Abstract

The mycotoxin rubratoxin B binds to some component(s) of the microsomal fraction of mouse liver. The α-β unsaturated lactone ring was a structural requirement for binding. The rubratoxin B-microsomal adduct was not separated by Sephadex G-25 filtration. The binding occurred rapidly and persisted for at least 2 hr. Sulfhydryl reagents, dithiothreitol and cysteine partially protected from and/or reversed binding. Rubratoxin B inhibited the in vitro activity of an enzyme located in the endoplasmic reticulum associated with binding of the ribosomes to the membrane.
