The release of nanoplastics (NPs) into the environment is growing due to the extensive use of plastic products. Numerous studies have confirmed the negative effects of NPs on microorganisms, which poses uncertainties concerning their impact on nanofiltration (NF) membrane biofouling. This study investigated the initial cell adhesion process, NF membrane biofouling kinetic processes and bacterial responses of Pseudomonas aeruginosa (P. aeruginosa) exposed to varied NPs concentrations (0–50 mg·L−1). Transcriptome analysis demonstrated that low concentration of NPs (0.1 mg·L−1) promoted bacterial quorum sensing, energy metabolism, exopolysaccharide biosynthesis and bacterial secretion systems. Correspondingly, the polysaccharide content increased remarkably to 2.77 times the unexposed control, which served as a protective barrier for bacteria to avoid the impact of NPs-induced stress. Suppressed homologous recombination, microbial metabolic potentials and flagellar assembly were detected in bacteria exposed to a high concentration (50 mg·L−1) of NPs, mainly due to the triggered reactive oxygen species (ROS) generation, genomic DNA damage, and decreased energy production. Overall, enhanced formation of the extracellular polymeric substances (EPS) and aggravated membrane flux decline were observed when NPs interacted with the membrane surface by cell secretions (low NPs levels) or cell lysis (high NPs levels). These findings shed light on understanding the microbial metabolism mechanism and membrane biofouling propensity with NPs stress at both the molecular and gene levels. • The impact of antibiotic resistance on initial bacterial adhesion and biofilm formation on plastics was explored. • Single-cell adhesion spectroscopy was used to probe the initial cell adhesion on plastics. • Antibiotic resistance promoted biofilm growth on plastics by enhancing initial bacterial adhesion in antibiotic environments. The microbial biofilm formed on plastics, is ubiquitous in the environment. However, the effects of antibiotic resistance on the development of the biofilm on plastics, especially with regard to initial cell attachment, remain unclear. In this study, we investigated the initial bacterial adhesion and subsequent biofilm growth of a rifampin (Rif) resistant E. coli (RRE) and a normal gram-positive B. subtilis on a typical plastic (polyethylene, PE). The experiments were conducted in different antibiotic solutions, including Rif, sulfamethoxazole (SMX), and kanamycin (KM), with concentrations ranging from 1 to 1000 μg/L to simulate different aquatic environments. The AFM-based single-cell adhesion force determination revealed that Rif resistance strengthened the adhesion force of RRE to PE in the environment rich in Rif rather than SMX and KM. The enhanced adhesion force may be due to the higher secretion of extracellular polymeric substances (EPS), particularly proteins, by RRE in the presence of Rif compared to the other two antibiotics. In addition, the higher ATP level of RRE would facilitate the initial adhesion and subsequent biofilm growth. Transcriptome analysis of RRE separately cultured in Rif and SMX environments demonstrated a clear correlation between the expression of Rif resistance and the augmented bacterial adhesion and cellular activity. Biofilm biomass analysis confirmed the promotion effect of Rif resistance on biofilm growth when compared to non-resistant biofilms, establishing a novel association with the augmentation of microbial adhesion force. Our study highlights concerns related to the dissemination of antibiotic resistance during microbial colonization on plastic that may arise from antibiotic resistance. Abstract: The work provides a method on the basis of flow cytometry to evaluate the performance of denitrification biofilm during the preservation, reactivation and pilot-scale operation process. The viable cell ratio of denitrification biofilm significantly reduced and further led to the decrease of denitrification capacity after long-term preservation for 5 months. Protein component in tightly bound extracellular polymeric substances (TB-EPS) could serve to enhance microbial adhesion and promote denitrification biofilm formation. With the significant correlation of viable cell ratio and microbial characteristics, 4 °C was more appropriate for preserving denitrification biofilm and conducive to maintain the relatively high denitrification capacity. A maximum denitrification rate of 5.80 gNO 3 --N/m2·d was obtained in pilot-scale anoxic-oxic (AO) process and Dechloromonas became greater prevalence in denitrification suspended carriers. Furthermore, the enrichment of Pseudomonas , Parcubacteria , Acidovorax , Aquabacterium and Unclassified_Flavobacteriaceae enhanced biofilm formation and nutrient conservation. The significantly positive correlation between viable cell ratio and the ratio of nitrate reduction to COD consumption was discovered, and the indices of Chao, ACE, Shannon and Simpson of denitrification biofilm were positively correlated with viable cell ratio, meaning that flow cytometry analysis was reasonable and suitable to evaluate the performances of denitrification biofilm. Abstract Fecal indicator bacteria (FIB) are used to assess fecal pollution levels in surface water and are among the criteria used by regulatory agencies to determine water body impairment status. While FIB provide no information about pollution source, microbial source tracking (MST) does, which contributes to more direct and cost effective remediation efforts. We studied a watershed in Florida managed for wildlife conservation that historically exceeded the state regulatory guideline for fecal coliforms. We measured fecal coliforms, enterococci, a marker gene for avian feces (GFD), and a marker gene for human-associated Bacteroides (HF183) in sediment, vegetation, and water samples collected monthly from six sites over two years to: 1) assess the influence of site, temporal factors, and habitat (sediment, vegetation, and water) on FIB and MST marker concentrations, 2) test for correlations among FIB and MST markers, and 3) determine if avian feces and/or human sewage contributed to FIB levels. Sediment and vegetation had significantly higher concentrations of FIB and GFD compared to water and thus may serve as microbial reservoirs, providing unreliable indications of recent contamination. HF183 concentrations were greatest in water samples but were generally near the assay limit of detection. HF183-positive results were attributed to white-tailed deer (Odocoileus virginianus) feces, which provided a false indication of human sewage in this water body. FIB and GFD were positively correlated while FIB and HF183 were negatively correlated. We demonstrated that birds, not sewage, were the main source of FIB, thus avoiding implementation of a total maximum daily load program (TMDL). Our results demonstrate that the concomitant use of FIB and MST can improve decision-making and provide direction when water bodies are impaired, and provides a strategy for natural source exclusion in water bodies impacted by wild animal feces. Abstract: Tightly bound extracellular polymeric substances (TB-EPS) play a substantial role on microbial aggregates, which can promote microbial cells to aggregate and adhere onto the carrier in bioreactor. However, the attachment and adsorption of TB-EPS on different surfaces were awaited to be elucidated. In this study, four self-assembled monolayers (SAMs) carrying methyl (CH3-SAM), amino (NH2-SAM), hydroxyl (OH-SAM), and carboxyl (COOH-SAM) terminal groups were prepared to model different surfaces. TB-EPS adsorption on these surfaces under different pH conditions and additional cations were investigated using surface plasmon resonance. The adsorption of TB-EPS dramatically decreased with the decreasing pH values. CH3-SAM surface achieved the maximum adsorption at the same condition. Na+ promoted the TB-EPS adsorbed on COOH-SAM surface. The Ca2+-mediated complexes were attracted by COOH-SAM and repelled by NH2-SAM, respectively. The adsorptions of TB-EPS on the four SAM surfaces were significantly increased by adding Fe3+. These results demonstrated that the TB-EPS adsorption on the organic surfaces were dependent on the pH and cation of solution. [Copyright &y& Elsevier]

Association with particles in the water column can have a significant impact on microbial fate and transport. This study analyzed multiple stormwater samples taken throughout the duration of three separate storms (at two different sites) to evaluate the fraction of microbes partitioning to denser “settleable” particles and to examine how partitioning behavior varied over the course of a storm. Intra-storm sampling also allowed for estimates of microbial loading rates (both total and particle-associated) and cumulative storm-induced microbial load. Five different indicator organisms were examined, with the fraction of microbes associated with settleable particles assessed via a calibrated centrifugation method. Partitioning behavior varied across microorganism type, with an average of 40% of fecal coliforms, Escherichia coli, and enterococci associating with settleable particles, compared to approximately 65% of Clostridium perfringens spores and only 13% of total coliphage. Partitioning remained fairly constant for each type of organism throughout storm events. Nonetheless, higher concentrations of both settleable particles and microbes entering the water column soon after the onset of a storm led to higher loading rates of settleable microbes in the storm''s earliest stages, a trend that could have important implications for the design of stormwater management structures (e. G. , detention basins). Estimates of cumulative storm-induced microbial loading suggested that one day''s worth of storm loading can be the equivalent of months, or even years, of dry-weather loading. Bacterial biofilm growth on a pilot lab scale anaerobic fluidized bed bioreactor under mesophilic conditions is analyzed in this paper. Biolite was used as substratum and the system was fed with acetic acid as sole carbon source.Bed expansio´n was maintained around 40%. It was observed that the decrease of the expansion provoked a biomass increase in the reactor and the decrease of the removal rate of organic matter. This can be explained by a mass transfer limitation phenomenon. Volatile fatty acids accumulated within the system when the expansion of the fluidized bed was lower than 10%.An initial bed segregation was observed. Biomass adhesion and growth influenced bioparticle size and density, which contributed to further bed segregation. Biomass concentration in the bottom part of the bed showed higher values than in the upper levels. On the other hand, biofilm density increased towards the reactor bottom, wherein it showed the highest values. Methanogenic tests were conducted using acetic, propionic and butyric acids. These assays indicated that total activity was higher in bioparticle sampled from upper reactor levels. This was explained by the lower biofilm density values in this zone and mass transfer limitation phenomena. Moreover, biomass was unable to degrade acids different from acetic while performing the activity tests. This was probably due to the fact that acetic acid was the sole carbon source fed to the reactor. [Copyright &y& Elsevier]

Biofouling is the undesired deposition and growth of microorganisms on surfaces, forming biofilms. The definition is subjective and operational: not every biofilm causes biofouling - only if a given a subjective "threshold of interference" is exceeded, biofilms cause technical or medical problems. These range from the formation of slime layers on ship hulls or in pipelines, which increase friction resistance, to separation membranes, on which biofilms increase hydraulic resistance, to heat exchangers where they interfere with heat transport to contamination of treated water by eroded biofilm cells which may comprise hygienically relevant microorganisms, and, most dangerous, to biofilms on implants and catheters which can cause persistent infections. The largest fraction of anti-fouling research, usually in short-term experiments, is focused on prevention or limiting primary microbial adhesion. Intuitively, this appears only logical, but turns out mostly hopeless. This is because in technical systems with open access for microorganisms, all surfaces are colonized sooner or later which explains the very limited success of that research. As a result, the use of biocides remains the major tool to fight persistent biofilms. However, this is costly in terms of biocides, it stresses working materials, causes off-time and environmental damage and it usually leaves large parts of biofilms in place, ready for regrowth. In order to really solve biofouling problems, it is necessary to learn how to live with biofilms and mitigate their detrimental effects. This requires rather an integrated strategy than aiming to invent "one-shot" solutions. In this context, it helps to understand the biofilm way of life as a natural phenomenon. Biofilms are the oldest, most successful and most widely distributed form of life on earth, existing even in extreme environments and being highly resilient. Microorganisms in biofilms live in a self-produced matrix of extracellular polymeric substances (EPS) which allows them to develop emerging properties such as enhanced nutrient acquisition, synergistic microconsortia, enhanced tolerance to biocides and antibiotics, intense intercellular communication and cooperation. Transiently immobilized, biofilm organisms turn their matrix into an external digestion system by retaining complexed exoenzymes in the matrix. Biofilms grow even on traces of any biodegradable material, therefore, an effective anti-fouling strategy comprises to keep the system low in nutrients (good housekeeping), employing low-fouling, easy-to-clean surfaces, monitoring of biofilm development, allowing for early intervention, and acknowledging that cleaning can be more important than trying to kill biofilms, because cleaning does not cut the nutrient supply of survivors and dead biomass serves as an additional carbon source for "cannibalizing" survivors, supporting rapid after growth. An integrated concept is presented as the result of a long journey of the author through biofouling problems. This study aims to enhance biological nitrogen removal performance by the innovative carbon-based carriers. The new carriers were produced based on recycling waste materials. In these carriers, the advantages of the hybrid system and physicochemical properties of activated carbon were integrated to promote microbial attachment. To verify the performance of the new carriers compared to the conventional moving carriers, the experiments were conducted in two parallel laboratory-scale sequencing batch reactors under various operating conditions. The analysis revealed that the specific surface area of the new carrier with a total pore volume of 0.0015cm3/gr was 10.9 times the specific surface area of a conventional carrier. Further, the comparative results indicated that the new highly porous carriers made a major contribution to increasing the attached active biomass up to 20.2%. From the data analysis (DO, ORP, and pH), it was also confirmed that the new carriers had a positive effect on the creation of a greater anoxic zone within the biofilm. Consequently, the simultaneous nitrification-denitrification and total nitrogen removal efficiencies enhanced significantly up to 14.3% and 16.8%, respectively. From the environmental and economic viewpoints, the benefits of the novel carrier showed that it is a practical alternative for the conventional carrier providing a cost-effective wastewater treatment technology.
