Objective To evaluate the effects of semen storage temperature and storage duration on mitochondrial membrane potential(MMP). Methods Thirty semen samples from healthy men were collected, and divided into 14 parts respectively, one part was immediately determined (within 1 h), two parts were stored at 4 T;, four parts were stored at room temperature, four parts were stored at 37 T, and three parts were immediately frozen at 一 80 戏.Semen stored at room temperature and 37 七 for 2, 4, 8, 24 h, refrigerated at 4 for 1, 3 d, and frozen at -80 for 1, 3, 7 d, respectively. Sperm mitochondrial membrane potential (MMP) was analyzed immediately after the storage time was reached. Results At room temperature and 37 兀,speim MMP decreased gradually with the extension of storage time. Compared with the real-time measurement, the MMP of sperm stored for 8, 24 h was significantly decreased (PvO.Ol). At 4, -80, the MMP of sperm stored for 1, 3, 7 d decreased significantly (P <0・ 01)・ Conclusion Storage conditions and storage time of semen in vitro are the important factors affecting mitochondrial fimction of sperm. At room temperature and 37 °C, semen analysis should be completed within 4 hours, and internal quality control before analysis is crucial to MMP test. [ABSTRACT FROM AUTHOR]
