Objective To explore the effect of omeprazole combined with different probiotics on regulating intestinal flora in reducing functional dyspepsia (FD) in children. Methods Two hundreds children with FD admitted to the Pediatric Department of Foshan Maternal and Child Health Hospital from January 2022 to February 2023 were selected as the study subjects. They were randomly divided into omeprazde(omep) group, groups of omeprazole +yeast(yeast group), +clostridium butyricum(clos group), and +bifidobacterium(bifi group) respectively. Results After treatment, serum level of IL-6, TNF-α, IL-1β, hs-CRP, VIP, SS, Enterobacter and Enterococcus in all groups significantly decreased as compared with the finding before treatment (P＜0.05). Those targets in the three combined treatment groups were significantly lower compared to the ome group; After treatment, the serum MOT level,bifidobacteria, and lactobacilli in each group were significantly increased (P＜0.05), and the results from three combined treatment groups demonstrated notably higher levels compared to the omep group(P＜0.05); The scores of symptoms in all groups showed a significant alleviation after the treatment(P＜0.05). Additionally, the three combined treatment groups exhibited significantly lower symptom scores than the group treated with omeprazole alone (P＜0.05). There was no difference in the incidence of adverse reactions during treatment among the groups. Conclusions Omeprazole combined with different probiotics have achieved good results in the treatment of FD in children.

In view of problem that it is hard to precisely measure time of pulse arriving at receiving sensor due to presence of start-up process when ultrasonic sensor receives ultrasonic pulse signal, the paper proposed a method of precise measurement of air speed in mine ventilation. The method fits the curve and envelope curve of sinusoidal signals received by receiving sensor, and calculates time of the ultrasonic pulse signal arriving at the sensor. The results of calculation and analysis of simulate sampling data show that the method can achieve high measuring precision, which can improve measurement accuracy of mine tunnel air speed.

Objective: To explore the genetic etiology of a child with Cowden syndrome 1 (CS1)., Methods: A child who had visited the Ningbo Women and Children's Hospital on August 26, 2022 was selected as the study subject. Clinical information of the child was collected. Genomic DNA was extracted from peripheral blood samples of the child and his family members and subjected to whole exome sequencing (WES). Candidate variant was verified by Sanger sequencing., Results: The child, a 13-year-old boy, had manifested with severe mental retardation, hyperactivity, autistic behavior, sparse and prominent teeth, macrocephaly, and skin freckles on the penis. His mother had presented with multiple papules, hamartomatous polyps, thyroid adenoma and macrocephaly. WES results revealed that the child has harbored a nonsense c.781C>T (p.Q261*) variant of the PTEN gene, which was inherited from his mother. Based on the guidelines from the American College of Medical Genetics and Genomics, the c.781C>T variant was classified as likely pathogenic (PVS1+PM2_Supporting)., Conclusion: The c.781C>T variant of the PTEN gene probably underlay the pathogenesis in the child and his mother. Above finding has facilitated genetic counseling for this family.

Objective: To explore the genetic basis for a fetus with Cardiac valvular dysplasia type 1 (CVDP1)., Methods: A CVDP1 fetus identified at the Ningbo Women and Children's Hospital on July 7, 2022 was selected as the study subject. Clinical data of the fetus was collected. The fetus and its parents were subjected to trio-whole exome sequencing (trio-WES), and candidate variants were verified by Sanger sequencing., Results: The fetus had exhibited generalized edema, complex cardiac malformation, abdominal effusion, and enhanced intestinal and renal parenchymal echoes. Trio-WES revealed that it has harbored compound heterozygous variants of the PLD1 gene, namely c.2977C>T (p.R993*) and c.1460G>A (p.W487*), which were respectively inherited from its father and mother. Neither variant was reported previously. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), the c.2977C>T (p.R993*) variant was evaluated to be likely pathogenic (PVS1_Moderate+PM2_Supporting+PM3+PP4), whilst the c.1460G>A (p.W487*) variant was evaluated to be pathogenic (PVS1+PM2_Supporting+PP4)., Conclusion: The c.2977C>T (p.R993*) and c.1460G>A (p.W487*) compound heterozygous variants of the PLD1 gene probably underlay the CVDP1 in the fetus. Above discovery has enriched the mutational spectrum of the PLD1 gene and provided a guidance for genetic counseling and prenatal diagnosis in this family.

Objective: To explore the genetic etiology for a fetus with hydrocephalus and intraventricular hemorrhage., Methods: Trio whole exome sequencing was carried out. Candidate variants were verified by Sanger sequencing of the fetus and its parents., Results: The fetus was found to harbor c.818G>A (p.W273X) and c.833T>C (p.L278P) compound heterozygous variants of the PROC gene, which were respectively inherited from its mother and father. Based on the guidelines of the American College of Medical Genetics and Genomics (ACMG), both variants were predicted to be likely pathogenic (PVS1_Strong+PM2_Supporting+PP4; PM2_Supporting+PM3+PP1+PP3+PP4)., Conclusion: The fetus was diagnosed with Protein C deficiency due to the c.818G>A (p.W273X) and c.833T>C (p.L278P) compound heterozygous variants of the PROC gene. Above finding has enriched the spectrum of PROC gene variants and enabled genetic counseling and prenatal diagnosis for the family.

Objective: To explore the genetic etiology of a Chinese pedigree featuring non-simplex blepharocheilodontic syndrome., Methods: Whole exome sequencing was carried out to detect genetic variant and copy number variations (CNVs) in the pedigree. Suspected variants were verified by Sanger sequencing and qPCR., Results: The fetus and its elder brother, father and grandfather were found to harbor a heterozygous c.83delG (p.A29Rfs*55) variant of the CTNND1 gene, which was unreported previously. In addition, its elder brother was also found to be a double heterozygote for a c.235delC (p.L79Cfs*3) variant of GJB2 gene and a c.538C>T (p.R180X) variant of GJB3 gene, which were respectively inherited from his mother and father. CNVs analysis revealed a de novo heterozygotic deletion (1.46 Mb) at 17q12 in the mother, which was confirmed by qPCR. Based on American College of Medical Genetics and Genomics guidelines, the c.83delG variant, the c.235delC variant and the 17q12 microdeletion were predicted as pathogenic, while the c.538C>T variant was of uncertain significance., Conclusion: The c.83delG (p.A29Rfs*55) variant of the CTNND1 gene probably underlay the pathogenesis of non-simplex blepharocheilodontic syndrome in this pedigree. The double heterozygous variants of c.235delC (p.L79Cfs*3) of GJB2 gene and c.538C>T (p.R180X) of GJB3 gene probably underlay the hearing loss in the elder brother. The bilateral renal cysts in the mother may be attributed to the 17q12 microdeletion. Above results have provided guidance for genetic counseling and prenatal diagnosis for this pedigree.

Objective: To delineate the clinical and genetic features of a fetus with micrognathia, low-set ears, microtia, polyhydramnios and anechoic stomach by ultrasonography., Methods: Whole exome sequencing (WES) was carried out to detect genetic variant in the fetus, for which routine chromosomal karyotyping and chromosomal microarray analysis (CMA) yielded no positive finding. Candidate variants were verified by Sanger sequencing and bioinformatic analysis., Results: WES revealed that the fetus has carried a de novo nonsense c.2302C>T (p.Q768X) variant in exon 23 of the EFTUD2 gene, which was detected in neither parent. The variant was unreported previously and may lead to premature termination of the translation of EFTUD2 protein at the 768th amino acid. Bioinformatic analysis predicted the amino acid to be highly conserved and may alter the structure and function of the EFTUD2 protein., Conclusion: The c.2302C>T variant of the EFTUD2 gene probably underlay the mandibulofacial dysostosis Guion-Almeida type in the fetus. Discovery of the novel variant has enriched variant spectrum of the EFTUD2 gene and provided a basis for genetic counseling and prenatal diagnosis for the family.

Objective: To predict and verify the target gene of miR-200c-3p and evaluate the inhibitory effect of miR-200c-3p on the proliferation of nephroblastoma cells., Methods: The putative target genes of miR-200c-3p were predicted by bioinformatics approach. Nephroblastoma cell models with miR-200c-3p overexpression or knockdown were established in SK-NEP-1 and G401 cells with corresponding control groups. The expressions of CCNE2 in SK-NEP-1 and G401 cells in different groups were detected by RT-PCR and Western blotting. A luciferase reporter assay was used to determine the targeting relationship between miR-200c-3p and CCNE2. The effects of miR-200c-3p overexpression or knockdown on cell proliferation was detected by cell counting kit-8 (CCK-8) assay and soft agarose assay., Results: CCNE2 was one of the target genes of miR-200c-3p as predicted by bioinformatics methods. Transfection of the two nephroblastoma cell lines with miR-200c-3p mimic resulted in significantly lowered CCNE2 mRNA and protein expressions ( P < 0.05). The results of dual-luciferase assay confirmed that miR-200c-3p bound to the 3'UTR of CCNE2. CCK-8 assay and soft agarose assay demonstrated that overexpression of miR-200c-3p significantly inhibited the proliferation of the nephroblastoma cells ( P < 0.01), and knocking down miR-200c-3p in the cells produced the opposite effects., Conclusions: miR-200c-3p overexpression inhibits the proliferation of nephroblastoma cells by down-regulating its target gene CCNE2.

Objective: To study the pathology, imaging and clinical features of a child with trisomy 21 syndrome associated interstitial lung disease., Method: Data of a case with trisomy 21 syndrome associated interstitial lung disease confirmed by lung imaging and pathology were collected, analyzed and the related reports in literature were reviewed., Result: The patient was a one year and 7 months old boy who suffered from severe pneumonia and recurrent infection during his hospital stay. When his disease was stable, he did not have shortness of breath and cyanosis, but a chest computed tomography (CT) showed ground-glass opacity, regional emphysema, band-like change in lung parenchyma, which indicated interstitial lung diseases. Unequal air inflation in bilateral lungs and diffuse over-distension of peripheral air spaces in lung surface were seen through thoracoscope. Pathological examination indicated that alveolar, alveolar ducts and alveolar sac were enlarged, alveolar septa was expanded. There were two reports in lung pathology of trisomy 21 syndrome, alveolar growth abnormalities was seen in 86%-88% cases. The multiple subpleural cysts in chest CT was characteristic. Clinically, trisomy 21 syndrome had high morbidity of respiratory tract infection and progress to respiratory failure frequently. Prolonged postoperative desaturation was constant which required long duration of respiratory support., Conclusion: Trisomy 21 syndrome associated alveolar growth abnormalities were confirmed, which manifest as alveolar simplification in pathology and interstitial lung diseases in imaging. The risk of respiratory failure in these cases caused by infection and surgery should be considered.

Compound L, as the procedural sensor for the detection of Cu2+ and Hg2+, was designed and synthesized based on the coumarin-modified rhodamine derivative. The structure of compound L was characterized by NMR, high resolution mass spectrometry and infrared method. Its sensing behavior toward various metal ions was investigated with absorbance methods. The study found that L had good selectivity and sensitivity for Cu2+. When addition of various metal ions (Zn2+, Hg2+, Cu2+, Fe3+, Cd2+, CO2+, Ni2+, Mg2+, Ca2+, Al3+, La3+, K+, Na+, Mn2+, Pb2+ and Ag+), only Cu2+ could induce a visible change of solution from colourless to pink and a new absorption band centered at 534 nm appear, which indicated that compound L could be used for the naked eye detection of Cu2+. From UV titration, the detection limit was about 1.9 X 10(-8) mol x L(-1). Test strips based on L were fabricated, and this test strips could act as a convenient and efficient Cu2+ test kit. The binding ratio of the complex of L-Cu2+ was 1:1 according to the Job's plot and high resolution mass spectrometer (HRMS) experiments. Moreover, Upon addition of 1 equiv. EDTA to the mixture of L and Cu2+ in DMSO solution, colour changed from pink to almost colourless, indicating that the EDTA replaced the receptor L to coordinate with Cu2+. Therefore, L could be classified as a reversible sensor for Cu2+. In addition, when adding Hg2+ to L-Cu2+ complexes, a visible change of solution from pink to colourless was observed, while other metal ions didn't cause this change. Thus, L-Cu2+ complex also could be used for the naked eye recognition of Hg2+, and the detection limit was calculated about 2.9 x 10(-1) mol x L(-1) according to the UV titration. Consequently, this procedural sensor L could be use for the orderly naked eye recognition of Cu2+ and Hg2+.

Objective: To establish a HPLC-FP for the determination of honey-fried Farfarae Flos., Methods: HPLC analysis was performed on a C18 column (4.6 mm x 250 mm, 5 microm), the mobile phase consisted of 0.1% phosphoric acid solution and acetonitrile as gradient elution with the flow rate of 1.0 mL/min. The wavelength was 240 nm., Results: HPLC fingerprint of honey-fried Farfarae Flos was established and the results of methodological study met the technical requirements for fingerprint., Conclusion: The established method can be used for quality control of honey-fried Farfarae Flos.

Objective: To analyse the lipid-soluble components of Amomum villosum fruits by GC/MS., Methods: Lipid-soluble components in pericarps and seeds of A. villosum were extracted by Soxhlet extraction. The Lipid-soluble components were esterified and then analyzed by gas chromatography-mass spectrometry (GC-MS)., Results: Twenty-five and thirteen compounds were identified in pericarps and seeds of A. villosum, respectively, mainly including fatty acids and their esters., Conclusion: The results indicate that it is highly potential and significant to exploit its value in foods, medicines and healthy care.

Objective: To optimize the extracting process of Folium Nelumbinis by central composite design and response surface method., Methods: Independent variables were ethanol concentration, solvents ratio and pH,dependent variable was extracting rate of alkaloid. Composite design and response surface method were used to optimize the extracting process. Resultant model was used to predict the response in the optimal region., Results: The optimum extraction conditions were 80% aqueous ethanol, 75 fold solvent, pH value 5., Conclusion: The optimum process is simple and more convenient for extracting Folium Nelumbinis. The observed and predicted values are close to each other.

Objective: To analyze the effect of arteriovenous impulse system (AVIS) combined with low-molecular-weight heparins calcium (LMWHC) for prophylaxis of deep vein thrombosis (DVT) following total knee arthroplasty (TKA)., Methods: From March 2006 to March 2008, 76 cases of osteoarthritis patients (76 knees) accepted TKA, including 25 males and 51 females with an average age of 66.6 years (range, 58-79 years). The affected knees were left side in 41 cases and right side in 35 cases. They were randomly divided into experimental group and control group before surgery. Then LMWHC and rehabilitation training were routinely given in two groups before and after surgery. However, only experimental group was treated with AVIS continually during the first four days and then two times a day for 30 minutes one time during 5-7 days. At 7 days after operation, color Doppler ultrasound was used to detect the occurrence condition of DVT., Results: Five cases (13.16%) had thrombosis of calf and recovered after treated with urokinase and salvia in the experimental group. Eleven cases had thrombosis of calf and 3 cases had thrombosis of whole low extremities (36.84%), and improved after treated with urokinase and salvia in the control group. There was significant difference in DVT incidence rate between two groups (P < 0.05). No pulmonary embolism or death was found in both groups., Conclusion: AVIS can effectively accelerate the venous blood return velocity, a combination of AVIS and LMWHC has a better effect in the prevention of DVT following TKA.

Objective: To investigate the effects of leptin on airway inflammation and the expression of Th1/Th2 cytokines., Methods: The obesity and acute asthma models were established in 40 female SD rats, which were randomly divided into a normal weight control group (group A), a normal weight asthmatic group (group B), a normal weight intervention group (group C), an obese control group (group D) and an obese asthmatic group (group E). The airway resistance and airway responsiveness were calculated by transpulmonary pressure and gas flow rate. The numbers of leukocytes, eosinophils (EOS) and neutrophils (N) in bronchoalveolar lavage fluid (BALF) were counted. The concentrations of interleukin-4 (IL-4), interferon-gamma (IFN-gamma) and leptin in serum and BALF were determined by ELISA. The protein and mRNA expression of leptin was measured by Western blot and RT-PCR respectively., Results: The airway resistance in group C and E [(0.890 +/- 0.106) cm H2Oxml(-1)xs(-1), (1.024 +/- 0.096) cm H2Oxml(-1)xs(-1), (1.129 +/- 0.107) cm H2Oxml(-1)xs(-1), (0.946 +/- 0.104) cm H2Oxml(-1)xs(-1), (1.124 +/- 0.095) cm H2Oxml(-1)xs(-1), (1.135 +/- 0.105) cm H2Oxml(-1)xs(-1), respectively.] was increased significantly compared to group B [(0.638 +/- 0.128) cm H2Oxml(-1)xs(-1), (0.745 +/- 0.073) cm H2Oxml(-1)xs(-1), (0.773 +/- 0.090) cm H2Oxml(-1)xs(-1)] (q = 7.128, 8.712, 8.318, 11.300, 11.258, 11.447, all P < 0.05). The numbers of leukocyte and neutrophils in group C and E [(91 +/- 9) x 10(4)/ml, (108 +/- 21) x 10(4)/ml, (12.4 +/- 4.0) x 10(4)/ml, (14.2 +/- 5.9) x 10(4)/ml, respectively.] were increased significantly compared to group B [(79 +/- 7) x 10(4)/ml, (2.4 +/- 1.1) x 10(4)/ml] (q = 2.923, 7.063, 8.629, 10.182, all P < 0.05). The concentrations of IFN-gamma were [(42.3 +/- 3.5) ng/L, (45.1 +/- 4.8) ng/L, (19.2 +/- 1.8) ng/L, (20.3 +/- 1.5) ng/L] in group C and E respectively, which were significantly higher than those of group B [(16.5 +/- 1.4) ng/L, (9.3 +/- 1.0) ng/L] (q = 21.607, 23.952, 16.919, 18.799, all P < 0.05). The protein and mRNA expression of leptin in lung tissue in group C and E [(0.40 +/- 0.07) ng/L, (0.44 +/- 0.05) ng/L, (0.34 +/- 0.06) ng/L, (0.38 +/- 0.04) ng/L, respectively.] were remarkably higher than those of group B [(0.31 +/- 0.03) ng/L, (0.21 +/- 0.04) ng/L] (q = 4.648, 6.713, 8.222, 10.752, all P < 0.05)., Conclusion: Leptin could aggravate airway inflammation featured by infiltration of neutrophils and enhancement of Th1 type inflammation.

Infrared vibration spectra of nanosize (SiO2)n,O2H4 with line, ring and cage structures were calculated using the density-functional theory (B3LYP) at 6-31G(d) level. For quasi 1D (one dimension) line and ring structure, the frequencies of stronger infrared vibration modes monotonically change as a function of the length, which shows strong size effect. The IR vibration modes in directions parallel and perpendicular to the axis of nanoline or nanoring have opposite size dependence trends, showing the real vibration anisotropy property of nano scale material. Due to constructional complexity, cage structure does not have quasi 1D character. Morever, IR vibration modes increase as structural symmetry decreases. Vibrations of two-membered ring, isolated hydroxyls of three structures and framework SiO4 tetrahedron in cage structure agree well with experimental data. Theoretical calculation result of IR spectra can be used to guide characterization of silicon oxide nano scale materials fabricated in experiment.
