Abstract: Functionalized 6-arylsalicylate substructures occur in a variety of pharmacologically relevant natural products and bioactive compounds. They are also broadly used as important intermediates in organic synthesis. Traditional synthetic methods have suffered from some drawbacks, such as relatively harsh reaction conditions, narrow range of substrates, and poor yields. Utilizing microwave irradiation, the synthesis of functionalized 6-arylsalicylates via a Suzuki cross-coupling has been developed with a wide range of substrates. Almost all the reactions proceeded smoothly and afforded moderate to excellent yields of products, which indicated that electronic effects and steric modifications have little effect on this reaction. Abstract: C 1-symmetric cyclotriveratrylene analogues 2–8 with various functional groups at one benzene moiety were prepared starting from the selectively demethylated compound 1. Through the chemical resolution, a pair of enantiomers of C 1-symmeric compound 1 could be separated with gram scale. Compound 8, which possessed an N-linked imidazolium unit at the upper rim of the macrocyclic skeleton through a methylene linker, was successfully applied to 1,4-Michael addition reaction of alcohol to unsaturated aryl ketone. Its supramolecular catalytic activity as an NHC precursor was demonstrated by the chem-selectivity to aromatic alcohol than alkyl alcohol. [Copyright &y& Elsevier]

Abstract: Halogen substituent plays an important role in the crystalline packing of aromatic compounds. The [2+2] photocycloaddition of (E)-3-benzylidene-4-chromanones in the crystalline state was investigated, and halogen substitution has been adopted to organize molecules with proper arrangement for photodimerization. Not halogen bonds, but the electron-withdrawing property of halogen atoms can enhance the face-to-face π–π interactions. Therefore, F, Cl or Br substitution at the para position of phenyl gave rise to almost the same β-structures with face-to-face π-stacking. Only resulted β-structures can undergo photodimerization, which gave the syn-HH (syn-head-to-head) products with high regio-/stereoselectivity. [Copyright &y& Elsevier]

Abstract: Zwitterionic 4-hydroxy-2(1H)-quinolinone derivatives were synthesized through a unique reaction of 4-hydroxy-2(1H)-quinolinone with p-benzoquinone and N-heterocyclic aromatics. Abstract: A copper-catalyzed dehydrogenative cross-coupling reaction between unactivated ethers and simple ketones mediated by pyrrolidine has been developed. Under the catalysis of CuBr2 and in the presence of pyrrolidine, either tetrahydrofuran 2a or tetrahydropyran 2b can react smoothly with a series of methyl aryl ketones 1a–m to give desired coupling products 3aa–mb using TBHP as an oxidant. The advantages of the dehydrogenative cross-coupling reaction are adoption of unmodified ethers as substrates, good tolerance of many functional groups and use of cheap copper salt as a catalyst. Heterocyclic ketene aminals having at least one secondary amino group underwent the aza–ene reaction with ethyl propiolate under various conditions to yield the corresponding adducts, which were readily transformed into the δ-lactam fused heterocyclic products with or without the aid of sodium ethoxide in refluxing ethanol, while the ester- and cyano-substituted tertiary enediamines acted as the strong Michael donor to add to ethyl propiolate in a polar or a protic solvent. We reported a novel mono-β-cyclodextrin derivative, mono-6-deoxy-6-biguanidino-β-cyclodextrin (β-biGCD), which was investigated as a mimic of ADP/ATP carrier (AAC). Its affinity toward AMP, ADP, and ATP was evaluated by means of isothermal titration calorimetry (ITC). The association constants (K a) of β-biGCD binding to AMP, ADP, and ATP were determined to be (1.07±0.04)×106, (5.86±0.02)×106, and (4.33±0.06)×106 L mol−1, respectively, which were 100-fold higher than mono-guanidino-β-cyclodextrin (ca. 104 L mol−1). UV spectroscopic titrations further confirmed the above results. The interaction between β-biGCD and nucleotides was probed by docking simulation. These results reveal that the biguanidinium moiety mimics the arginine residues of mitochondrial AAC protein. [ABSTRACT FROM AUTHOR]

Abstract: A mild and efficient one-pot protocol for the preparation of 8,13-dihydro-7H-indolo[2′,3′:3,4]pyrido[1,2-b]indazole and 5,6-dihydroindazolo[3,2-a]isoquinoline via the reductive cyclization of nitro-aryl substrates mediated by a low-valent titanium reagent has been developed. The attractive features of the current method include an N–N bond formation and the selective reduction of the C N bond and nitro group, both of which were easily achieved in one-pot by controlling the pH of the reaction mixture. Abstract: A series of disubstituted bis(vinylquinolinium)benzene derivatives were designed, which were prepared through a facile three-component one-pot reaction in good yield. FRET results showed that 1,3-disubstituted benzene derivatives had much stronger stabilization effect on G-quadruplex DNA than that of 1,4-disubstituted benzene derivatives. The introduction of substituted amine side chain at quinolinium obviously increased the binding affinity of compounds to G-quadruplex DNA. It was also found that 1,3-disubstituted benzene derivatives and 1,4-disubstituted benzene derivatives had different effects on the conformation of G-quadruplex DNA by CD spectroscopy analysis. The differences for the interactions of these two classes of compounds with G-quadruplex were further studied and elaborated through molecular modeling experiments. [Copyright &y& Elsevier]

Abstract: An efficient and improved synthesis of 2H-indazoles via reductive cyclization of 2-nitrobenzylamines induced by low-valent titanium reagent (TiCl4/Zn) is described. In this reaction triethylamine (TEA) was used to control the pH value. This method has the advantages of easily accessible starting materials, convenient manipulation, higher yield, shorter reaction time, and wider substrate scope. [Copyright &y& Elsevier]

Abstract: A novel route for the efficient synthesis of a target psoralen moiety, 4,4′-dimethylxanthotoxol, has been developed, which need only four steps using cheap pyrogallol as a starting material. Subsequently, a range of new glycosylated psoralen derivatives were synthesized in good yields with simple procedures and mild reaction conditions. The experiment of biological activity shows that some of the glycosylated psoralen derivatives have antiproliferative activities against human cancer cell lines. A strong photo-induced antiproliferative effects were found under UVA. All of the glycosylated psoralen derivatives exhibited antioxidant activities against the oxidation of DNA induced by Cu2+/glutathione (GSH). Further experiment also demonstrates that the introduction of sugar moieties in some glycosylated psoralen derivatives can improve their antioxidant activities significantly. Abstract: Two optical pure m-dimethylamino substituted inherently chiral calix[4]arene derivatives 8a and 8b bearing an l-prolinamido group have been synthesized by two routes, and structurally studied by the usual spectroscopic methods and X-ray crystallographic analysis. It was found that both of 8a and 8b could be utilized as bifunctional organocatalysts to efficiently promote the aldol reactions between aromatic aldehydes and ketones in the presence of acetic acid. Especially, with 8a as the catalyst, the reaction between 4-nitrobenzaldehyde and cyclopentanone at −20°C gave the anti-aldol product up to 94% ee, while the anti-aldol product in up to 94:6 dr and 79% ee was obtained when 4-cyanobenzaldehyde was used as the aldol donor. Moreover, it was also demonstrated that the inherently chiral calixarene skeleton with (cS)-conformation in 8a was identified as the matched configuration of the stereogenic elements, and the inherently chiral moiety might play an important role in helping to stereocontrol the reaction. Abstract: A novel approach to one-pot synthesis of dihydrofuro[3′,4′:5,6]pyrido[2,3-c]pyrazole and indeno[2′,1′:5,6]pyrido[2,3-c]pyrazole derivatives have been investigated using organocatalysts that are recyclable. This new protocol has the advantages of environmental friendliness, higher yield, low cost, as well as convenient operation. The catalytic efficiency of various small organocatalysts such as l-proline, trans-4-hydroxy-l-proline, l-thiaproline, cinchonidine, (+)-cinchonine, and dl-2-phenylglycine has also been studied. Abstract: The strategy of bridging the anisole units at the upper rim of calix[6]arene has been applied to strain the conformations of calix[6]arene. Based on the selective formylation of the 1,3,5-tri-p-tert-butylcalix[6]arene, several new calix[6]arene derivatives with different 1,3-bridged chains or a 1,3,5-tripod bridge at the upper rim have been prepared with moderate yields. The 1H NMR spectra indicate that these calix[6]arene derivatives adopt a cone conformation, which has also been confirmed by the theoretical calculation at AM1 level. X-ray crystal structure of 1,3,5-tripod bridged compound 5 discloses that the calix[6]arene host stands in a cone conformation with approximate C 3v symmetry, and that a methanol molecule is enclosed in its hydrophobic cavity and stabilized by multi hydrogen bonds. Abstract: A novel procedure to construct o-naphthothiophenequinones has been achieved from readily available o-benzothiophenquinones and N-dienes via Diels–Alder reaction-aromatization sequence as key steps. Abstract: A series of tri-O-alkylated inherently chiral fluorescent calix[4]crowns in the cone conformations and a series of tetra-O-alkylated inherently chiral fluorescent calix[4]crowns in the partial cone conformations have been synthesized. By condensing with chiral auxiliary (S)-BINOL, the resulting diastereomers could be separated via preparative TLC. We found that the size of the crown moiety effected the separation of the diastereomers. Further, removal of the BINOL unit by hydrolysis furnished pairs of enantiomers with optical purity. Moreover, we found that a tetra-O-alkylated inherently chiral fluorescent calix[4]crown-6 in the partial cone conformation 6c showed considerable enantioselective recognition capability towards chiral leucinol. Abstract: The synthesis, structure and ring-closing metathesis (RCM) reactions of polyether bridged biscalix[4]arenes 6 in the partial cone conformation with upper rim allyl substituents are reported. The RCM reaction modes depend on the length of polyether chain. Diethylene glycolic chain produced the dimer 7a and linear oligomer 7a′ with multi-cavities, whereas triethylene and tetraethylene glycolic chains allowed direct cyclization through intramolecular head-to-tail pattern to yield novel bridged biscalix[4]arenes 7b–c. A family of novel calix[4]arene derivatives containing nucleoside bases were designed and synthesized. Coupling reaction between para mono- or bis-amino calix[4]arenes 5, 6 or 7 and thymin-1-ylacetic acid in the presence of DCC afforded mono- or bis-thymine-substituted calix[4]arenes 8, 9 or 10 in over 70% yield. Owing to the low solubility of adenine-N9-ylacetic acid in DMF and DMSO and the weak nucleophilicity of aminocalix[4]arene derivatives, alternatively, the substitution reaction of bromoacetylated aminocalix[4]arenes derivatives 11, 12, 13 with adenine in the presence of sodium hydride was carried out to synthesize mono- or bis-adenine-substituted calix[4]arenes. Two kinds of isomers 15 and 16 or 17 and 18 were obtained due to the non-regiospecific alkylation of adenine, and their structures have been confirmed by 13C NMR and 1H NMR spectra. Five-membered ring C-alkylated heterocyclic ketene aminals react with diethyl azodicarboxylate to give the quaternary carbon-containing adducts in moderate to excellent yields while the six-membered analogs afford imidazo[1,5-a]pyrimidin-6-one derivatives. Chloromethylation of 5,17-di-tert-butyl-25,27-dihydroxy-26,28-dimethoxycalix[4]arene with chloromethyl methyl ether in the presence of zinc chloride led to the formation of macrocyclic and linear oligomers containing three to five calix[4]arenes bridged via methylenes at the upper rims. Under the identical conditions, however, 5,17-di-tert-amyl-25,27-dihydroxy-26,28-dimethoxycalix[4]arene was converted into a mixture of linear dimer and trimer in yields of 39.5 and 34%, respectively. Only dimer was obtained as the sole product in 41% yield when 25,27-dihydroxy-26,28-diethoxycalix[4]arene was used. Efficient synthesis of linear or starlike polycalixarenes was achieved utilizing the Friedel–Crafts reaction of debutylated or partially debutylated calix[4]arenes with chloromethylated calix[4]arene promoted by anhydrous zinc chloride.
