Mannan is a predominant constituent of cork hemicellulose and is widely distributed in various plant tissues. β-Mannanase is the principal mannan-degrading enzyme, which breaks down the β-1,4-linked mannosidic bonds in mannans in an endo-acting manner. Microorganisms are a valuable source of β-mannanase, which exhibits catalytic activity in a wide range of pH and temperature, making it highly versatile and applicable in pharmaceuticals, feed, paper pulping, biorefinery, and other industries. Here, the origin, classification, enzymatic properties, molecular modification, immobilization, and practical applications of microbial β-mannanases are reviewed, the future research directions for microbial β-mannanases are also outlined. Plastic pollution in the form of microplastics (MPs), poses a significant threat to natural ecosystems, with detrimental ecological, social, and economic impacts. This review paper aims to provide an overview of the existing research on the interaction between microbial biofilms and MPs in natural environments. The review begins by outlining the sources and types of MPs, emphasizing their widespread presence in marine, freshwater, and terrestrial ecosystems. It then discusses the formation and characteristics of microbial biofilms on MPs surfaces, highlighting their role in altering the physicochemical properties of MPs and facilitating processes such as vertical transport, biodegradation, dispersion of microorganisms, and gene transfer. Different methods used to assess these interactions are discussed, including microbiological and physicochemical characterization. Current gaps and challenges in understanding the complex relationships between biofilms and MPs are identified, highlighting the need for further research to elucidate the mechanisms underlying these complex interactions and to develop effective mitigation strategies. Innovative solutions, including bioremediation techniques and their combination with other strategies, such as nanotechnology, advanced filtration technologies, and public awareness campaigns, are proposed as promising approaches to address the issue of MPs pollution. Overall, this review underscores the urgent need for a multidisciplinary approach to combating MPs pollution, combining scientific research, technological innovation, and public engagement to safeguard the health and integrity of natural ecosystems. The traditional way of dealing with plant diseases has been the use of chemical products, but these harm the environment and are incompatible with the global effort for sustainable development. The use of Bacillus and related species in the biological control of plant diseases is a trend in green agriculture. Many studies report the positive effect of these bacteria, but a synthesis is still necessary. So, the objective of this work is to perform a meta-analysis of Bacillus biocontrol potential and identify factors that drive its efficacy. Data were compiled from articles published in journals listed in two of the main scientific databases between 2000 and 2021. Among 6159 articles retrieved, 399 research papers met the inclusion criteria for a systematic review. Overall, Bacilli biocontrol agents reduced disease by 60% compared to control groups. Furthermore, experimental tests with higher concentrations show a strong protective effect, unlike low and single concentration essays. Biocontrol efficacy also increased when used as a protective inoculation rather than therapeutic inoculation. Inoculation directly in the fruit has a greater effect than soil drenching. The size of the effect of Bacillus-based commercial products is lower than the newly tested strains. 1,2,4-butanetriol (BT) is a polyol with unique chemical properties, which has a stereocenter and can be divided into D-BT (the S-enantiomer) and L-BT (the R-enantiomer). BT can be used for the synthesis of 1,2,4-butanetriol trinitrate, 3-hydroxytetrahydrofuran, polyurethane, and other chemicals. It is widely used in the military industry, medicine, tobacco, polymer. At present, the BT is mainly synthesized by chemical methods, which are accompanied by harsh reaction conditions, poor selectivity, many by-products, and environmental pollution. Therefore, BT biosynthesis methods with the advantages of mild reaction conditions and green sustainability have become a current research hotspot. In this paper, the research status of microbial synthesis of BT was summarized from the following three aspects: (1) the biosynthetic pathway establishment for BT from xylose; (2) metabolic engineering strategies employed for improving BT production from xylose; (3) other substrates for BT production. Most biosynthetic gene clusters (BGCs) of Actinobacteria are either silent or expressed less than the detectable level. The non-genetic approaches including biological interactions, chemical agents, and physical stresses that can be used to awaken silenced pathways are compared in this paper. These non-genetic induction strategies often need screening approaches, including one strain many compounds (OSMAC), reporter-guided mutant selection, and high throughput elicitor screening (HiTES) have been developed. Different types of genetic manipulations applied in the induction of cryptic BGCs of Actinobacteria can be categorized as genome-wide pleiotropic and targeted approaches like manipulation of global regulatory systems, modulation of regulatory genes, ribosome and engineering of RNA polymerase or phosphopantheteine transferases. Targeted approaches including genome editing by CRISPR, mutation in transcription factors and modification of BGCs promoters, inactivation of the highly expressed biosynthetic pathways, deleting the suppressors or awakening the activators, heterologous expression, or refactoring of gene clusters can be applied for activation of pathways which are predicted to synthesize new bioactive structures in genome mining studies of Acinobacteria. In this review, the challenges and advantages of employing these approaches in induction of Actinobacteria BGCs are discussed. Further, novel natural products needed as drug for pharmaceutical industry or as biofertilizers in agricultural industry can be discovered even from known species of Actinobactera by the innovative approaches of metabolite biosynthesis elicitation. The objective of this paper is to explore the function of the AOL-s00215g415 (Aog415) gene, which encodes for the synthesis of siderophore in the nematode trapping fungal model strain A. oligospora, in order to understand the relationship between siderophore biosynthesis and nematode trapping activity. After a through sequence analysis, it was determined that Aog415 is a siderophore-synthesizing NRPS. The product of this gene was then identified to be the hydroxamate siderophore desferriferrichrome, using mass spectrometry analysis. When compared to the WT strains, the Aog415 knockout strain exhibited a 60% decrease in siderophore content in fermentation broth. Additionally, the number of predatory rings of decreased by 23.21%, while the spore yield increased by 37.34%. The deletion of Aog415 did not affect the growth of A. oligospora in diverse nutrient medium. Lipid metabolism-related pathways were the primary targets of Aog415 disruption as revealed by the metabolomic analysis. In comparison to the WT, a significant reduction in the levels of glycerophospholipids, and glycolipids was observed in the mutation. The metabolic alteration in fatty acyls and amino acid-like molecules were significantly disrupted. The knockout of Aog415 impaired the biosynthesis of the hydroxamate siderophore desferriferrichrome, remodeled the flow of fatty acid in A. oligospora, and mainly reprogrammed the membrane lipid metabolism in cells. Desferriferrichrome, a hydroxamate siderophore affects the growth, metabolism and nematode trapping ability of A. oligospora by regulating iron intake and cell membrane homeostasis. Our study uncovered the significant contribution of siderophores to the growth and nematode trapping ability and constructed the relationship among siderophores biosynthesis, lipid metabolism and nematode trapping activity of A. oligospora, which provides a new insight for the development of nematode biocontrol agents based on nematode trapping fungi. Pyrroloquinoline quinone (PQQ) is a peptide-modified natural product. PQQ has important physiological functions such as anti-oxidation, anti-aging, and immunity enhancement. However, due to the lack of in-depth understanding of PQQ biosynthesis and regulation, inefficient PQQ production level limits its wide application. Accordingly, there is still an urgent need to develop high-yielding strains for synthesis of PQQ. This paper reviewed the research and development trends on the PQQ biosynthetic pathways, catalytic reaction mechanism of key enzymes, and the selection of high-yielding strains, which also prospects for the future construction of PQQ biosynthetic microbial cell factories. Nowadays, emerging pollutants are widely used and exist in wastewater, such as antibiotics, heavy metals, nanoparticle and microplastic. As a green alternative for wastewater treatment, microalgal-bacterial symbiosis has been aware of owning multiple merits of low energy consumption and little greenhouse gas emission. Thus, the responses of microalgal-bacterial symbiosis to emerging pollutants in wastewater treatment have become a hotspot in recent years. In this review paper, the removal performance of microalgal-bacterial symbiosis on organics, nitrogen and phosphorus in wastewater containing emerging pollutants has been summarized. The adaptation mechanisms of microalgal-bacterial symbiosis to emerging pollutants have been analyzed. It is found that antibiotics usually have hormesis effects on microalgal-bacterial symbiosis, and that microalgal-bacterial symbiosis appears to show more capacity to remove tetracycline and sulfamethoxazole, rather than oxytetracycline and enrofloxacin. Generally, microalgal-bacterial symbiosis can adapt to heavy metals at a concentration of less than 1 mg/L, but its capabilities to remove contaminants can be significantly affected at 10 mg/L heavy metals. Further research should focus on the influence of mixed emerging pollutants on microalgal-bacterial symbiosis, and the feasibility of using selected emerging pollutants (e.g., antibiotics) as a carbon source for microalgal-bacterial symbiosis should also be explored. In the field of wastewater treatment, nitrate nitrogen (NO3−-N) is one of the significant contaminants of concern. Sulfur autotrophic denitrification technology, which uses a variety of sulfur-based electron donors to reduce NO3−-N to nitrogen (N2) through sulfur autotrophic denitrification bacteria, has emerged as a novel nitrogen removal technology to replace heterotrophic denitrification in the field of wastewater treatment due to its low cost, environmental friendliness, and high nitrogen removal efficiency. This paper reviews the advance of reduced sulfur compounds (such as elemental sulfur, sulfide, and thiosulfate) and iron sulfides (such as ferrous sulfide, pyrrhotite, and pyrite) electron donors for treating NO3−-N in wastewater by sulfur autotrophic denitrification technology, including the dominant bacteria types and the sulfur autotrophic denitrification process based on various electron donors are introduced in detail, and their operating costs, nitrogen removal performance and impacts on the ecological environment are analyzed and compared. Moreover, the engineering applications of sulfur-based electron donor autotrophic denitrification technology were comprehensively summarized. According to the literature review, the focus of future industry research were discussed from several aspects as well, which would provide ideas for the application and optimization of the sulfur autotrophic denitrification process for deep and efficient removal of NO3−-N in wastewater.
