This study aims to examine the effect of superfine powder and aqueous extract of Polygonati Rhizomaon on natural perimenopausal syndrome in rats and explore the underlying mechanism. To be specific, a total of 60 female SD rats(14-15 months old) with estrous cycle disorder were screened by the vaginal smear and randomized into model control group, β-estradiol 3-benzoate group(0.1 mg·kg~(-1)), superfine powder of Polygonati Rhizoma group(0.25, 0.5 g·kg~(-1)) and aqueous extract of Polygonati Rhizoma group(0.25, 0.5 g·kg~(-1)), and another 10 female SD rats(14-15 months old) were selected as the youth control group. The administration lasted 6 weeks. Then the perimenopausal syndrome-related indexes such as body temperature, microcirculatory blood flow of face and ear, vertigo period, salivary secretion, grip force, and bone strength were determined and open field test was conducted. The immune system-related indexes such as the wet weight and index of thymus and spleen, percentage of T lymphocytes and subgroups in peripheral blood, and hematological indexes were measured. In addition, the ovary-related indexes such as estrous cycle, the wet weight and index of uterus and ovary, ovarian tissue morphology, and cell apoptosis were determined. Moreover, hypothalamus-pituitary-ovary axis(HPO)-related indexes such as serum sex hormone levels, cytochrome P450 family 11 subfamily A member 1(CYP11A1), cytochrome P450 family 19 subfamily A member 1(CYP19A1), and cytochrome P450 family 17 subfamily A member 1(P450 17A1) in ovarian tissue were measured. The results showed that the superfine powder and aqueous extract of Polygonati Rhizoma significantly decreased body temperature(anal, facial and dorsal temperature), microcirculatory blood flow in the ear, and vertigo period, increased salivary secretion, grip force, bone strength, total distance and total speed in the open field test, wet weight and index of thymus and spleen, lymphocyte ratio, CD3~+ level, and CD4~+/CD8~+ ratio, reduced neutrophil number and ratio, estrous cycle disorder ratio, and number of ovarian apoptotic cells, raised wet weight and index of uterus, wet weight of ovary, levels of inhibin B(INHB), estradiol(E_2), anti-müllerian hormone(AMH), and ovarian CYP11A1 and CYP19A1, decreased follicle-stimulating hormone(FSH) and luteinizing hormone(LH) content, and improved ovarian tissue morphology. It is suggested that the superfine powder and aqueous extract of Polygonati Rhizoma can improve the symptoms associated with natural perimenopausal syndrome in rats and enhance ovarian function and immune function. The mechanism is that they regulate HPO axis function by increasing estrogen synthesis.

Objective: To investigate the relationship between blood aryl hydrocarbon receptor(AhR)mRNA and cytochrome P450(CYP)1A1 mRNA expression and corrected QT interval among residents exposed to arsenic via drinking water., Methods: Arsenic exposure area in Bayannao'er city of Nei Monggol Autonomous Region was selected as the survey point, and the residents living more than 10 years in this area were investigated from December 2012 to January 2015.A total of 233 residents were divided into four groups according to drinking water arsenic concentration (ranged from 0.8 to 824.7 μg/L): control group (drinking water arsenic concentration <10.0 μg/L, n=55), low exposure group (drinking water arsenic concentration 10.0-99.9 μg/L, n=47), middle exposure group (drinking water arsenic concentration 100.0-199.9 μg/L, n=45), high exposure group (drinking water arsenic concentration ≥200.0 μg/L, n=86). Epidemiological investigation was performed.Real-time PCR technology was used to detect the expression levels of blood AhR mRNA and CYP1A1 mRNA, and the relationship between expression levels of blood AhR mRNA and CYP1A1 mRNA and corrected QT interval was analyzed., Results: (1) Blood AhR mRNA and CYP1A1 mRNA expression levels were similar among control group, low exposure group and middle exposure group (all P>0.05) while blood AhR mRNA (3.18×10(-3)(2.42×10(-3), 4.45×10(-3)) vs.2.30×10(-3)(1.53×10(-3), 3.20×10(-3)), P<0.05) and CYP1A1 mRNA (1.58×10(-3)(0.80×10(-3), 2.73×10(-3))vs.1.00×10(-3)(0.59×10(-3), 2.09×10(-3)), P<0.05) expression levels were significantly higher in high dose group than in control group.(2) AhR mRNA expression level was similar between residents with longer corrected QT interval and residents with normal corrected QT interval (2.89×10(-3)(1.90×10(-3), 3.71×10(-3)) vs.2.58×10(-3)(1.85×10(-3), 3.57×10(-3)), P>0.05). CYP1A1 mRNA expression level was significantly higher in residents with longer corrected QT interval than in residents with normal corrected QT interval (1.50×10(-3)(0.78×10(-3), 2.63×10(-3)) vs.1.16×10(-3)(0.64×10(-3), 2.36×10(-3)), P<0.05). (3) AhR mRNA expression level was similar between residents with sinus tachycardia or bradycardia or normal heart rate(2.89×10(-3)(1.71×10(-3), 4.45×10(-3)) vs.2.64×10(-3)(1.86×10(-3), 3.52×10(-3)), P>0.05). CYP1A1 mRNA expression level was significantly higher in residents with sinus tachycardia or bradycardia than in residents with normal heart rate (1.47×10(-3)(0.87×10(-3), 2.77×10(-3)) vs.1.24×10(-3)(0.64×10(-3), 2.31×10(-3)), P<0.05)., Conclusions: Long-term exposure to arsenic is associated with upregulated blood AhRmRNA and CYP1A1 mRNA expression.Blood CYP1A1 mRNA expression, but not AhRmRNA expression, is associated with prolonged corrected QT interval.

Objective: To investigate the correlation between the combination of smoking and the polymorphisms of cytochrome P450 (CYP) 1A1-Msp I and glutathione S-transferase (GST) T1 genes and oral cancer., Methods: The genetic polymorphisms of CYP1A1-Msp I and GSTT1 were detected by polymerase chain reaction (PCR) technique in peripheral blood leukocytes of 300 oral cancer cases and 300 non-cancer controls, and the correlation between smoking, the two metabolic enzymes genetic polymorphisms and oral cancer were analyzed., Results: The frequencies of CYP1A1-Msp I (m2/m2) and GSTT1(-) were 38.33% and 69.33% in oral cancer cases and 21.00% and 44.33% in healthy controls respectively. Statistical tests showed significant difference in the frequencies between the two groups (P<0.01). The risk of oral cancer with CYP1A1-Msp I (m2/m2) was significantly higher than that of controls (OR= 2.34, 95%CI 1.76-4.07). The individuals who carried with GSTT1(-) had a high risk of oral cancer(OR=2.84, 95% CI 1.98-4.54). Combined analysis of the polymorphisms showed that percentage of CYP1A1-Msp I (m2/m2)/GSTT1(-) in oral cancer and control groups was 30.67% and 6.67% respectively (P<0.01). The people who carried with CYP1A1-Msp I (m2/m2)/GSTT1(-) had a high risk of oral cancer(OR=8.27, 95%CI 3.63-11.29). The smoking rate of the case group was significantly higher than that in the control group (OR=2.71, 95%CI 1.31-4.52, P<0.01), and statistic analysis suggested an interaction between smoking and CYP1A1-Msp I (m2/m2)/GSTT1(-) genotypes polymorphisms which increased risk of oral cancer (OR=25.00, 95%CI 11.87-35.64)., Conclusion: CYP1A1-Msp I (m2/m2) and GSTT1(-) are the risk factors in oral cancer. Smoking is also related to the susceptibility to oral cancer. There may be a synergetic interaction among CYP1A1-Msp I (m2/m2), GSTT1(-) and smoking on the elevated susceptibility of oral cancer.
