Herein we describe an efficient one pot strategy toward highly functionalized isoquinolone fused azabicycles having great synthetic potential via C–H activation of N -pivaloyloxy benzamides under very mild conditions. The reaction is accomplished at room temperature within one hour in good to excellent yields and is found to be compatible with a range of diazabicyclic olefins and benzamides. The present strategy offers an easy route for the synthesis of biologically relevant compounds which possess multiple points for divergent synthesis. N–N bond cleavage of synthesized compounds may enable their significant role as effective precursors for the preparation of diaminocyclopentane fused isoquinolones.

A new strategy for the synthesis of novel zerumbone derivatives via a regioselective palladium catalyzed decarboxylative coupling reaction using arene carboxylic acids is described. The current methodology involves the repositioning of endocyclic double bond to exocyclic double bond of zerumbone. Preliminary in vitro analysis revealed that most of the newly synthesized derivatives are potent α-glucosidase enzyme inhibitors.

An efficient strategy for the stereoselective synthesis of functionalized cyclopentenes and spiro[2.4]heptenes from strained olefins via C–H activation of aryl ketone O-acetyl ketoximes using [RhCl2Cp∗]2 catalyst is described. The results revealed that a wide range of readily accessible aryl and heteroaryl ketoximes are compatible in this method for the ring opening of bicyclic and spirotricyclic olefins.

Synthesis and biological evaluation of novel alkylidenecyclopentene glycohybrids via palladium and click chemistry are described. Out of the six molecules synthesized, four molecules were screened against α-glucosidase, α-amylase, and glycation reactions. The results showed that the compound 7f with a galactose appendage is a good α-glucosidase inhibitor and a potent anti-glycation agent under in vitro conditions with an IC50 value of 0.075 μM and 0.022 μM, respectively. The toxicity of the compounds against normal cell lines was checked by MTT assay.

Various Lewis acids have been examined as agents which react with N -iodosuccinimide to release iodonium ion for activating n -pentenylorthoester (NPOE) donors, particularly for coupling to acceptors that have acid-labile protecting groups. Although many of the reagents do not tolerate cyclic acetals, BF 3 ·Et 2 O does. However, the latter cleaves p -methoxybenzyl (PMB), making it possible to use this Lewis acid to simultaneously promote coupling of NPOEs and remove PMB groups. Of several transition metal salts investigated, ytterbium triflate is outstanding in its ability to promote NPOE coupling with complete preservation of acid-labile protecting groups.
