New polystyrene-based resins containing benzyl and aryl vinyl sulfone groups are described. The vinyl sulfone group reacts efficiently with 2° amines, via conjugate addition, and the resin-bound 3° amine products can be quatermised through alkylation. Subsequent deamination to give 3° amines and the regenerated vinyl sulfone occurs in moderate to good yield. Both systems can be recycled and show moderate stability to acids and high stability to nucleophiles including Grignard reagents.

The REM resin for solid phase synthesis is described. Its use is illustrated by preparing a small array of tertiary amines using a Hofmann elimination reaction. No functional group is required for linking these compounds onto the resin other than the amine constructed during the synthesis.

A new method for the synthesis of amino amides using Wang or Tentagel-PHB resins is described. The method uses aluminium chloride to promote the conversion of resin-bound benzylic esters to amides under ambient conditions. The reactions were monitored in ‘real-time’ using 13C gel phase NMR.

We utilize a 1,6-disubstituted indane as a template onto which two amino acid side-chains are appended in an orientation which mimics that found in a protein alpha-helix motif.

Synthetic methodology for the introduction of a 2,5,5-trisubstituted imidazoline ring as an amide replacement into dipeptide derivatives of Phe, Trp, Lys(N 6 -CBZ) and Nle is described. This replacement is also incorporated into the Trp-Nle bond of the tetrapeptide CCK-4 derivative Trp-Nle-Asp-Phe-NH 2 and the pentagastrin derivative Gly-Trp-Nle-Asp-Phe-NH 2

Reaction of cyclohexadienylium-Fe(CO) 3 complexes (7) and (8) with sodiomalono-nitrile gives the carbo-spirocyclic derivatives (9) and (10), whilst reaction with benzylamine and removal of iron leads to the azaspirocyclic enones (15) and (16), respectively.

A method of constructing the quaternary carbon, corresponding to C-20 of Aspidosperma alkaloids, via tricarbonylcyclohexadienyliumiron salts is described.
