A new dual-modality immunosensor based on molecularly imprinted polymer (MIP) and a nanostructured biosensing layer has fabricated for the simultaneous detection of two important markers including prostate-specific antigen (PSA) and myoglobin (Myo) in human serum and urine samples. In the first step, 3,3′-dithiodipropionic acid di(N-hydroxysuccinimide ester) (DSP) was self-assembled on a gold screen printed electrode (SPE). Then, the target proteins were attached covalently to the DSP-SPE. The imprinted cocktail polymer ((MIP(PSA, Myo)-SPE)) was synthesized at the SPE surface using acrylamide as monomer, N,N′-methylenebisacrylamide as a crosslinker, and PSA and Myo as the templates, respectively. The MIP-SPE was specific for the impedimetric sensing of PSA and Myo. After that, a nanocomposite (NCP) was synthesized based on the decorated magnetite nanoparticles with multi-walled carbon nanotube, graphene oxide and specific antibody for PSA (Ab). Then, NCP incubated with (MIP(PSA, Myo)-SPE. The modified electrodes and synthesized nanoparticles were characterized using electrochemical impedance spectroscopy, dynamic light scattering, surface plasmon resonance and scanning electron microscopy. The limits of detections were found to be 5.4 pg mL−1 and 0.83 ng mL−1 with the linear dynamic ranges of 0.01–100 and 1–20000 ng mL−1 for PSA and Myo, respectively. The ability of proposed biosensor to detect PSA and Myo simultaneously with high sensitivity and specificity offers a powerful opportunity for the new generation of biosensors. This dual-analyte specific receptors-based device is highly desired for the integration with lab-on-chip kits to measure a wide panel of biomarkers present at ultralow levels during early stages of diseases progress.

Natural flavouring materials are in high demand, and a premium price is paid for all-natural flavourings, making them vulnerable to fraud. At present, compound-specific isotope analysis (CSIA) is perhaps the most sophisticated tool for determining flavour authenticity. Despite promising results, the method is not widely used, and the results are limited to the most common volatile organic compounds (VOCs). This paper describes a robust protocol for on-line measurements of δ13C and δ2H using HS-SPME coupled with GC-C-IRMS and GC-HTC-IRMS for common fruit VOCs. To achieve reproducible and accurate results, a combination of a peak size/linearity correction with drift correction were used. Finally, the results were normalised by multiple point linear regression using the known and measured values of reference materials. Special care was taken to avoid irreproducible isotopic fractionation and the effects of equilibration, adsorption, desorption times and temperatures on δ13C or δ2H values were examined. Method validation was performed, and the average combined measurement uncertainty (MU) was 0.42‰. All the δ13CVPDB values were below ±3*MU, regardless of analytical conditions. In contrast, for δ2HVSMOW-SLAP values, only low temperature (30 °C) with equilibration time (15 min) and shorter adsorption time (between 10 and 20 min) can produce an isotopic difference of

Among the organic contaminants that could pass from waste to polyhydroxyalkanoates (PHAs), there are the polycyclic aromatic hydrocarbons (PAHs). For this reason, we have developed a rapid analytical method for the determination of sixteen PAHs in PHAs. PAHs were extracted by n-hexane, after matrix dispersion and crumbling into sand; the extract was purified by solid phase extraction using florisil as adsorbent. Recoveries in the range of 89–101% were obtained for the deuterated analytes, except for the two with the lowest molecular weight. Trueness between 92% and 108% and within-laboratory precision (expressed as relative standard deviation) ≤18% were estimated for all the analytes. Gas chromatography/mass spectrometry was used for analyte determination. Method limits of quantification were suitable to assure that PAH presence in PHA biolpolymers is much below the limits set by European law for plastic materials. Indeed, analysis of two different PHA samples showed that contamination is limited to few compounds at non-concerning levels.

Nowadays, environmentally friendly synthesis pathways for preserving the environment and minimizing waste are strongly required. Herein, we propose filter paper as a convenient scaffold for chemical reactions. To demonstrate this novel approach, Prussian Blue Nanoparticles (PBNPs) were synthesized on filter paper by utilizing few μL of its precursors without external inputs, i.e. pH, voltage, reducing agents, and without producing waste as well. The functional paper, named “Paper Blue”, is successfully applied in the sensing field, exploiting the reduction of hydrogen peroxide at low applied potential. The eco-designed “Paper Blue” was combined with wax- and screen-printing to manufacture a reagentless electrochemical point-of-care device for diabetes self-monitoring, by using glucose oxidase as the biological recognition element. Blood glucose was linearly detected for a wide concentration range up to 25 mM (450 mg/dL), demonstrating its suitability for management of diabetes and glucose-related diseases. The Paper Blue-based biosensor demonstrated a correlation coefficient of 0.987 with commercial glucose strips (Bayer Contour XT). The achieved results demonstrated the effectiveness of this approach, which is also extendible to other (bio)systems to be applied in catalysis, remediation, and diagnostics.

In this paper, a novel voltammetric/amperometric approach for the direct determination of free acidity (FFA, expressed as mass percentage of free oleic acid) in olive oil samples is presented. The method is based on the reduction processes occurring at a platinum microdisk electrode involving the free fatty acids present in the matrices. To overcome problems related to the low conductivity of the samples investigated, olive oils were mixed with suitable amounts of the room temperature ionic liquid (RTIL), tri-hexyl(tetradecyl)phosphonium bis (trifluoromethylsulfonyl) imide ([P14,6,6,6]+[NTf2]-), which acted as a supporting electrolyte. Conditions for a reliable quantification of the acids were preliminarily investigated by performing voltammetric and chronoamperometric measurements in RTIL solutions containing oleic acid at different concentrations. Oleic acid (OA) was chosen as a model compound as it is the main component of the FFA content in olive oils. In order to establish the effect of oxygen on the electroanalytical responses, the reduction process of OA was investigated under both deoxygenated and oxygenated conditions. It was found that, in both situations, the current arising from the electrode process of OA depended linearly on the OA concentration over a wide range varying from 0.1% to 8% OA (w/w). This range includes FFA values which can be found on all categories of commercially available oil samples, including extra-virgin, virgin and lampante oils. Voltammetric and chronoamperometric experiments were also performed in oil/RTIL samples artificially acidified (extra-virgin olive oils with known addition of oleic acid) and in natural olive oils from some commercial categories. The results obtained indicated that the electrochemical procedure developed was satisfactory in terms of both sensitivity and detection limits. The reliability of the proposed approach for the detection of FFA was finally assessed by comparison of the voltammetric/chronoamperometric values with those obtained by the official method for quantification of olive oil acidity, which is an acid/base volumetric titration.

The cyclic voltammetric behaviour of propionaldehyde (PA) and hexanaldehyde (HA), in 1-butyl-3-methylimidazolium bis(trifluoromethyl-sulfonyl) imide ([BMIM][NTF2]), 1-butyl-3-methylimidazolium hydrogen sulphate ([BMIM][HSO4]) and 1-butyl-3-methylimidazolium hydroxide ([BMIM][OH]) was investigated at a platinum microelectrode. A clear oxidation process for both aldehydes was recorded only in [BMIM][OH]. On the basis of these evidences, an electrochemical microprobe (EMP), incorporating [BMIM][OH] as electrolyte, was assembled for sensing these aldehydes in gaseous phases. The EMP exposed in the headspace of the liquid aldehydes displayed voltammetric and amperometric responses, which depended on the aldehyde vapour pressures and, consequently, on the temperature employed. The usefulness of the [BMIM][OH] coated EMP for practical applications was assessed in the detection of HA vapour released from squalene (i.e., a lipid simulant matrix) samples spiked with known amounts of the aldehyde. Calibration plots were constructed at 40 °C, 50 °C and 60 °C, using both voltammetry and chronoamperometry. In both cases, good linearity between current and HA concentration in squalene was obtained over the range 3–300 ppm, with correlation coefficients higher than 0.991. Reproducibility, evaluated from at least three replicates, was within 5%. Detection limits, evaluated for a signal-to-noise ratio of 3, were in any case lower than 1.7 ppm. These analytical performances are suitable for monitoring VAs coming from lipid oxidation processes in food. An application concerning the determination of VAs in headspace of sunflower oil during an induced oxidative test to establish its thermal stability was also performed.

Bioassays based on enzyme inhibition are analytical tools widely employed for inhibitor analysis. Beside the conventional analytical techniques such as chromatography and mass spectrometry, these bioassays are cost-effective, easy to use, and suitable for in situ measurement but they are often characterised by a quite narrow linear range. Herein, we report a novel graphical method based on integrated Michaelis-Menten equation, valid for all types of reversible inhibition, which provides an extended linear range. The suitability of this innovative approach was demonstrated in the case of fluoride quantification using a colorimetric bioassay based on acetylcholinesterase inhibition. The “half time reaction”, estimated by the progress curve of cholinesterase inhibition, was plotted versus the fluoride inhibitor concentration, observing an extended linear range up to 5 mM, instead of 0.6 mM using initial rate measurements. The applicability of this new concept was further demonstrated in the case of catalase enzyme inhibited by cyanide. Furthermore, it was demonstrated that fixed substrate conversion at level of 10–50% allows determination of inhibitor concentration in a wide linear range with high precision and in short time of analysis. This novel theoretical and practical approach allows for the extension of the linear range without any further experiments, with several advantages including low reagent consumption, reduced waste generation and time of measurement.

Honey, in particular monofloral varieties, is a valuable commodity. Here, we present proton transfer reaction-time of flight-mass spectrometry, PTR-ToF-MS, coupled to chemometrics as a successful tool in the classification of monofloral honeys, which should serve in fraud protection against mispresentation of the floral origin of honey. We analyzed 7 different honey varieties from citrus, chestnut, sunflower, honeydew, robinia, rhododendron and linden tree, in total 70 different honey samples and a total of 206 measurements. Only subtle differences in the profiles of the volatile organic compounds (VOCs) in the headspace of the different honeys could be found. Nevertheless, it was possible to successfully apply 6 different classification methods with a total correct assignment of 81-99% in the internal validation sets. The most successful methods were stepwise linear discriminant analysis (LDA) and probabilistic neural network (PNN), giving total correct assignments in the external validation sets of 100 and 90%, respectively. Clearly, PTR-ToF-MS/chemometrics is a powerful tool in honey classification.

We report a sensor for phosphate detection based on screen-printed electrodes modified with carbon black nanoparticles. The phosphate was measured in amperometric mode via electrochemical reduction of molybdophosphate complex. Carbon black nanoparticles demonstrated the ability to quantify the molybdophosphate complex at a low applied potential. Some analytical parameters such as the working solution (sulfuric acid 0.1M), applied potential (0.125V vs Ag/AgCl), and molybdate concentration (1mM) were optimized. Using these conditions, a linear range of 0.5-100µM was observed with a detection limit of 0.1µM, calculated as three times the standard deviation of the blank divided by the slope of calibration curve. The system was challenged in drinking, river, aquarium, and waste water samples yielding satisfactory recovery values in accordance with a spectrophotometric reference method which demonstrated the suitability of the screen-printed electrode modified with carbon black nanoparticles coupled with the use of molybdate to detect phosphate in water samples.

Cortisol is a steroidal hormone and an important stress marker. Free serum cortisol concentration has been identified to correlate well with free salivary cortisol. In this present work an electrochemical immunosensor was developed to determine cortisol concentration within the physiological concentration range found in human saliva. The immunosensor is based on a direct competitive enzyme linked immunoassay using a home-made cortisol-alkaline phosphatase (AP) conjugate synthesized in our laboratory with disposable graphite screen-printed electrodes (SPEs). 1-nalphtyl phosphate (1-NP) was used as an enzymatic substrate and a square wave voltammetry (SWV) for electrochemical detection. To study method suitability for use with saliva samples, calibration curves were performed both in buffer and saliva. In buffer standard samples showed a limit of detection (LOD) of 0.6 ng/ml and working range (WR) of 0.2–44.6 ng/ml with good reproducibility (RSD 10%). Saliva matrix effect was removed effectively with Salivette Cortisol collection device (polyethylene) and a calibration curve showed similar characteristics as in buffer with LOD 1.7 ng/ml and WR 0.5–55.1 ng/ml (RSD 8%) demonstrating the possibility to determine human salivary cortisol within the desired human physiological range. Spiked saliva samples were analyzed with the developed immunosensor presenting excellent 92–114% recovery. Comparison to liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) method showed strong 0.90 correlation between methods indicating good accuracy of the developed immunosensor.

The recent goal of sustainability in analytical chemistry has boosted the development of eco-designed analytical tools to deliver fast and cost-effective analysis with low economic and environmental impact. Due to the recent focus in sustainability, we report the use of low-cost filter paper as a sustainable material to print silver electrodes and to load reagents for a reagent-free electrochemical detection of chloride in biological samples, namely serum and sweat. The electrochemical detection of chloride ions was carried out by exploiting the reaction of the analyte (i.e. chloride) with the silver working electrode. During the oxidation wave in cyclic voltammetry the silver ions are produced, thus they react with chloride ions to form AgCl, while in the reduction wave, the following reaction occurs: AgCl + e- -->Ag + Cl-. These reactions at the electrode surface resulted in anodic/cathodic peaks directly proportional to the chloride ions in solution. Chloride ions were detected with the addition of only 10 μL of the sample on the paper-based electrochemical cell, obtaining linearity up to 200 mM with a detection limit equal to 1 mM and relative standard deviation lower than 10%. The accuracy of the sensor was evaluated in serum and sweat samples, with percentage recoveries between 93 ± 10 and 108 ± 8%. Moreover, the results achieved with the paper-based device were positively compared with those obtained by using the gold standard method (Ion Selective Electrode) adopted in routine clinical analyses.

In this paper we present an electrochemical approach to prepare standard solutions of metal ions in a room temperature ionic liquid (IL), which can find useful application for analysis in hydrophobic matrices. The method, developed here for the case of lead ions, is based on the galvanostatic dissolution of a lead anode dipped directly in a suitable IL, namely tri-hexyl(tetradecyl)phosphonium bis (trifluoromethylsulfonyl) imide ([P 14,6,6,6 ] + [NTf 2 ] - ). After each oxidation step, the metal dissolution process in the IL solutions was monitored by cyclic voltammetric measurements at a glassy carbon disk electrode. The results indicated that the peak current relevant to the reduction of the electro-generated Pb(II) increased linearly while increasing the oxidation time. By varying the oxidation time from 200 to 6000 s, a set of Pb (II) /[P 14,6,6,6 ] + [NTf 2 ] - solutions at concentrations ranging between 10 and 300 μg g −1 was prepared. To validate the efficiency of the electrochemical procedure to produce metal ion standard solutions, the Pb content was quantified by developing a microwave digestion procedure specifically suitable for the IL medium, followed by ICP-QMS analysis in the digested standards. The results indicated a satisfactory agreement between concentrations found by ICP-QMS and calculated from electrochemical data, with a coulometric efficiency of Pb(II) generation in ionic liquid ≥95.6%. Finally, the applicability of the Pb (II) /IL solutions as standards for analyses in hydrophobic media was tested by determining, by ICP-QMS, the Pb content in an extra-virgin olive oil spiked with known amounts of a Pb (II) /IL standard. Satisfactory Pb recoveries, ≥96%, were measured.

The oxidative deterioration of unsaturated lipids fatty acids is a great concern for the food industry as it is associated with the development of rancid off-odors and the reduction of food quality. This work describes the potential use of isothermal microcalorimetry to monitor the oxidation of linoleic acid, an important conjugated fatty acid responsible for the development of rancidity. The heat flow signal developed during the oxidation process reflects a multistep mechanism typical of radical chain reactions. From the analysis of the thermograms, it is possible to identify a period of the reaction that occurs with an order equal to 0.5. This period is correlated with the propagation period of the radical chain reaction. In addition, it allows detecting the early appearance of rancidity notes, as confirmed with the analysis of the samples headspace by Proton-Transfer-Reaction Mass Spectrometry (PTR-MS). The proposed approach was finally applied to compare the capacity of natural and synthetic antioxidants to inhibit the oxidation process of linoleic acid and the occurrence of rancidity. The results presented here show the advantages of microcalorimetry to study oxidation reactions and their control.

Heavy metals, as cadmium, attract a rising attention in environmental studies due to their increasing release by human activities and acute toxicity. In situ analytical methods are needed to minimize current uncertainties caused by the transport and conservation of samples. Here, we present the completely automatic determination of Cd in natural waters using a newly developed screen printed electrode sensor (SPE), inserted in a homemade purpose-built flow cell coupled to a Multi-Syringe Flow Injection Analysis system (MSFIA). The working electrode of SPEs was constituted by a carbon film modified with Nafion. Cd was plated on an in situ bismuth film and determined using Square Wave Anodic Stripping Voltammetry. Different chemical conditions of deposition and stripping were studied. A sample/acetic buffer mixture was found to be a well suited medium to form the Bi film and perform the analysis. Cd was quantified via calibration by on line standard additions. The limit of detection was found to be 0.79μgL(-1), well below the limit stipulated by the European directive (5μgL(-1)). Good sample throughput (14h(-1)) and low consumption of reagent and sample (1.3mL) were also obtained in line with previous works in Cd flow analysis.

The use of a Dynamic Headspace System (DHS) device combined with a Thermal Desorption Unit (TDU) interfaced to a Gas Chromatography/Mass Spectrometry (GC/MS) system is proposed for the determination of furfurals in oenological products. An experimental design protocol has been employed for the optimization of the instrumental settings concerning DHS and TDU extraction and desorption steps. It has been possible to individuate the following optimized conditions: incubation temperature 40 °C, purge volume 800 mL, dry volume 1500 mL, TDU hold time 5 min and incubation time 10 min. The performance of two different SPE sorbents, namely Tenax TA and Tenax GR used for the furfurals trapping, was investigated too. The developed DHS sampling procedure showed good reproducibility values with a RSD% lower than 10% for all the monitored species. The optimized experimental settings have been used to determine furfurals in several vinegar samples obtained by traditional procedure starting from cooked grape musts, i.e. in Aceto Balsamico Tradizionale di Modena (ABTM). In fact, the control of these species is extremely important for quality and safety issues.

The formation of open chain polyammonium cation-fluoride and -carbonate complexes was studied by potentiometric and calorimetric techniques at t=25 degrees C. Several species of H(i)AL (A=amine; L=F(-), CO(3)(2-)) are formed in both systems with a mean stability log K=1.0zeta (zeta=|z(anion)xz(cation)|) and log K=2.0zeta for fluoride and carbonate, respectively. The comparison with analogous systems (chloride and acetate for fluoride and hydrogenphosphate, sulfate and malonate for carbonate) showed that fluoride and carbonate form the most stable species with open chain polyammonium cations, among low molecular weight anions. The N-alkyl substitution does not play negligible role in the stability of these complexes, the species formed by substituted amines being more stable.

Due to worker׳s exposure, solvent and stationary phases׳ consumption, sample purification is one of the most polluting steps in analytical procedures for determination of organic pollutants in real samples. The use of photochemical sample treatment represents a valid alternative methodology for extracts clean up allowing for a reduction of the used amount of organic solvents. In this paper we report the first application on the photolytic destruction of organic substances to eliminate some of the interferences in the analysis of Chlorobenzenes in sediment samples. The method׳s efficiency and robustness were compared with classic silica column purification process currently used in clean up procedures in sediment analysis. Quality parameters such as recovery, linearity and reproducibility were studied. The entire procedure was validated by three replicate analysis of spiked real sediment sample. The quantification limits (LOQ) obtained by us ranged from 1.0 to 2.3 ng g−1, while the detection limits (LOD) were of 1.0 ng g−1. The RSD for each congener was below 10% and recoveries were in the range 95–130%. Results based on the analysis of real samples showed similar or improved detection thresholds and pointed out the advantages of the photochemical methodology in terms of costs, use of chemical substances and operator׳s safety according to Green Analytical Chemistry principles.

An increasing number of studies have concerned the profiling of polyphenol microbial metabolites, especially in urine or plasma, but only a few have regarded their accurate quantification. This study reports on a new ultra-performance liquid chromatography tandem mass spectrometry method with electrospray ionisation (UHPLC-ESI-MS/MS) using a simple clean-up step with solid phase extraction (SPE) and validation on different biological matrices. The method was tested with spiked samples of liver, heart, kidneys, brain, blood and urine. The purification procedure, after the evaluation of three different cartridges, makes it possible to obtain cleaner samples and better quantification of putative trace metabolites, especially related to dietary studies, with concentrations below ng/g in tissue and for urine and blood, starting from ng/ml. Limits of detection and linear range were also assessed using mixed polyphenol metabolite standards. Short chromatographic separation was carried out for 23 target compounds related to the polyphenol microbial metabolism, coupled with a triple quadrupole mass spectrometer for their accurate quantification. By analysing different spiked biological samples we were able to test metabolite detection in the matrix and validate the overall recovery of the method, from purification to quantification. The method developed can be successfully applied and is suitable for high-throughput targeted metabolomics analysis related to nutritional intervention, or the study of the metabolic mechanism in response to a polyphenol-rich diet.

The use of photochemical sample treatment represents an alternative methodology for extracts clean up allowing for a reduction of the used amount of organic solvents. The first application of a photochemical sample treatment for the selective removal or reduction of organic substances interfering with PCBs analyses in sediments is reported. The method's efficiency and robustness were compared with currently used chromatographic purification. Quality parameters such as recovery, linearity and reproducibility were studied. The entire procedure was validated by four replicate analysis of certified reference sediment. The quantification limits (LOQ) obtained by us ranged from 1 to 3.1 ng g(-1). The RSD for each congener was below 15\% and recoveries were in the range 40-130\%. Results based on the analysis of real and certified samples showed similar or improved detection thresholds and pointed out the advantages of the photochemical methodology in terms of costs and environmental friendly conditions. Microdialysis probes have been tested to evaluate the influence of flow-rate, probe dimensions and temperature when used as sampling systems inserted subcutaneously for in vivo monitoring of glucose. The probe was coupled with a glucose biosensor obtained from a thin layer electrochemical cell generally used as detector for liquid chromatography. Glucose oxidase was immobilized on a nylon net membrane and it was placed over an acetate cellulose membrane into the cell in contact with the platinum anode.
