Present investigation was undertaken on three buffalo bulls (Murrah) to study the leakage of phosphatases during cryopreservation. A total of 18 collections from three bulls (six collections per bull) were utilized for the study. After initial evaluation, each semen sample was diluted in two different extenders viz. Tris egg yolk extender which acted as control and Soya-milk extender as experimental. At the equilibration and post-thaw stages leakage of phosphatases viz. alkaline phosphatase (AKP) and acid phosphatase (ACP) were studied. There was a significant difference in the levels of AKP and ACP at post thaw stages in between the extenders. The leakage of phosphatases was found to increase from pre-freeze to post-freeze stage in both the extenders with better results in Tris egg yolk. [ABSTRACT FROM AUTHOR]
