast and goes into completion instantaneously at ambient temperature to produce exclusively the corresponding polybrominated product. The electrochemical bromination of 4-methoxy toluene by two-phase electrolysis yields 3-bromo 4-methoxy toluene at first, which subsequently undergoes side-chain bromination to give 3-bromo 4-methoxy benzyl bromide as a final product in 86% yield. The two-phase electrolysis consists of 25–50% NaBr as aqueous electrolyte and CHCl3containing aromatic compound as organic phase. The reaction temperature is maintained at 10–25 °C. The main goal of this article is to the present research on the development of ketamine derivatives. The target molecule was a fluoroderivative of ketamine, for which a multistep synthesis has been reported. This novel ketamine derivative, 2-(2-fluorophenyl)-2-methylamino-cyclohexanone, has been called fluoroketamine by our research group. The starting fluorobenzonitrile was reacted with the appropriate Grignard reagent followed by the bromination reaction to obtainα-bromocyclopentyl-(2-fluorophenyl)-ketone. The reaction of the obtained ketone with methylamine at −40 °C then resulted in the formation ofα-hydroxycyclopentyl-(2-flourophenyl)-N-methylamine. Finally, the five-memberd ring cyclopentanol was expanded to the cyclohexylketone by a thermal rearrangement reaction. The HCl salt of the target molecule, which is soluble in water, was obtained by the acidification of the free fluoroketamine with HCl. Preliminary animal tests on mice have shown that the resulting fluoroketamine has some advantages over ketamine in terms of the effective dose and the recovery time. [ABSTRACT FROM AUTHOR]

Two literature procedures for selective bromination of 2-nitrobenzaldehyde usingN-bromosuccinimide (NBS) or NaBr-NaIO4in sulfuric acid were examined. In contrast to the reports that 4-bromo-2-nitrobenzaldehyde is formed as a single product in good yield, reactions using NBS gave a number of isomeric mono- and dibrominated products identified by spectroscopical methods and by sodium borohydride reduction to the corresponding benzyl alcohol, and reactions using NaBr-NaIO4did not furnish any product. A series of 15 new polyfunctional drimanic compounds have been obtained by synthesis via dye-sensitized photooxygenation, bromination with N-bromosuccinimide, and electrochemical transformation of drim-7,9(11)-diene1and drim-8-en-7-one2. Three alternative syntheses of 12-acetoxy-drim-7,9(11)-diene8and two of 11-bromo-drim-8(9)-en-7-one10have been accomplished. For the first time a drimanic compound12with an aromatic ring B has been synthesized and the experimental conditions for the selective bromination at C11and C12methyl groups of drim-8-en-7-one2have been optimized. An original one-pot synthesis of bromomethyl ketones is achived using the Petasis reagent (dimethyltitanocene) as a key for enamine generation. Several amides were used to test the limits of the procedure by changing either the alkyl chain R or the amino portion of the starting materials. The enamines generated in situ were allowed to react with bromine at low temperature followed by hydrolysis to yield bromomethyl ketones in excellent yields (85 to 95%). Mechanistic details and optimum conditions for the reaction are briefly discussed. Unprecedented results are obtained from bromination of the normal/non-bridged and abnormal/bridged Biginelli compound of salicyldehyde. Bromination with 2,4,4,6-tetrabromocyclohexa-2,5-dienone yields a bridged monobromo-derivative and a bridged dibromo-derivative, respectively, whereas bromination with bromine and acetic acid gives a mixture of those two products in each case. Bromination with N-bromosuccinimide gives a third product, namely a tribromo-derivative, in addition to those two products. In this study, we discussed an efficient approach for the synthesis of camphorquinone from camphor by the continuous reaction of bromination and oxidation. The oxidation of 3-bromocamphor was catalyzed by Fe-porphyrins with air. The catalytic activity of iron-metallated functional porphyrins was investigated under optimization conditions. The results showed that this method was a milder alternative to the reported methods, with a simple, greener procedure in which the advantages were the utilization of air as the oxidant and the use of metalloporphyrins as catalysts. Three synthetic routes to 9-substituted 8-oxoadenines have been studied: bromination of adenine followed by N-9-alkylation/arylation and finally hydrolysis; bromination of adenine, hydrolysis, and N-functionalization as the last step; and N-9-alkylation of adenine, halogenation, and finally hydrolysis. As long as the N-9-functional group is compatible with conditions required for introduction of the halogen, the latter strategy was the most efficient. Also, a strategy starting from 5-amino-4,6-dichloropyrimidine was found to be a very good alternative for synthesis of 9-substituted 8-oxoadenines. A simple one-pot process for in situ bromination of (ethoxycarbonylmethylene)-triphenylphosphorane1was carried out. This was followed by oxidation of alcohol using SO3. We have described an elegant synthesis of 2-benzoylfuro[2,3-b]quinolines from quinolinyl chalcones via bromination and epoxidation. Interestingly, we found that during the bromination, the chalcones were cyclized to gave monobromodihydrofuroquinolines, which were dehydrobrominated with 1,8-diazabicyclo[5,4,0]undec-7-ene. During the epoxidation, chalcones were not cyclized and gave epoxides, which were treated with polyphosphoric acid to give the title compound. We have differentiated the addition of bromine and urea hydrogen peroxide to α,β-unsaturated carbonyl of quinolinyl chalcones by this new mechanism. [ABSTRACT FROM PUBLISHER]

Selective and efficient α-bromination of β-ketoesters and cyclic and acyclic ketones is achieved by reaction with N-bromosuccinimide (NBS) catalyzed by silica-supported sodium bicarbonate (NaHCO3 · SiO2) under mild reaction conditions and with short reaction times. With 100% selectivity with all substrates, after 45 min at room temperature (20 ± 2 °C), conversions for ethylacetoacetate were 67% and for acetophenone, cyclohexanone, and cycloheptanone were 58, 50, and 55%, respectively. Acetyl acetone recorded 65% conversion with 100% selectivity. Although cyclopentanone and toluene had only 20% conversion, both reactions showed 100% selectivity toward α-bromination. A convenient procedure for the synthesis of 1-bromo-3-buten-2-one, 4, from commercially available 2-ethyl-2-methyl-1,3-dioxolane, 1, is described. The procedure involves three reaction steps: (1) The acetal 1 is converted to 2-(1-bromoethyl)-2-bromomethyl-1,3-dioxolane, 2, by reacting 1 with elemental bromine in dichloromethane to yield 98% of 2. (2) Dehydrobromination of 2 with potassium tert-butoxide in tetrahydrofuran gives 2-bromomethyl-2-vinyl-1,3-dioxolane, 3, in 84-93% yield. (3) Removal of the acetal protection from 3 by formolysis for 6-10 h afforded 1-bromo-3-buten-2-one, 4, in 85-94% yield. An efficient and environmentally friendly synthetic approach for the preparation of fexofenadine was developed from commercially available cumene.
