A large number of hadiths in Shia narrative texts contain the word " Nafi-e- ba~s". In jurisprudence and principles courses, Imami jurists have provided explanations regarding the meaning of this phrase; But there is no independent research about discovering the semantic truth of this phraze. Identifying the concept of "Nafi"(negation) and the scope of its implications play a significant role in explaining the contents of these narrations and choosing the correct ruling in the inference position. By looking at the literal meaning of "Ba~s", the Qur'anic usage of this word and the statements of Imami jurists about it, it is concluded that the meaning of "negation of B~as" is the exclusion of Tahrimi prohibition and punishment in the area where they are suspected to exist. miRNA genes consider as an important class of gene expression regulators that have diverse responses to environmental stresses and play a key role in regulating gene expression during post-transcriptional regulation. Hypericum perforatum, is well-known medicinal plant for its application in the treatment of depression due to the effects of its bioactive compounds named hypericin and hyperforin. The present study was performed to identify the conserved miRNAs and their target genes in the transcriptome of H. perforatum. First, RNA-seq data were obtained from the European Nucleotide Archive (ENA) of EMBL-EBI database and then the transcriptome (RNA) sequences were assembled. The non-coding transcripts were identified and considered as miRNA precursors candidate sequences. Finally, six miRNAs named HpmiR395, Hp-miR845d, Hp-miR414, Hp-miR159, Hp-miR159e, and Hp-miR156c were identified from the candidate sequences using of C-mii software after applying strict filters. Investigation of the protein-protein interaction network determined the relationships between the target genes and target proteins especially in transcription factors. In the next step, to confirm the target genes for the identified miRNAs, the foliar application of Methyl Jasmonate (MJ) with concentrations of 0 (control) and 200 μM was performed on H. perforatum plants and the expression pattern of two target genes was investigated. In the analysis of qRT-PCR expression changes at 12, 24, 48 and 72 hours after applying MJ, the relative expression level of Hyp-1 (DN121523_c1_g4_i2) and HD-Zip (DN121003_c0_g1_i1) transcripts increased after 72 hours of application of MJ. In general, regarding to the regulatory role of the identified miRNAs in the present study, these genes could be used to better and more accurately identify the biosynthetic pathway of hypercin and hyperforin. Background: Currently, autologous and allogeneic adipose tissues represent a ubiquitous source of material for fat reconstructive therapies. However, these approaches are limited, and often accompanied by a 40-60% reduction in graft volume following transplantation, limited proliferative capacity of mature adipocytes for ex vivo expansion, and extensive adipocyte damage encountered when harvested with conventional liposuction techniques. Recently, cell-based approaches utilizing adipogenic progenitor cells for fat tissue engineering have been developed and were reported to promote both short-term in vivo adipogenesis and to repair defect sites. The aim of this study was to isolate stem cells from fat tissue than examine the growth of stem cells by invitro tests. Methods: For human adipose stem cell isolation (hASC), subcutaneous adipose tissue sites were obtained from female subjects undergoing elective procedures. Tissues were washed 3-4 times in phosphate buffered saline (PBS) and suspended in an equal volume of PBS supplemented with 1% FCS and 0.1% collagenase type I. The tissue was placed in an agitated water bath at 37 1C. The supernatant containing mature adipocytes, was aspirated. Portions of the SVF were suspended in DMEM medium. hASCs were selected based on their ability to adhere to tissue culture plastic and subsequently expanded to 75-90% confluence. Adipose stem cells were isolated and cultured on DMEM. To assess mesenchymal origin of stem cells we used flow-cytomery technique as well as differentiation to osteocyte and chondrocyte lines. Results: The nature of the mesenchymal cells was confirmed by flow -cytometry techniques, based on the expression of CD90, CD105, CD166, and lack of expression of hematopoietic markers of CD34, CD31, and CD45. The successful differentiation of our stem cells to osteocyte, chondrocyte had been showed by specific Alizarin-Red and Toluidine-blue staining of cells. Conclusion: Although we have not the results of in vivo tests to support in vivo adipogenesis either alone or in combination with natural or synthetic matrix, the results showed that stem cells isolation from adipose tissue was successful, and we provided an environment for differentiation of stem cells. Endodontic treatment of immature permanent teeth accompanies with several issues. The primary goal when treating such teeth is to maintain pulp vitality so that root development can occur normally. Indications and requirements for vital pulp therapy include asymptomatic and reversible pulpitis. Also there are controversial opinions regarding the ultimate clinical treatment of the vital pulp therapy techniques. In this manuscript we report 3 cases of immature symptomatic permanent molars with irreversible pulpitis caused by caries exposure of the pulp that have been undergone vital pulp therapy successfully. Background and aim: The ability to maintain upright stance is essential in gait and initiation of voluntary movements that are vital not only for sport but also for activity of daily living. Lateral ankle sprains are common in athletes and are most prevalent in jumping sports such as volleyball , football and basketball .Although these injuries are common after direct contact. It may occur in non contact mechanism such as landing from a jump. Neuromuscular control plays a major role in dynamic joint stability. Most of ankle joint injuries occur in jump-landing task and at the end of sport activities particularly when the athlete is fatigued. Our purpose was to evaluate the effects of ankle muscle fatigue on dynamic postural stability in jump landing task. Material and methods: Twenty two healthy women athletes (age:22.8±1.7 years, height:160.3±4.9 cm and weight:56.2±5.8 kg) volunteered to participate in this study. Two test sessions were done with a rest period of one week between them. During each session, effect of one group of ankle muscles (sagital or frontal movers) on dynamic postural stability was assessed. Postural stability was evaluated for 5 seconds after landing on force plate by postural stability indices ( Medial/Lateral, Anterior/Posterior, Vertical and Dynamic Postural Stability Indices) before and after isokinetic fatigue of ankle muscles. Results: The finding indicate significant increase in all stability indices after isokinetic fatigue of ankle muscles (p<0.05). Conclusion: These results indicate that isokinetic fatigue of ankle muscles significantly decrease postural control ability in healthy women athlete. [ABSTRACT FROM AUTHOR]
