An inexpensive and commercially available Tf2NH-catalyzed 1,6-conjugate addition of imidazopyridine (IMPY) heterocycles to para-quinone methides (p-QMs) is reported. The present transformation provides a diverse class of C3-functionalized triarylmethanes heterocyclic derivatives of imidazopyridine. These metal-free transformations provided a very broad substrate scope of conjugate addition product with a high yield up to 97% within a short duration.

2H-Pyran-2-ones are widely present in nature and found as either isolated or in fused form. Depending on the fusion of benzene ring, pyran can give coumarins, isocoumarins, dibenzopyrans etc. This class of molecules exhibit excellent biological activity and photophysical properties. Apart from these, 2H-pyran-2-one also acts as very good Michael acceptor, therefore act as very important synthetic precursor. Suitably functionalized 2H-pyran-2-one has been widely explored for their synthetic potential. In this review, we have focused on use of different nucleophiles based on carbon, nitrogen, oxygen and sulfur to generate vast molecular diversity. The functionalized 2H-pyran-2-ones exhibit three electrophilic centres with different electrophilicity. Use of suitable nucleophiles can act on either of the position based on its reactivity to generate new chemical entity. 2H-Pyran-2-one has been explored as precursor for the synthesis of various biologically important nucleuses like pyridine, pyrimidine, quinolines, isoquinolines, fused quinolines, pyrazole, pyrroles, imidazopyridine, thiazole, oxazole, fused and isolated pyrans, coumarins, isocoumarins, benzothiophenes, dibenzohiophenes, dibenzofurans, diazapines, oxahelicines, thia-oxa helicines, and various other heterocycles under base mediated conditions. Use of suitably substituted 2H-pyran-2-one and nucleophile can generate new molecular entity. In this review, we have discussed the properties, synthesis and their synthetic applications to generate various classes of heterocycles.

A fused heterocyclic scaffold combining the imidazo[1,2-a]pyridine with a substituted chromone was synthesized in a one-pot procedure. The reaction proceeds by intramolecular cyclization of 1-(2-imino-2H-chromen-3-yl)pyridinium chloride in ethanol and in the presence of DABCO. A detailed study of the experimental conditions allowed a clear understanding of the reaction pathway.

Small heteroaromatic rings are valuable monomers in drug discovery that can enable rapid access to novel and desirable chemical space. Installation of a synthetic handle on a heteroaromatic core may be difficult if steric and electronic factors are not in alignment with the desired transformation. Described are practical routes for the construction of 5-bromo-6-methyl imidazopyrazine (1) as well as 5-bromo and 5-chloro-6-methyl imidazopyridines (2a and 2b), which were developed using electron density surface maps encoded with ionization potential to guide synthetic strategy.

A second generation total synthesis of the marine sponge metabolite ageladine A utilizing a biogenetically inspired 6π-2-azaelectrocyclization of triene 34 as the key step is performed to construct the imidazopyridine core of the metabolite.

Annelation of pentafluoropyridine via an intramolecular nucleophilic aromatic substitution process with benzamidine gave an imidazopyridine system in high yield in a two step process whilst alkyl amidines gave 4-aminotetrafluoropyridine by a competing elimination reaction. 4-Phenylsulfonyl tetrafluoropyridine reacts with amidines to give the corresponding imidazo[4,5-b]pyridine systems. In contrast, 4-cyanotetrafluoropyridine gave a [6,6]-fused pyrimidinopyridine system arising from initial nucleophilic substitution at the C-3 position of the pyridine ring followed by intramolecular cyclization onto the pendant cyano group. The systems prepared by this annelation methodology further demonstrate the utility of perfluorinated heterocyclic substrates for the synthesis of heterocyclic scaffolds that possess multiple functionality and have potential applications in the drug discovery arena.

The dynamic NMR analysis of 2, a subunit of a new class of cyclic-dependent kinase inhibitors, reveals that the compound exists as two conformational isomers, Z and E, in acetone, as a consequence of the restricted rotation about the imidazopyridine–carbonyl bond. The less hindered Z-rotamer is the most abundant conformer (85:15 Z/E at 233 K) and the free energy of activation of the interconversion is 13.2 kcal mol−1. The rotamer ratio and the interconversion barrier are similar in other solvents, such as CD3OD and CDCl3.

A synthesis of all possible N-alkylated 2-n-butyl-imidazo[4,5]pyridine isomers is described as well as their structural assignment by 1H NMR spectroscopy. One of these derivatives, 2,-n-butyl-3-[2′- (1H-tetrazol-5-yl)-4-biphenylylmethyl]-3H-imidazo[4,5-b]pyridine 9 is a potent angiotensin II receptor antagonist.
