The four serotypes of dengue virus (DENV) are the leading etiologic agent of disease caused by arthropod-borne viruses (arboviruses) in the world, with billions at risk of DENV infection spread by infected mosquitoes. DENV causes illness ranging from dengue fever (DF) to life-threatening dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS). DENV proliferates well in two different host systems, an invertebrate mosquito vector and vertebrate primate host, which have a distinct difference in their preference of codon pairs (CP) for translation (different “codon pair bias”). Consequently, arboviruses must delicately balance the use of codon pairs between mammals and arthropods, which presents an Achilles’ heel that we have exploited by specifically shifting the codon pair preference in the E and NS3 ORFs away from mammals while keeping the CPB favorable for mosquito ORFs. Here we report that recoding of the ORFs has led to variants that were over-attenuated in rhesus macaques although induction of protective antibodies in animals vaccinated with the smallest recoded ORF (E) was observed. The flexibility of our synthetic vaccine design (by decreasing the number of unfavorable CPs in the E ORF), allowed us to construct two new vaccine candidates (EhminA and EhminB) with intermediate attenuation in cell culture and neonatal mice, a result demonstrating proof of concept. New DENV vaccine candidates are being developed based on selective attenuation by dramatic recoding, with flexibility in balancing the attenuation and immunogenicity by marrying rational design and empirical modification.

Several mouse cell lines expressing hybrid human poliovirus receptors (hPVRs) bearing mutations in the first immunoglobulin-like domain were previously characterized for their defective binding and replication of poliovirus type 1 Mahoney (G. Bernhardt, J. Harber, A. Zibert, M. DeCrombrugghe, and E. Wimmer,Virology,203, 344–356, 1994). Here we report that these mutant hPVRs were utilized to explore differences in the binding behavior of the three serotypes of poliovirus. Type 3 polioviruses (both Sabin and the neurovirulent Leon strain) clearly bound to the hPVR mutant Q130G/GD, but were incapable of initiating infection. Also, binding at 25° of poliovirus types 2 and 3 to cell lines expressing the hPVR mutants P84SYS/HPGA, L99GAE/AAAA, and D117F was greater than type 1 poliovirus. Further study of the serotype-specific interaction with mutant hPVRs was accomplished with antigenic hybrid viruses. Improved binding by antigenic hybrid viruses demonstrated that serotype-specific binding to mutant hPVRs is, in part, determined by the amino acid sequence of neutralization antigenic sites (NAgs) and the probable conformational rearrangement of amino acids adjacent to the NAg sites. Finally, site-directed mutants of poliovirus were utilized to determine the relative contributions, to hPVR interactions, of individual amino acids with solvent accessible side chains in the viral canyon. Of the 18 viable virus mutants produced, 3 (D1226A, I1089A, and VPEK1166HPGA) expressed impaired replication phenotypes on the mutant hPVR cell lines P84SYS/HYSA and D117F. A location at the rim of the poliovirus canyon was implicated for the interaction of the amino terminal domain of the poliovirus receptor with conserved and serotype-specific viral surface amino acids. The possible involvement of elements of neutralization antigenic sites in receptor binding may explain, in part, why poliovirus exists in only three serotypes.

Translation initiation of encephalomyocarditis virus (EMCV) mRNA occurs by ribosomal entry into the 5′ untranslated region. Internal ribosome binding to EMCV mRNA requires a viral cis -acting element, termed the internal ribosomal entry site (IRES), and cellular trans -acting factors. The polypyrimidine tract binding protein (PTB) has been identified as one such trans -acting factor required for EMCV IRES-dependent translation. Using a dicistronic mRNA and an in vitro translation system, we have identified cis -acting elements of the EMCV IRES required for IRES-dependent translation. The results identify several regions of the IRES that are required for efficient IRES-dependent translation, including the PTB binding site. Other regions of the IRES may act only as spacer sequences, analogous to the spacer sequences found in rhinovirus and enterovirus IRES elements, to link essential regions of the IRES together. The flexibility of one region of the EMCV IRES was demonstrated by an insertion of 125 nucleotides that had little effect on IRES function while the constraint imposed on another region was demonstrated by a 3-nucleotide deletion that nearly abolished IRES-dependent translation.

We report the construction of a poliovirus genome [pPVM-VPg(3F4A)] harboring a double mutation in VPg. This mutant, in which the tyrosine and the threonine at residues 3 and 4 of the VPg region were replaced by phenylalanine and alanine, respectively, is lethal, that is, all RNA synthesis was abolished and no revertants could be isolated. Using the properties of dicistronic polioviruses (with the general genotype PV 5′NTR-3AB-EMCV IRES-PV ORF-3′NTR), we have observed that the defect in RNA synthesis of the VPg(3F4A) mutant could be rescued by providing wild-type protein 3AB from the first open reading frame in trans. We conclude that the 3AB provided by the first cistron of the dicistronic construct was capable of "intragenomic complementation." Intragenomic complementation, however, was inefficient. Thus, the dicistronic RNAs were only quasi-infectious, and even first-passage viruses were found to have reverted to a functioning VPg in the polyprotein. This phenomenon underlines the role of polypeptide 3AB in multiple functions of viral proliferation. First-passage viruses, all of which expressed a small-plaque phenotype, had retained the original dicistronic genotype. At the fourth passage, however, all isolates were monocistronic, and they displayed complex genetic rearrangements revealing interesting information regarding IRES function.

Poliovirus translation is initiated at AUG743, 154 nt downstream of a conserved heptanucleotide CUUAUGG at the 3' border of the internal ribosome entry site. AUG586 is part of this motif and is normally not an initiation codon, but was activated following alteration of its context from CUUAUGG to ACCAUGG. Initiation at AUG586 was efficient and yielded a 7.2-kDa polypeptide translated in an open reading frame that overlapped AUG743 by 38 nt, but the presence of this activated codon reduced initiation at AUG743 by only 50%. Growth of a mutant poliovirus W1-5NC-1 containing the CUU-->ACC substitutions was impaired and was not alleviated by a termination codon placed four triplets downstream of AUG586 in the virus W1-5NC-2. The virus W1-5NC-6 contained the substitution U584A and had a similar sp phenotype; the phenotype of W1-5NC-1 is thus probably due to substitution within the conserved CUUAUGG motif per se rather than to activation of AUG586. A sp mutant virus W1-5NC-3 was derived from W1-5NC-1 by deletion of nt 588-745, indicating that AUG586 could initiate translation in vivo. These observations indicate that although AUG586 can be activated by upstream substitutions, it is nevertheless readily bypassed by ribosomes in mRNAs containing wt downstream elements, resulting in initiation at AUG743.

The N-terminal domain 1 of the human poliovirus receptor (hPVR), a three-domain, immunoglobulin-like molecule, was previously shown to be necessary and sufficient to confer poliovirus (PV) susceptibility to mouse cells. However, studies with truncated versions of hPVR suggested that the C-terminal hPVR domains may contribute to receptor function. We describe sets of hybrid receptors, constructed between hPVR and hICAM-1 (human intercellular adhesion molecule-1) that were tested in mouse cells for hPVR functionally. Whereas the context in which hPVR is expressed is of minor importance, all three domains of hPVR are required to reach wild-type function. Single and multiple amino acid exchanges were introduced into the first hPVR domain in order to localize regions that were involved in virus-receptor interactions. The mutations were analyzed for their ability to bind PV1 (Mahoney) or monoclonal antibodies as well as their ability to support viral replication in either the hPVR alpha or hybrid hPVR-hICAM-1 receptor context. When placed into a model of the V domain of hPVR, the effect of the mutations indicated that the C'C"D as well as the DE region harbored amino acids that contacted the PV1(M) surface in the process of receptor-virus complex formation. The binding of the virus to the receptor and subsequent uptake into the cells were linked; no hPVR mutants were observed that bound the virus but blocked infection. N-glycosylation of the four sites in domains 1 and 2 is not required for hPVR function, but glycosylation in domain 1 has a greater effect on receptor function than that of domain 2.

Poliovirus protein 2C contains near its N-terminus a putative amphipathic helix which is well conserved among picornaviruses. Three mutants were constructed within this region by site-directed mutagenesis. In the first mutant (pT7XL2-2C-N1) two glutamic acids were replaced with valines at the boundary of the charged and uncharged faces of the helix. The second mutant (pT7XL2-2C-N2) contains an isoleucine to lysine change in the hydrophobic half; in the third mutant (pT7XL2-2C-N3) two lysines were replaced with threonines in the hydrophilic half of the helix. Upon transfection of HeLa cells with RNA transcripts made from these plasmids only pT7XL2-2C-N1 yielded viable virus (W1-2C-N1) which had a small-plaque phenotype. A large-plaque revertant of this virus, W1-2C-N1R, was found to contain the original glutamic acid at one of the mutated sites (E19). There is no detectable minus-stranded RNA synthesis following transfection of HeLa cells with transcript RNAs of the other two plasmids, pT7XL2-2C-N2 and -N3. In vitro translation of these two mutant RNA transcripts in HeLa extracts revealed processing abnormalities in the P2/P3 region of the polyprotein. This leads to a nearly complete absence of 2C and 3AB, which might be the primary cause of defective viral RNA synthesis. The putative amphipathic helix was found to overlap a consensus binding site for double-stranded RNA.

Poliovirus 2C is a nonstructural polypeptide proposed to function in viral RNA replication. Poliovirus 2C is a member of a rapidly expanding family of proteins containing a consensus for nucleotide binding (NTP-B). Site-directed mutagenesis of conserved residues in the consensus A and S sites have suggested a functional role for the NTP-B motif in viral RNA replication and proliferation of poliovirus. We have expressed wildtype 2C and a 2C mutant, carrying a single amino acid exchange in the NTP-B motif A (Lys135Gln) using the baculovirus system. Both wildtype and mutant proteins are membrane associated. Following membrane solubilization, we have purified wildtype and mutant proteins to near homogeneity using conventional chromatography. We present biochemical evidence that wildtype 20 copurifies with an ATPase activity that is absent in the mutant preparation.

A dicistronic poliovirus W1-P1/E/P2,3-1 with the genotype [PV]5'NTR-P1-[EMCV]IRES-[PV]P2,3-3'NTR (Molla, Jang, Paul, Reuer, and Wimmer, 1992, Nature 356, 255) was used to investigate whether the viral proteinase 2Apro, whose primary function in proteolytic processing was erased through the insertion of an internal ribosomal entry site (IRES) element into the ORF of the polyprotein, had other function(s) in viral replication. Deletion of 2Apro from W1-P1/E/P2,3-1 rendered the corresponding transcripts unable to replicate whereas partial deletion of 2Apro or an exchange of Cys109 (an amino acid of the catalytic triad of the proteinase) to Ala reduced RNA replication. No cytopathic effects were observed after transfection with any of the three dicistronic constructs containing mutant 2A, and no virus was recovered after attempts to expand a possibly low yield of mutant virus. In contrast, insertion of the IRES of encephalomyocarditis virus (EMCV) into the ORF of the poliovirus polyprotein at the cleavage site between 2Apro and 2B yielded the novel dicistronic virus W1-P1,2A/E/2BC,P3-1 with the genotype [PV]5'NTR-P1-2A-[EMCV]IRES-[PV]2BC-P3-3'NTR, expressing a small plaque phenotype. These results indicate that neither the intact P2 polypeptide nor the cleavage fragment 2AB of P2 is required for viral proliferation. On the other hand, 2Apro appears to be an essential component in RNA replication as no viral RNA synthesis can be observed by reverse transcription/PCR in cells transfected with dicistronic RNA lacking this viral polypeptide.

Poliovirus polypeptide 2C is a nonstructural protein involved in replication of the viral genome. Analysis of the primary amino acid sequence of 2C shows homology to a family of proteins which contain a nucleoside-triphosphate (NTP)-binding motif. This motif consists of elements “A” ( 2 5 hydrophobic stretch) G/AXXGXGKS/T, where X stands for any amino acid, and “B” ( 3 5 hydrophobic stretch) D or DD/E. To assess the significance of the consensus sequence in 2C, we have engineered point mutations into the most conserved residues in the A and B sites and tested their effect on viral RNA replication in vivo and translation in vitro. Whereas in vitro translation of synthetic RNAs carrying mutations in the NTP-binding motif showed efficient processing of all viral proteins, indistinguishable from that of the parental strain, transfection of the RNAs into HeLa cells did not give rise to infectious virus. No viral RNA replication could be detected in cells transfected with mutant RNAs. However, revenants to the wild-type genotype in the A and B sites were obtained which gave rise to wild-type RNA synthesis, but pseudorevertants or second-site suppressors were not observed. Thus, viral RNA synthesis is greatly reduced but not entirely abolished in cells transfected with mutant RNAs. These results strongly suggest a functional role for the proposed NTP-binding motif of 2C in RNA replication and proliferation of poliovirus.

The primary functions of viral capsids are (a) to protect the encapsidated nucleic acids from degradation, (b) to interact with specific cellular receptors on susceptible host cells in a manner that results in internalization of the genome, (c) to selectively package the newly synthesized genome, and (d) to facilitate its eventual exit. Poliovirus provides an attractive model to study the relationship between these different biological functions and the structural rearrangements that virions undergo to accommodate them at various points in the viral life cycle. Poliovirus has a relatively simple composition (Ruecker-t, 1990) several assembly precursors and intermediates have been identified and characterized (Rueckert, 1990; Putnak and Phillips, i 981) and the three dimensional structure of the mature virion has been determined by X-ray crystallography (Hogle et al., 1985; Filman et al., 1989). Two recent developments have accelerated progress in characterizing the complementary processes of poliovirus assembly and disassembly. First, the availability of infectious cDNA copies of the poliovirus genome (Racaniello and Baltimore, 1981) and of synthetic infectious RNA transcripts thereof (van der Werf et al., 1986) has allowed detailed characterization of individual steps in these two processes following genetic and biochemical analysis of defined mutants, and second, new means of analyzing intermediates have been developed (Flore et al., 1990; Fricks and Hogle, 1990).

In order to study the properties of foreign antigenic sites expressed on poliovirus a hybrid was constructed in which neutralization antigenic site IA of poliovirus type 1 strain Mahoney [PV1(M)] was replaced by neutralization immunogenic site IA (NImIA) of human rhinovirus 14 (HRV14). The resulting hybrid was viable, but growth was impaired in comparison to PV1(M). The hybrid expressed both PV1(M) and HRV14 antigenic determinants. When inoculated into rabbits it elicited neutralizing antibodies against both PV1(M) and HRV14. Furthermore, the hybrid was efficiently neutralized by polyclonal antisera specific for either PV1(M) or HRV14 and by three out of five monoclonal antibodies directed to NImIA. The monoclonal antibodies also blocked binding of the hybrid to the cellular receptor for poliovirus. One of them is thought to neutralize rhinovirus in this manner, and it appears that NImIA is expressed in a sufficiently favorable context on the hybrid for the same mechanism to be effective. This can be interpreted to mean that the interactions between the parental viruses and their respective cellular receptors are very similar.

We have used cDNA encoding the cellular receptor for poliovirus (PVR) to prepare polyclonal antisera against beta-galactosidase PVR fusion proteins. One of these antisera allowed identification of a glycoprotein doublet band of about 67 kDa in membrane preparations of HeLa cells and in a PVR cosmid-bearing mouse cell line. In vitro translation of PVR-specific transcripts gave rise to a protein of 46 kDa; the product had a molecular weight of 67 kDa when microsomal membranes were added to the cell-free extract. Overexpression of PVR cDNA in mouse L-cells by means of a recombinant vaccinia virus led to the synthesis of a glycoprotein having a molecular weight identical to that of the glycosylated in vitro product. The vaccinia virus-mediated protein was also recognized by a monoclonal antibody that blocks poliovirus infection. Its biological activity was demonstrated by poliovirus binding and infectivity assays. The data show that PVR is a glycoprotein of 67 kDa and that this protein is sufficient to confer poliovirus susceptibility to mouse cells.

The human receptor for poliovirus (CD155) is an immunoglobulin-like molecule with unknown normal function(s). Here we provide evidence that CD155 binds specifically to vitronectin with a dissociation constant ( K d ) of 72 nM as determined by surface plasmon resonance. Based on sequence homology to the CD155 gene, three poliovirus receptor-related genes ( PRR1, PRR2, and PRR3 ) were cloned recently. PRR proteins were reported by others to mediate homophilic cell adhesion. Neither PRR1 nor PRR2 binds poliovirus and it is assumed that their physiological functions differ from that of CD155. Indeed, mPRR2 was found to bind to vitronectin only weakly, while its self-adhesion activity is characterized by a K d of 310 nM. Moreover, there is no evidence for CD155 self-adhesion. Both CD155 and vitronectin colocalize to follicular dendritic cells and B cells inside the germinal centers of secondary lymphoid tissue (tonsils)—an observation suggesting that the CD155/vitronectin interaction is required for the establishment of a proper immune response in this particular context.

Based on previous studies of dicistronic polioviruses carrying two internal ribosomal entry sites (IRESes), we performed a novel experiment of IRES scanning through a polypeptide by inserting sequentially the IRES of encephalomyocarditis virus into the open reading frame (ORF) of the poliovirus polyprotein at selected 3C pro -specific Q*G cleavage sites. No cytopathic effects were observed after transfection of HeLa cells with any of the dicistronic constructs, and no virus was recovered. In vitro translation of the dicistronic RNA transcripts in HeLa cell-free extracts revealed that multiple defects in the processing of the P2–P3 domain of the polyprotein is the primary reason for the lethal phenotypes. Surprisingly, the interruption of 3C pro -catalyzed cleavages downstream of 2C interfered with the 2A pro -catalyzed, primary cleavage between P1 and P2. In contrast, insertion of a foreign coding sequence (V3 loop of human immunodeficiency virus type 1 gp120) into the ORF of the polyprotein at the 2C–3A junction yielded a viable virus that appeared to be genetically stable over several passages. The results of these experiments, which are generally applicable to analyses of viral polyproteins or multidomain polypeptides, suggest that processing of the P2–P3 domain by 3C–3CD pro is rapid and accurate only in the context of the unperturbed P2-P3 precursor; this is consistent with cleavages occurring in cis. Moreover, an intact 2C–3A precursor is not required for viral proliferation.

The expression of the human poliovirus receptor (hPVR) in several cultured cell lines was studied with the use of different antibodies directed against hPVR proteins. Immunoprecipitations of metabolically labeled cell lysates revealed that membrane-bound glycoforms of hPVR proteins have a molecular weight of about 80 kDa. By applying inhibitors of the glycosylation pathway (deoxymannojirimycin and swainsonine) we were able to monitor the modification of the hPVR glycoproteins when passing through the processing pathway. We show that a 67-kDa hPVR protein identified earlier (using a vaccinia virus expression system) is an intermediate glycoform probably located in the endoplasmic reticulum or cis-Golgi. Further modification of this glycoform is blocked by vaccinia virus infection or by the inhibitor deoxymannojirimycin. We, therefore, conclude that the 80-kDa glycoforms identified here are the fully processed hPVR isoforms. Surface iodination confirms that only the 80-kDa glycoforms are expressed on the cell surface. Treatment with various deglycosylating enzymes (N-Glycanase, O-Glycanase, Endo-H, and neuraminidase) demonstrates that the hPVR proteins bear sialylated complex-type oligosaccharides. Deglycosylation of the hPVR proteins also reveals the presence of both hPVR membrane-bound forms in cultured cells. Their relative expression levels, with respect to each other, vary considerably. The distribution of these hPVR isoforms in tissues may help explain the natural function of the hPVR proteins.

In order to determine whether any primary human blood cells have the ability to replicate poliovirus (PV), peripheral blood cell components were isolated and analyzed for their cell surface expression of the poliovirus receptor (PVR). Following two-color immunfluorescence staining with lineage-specific markers, the cells were analyzed by flow-cytometric methods. PVR cell surface expression was detected on most mononuclear cells expressing CD14, a marker for mononuclear phagocytes. There was no PVR cell Surface expression oh platelets ned extremely low levels on polymorphonuclear leukocytes. Mononuclear leukocytes from Ficoll density centrifugation were found to support PV replication. The finding of PVR on mononuclear phagocytes and the ability of primary human blood cells to support PV replication in the absence of cultivation has implications for both the normal and pathogenic role of PVR.

Poliovirus and foot-and-mouth disease virus (FMDV) initiate infection by binding to specific cell surface receptors, which is followed by a poorly understood disassembly process. To probe these early steps of infection, the ability of poliovirus and FMDV to infect cells following binding through an alternative receptor was examined. For these studies, a Chinese hamster ovary (CHO) cell line expressing the B2 isoform of the murine Fc receptor (FcR) was used. Both viruses were able to bind to this cell line in an antibody-dependent manner, but only FMDV was able to productively infect these cells following binding through the FcR. These results suggest that the natural poliovirus receptor has dual functions in binding and destabilizing the virus particle, whereas the putative FMDV receptor may only be necessary for virion binding. These findings are consistent with differences in virion architecture which predict a more intimate virion-receptor association for poliovirus than for FMDV.

By in vitro recombination between the wild-type full-length infectious cDNA of poliovirus and a clone generated by the construction of a cDNA bank from a chemically derived temperature-sensitive plurimutant, we obtained a mutant cDNA with a T to C change at nucleotide 5658. This mutation replaces the isoleucine at residue 74 of the viral protease 3C by a threonine. The mutant virus recovered after transfection exhibited a small-plaque phenotype, and was deficient for viral RNA synthesis. Both these defects were more marked at 39 than at 37°. The mutation was introduced into a bacterial plasmid which expresses the 3C protease along with its flanking autocatalytic cleavage sites. Analysis of the cleavage products expressed in Escherichia coli provided direct evidence that the modification impaired cleavage at the COOH-terminus of 3C. Cleavage at this same site was partially defective in mutant virus-infected HeLa cells, reducing the production of mature 3C and the viral replicase, 3D. Cleavage of P1, the precursor to the capsid polypeptides, was apparently unaffected by this defect, whereas cleavage events within the P2 region of the genome occurred inefficiently. This is indicative of differential strategies for 3C-specific cleavage events in vivo .

Fingerprints have been prepared of RNase T1- and RNase A-resistant oligonucleotides of poliovirus type 1 (Mahoney) and its attenuated derivative, the Sabin vaccine strain LSc 2ab. Changes in the position of seven oligonucleotides have been observed and correlated by sequence analysis with transversions and transitions. Based on mapping procedures ( A. Nomoto, A. Jacobson, Y. F. Lee, J. J. Dunn, and Eckard Wimmer, 1979 , J. Mol. Biol. 128 , 179–196) and sequence studies ( N. Kitamura and E. Wimmer, 1980 , Proc. Nat. Acad. Sci. USA 77 , 3196–3200) the mutations have been localized within the viral genome. Assuming random distribution of the mutations our results imply that approximately 35 base changes may have occurred during attenuation of poliovirus type 1 (Mahoney). A catalogue of sequences of 55 RNase T1- and 16 RNase A-resistant oligonucleotides has been established.

Immuneprecipitation analyses of various picornavirus-infected cell lysates were performed using antisera to poliovirus type 1-specific structural and nonstructural proteins. The results showed differing patterns of antigenic conservation and divergence. However, the VP3 and 2C polypeptides were strongly antigenically conserved among the large majority of these viruses. This conservation was especially notable given the degree of divergence exhibited by the other viral proteins and may be due to environmental pressure exerted by interaction with the host cell. The results, furthermore, allowed for an analysis of the evolutionary relationship of the tested viruses. This analysis showed a particularly strong antigenic relationship between the proteins of the poliovirus group and coxsackievirus A21 as well as a weaker, but significant, relationship with coxsackieviruses B1 and B3.

Independent substitution mutations have been detected in capsid polypeptide VP1 of the type 1 oral poliovirus vaccine isolated from normal infant vaccine recipients. These mutations map at amino acid residues 142 and 147 of VP1, a region only minimally hydrophilic. A synthetic peptide, corresponding to residues 141 to 147 of VP1 was synthesized, conjugated to a carrier polypeptide of bovine serum albumin. The conjugate was found to elicit a weak poliovirus neutralizing antibody response. It was also capable of priming the immune system for the production of IgG-type antibodies able to neutralize greater than 99.999% of infectious type 1 virus. It is suggested that region 141 to 147 of VP1 may be involved in neutralization of the virus and that the mutants may have accumulated by antibody selection.

Poliovirus protein 2A contains a short amino acid sequence that also occurs in the putative active site of the known viral proteinase, 3C, previously shown to be responsible for glutamine/glycine cleavages in the poliovirus polyprotein precursor. Experimental evidence indicates that 2A is a second viral proteinase that mediates the cleavage of two tyrosine/glycine cleavages in the generation of virus-specific proteins. Since poliovirus inhibition of host cell protein synthesis correlates with the specific cleavage of the 220,000-Da component of the cap binding protein complex, we have tested whether viral protein 2A contains the p220 cleavage activity. The results show that (1) 2A does not copurify with p220 cleavage activity, (2) partially purified fractions containing high p220 cleavage activity contain no detectable 2A sequences in the form of either mature or precursor protein, and (3) anti-2A serum or IgG does not inhibit p220 cleavage in vitro .

VP4, one of the poliovirus structural proteins, was purified and used to prepare rabbit anti-VP4 serum. In addition to the anti-VP4 activity, this serum was also found to contain significant anti-VP3 and antivirion activities. The serum also effectively neutralized viral infectivity. The ease with which nonneutralizable variants were obtained indicated that neutralization was due to an antibody population which bound to a single virion epitope. Antigen saturation and immunoprecipitation experiments demonstrated that antibody to this epitope was also responsible for the serum's antivirion and anti-VP3 activities, as well as for a part of the anti-VP4 activity. The identification of a neutralization epitope most probably present on VP3, which cross-reacts with a site on denatured VP4, is the first report of such an epitope on a poliovirus structural protein other than VP1.
